Testing Viability Of Oligonucleotide Primers On Microsatellite Markers Amplified By Polymerase Chain Reaction On Dermacentor Variabilis Samples From The Bulloch County Region Of Georgia, Usa, Danielle Souza

Honors College Theses

Dermacentor variabilis, more commonly known as the wood tick, or the American dog tick, can be found all over North America. D. variabilis is a common tick in Bulloch county, Georgia, USA, and little is known about the population structure of this population of the species. Microsatellite markers are able to locate sections of the genome and can aid in estimating relatedness among individuals in a population. Six different oligonucleotide primer pairs were published for D. variabilis, however, published and non-published works indicated that not all of those primer pairs were working equally well for PCR. This study is …

The Activity Of Saccharolytic Enzymes In Collembola Is Associated With Species Affinity For Caves, Andrea Parimuchová, Vladimír Šustr, Miloslav Devetter, Ondřej Vošta, Ionuţ Popa, Ľubomír Kováč

International Journal of Speleology

The activity of enzymes associated with digestion can reflect food availability and feeding preferences of invertebrates in a particular habitat. Caves are mostly nutrient-poor habitats lacking primary production. In the present study the enzymatic activity of cellulases, trehalases and chitinases was measured in eight collembolan species differently associated with the cave environment: the troglobionts (obligate cave species) Pseudacherontides spelaeus and Protaphorura janosik; the eutroglophiles Ceratophysella denticulata, Folsomia candida and Heteromurus nitidus; the subtroglophiles Hypogastrura aequepilosa and Orthonychiurus rectopapillatus; and the trogloxene (not associated with caves) Megaphorura arctica. Qualitative enzymatic patterns and quantitative differences in species activity were …

Foraging Distance Of The Argentine Ant In California Vineyards., Brian N. Hogg, Erik H. Nelson, James R. Hagler, Kent M. Daane

Natural Sciences and Mathematics | Faculty Scholarship

Argentine ants, Linepithema humile (Mayr) (Hymenoptera: Formicidae), form mutualisms with hemipteran pests in crop systems. In vineyards, they feed on honeydew produced by mealybugs and soft scales, which they tend and protect from natural enemies. Few options for controlling Argentine ants are available; one of the more effective approaches is to use liquid baits containing a low dose of an insecticide. Knowledge of ant foraging patterns is required to estimate how many bait stations to deploy per unit area. To measure how far ants move liquid bait in vineyards, we placed bait stations containing sugar water and a protein marker …

Impact Of Sorghum Cultivar, Phenology, Nitrogen, And Silicon Fertilization On Melanaphis Sacchari Biology, Luna Lama

LSU Master's Theses

The sugarcane aphid, Melanaphis sacchari (Zehntner) (Hemiptera: Aphididae), is an invasive pest of grain sorghum, Sorghum bicolor (L.) Moench. Since its first outbreak in sorghum in 2013, severe infestations have spread throughout the southern U.S. causing high economic losses. While insecticidal control has mitigated some of the impacts of this pest, a sustainable ecology-based management program is needed to reduce reliance on chemical control. We studied the effects of silicon (rates equivalent to 0 and 3360 kg Si/ha) and nitrogen (rates equivalent to 0, 110, and 220 kg N/ha) on M. sacchari growth and reproduction on resistant (DKS 37-07) and …

An Inordinate Fondness For Beetles: A Study Of Insect Species Diversity And Abundance In Mazumbai Forest Reserve Versus Nearby Agricultural Areas, Emma Weisner

Independent Study Project (ISP) Collection

This study investigates how human disturbance of ecosystems alters insect diversity and abundance, specifically exploring how insect communities inside Mazumbai Forest Reserve in Tanzania differ from insect communities in agricultural areas near the reserve. Following methods of previous studies on the effect of disturbance on insect populations (Bellamy et al. 2018; McLaughlin & Mineau 1995; Perry et al. 2016), this research utilizes pitfall traps and yellow bowl traps in multiple locations throughout the two study areas to catch insects, which are then identified to their specific order. The collected data support the hypothesis that insect order diversity and abundance vary …

Diurnal And Developmental Differences In Gene Expression Between Adult Dispersing And Flightless Morphs Of The Wing Polymorphic Cricket, Gryllus Firmus: Implications For Life-History Evolution, Anthony J. Zera, Neetha Nanoth Vellichirammal, Jennifer A. Brisson

Anthony Zera Publications

The functional basis of life history adaptation is a key topic of research in life history evolution. Studies of wing polymorphism in the cricket Gryllus firmus have played a prominent role in this field. However, prior in-depth investigations of morph specialization have primarily focused on a single hormone, juvenile hormone, and a single aspect of intermediary metabolism, the fatty-acid biosynthetic component of lipid metabolism. Moreover, the role of diurnal variation in life history adaptation in G. firmus has been understudied, as is the case for organisms in general. Here, we identify genes whose expression differs consistently between the …

Stingless bees are important pollinators of plants, and also producers of honey. Species within the African stingless bee genus Hypotrigona are difficult to differentiate due to morphological similarities. Chemical profiles of whole head extracts from workers of three Hypotrigona species: H. gribodoi, H. araujoi and H. ruspolii were studied by gas chromatography–mass spectrometry. A total of 50 components belonging to six chemical classes: hydrocarbons, aldehydes, alcohols, terpenoids, steroids and fatty acids were identified. Twenty-nine compounds were found in H. araujoi, 26 in H. gribodoi and 33 in H. ruspolii head extracts. Hydrocarbons, alcohols and fatty acids were the major classes, …

Edible insects are part of the diets of a significant proportion of rural populations in the tropics especially Africa and Asia, and their use as source of key nutrients for better nutrition is re-emerging. Indigenously, elemental methods are used to process the insects before they are consumed or sold in retail outlets. In recent years, better knowledge of processing, packaging and storage has become necessary because of commercialisation needs. A common processing approach involves drying after a brief heat-treatment step, and then milling into a powdered product which is sold to manufacturers or consumers as ingredient for processing final products. …

It is largely accepted that larval experience influence host-plant preference of larvae. In general, larval experience with a host plant induces both an odour and feeding preference for that plant. In a previous study, exposure of three noctuid stem borer species, i.e. Sesamia nonagrioides, Busseola fusca and B. nairobica, to a vanillin-enriched medium induced an olfactory preference for the odours of this medium in the larvae of these species. Here, we checked if the exposure to a vanillin-enriched medium also induced a feeding preference for this diet. Different life stages (i.e. neonate larvae to adults) of the three species were …

The potato aphid, Macrosiphum euphorbiae, is an important agricultural pest that causes economic losses to potato and tomato production. To establish the transcriptome for this aphid, RNA-Seq libraries constructed from aphids maintained on tomato plants were used in Illumina sequencing generating 52.6 million 75±105 bp paired-end reads. The reads were assembled using Velvet/Oases software with SEED preprocessing resulting in 22,137 contigs with an N50 value of 2,003bp. After removal of contigs from tomato host origin, 20,254 contigs were annotated using BLASTx searches against the non-redundant protein database from the National Center for Biotechnology Information (NCBI) as well as IntereProScan. …

Characterizing The Physiological Role Of Inward Rectifier Potassium (Kir) Channels In Tick Salivary Gland Function, Zhilin Li Mar 2018

Characterizing The Physiological Role Of Inward Rectifier Potassium (Kir) Channels In Tick Salivary Gland Function, Zhilin Li

LSU Master's Theses

The tick salivary gland is a critical tissue that enables blood feeding, maintenance of the ionic gradients, and facilitates pathogen transmission. Therefore, the broad objective of this investigation was to leverage pharmacological approaches to investigate the role of potassium ion channels to salivary gland function of Amblyomma americanum in an effort to identify a tractable target site for therapeutic development.

Data collected in Chapter 2 clearly illustrated that the A. americanum salivary gland is reliant upon inward rectifier potassium (Kir) channels for fluid secretion, which was similar to previous work performed by our laboratory on Drosophila salivary gland. Therefore, the …

Spinal muscular atrophy (SMA) is caused by homozygous mutations in human SMN1. Expression of a duplicate gene (SMN2) primarily results in skipping of exon 7 and production of an unstable protein isoform, SMNΔ7. Although SMN2 exon skipping is the principal contributor to SMA severity, mechanisms governing stability of survival motor neuron (SMN) isoforms are poorly understood. We used a Drosophila model system and label-free proteomics to identify the SCFSlmb ubiquitin E3 ligase complex as a novel SMN binding partner. SCFSlmb interacts with a phosphor degron embedded within the human and fruitfly SMN YG-box oligomerization domains. …

Recent study has shown that RNA interference (RNAi) is efficient in emerald ash borer (EAB), Agrilus planipennis, and that ingestion of double-stranded RNA (dsRNA) targeting specific genes causes gene silencing and mortality in neonates. Here, we report on the identification of highly effective target genes for RNAi-mediated control of EAB. We screened 13 candidate genes in neonate larvae and selected the most effective target genes for further investigation, including their effect on EAB adults and on a non-target organism, Tribolium castaneum. The two most efficient target genes selected, hsp (heat shock 70-kDa protein cognate 3) and shi (shibire), …

The preference-performance hypothesis states that ovipositing phytophagous insects will select host plants that are well-suited for their offspring and avoid host plants that do not support offspring performance (survival, development and fitness). The mountain pine beetle (Dendroctonus ponderosae), a native insect herbivore in western North America, can successfully attack and reproduce in most species of Pinus throughout its native range. However, mountain pine beetles avoid attacking Great Basin bristlecone pine (Pinus longaeva), despite recent climate-driven increases in mountain pine beetle populations at the high elevations where Great Basin bristlecone pine grows. Low preference for a potential host plant species may …

The southern cattle fever tick, Rhipicephalus microplus Canestrini, is widely distributed in tropical and subtropical regions, causing huge economic losses to milk and meat production. Application of acaricides is the most widely used control strategy, but because of increasing evidence of resistance to acaricide and environmental concerns, novel strategies are needed. Use of entomopathogenic nematodes is a promising alternative for tick control. Six species of nematodes, Steinernema riobrave Cabanillas, Poinar, and Raulston (355 strain), S. carpocapsae (Weiser) (All strain), S. feltiae Filipjev (SN strain), Heterorhabditis bacteriophora Poinar (VS strain), H. indica Poinar, Karunakar, and David (HOM1 strain), and H. floridensis …

Displacement rates of tsetse affect performance of targets during vector control. Fly size, one of the indicators of population structure usually obtained from wing measurement, is among the determinants of displacement rates. Although recovery of tsetse in previous intervention areas has been widely reported, the population structure of tsetse that recover is rarely evaluated despite being associated with displacements rates. Previously, intervention trials had reduced tsetse densities by over 90% from >3 flies/trap/day to <1fly/trap/day on Big Chamaunga and Manga islands of Lake Victoria in western Kenya. In this study, we assessed the recovery in densities of Glossina fuscipes fuscipes on the two islands and evaluated the effects vector control might have on the population structure. A before and after intervention study was undertaken on four islands of Lake Victoria in western Kenya; Small and Big Chamaunga, Manga and Rusinga Islands, two of which tsetse control intervention had previously been undertaken. Three years after intervention average G. f. fuscipes catches in biconical traps were estimated on each island. Wing centroid size (CS) (a measurement of fly size) and shape, indicators of the population structure of flies from the four islands were compared using geometric morphometric analyses. CS and shape of available female but not male tsetse wings obtained before the intervention trial on Big and Small Chamaunga islands were compared with those from the same islands after the intervention trial. G. f. fuscipes apparent density on the previous intervention islands were>9 flies/trap/day. Irrespective of sex, wing shape did not isolate tsetse based on their islands of origin. The fly size from Big and Small Chamaunga did …

Fall armyworm, Spodoptera frugiperda (J E Smith), an economically important pest native to tropical and sub-tropical America has recently invaded Africa, causing substantial damage to maize and other crops. We evaluated functionality of a companion cropping system, ‘climate-adapted push-pull’, developed for control of cereal stemborers in drier agro-ecologies, as an added tool for the management of fall armyworm. The technology comprises intercropping maize with drought-tolerant greenleaf desmodium, Desmodium intortum (Mill.) Urb., and planting Brachiaria cv Mulato II as a border crop around this intercrop. Protection to maize is provided by semiochemicals that are emitted by the intercrop that repel (push) …
