Role Of Vegf-C In Proliferation And Migration In A Cancer Model, Emily Marie Benke
Role Of Vegf-C In Proliferation And Migration In A Cancer Model, Emily Marie Benke
Theses and Dissertations
Head and neck cancer ranks high among the most common cancers world wide. In addition, there is a high recurrence rate, as well as a high prevalence of loco-regional tumor spread. Among many factors contributing to metastasis is vascular endothelial cell growth factor C. VEGF-C is primarily an inducer of new lymph vessel formation, typically during embryogenesis; however, some advanced cancers show a significant increase in VEGF-C expression, suggesting a role in metastasis. In the current study, the effects of VEGF-C expression were tested in HN12 cells, which are highly metastatic and known to express high levels of the chemokine …
Augmentation Of T Cell Expansion For Adoptive Immunotherapy By Alternate Gamma Chain Cytokines And By Gemcitabine Mediated Inhibition Of Myeloid Derived Suppressor Cells, Hanh Le
Theses and Dissertations
Successful treatment of cancer with adoptive immunotherapy (AIT) is dependent on the ability to produce large numbers of tumor-specific, functional T cells. The purpose of this thesis is to explore ways in which T cell expansion could be augmented. We have focused on exploring alternate gamma chain cytokines as stimulators of T cell proliferation and differentiation in addition to investigating the potential usefulness of gemcitabine (GEM) in abrogating the immunosuppressive effects of myeloid derived suppressor cells (MDSCs). B16 melanoma sensitized draining lymph node cells that have been activated in vitro with bryostatin-1 and ionomycin (B/I) were expanded in either IL-7/15 …
Camk-Ii Promotes Beta-Catenin-Dependent Transcription By Binding Flightless-I, Jamie Mcleod
Camk-Ii Promotes Beta-Catenin-Dependent Transcription By Binding Flightless-I, Jamie Mcleod
Theses and Dissertations
Transient intracellular elevations of Ca2+ are common signaling mechanisms used to allosterically regulate proteins. One potential target of Ca2+ is Ca2+/calmodulin dependent protein kinase type II (CaMK-II). CaMK-II is a multi-functional protein kinase known to influence cellular pathways such as cell motility and cell cycle progression. Within the cell cycle, CaMK-II promotes the expression of the regulator protein cyclin D1, which is necessary for cell cycle progression. To further understand CaMK-II’s role in cyclin D1 expression, the binding partners of cytosolic CaMK-II were studied using mass spectrometry. Several proteins were identified including β actin, β tubulins, tropomodulin- 3 and Flightless-I. …
Fibronectin-Dependent Activation Of Camk-Ii Promotes Focal Adhesion Turnover By Inducing Tyrosine Dephosphorylation Of Fak And Paxillin, Charles Easley Iv
Fibronectin-Dependent Activation Of Camk-Ii Promotes Focal Adhesion Turnover By Inducing Tyrosine Dephosphorylation Of Fak And Paxillin, Charles Easley Iv
Theses and Dissertations
Transient elevations in Ca2+ have previously been shown to promote focal adhesion disassembly and cell motility. Yet the targets of these Ca2+ transients have not been fully examined. In this study, we demonstrate that CaMK-II, a Ca2+/calmodulin dependent protein kinase, is activated in response to β1 integrin engagement with fibronectin to influence fibroblast adhesion and motility. We also show that CaMK-II is dynamically localized to the cell surface using Total Internal Reflection Fluorescence microscopy (TIRFm) and that inhibition of CaMK-II with two mechanistically distinct, membrane permeant inhibitors accelerates spreading on fibronectin, enlarges paxillin-containing focal adhesions and …
Regulation Of Platelet-Activating Factor Acetylhydrolase By Oxidized Phospholipids And Proinflammatory Cytokines, Muralikrishna Mukkamala
Regulation Of Platelet-Activating Factor Acetylhydrolase By Oxidized Phospholipids And Proinflammatory Cytokines, Muralikrishna Mukkamala
Theses and Dissertations
Platelet-Activating Factor Acetylhydrolase (PAFAH) is a monocyte-derived phospholipase A2 that catalyzes the hydrolysis of platelet-activating factor (PAF) and has been implicated in atherosclerosis. Although PAF and other proinflammatory stimuli are postulated to induce the enzyme, mechanisms controlling PAFAH expression are largely unknown at present. We have shown that PAFAH induction in monocytes is increased in response to oxidized phospholipids. The PAFAH 5' flanking region has at least 10 putative Stat elements, and IL-6 has been shown to be downstream from the prostaglandin receptor, EP2, which has been shown to bind oxidized phospholipids, prompting the hypothesis that Stat proteins might regulate …
Role Of The Small Terminase Subunit Encoded By Staphylococcus Aureus Pathogenicity Island Sapi1 In Formation Of Sapi1 Transducing Particles, Nicholas Paul Olivarez
Role Of The Small Terminase Subunit Encoded By Staphylococcus Aureus Pathogenicity Island Sapi1 In Formation Of Sapi1 Transducing Particles, Nicholas Paul Olivarez
Theses and Dissertations
Staphylococcus aureus pathogenicity island SaPI1 is a genomic element that is mobilized and transduced at high frequency by helper phage 80α. SaPI1 encodes a small terminase protein that belongs to the phage small terminase subunit family. The presence of SaPI1-encoded small terminase suggests that it plays a role in SaPI1-specific packaging into transducing particles by complexing with the 80α large terminase subunit and redirecting recognition to a pac site on SaPI1 DNA from 80α DNA. The effects of deleting the small terminase genes in SaPI1 and in a prophage copy of 80α are consistent with this hypothesis. Induction of the …
Inflammation-Associated Gene Regulation In Primary Astrocytes, Glial Tumors And Cellular Differentiation, Katarzyna Marta Wilczynska
Inflammation-Associated Gene Regulation In Primary Astrocytes, Glial Tumors And Cellular Differentiation, Katarzyna Marta Wilczynska
Theses and Dissertations
This dissertation elucidates several independent molecular mechanisms that function in astrocytes and glial tumor cells, and suggest that developmental and inflammatory signals may contribute to the development of brain tumors. First, we analyzed the mechanism of TIMP-1 activation in astrocytes and glioblastoma cells. TIMP-1 expression is activated by IL-1, which is the major neuroinflammatory cytokine, via simultaneous activation of IKK/NF-kB and MEK3/6/p38/ATF-2 pathways in primary human astrocytes. In contrast to astrocytes, TIMP-1 is expressed at lower levels in glioblastomas, and is not regulated by IL-1 due to either dysfunctional IKK/NF-kB or MEK3/6/p38/ATF-2 activation. Thus, we propose a novel mechanism of …
Importance And Effect Of Nitrogen On Crop Quality And Health, Jürg Blumenthal, David D. Baltensperger, Kenneth G. Cassman, Stephen Mason, Alexander Pavlista
Importance And Effect Of Nitrogen On Crop Quality And Health, Jürg Blumenthal, David D. Baltensperger, Kenneth G. Cassman, Stephen Mason, Alexander Pavlista
Department of Agronomy and Horticulture: Faculty Publications
Nitrogen (N) is often the most limiting factor in crop production. Hence, application of fertilizer nitrogen results in higher biomass yields and protein yield and concentration in plant tissue is commonly increased. Nitrogen often affects amino acid composition of protein and in turn its nutritional quality. In cereals, abundant supply of nitrogen decreases the relative proportion of lysine and threonine, thus, reducing the biological value of the protein. Increasing nitrogen supply generally improves kernel integrity and strength, resulting in better milling properties of the grain. In oil seed crops, protein levels are increased upon nitrogen fertilization, whereas oil concentration is …
Harvester Ant Nests Improve Recovery Performance Of Drought Impacted Vegetation In Grazing Regimes Of Semiarid Savanna, Texas, Nancy Nicolai, Fred Smeins, Jerry Cook
Harvester Ant Nests Improve Recovery Performance Of Drought Impacted Vegetation In Grazing Regimes Of Semiarid Savanna, Texas, Nancy Nicolai, Fred Smeins, Jerry Cook
Department of Agronomy and Horticulture: Faculty Publications
Vegetation composition is often dictated by grazing intensity in semiarid savannas; recovery following drought may depend on pre drought species composition. Nests of the red harvester ant, Pogonomyrmex barbatus, affect the dynamics, composition and recovery of post drought communities due to their larger size, greater seed production and higher perennial grass richness. We hypothesized that vegetation at ant nests would survive drought and recover faster than vegetation in the surrounding grasslands, but that individual and population recovery would depend on plant species composition, which, in turn, would be influenced by grazing intensity. We assessed nest influence on density, cover, …

Agricultural Systems: Agroecology And Rural Innovations For Development [Book Review], Charles A. Francis
Department of Agronomy and Horticulture: Faculty Publications
To welcome yet another book on sustainable development, a reviewer must look for what is unique and what adds value to the volumes already published. In Agricultural Systems, editors Sieglinde Snapp and Barry Pound have assembled a collection of chapters that goes beyond the usual praise and criticism of the green revolution and the focus on economic development. Their book provides a first-hand story by people with lengthy experiences in the field, especially in Africa, who have developed and tested grassroots, participatory development approaches. The result is a valuable set of principles, strategies and case studies that paint a new …

Characterization Of The Folate Salvage Enzyme P-Aminobenzoylglutamate Hydrolase In Plants, Gale G. Bozzo, Gilles J. C. Basset, Valeria Naponelli, Alexandre Noiriel, Jesse F. Gregory Iii, Andrew D. Hanson
Department of Agronomy and Horticulture: Faculty Publications
Folates break down in vivo to give pterin and p-aminobenzoylglutamate (pABAGlu) fragments, the latter usually having a polyglutamyl tail. Pilot studies have shown that plants can hydrolyze pABAGlu and its polyglutamates to p-aminobenzoate, a folate biosynthesis precursor. The enzymatic basis of this hydrolysis was further investigated. pABAGlu hydrolase activity was found in all species and organs tested; activity levels implied that the proteins responsible are very rare. The activity was located in cytosol/vacuole and mitochondrial fractions of pea (Pisum sativum L.) leaves, and column chromatography of the activity from Arabidopsis tissues indicated at …

Phylloquinone (2-methyl-3-phytyl-1,4-naphthoquinone; vitamin K1) is vital to plants. It is responsible for the oneelectron transfer at the A1 site of photosystem I, a process that involves turnover between the quinone and semi-quinone forms of phylloquinone. Using HPLC coupled with fluorometric detection to analyze Arabidopsis leaf extracts, we detected a third redox form of phylloquinone corresponding to its fully reduced – quinol–naphthoquinone ring (PhQH2). A method was developed to quantify PhQH2 and its corresponding oxidized quinone (PhQ) counterpart in a single HPLC run. PhQH2 was found in leaves of all dicotyledonous and monocotyledonous species tested, …

Dental enamel forms through a protein-controlled mineralization and enzymatic degradation with a nanoscale precision that new engineering technologies may be able to mimic. Recombinant fulllength human amelogenin (rH174) and a matrix-metalloprotease (MMP-20) were employed in a pHstat titration system that enabled a continuous supply of calcium and phosphate ions over several days, mimicking the initial stages of matrix processing and crystallization in enamel in-vitro. Effects on the self-assembly and crystal growth from a saturated aqueous solution containing 0.4 mg/ml rH174 and MMP-20 with the weight ratio of 1:1000 with respect to rH174 were investigated. A transition from nanospheres to fibrous …

Cardiac fibroblasts produce and degrade extracellular matrix and are critical in regulating cardiac remodeling and hypertrophy. Fibroblasts are activated by factors such as transforming growth factor β and inhibited by agents that elevate 3′,5′-cyclic adenosine monophosphate (cAMP) levels. cAMP signal generation and response is known to be compartmentalized in many cell types in part through the colocalization of receptors and specific adenylyl cyclase isoforms in lipid rafts and caveolae. The present study sought to define the localization of key G protein-coupled receptors with adenylyl cyclase type 6 (AC6) in lipid rafts of rat cardiac fibroblasts and to determine if this …

Nixtamalization is the process of steeping dried corn in hot water with calcium hydroxide (lime) with subsequent removal of all or most of the pericarp through washing. The resulting product is called nixtamal. Approximately 60% of corn tortillas in Mexico are produced from nixtamal, with the remainder prepared from nixtamalized corn flour. Nixtamal was fortified with micronutrient premix containing iron, zinc, folic acid, niacin, riboflavin, and thiamin. Premix composition followed a proposed Mexican regulation for corn flour fortification, adjusted for moisture. Effects of premix on masa adhesiveness, hardness, and pH, as well as tortilla sensory properties, stretchability, rollability, and color …

Salt Marshes As A Source Of Chromophoric Dissolved Organic Matter (Cdom) To Southern California Coastal Waters, Catherine D. Clark, Liannea P. Litz, Stanley B. Grant
Biology, Chemistry, and Environmental Sciences Faculty Articles and Research
To determine chromophoric dissolved organic matter (CDOM) sources in Southern California coastal waters, optical properties of a river outlet and adjacent tidally flushed salt marshes were monitored (dry season; June July 2001). Average absorption coefficients doubled at ebb vs. flood tides (4.8 +/- 1.5 vs. 2.1 +/- 0.9 m(-1); 300 nm), suggesting significant salt marsh CDOM inputs into coastal waters. Average spectral slopes were not statistically different for any sites or tides (0.010 +/- 0.002 nm(-1)), consistent with salt marsh CDOM dominating coastal waters. Three-dimensional fluorescence excitation-emission matrices (EEMs) at ebb tide showed contributions from terrestrial, protein, and marine humic-like …
