BACKGROUND: Biopesticides and transgenic crops based on Bacillus thuringiensis (Bt) toxins are extensively used to control insect pests, but the rapid evolution of insect resistance seriously threatens their effectiveness. Bt resistance is often polygenic and complex. Mutations that confer resistance occur in midgut proteins that act as cell surface receptors for the toxin, and it is thought they facilitate its assembly as a membrane-damaging pore. However, the mechanistic details of the action of Bt toxins remain controversial.
RESULTS: We have examined the contribution of two paralogous ABC transporters and two aminopeptidases N to Bt Cry1Ac toxicity in the diamondback moth, …

The benefits of biopesticides and transgenic crops based on the insecticidal Cry-toxins from Bacillus thuringiensis (Bt) are considerably threatened by insect resistance evolution, thus, deciphering the molecular mechanisms underlying insect resistance to Bt products is of great significance to their sustainable utilization. Previously, we have demonstrated that the down-regulation of PxmALP in a strain of Plutella xylostella (L.) highly resistant to the Bt Cry1Ac toxin was due to a hormone-activated MAPK signaling pathway and contributed to the resistance phenotype. However, the underlying transcriptional regulatory mechanism remains enigmatic. Here, we report that the PxGATAd transcription factor (TF) is responsible for the …

The corn borer complex, consisting of Diatraea grandiosella (Dyar), Ostrinia nubilalis (Hübner), and D. saccharalis (F.), poses a risk to field corn that is not protected through the use of foliar-applied insecticides or Bacillus thuringiensis (Bt) proteins incorporated into the plant's genetics. In the southern U.S., corn borers have been adequately controlled through the widespread planting of Bt corn hybrids. Refuge systems have been implemented to prevent the selection of Bt-resistant populations in target species. Historically, structured refuge compliance among corn producers has been low, leading to the commercialization of seed blended refugia in areas of the U.S. where cotton …

Sugarcane aphid (SCA; Melanaphis sacchari Zehntner) is a key piercing-sucking pest of sorghum (Sorghum bicolor) that cause significant yield losses. While feeding on host plants, complex signaling networks are invoked from recognition of insect attack to induction of plant defenses. Consequently, these signaling networks lead to the production of insecticidal compounds or limited access of nutrients to insects. Previously, several studies were published on the transcriptomics analysis of sorghum in response to SCA infestation, but no information is available on the physiological changes of sorghum at the proteome level. We used the SCA resistant sorghum genotype SC265 for …

Greenhouses are technological advancements that have existed since the first century. Human life and the field of agriculture have greatly improved with their invention. However, greenhouses come with their own set of challenges. One of the biggest issues with greenhouse systems is the presence of large pest populations. This is due to the ample food and water supply along with the relative stability of greenhouses. Pests in greenhouses are usually dealt with through the application of chemical pesticides. However, integrated pest management (IPM) is gaining traction for use in greenhouse systems. This overview will cover the history of greenhouses and …
