Larval Environments Affect The Implementation Of The Sterile Insect Technique On Aedes Mosquitoes, Katherine G. Evans

Theses and Dissertations

Sterile Insect Technique (SIT) is an effective method for controlling pests and has been increasing in popularity to control Aedes mosquitoes. SIT involves inundating an area with sterilized male mosquitoes to reduce egg hatch and ultimately the number of wild adults. Previous research indicates that enacting mortality early in the larval life of Aedes mosquitoes can result in counter-productive effects, due to reduced competition among developing larvae. I hypothesize that the effectiveness of SIT at reducing adult populations is modified by densitydependent mortality and density-dependent growth among larvae. Tick-borne diseases are on the rise throughout the world, and there is a need to better understand tick behavior in order to identify potential new interventions. Ticks have a complex life history and can survive months off-host. There is a lack of large-scale data on off-host tick behavior, which leaves a gap in understanding of tick biology outside of tick-host interactions. Introducing undergraduate students to authentic research early in their studies can help prepare them for independent inquiry in upper-level classes. Investment in life history traits such as immune function and reproduction is constrained by finite available resources. A cost-of-immunity trade-off may occur in response to infection when resources are diverted away from reproductive effort and into an immune response. Alternatively, an infected individual may enhance reproductive effort to maximize terminal reproductive success in response to the survival threat inherent to infection (terminal investment). We measured male Aedes aegypti reproductive behavior following inoculations with: living bacteria; killed bacteria as an immune elicitor; and a sham control. In this dissertation I explore different ways that the environment impacts life history in mosquitoes in ways that may alter vectorial capacity. In chapter I, I tested if short-term sugar deprivation experienced after exposure to La Crosse virus altered infection rates in Aedes albopictus and if sugar treatment and virus infection status altered blood feeding behavior and fecundity. I found no evidence that sugar deprivation impacted infection rates or fecundity. Sugar deprivation did increase blood feeding. There was no effect of infection status on blood feeding or fecundity. The Asian tiger mosquito, Aedes albopictus (Skuse), is perhaps the most successful invasive mosquito species in contemporary history. In the United States, Ae. albopictus has spread from its introduction point in southern Texas to as far north as New Jersey (i.e., a span of approximate to 14 degrees latitude). This species experiences seasonal constraints in activity because of cold temperatures in winter in the northern United States, but is active year-round in the south. Estimating a mosquito's vector competence, or likelihood of transmitting disease, if it takes an infectious bloodmeal, is an important aspect of predicting when and where outbreaks of infectious diseases will occur. Vector competence can be affected by rearing temperature and inter- and intraspecific competition experienced by the individual mosquito during its larval development. This research investigates whether a new morphological indicator of larval rearing conditions, wing shape, can be used to distinguish reliably temperature and competitive conditions experienced during larval stages. Discarded vehicle tires are a common habitat for container mosquito larvae, although the environmental factors that may control their presence or abundance within a tire are largely unknown. We sample discarded vehicle tires in six sites located within four counties of central Illinois during the spring and summer of 2006 to determine associations between a suite of environmental factors and community composition of container mosquitoes. Our goal was to find patterns of association between environmental factors and abundances of early and late instars. Aedes albopictus (Skuse) is an invasive container-dwelling mosquito and an important disease vector that co-occurs with the native mosquito, Aedes triseriatus (Say), and the predatory midge, Corethrella appandiculata (Grabham). Larval Ae. triseriatus show significantly greater antipredatory responses when compared to larval Ae. albopictus in the presence of predation cues from C. appendiculata. The potential for evolution of antipredatory behavioral responses to C. appendiculata in Ae. albopictus is unknown. We used a controlled laboratory selection experiment to test whether Ae. albopictus could evolve antipredatory behavioral responses to C. appendiculata predation. Many studies of interspecific competition between Aedes albopictus (Skuse) and Aedes aegypti (L.) (Diptera: Culicidae) larvae show that Ae. albopictus are superior resource competitors to Ae. aegypti. Single-species studies indicate that growth and survival of Ae. albopictus and Ae. aegypti larvae are affected by the type of detritus present in containers, which presumably affects the amount and quality of microorganisms that the mosquito larvae consume. We tested whether different detritus types alter the intensity of larval competition by raising 10 different density/species combinations of Ae. albopictus and Ae. aegypti larvae under standard laboratory conditions, with one of four detritus types …

Climate differences across latitude can result in seasonal constraints and selection on life history characters. Since Aedes albopictus (Skuse) invaded North America in the mid-1980s, it has spread across a range of approximate to 14 degrees latitude and populations in the north experience complete adult mortality due to cold winter temperatures that are absent in the south. Compared with plant detritus, animal detritus yields higher growth rates, survival, adult mass, and population growth of container-dwelling mosquitoes. It is unclear whether the benefit from animal detritus to larvae results from greater microorganism growth, direct ingestion of animal detritus by larvae, or some other mechanism. We tested alternative mechanisms by which animal detritus may benefit the invasive container-dwelling mosquito Aedes albopictus (Skuse) (Diptera: Culicidae). Five species of psocids of the genus Amphigerontia Kolbe 1880, are recorded from the Rocky Mountains of the United States and Canada. Amphigerontia longicauda, n. sp., is described. Amphigerontia petiolata (Banks) is recorded from the Rocky Mountains for the first time. The holotype of Amphigerontia infernicola (Chapman) is re-examined and described in detail. New characters for the separation of females of Amphigerontia bijasciata (Latreille) and Amphigerontia montivaga (Chapman) resulted in many new records of the latter species, previously known in the Rocky Mountains from only a single locality. Feeding preferences of larval container-dwelling mosquitoes are not well understood. Primary production is often absent in container systems and external inputs of animal and plant detritus supply the energy base of container food webs by supporting microorganism prey for mosquitoes. We quantified the feeding preferences of Aedes albopictus (Skuse), a non-native invasive mosquito, and Ochlerotahts triseriatus (Say), a native mosquito, when given a choice of animal and plant detritus. We tested for interpopulational (Illinois versus Florida) differences in feeding preferences, and quantified each species' performance on these two detritus types. We tested for competitive advantage among larvae of Aedes albopictus (Skuse) and Culex pipiens L. in a laboratory experiment and determined the frequency and spatial and temporal patterns of co-occurrence in the field in East St. Louis, IL. In a laboratory competition experiment at multiple combined densities of Ae. albopictus and Cx. pipiens larvae, Ae. albopictus survivorship, and developmental times were significantly affected by conspecific densities but not by Cx. pipiens densities. In contrast, Cx. pipiens survivorship, and developmental times were significantly affected by both conspecific and Ae. albopictus densities. During the rainy season of 2001, the incidence of the dengue vectors Aedes aegypti and Ae. albopictus was examined in different habitats of two cities (Rio de Janeiro and Nova Iguacu) in Rio de Janeiro State, Brazil, and in two cities (Palm Beach and Boca Raton) in Florida. Oviposition trap collections were performed in urban, suburban, and rural habitats in both areas. Our hypothesis that the abundances and frequencies of occurrence of Ae. aegypti and Ae albopictus are affected in opposite ways by increasing urbanization was only partially supported. We investigated how temperature and precipitation regime encountered over the life cycle of Aedes albopictus (Skuse) affects populations. Caged populations of A. albopictus were maintained at 22, 26, and 30 degreesC. Cages were equipped with containers that served as sites for oviposition and larval development. All cages were assigned to one of three simulated precipitation regimes: (1) low fluctuation regime - water within the containers was allowed to evaporate to 90% of its maximum before being refilled, (2) high fluctuation regime - water was allowed to evaporate to 25% of its maximum before being refilled, and (3) drying regime - …

We investigated how constant temperatures of 22, 24, and 26 degreesC experienced across the full life cycle affected the dynamics of caged populations of Aedes albopictus (Skuse). All cages were equipped with plastic beakers that served as sites for oviposition and larval development. The intensity and prevalence of parasitism by Ascogregarina barretti (Vavra) in aedes triseriatus (Say) did not differ between tires and tree holes in field samples taken in September 1996. There was significant variation in the intensity of parasitism among containers that was not significantly cor-related with the pH, conductivity, or temperature of the container water. In an experiment manipulating habitat drying, treatments had a significant effect on A. barretti infection of Ae. triseriatus, only during midsummer in one of two years. Containers maintained at maximal volume had the lowest prevalence of parasitism, and containers that dried out had the greatest …
