Publications and Research

Retinal progenitor cells (RPCs) can be combined with nanostructured polymer scaffolds to generate compos- ite grafts in culture. One strategy for repair of diseased retinal tissue involves implantation of composite grafts of this type in the subretinal space. In the present study, mouse retinal progenitor cells (RPCs) were cultured on laminin- coated novel nanowire poly(e-caprolactone)(PCL) scaf- folds, and the survival, differentiation, and migration of these cells into the retina of C57bl/6 and rhodospsin −/−mouse retinal explants and transplant recipients were analyzed. RPCs were cultured on smooth PCL and both short (2.5μm) and long (27μ m) nanowire PCL scaffolds. Scaffolds with …

Recreation Visitor Attitudes Towards Management-Ignited Prescribed Fires In The Bob Marshall Wilderness Complex, Montana, Katie Knotek, Alan E. Watson, William T. Borrie, Joshua G. Whitmore, David Turner

United States Department of Agriculture, Forest Service / University of Nebraska-Lincoln: Faculty Publications

Research at the Bob Marshall Wilderness Complex in Montana explored differences in recreation visitors’ attitudes towards the use of management-ignited prescribed fires in the wilderness. A mail-back survey of visitors (n = 291) during the 2004 season revealed that over half of visitors would accept prescribed fires in wilderness. This support did not vary by ignition purpose: (a) to restore the natural role of fire or (b) to reduce hazardous fuels and potential for fire escaping to non-wilderness lands. Local visitors, however, were significantly more accepting of prescribed fires than non-local visitors across both ignition purposes. A smaller proportion …

Sex-Specific Feeding Rates And Provisioning Of Fruit To Nestling Bell's Vireo, Greg H. Farley Jan 2008

Sex-Specific Feeding Rates And Provisioning Of Fruit To Nestling Bell's Vireo, Greg H. Farley

The Prairie Naturalist

Provisioning of fruit to nestlings and possible sex-specific differences in feeding rate have not been reported for Bell's vireo (Vireo bellii; Brown 1993). While studying nesting ecology of Bell's vireo on Konza Prairie Biological Station, Geary and Riley counties, Kansas, I quantified feeding rate by sex and food type delivered to nestlings. Sex was determined by capturing adults with mist-nets and inspecting for a cloacal protuberance, as well as conducting behavioral observations of uniquely-marked individuals. Males sing regularly while moving around territories, as well as during incubation (Nolan 1960). I assigned "female" to the individual in these socially …

United States Army Corps of Engineers, Omaha District

Aboveground Hemiptera and Orthoptera communities were compared among three native and three restored mesic tallgrass prairies along the Platte River in central Nebraska to assess both the relative success of restored sites and the relationship between insect and plant communities. Hemiptera and Orthoptera were sampled using sweep nets in early June, mid-July, and mid-August 2000. Plant species composition was assessed in early June and mid-August. A total of 89 Auchenorrhyncha (71 Cicadellidae, 15 Fulgoroidea, and 3 Membracidae) and 23 orthopterans (15 Acrididae and 8 Tettigoniidae) were collected. Eighty-five plant species were observed in combined study sites. Shannon diversity was significantly …

The Wetlands Reserve Program Supports Migrating Waterfowl In Nebraska’S Rainwater Basin Region Jan 2008

The Wetlands Reserve Program Supports Migrating Waterfowl In Nebraska’S Rainwater Basin Region

United States Department of Agriculture, Forest Service / University of Nebraska-Lincoln: Faculty Publications

The Rainwater Basin region (RWB) consists of a 6,150 mi2 area of loess plains in south-central Nebraska (figure 1). The area is characterized by rolling plains formed by deep deposits of windblown silt with a high density of claypan playa wetlands. More than 200,000 acres of wetlands once existed in this region. As a result of agricultural and other development, only 17 percent of the original playa wetland area remains, most with hydrologic alterations. Siltation and colonization by invasive plant species (e.g., reed canary grass, narrow-leaved cattail, river bulrush) continue to threaten remaining wetland habitats in the RWB.

Despite historic …

Crosby, Kay L. Crabtree, Saul Phiri, Tendai J. M'Soka, Chipepo Kankasa, William J. Harrington, Charles D. Mitchell, Charles Wood

Nebraska Center for Virology: Faculty Publications

Human herpesvirus 8 (HHV-8), or Kaposi’s sarcoma (KS)-associated herpesvirus, has been linked to all forms of KS. The results of most current serological assays for the detection of HHV-8-specific antibodies have low levels of concordance among themselves. To establish a sensitive and specific testing strategy that can be used to screen for HHV-8-specific antibodies, three HHV-8 proteins, ORF65, ORF73, and K8.1A, were expressed by using baculoviral vectors in insect cells and incorporated into a monoclonal antibodyenhanced immunofluorescence assay (mIFA) termed the Sf9 three-antigen mIFA. The results obtained by this mIFA were compared to those obtained by a standard mIFA with …

Cpurter, Takahiro Soeta, Danny Ng, Liping Wang, Evan T. Brower, Shi-Hua Xiang, Young Do Kwon, Chih-Chin Huang, Richard Wyatt, Peter D. Kwong, Ernesto Freire, Amos B. Smith Iii, Joseph Sodroski

Nebraska Center for Virology: Faculty Publications

Human immunodeficiency virus (HIV-1) interaction with the primary receptor, CD4, induces conformational changes in the viral envelope glycoproteins that allow binding to the CCR5 second receptor and virus entry into the host cell. The small molecule NBD-556 mimics CD4 by binding the gp120 exterior envelope glycoprotein, moderately inhibiting virus entry into CD4-expressing target cells, and enhancing CCR5 binding and virus entry into CCR5-expressing cells lacking CD4. Studies of NBD-556 analogues and gp120 mutants suggest that: 1) NBD-556 binds within the Phe 43 cavity, a highly conserved, functionally important pocket formed as gp120 assumes the CD4- bound conformation; 2) the NBD-556 …

Varying Efficiency Of Long-Term Replication Of Papillomaviruses In Saccharomyces Cerevisiae, Adam J. Rogers, Malte Loggen, Karen Lee, Peter C. Angeletti Jan 2008

Varying Efficiency Of Long-Term Replication Of Papillomaviruses In Saccharomyces Cerevisiae, Adam J. Rogers, Malte Loggen, Karen Lee, Peter C. Angeletti

Nebraska Center for Virology: Faculty Publications

Human papillomaviruses (HPVs) replicate in mitotically active basal keratinocytes. Two virally encoded proteins, E1, a helicase, and E2, a transcription factor, are important players in replication and maintenance of HPV episomes. We previously showed that HPV16 could replicate stably in Saccharomyces cerevisiae [Angeletti, P.C., Kim, K., Fernandes, F.J., and Lambert, P.F. (2002)] and we identified cis-elements that mediate replication and maintenance [J. Virol. 76(7), 3350-3358.; Kim, K., Angeletti, P.C., Hassebroek, E.C., and Lambert, P.F. (2005)]. Here, we demonstrate that although multiple HPV genomes replicate stably in yeast, they do so with differing long-term efficiency; HPV6-Ura3 is replicated at the …

Wolf Body Mass Cline Across Minnesota Related To Taxonomy?, L. David Mech, William J. Paul Jan 2008

Wolf Body Mass Cline Across Minnesota Related To Taxonomy?, L. David Mech, William J. Paul

United States Geological Survey, Northern Prairie Wildlife Research Center: Publications

Recent genetic studies suggest that in northern Minnesota two species of wolves (Canis lupus L., 1758 or western wolf and Canis lycaon Schreber, 1775 (= Canis rufus Audubon and Bachman, 1851) or eastern wolf) meet and hybridize. However, little morphological information is available about these two types of wolves in Minnesota. We analyzed the mass of 950 female wolves and 1006 males older than 1 year from across northern Minnesota and found that it increased from 26.30 ± 0.56 kg (mean ± SE) for females and 30.60 ± 0.72 kg for males in northeastern Minnesota to 30.01 ± 0.43 …

Winter Cover Crops And Vinegar For Early-Season Weed Control In Sustainable Cotton, P. J. Moran, S. M. Greenberg Jan 2008

Winter Cover Crops And Vinegar For Early-Season Weed Control In Sustainable Cotton, P. J. Moran, S. M. Greenberg

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Weeds may be suppressed by winter cover crops and the use of organic herbicides such as vinegar. Black oat (Avena strigosa) and hairy vetch (Vicia villosa) winter cover crops were planted for 2 years as part of a sustainable production system for cotton in the Lower Rio Grande Valley of Texas, and were till-killed each spring prior to cotton planting. Palmer amaranth (Amaranthus palmeri), common purslane (Portulaca oleracea), and sunflower (Helianthus annuus) were frequently-encountered winter and spring weeds. Both cover crops controlled winter weeds as well as did winter tillage …

A Landscape Perspective In Managing Vegetation For Beneficial Plant-Pestnatural Enemy Interactions: A Foundation For Areawide Pest Management, Michael J. Brewer, Takuji Noma, Norman C. Elliott Jan 2008

A Landscape Perspective In Managing Vegetation For Beneficial Plant-Pestnatural Enemy Interactions: A Foundation For Areawide Pest Management, Michael J. Brewer, Takuji Noma, Norman C. Elliott

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

In the USA, Europe and increasingly in other regions, cropping systems designed for high production output are significant features of the landscape. Deployment of mechanized and high-input cropping systems over the last 50 years has resulted in substantial transformation and fragmentation of major grassland, shrubland and woodland systems throughout the world. These cropping systems are typically less diverse in species composition, structure and ecological functioning than those found in the original plant community (Altieri, 2004). Decreases in plant diversity of agroecosystems (i.e. the crops themselves and surrounding remnants of the original plant system) have negatively affected ecosystem functions (Freemark, 2005). …

Comparison Of Nitrogen Fertilization Methods And Rates For Subsurface Drip Irrigated Corn In The Semi-Arid Great Plains, D. D. Tarkalson, J. O. Payero

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

In semi‐arid areas such as western Nebraska, interest in subsurface drip irrigation (SDI) for corn is increasing due to restricted irrigation allocations. However, crop response quantification to nitrogen (N) applications with SDI and the environmental benefits of multiple in‐season (IS) SDI N applications instead of a single early‐season (ES) surface application are lacking. The study was conducted in 2004, 2005, and 2006 at the University of Nebraska‐Lincoln West Central Research and Extension Center in North Platte, Nebraska, comparing two N application methods (IS and ES) and three N rates (128, 186, and 278 kg N ha-1) using a …

Effects Of Shielding Adenoviral Vectors With Polyethylene Glycol On Vector-Specific And Vaccine-Mediated Immune Responses, Eric A. Weaver, Michael A. Barry Jan 2008

Effects Of Shielding Adenoviral Vectors With Polyethylene Glycol On Vector-Specific And Vaccine-Mediated Immune Responses, Eric A. Weaver, Michael A. Barry

Nebraska Center for Virology: Faculty Publications

Many individuals have been previously exposed to human adenovirus serotype 5 (Ad5). This prior immunity has long been known to hinder its use for gene therapy and as a gene-based vaccine. Given these immunogenicity problems, we have tested whether polyethylene glycol (PEG) can blunt immune effects against Ad5 during systemic and mucosal vaccination. Ad5 vectors were covalently modified with 5-, 20-, and 35-kDa linear PEG polymers and evaluated for their ability to produce immune responses against transgene antigen prod- ucts and the vector itself. We show that shielding Ad5 with different-sized PEGs generally reduces transduction and primary antibody responses by …

Krabbenhoft, Randall J. Hunt, John F. Walker, Paul F. Schuster, Ann Chalmers, Brent T. Aulenbach, Norman E. Peters, Mark Marvin-Dipasquale, David W. Clow, Martin M. Shafer

United States Geological Survey: Staff Publications

The small watershed approach is well-suited but underutilized in mercury research. We applied the small watershed approach to investigate total mercury (THg) and methylmercury (MeHg) dynamics in streamwater at the five diverse forested headwater catchments of the US Geological Survey Water, Energy, and Biogeochemical Budgets (WEBB) program. At all sites, baseflow THg was generally less than 1 ng L-1 and MeHg was less than 0.2 ng L-1. THg and MeHg concentrations increased with streamflow, so export was primarily episodic. At three sites, THg and MeHg concentration and export were dominated by the particulate fraction in association with …

Evolutionary And Structural Analyses Of Alpha-Papillomavirus Capsid Proteins Yields Novel Insights Into L2 Structure And Interaction With L1, John Lowe, Debasis Panda, Suzanne Rose, Ty Jensen, Willie Hughes, For Yue Tso, Peter C. Angeletti

Nebraska Center for Virology: Faculty Publications

Background: PVs (PV) are small, non-enveloped, double-stranded DNA viruses that have been identified as the primary etiological agent for cervical cancer and their potential for malignant transformation in mucosal tissue has a large impact on public health. The PV family Papillomaviridae is organized into multiple genus based on sequential parsimony, host range, tissue tropism, and histology. We focused this analysis on the late gene products, major (L1) and minor (L2) capsid proteins from the family Papillomaviridae genus Alpha-papillomavirus. Alpha-PVs preferentially infect oral and anogenital mucosa of humans and primates with varied risk of oncogenic transformation. Development of evolutionary associations between …

Kaposi’s sarcoma occurs at high incidence among Zambian adults and children, but there is a paucity of data on human herpesvirus 8 (HHV-8) incidence and routes of infection, especially in children. Between 1998 and 2004, the authors conducted a prospective study of viral transmission in a cohort of 684 children in Lusaka, Zambia, to estimate the annual incidence of HHV-8 from birth through 48 months of age. Maternal and pediatric human immunodeficiency virus type 1 (HIV-1) infection status was also determined. The results, based on 1,532 child-years of follow-up, showed that HHV-8 seroconversion occurs early in life. The incidence rate …

We assessed the utility of the modified Kigali combined (MKC) staging system for predicting survival in HIV-infected Zambian adults in a prospective, longitudinal, open cohort. From 1995 to 2004, HIV-discordant couples (one HIV-infected partner and one HIV-negative partner) were recruited from couples’ voluntary counseling and testing centers in Lusaka, Zambia and followed at 3-month intervals. MKC stage, which incorporates clinical stage with erythrocyte sedimentation rate (ESR), hematocrit, and body mass index (BMI), was determined at enrollment. Kaplan–Meier survival and Cox proportional hazard methods were used to calculate median survival and relative hazards. We enrolled 1479 HIV-discordant couples with a combined …

Biology And Biological Control Of Exotic True Thistles, Rachel Winston, Rich Hansen, Mark Schwarzlander, Eric Coombs, Carol Bell Randall, Rodney Lym Jan 2008

Biology And Biological Control Of Exotic True Thistles, Rachel Winston, Rich Hansen, Mark Schwarzlander, Eric Coombs, Carol Bell Randall, Rodney Lym

United States Department of Agriculture, Forest Service / University of Nebraska-Lincoln: Faculty Publications

“Thistle” is an old English name for a large variety of weedy, prickly plants that grow throughout the world. The most notable characteristics of thistles are the prickly stems and leaves and the bracts around the flower head. While many different plants have “thistle” in their common name, only certain plant species fit the taxonomic requirements of being considered “true thistles.” True thistle species fall within the family Asteraceae, the tribe Cardueae, and the subtribe Carduinae. Examples of plants that are not true thistles include yellow starthistle (subtribe Centaureinae), sow thistle (subtribe Sonchinae), and Russian thistle (family Chenopodiaceae). Only true …

Mosquito repellency data on acylpiperidines derived from the United States Department of Agriculture archives were modeled by using molecular descriptors calculated by CODESSA PRO software. An artificial neural network model was developed for the correlation of these archival results and used to predict the repellent activity of novel compounds of similar structures. A series of 34 promising N-acylpiperidine mosquito repellent candidates (4a–4q’) were synthesized by reactions of acylbenzotriazoles 2a–2p with piperidines 3a–3f. Compounds (4a–4q’) were screened as topically applied mosquito repellents by measuring the duration of repellency after application to cloth patches worn on the arms of human volunteers. Some …

Using The Solvent Retention Capacity Test When Breeding Wheat For Diverse Production Environments, Carl Walker, Kimberly A. Garland-Campbell, Brady Carter, Kimberlee Kidwell

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

The solvent retention capacity (SRC) test is used to predict commercial baking performance of soft wheat (Triticum aestivum L.) by measuring the capacity of flour to retain each of four solvents— water, Na2CO3, sucrose, and lactic acid— to assess overall absorption capacity, starch damage, pentosan and gliadin content, and glutenin quality, respectively. Our objectives were to determine sources of variation in the test, repeatability, and optimum scale and resource allocation needed to maximize efficiency. Duplicate SRC tests were conducted for each solvent using two flour sample sizes (5 and 0.2 g) from two field replications …

Hamulonema gen. nov. is proposed for Teladorsagia hamata and Ostertagia kenyensis in the ostertagiine nematode fauna found in artiodactyl hosts from Africa. Monomorphic species representing this genus are characterized by a bilaterally symmetrical and parallel synlophe in males and females, a 2-2-1 bursal formula, an accessory bursal membrane that is strongly cuticularized and reduced, a strongly reduced dorsal lobe and ray, and robust spicules with a simple, weakly pointed, ventral process, and curved, hooklike dorsal process. Species referred to Hamulonema nov. gen. are immediately distinguished from those of Camelostrongylus, Longistrongylus, Marshallagia, Orloffia, Ostertagia, and Pseudomarshallagia …
