Civil and Environmental Engineering Faculty Publications

In hydrologic research, there is a need to manage, archive, and publish data in a discoverable way to increase data reuse, transparency, and reproducibility. Multidimensional space-time data are commonly used in hydrologic research, and systems are needed for sharing and exchanging such data. Simply exchanging files may result in loss of metadata information and can be challenging when files are large. We developed an approach to manage, share, and publish multidimensional space-time data in HydroShare, a next generation hydrologic information system and domain specific repository. This paper presents the design, development, and testing of this approach. We selected the Network …

Characterization Of The Factors Influencing Retained Austenite Transformation In Q&P Steels, Derrik David Adams

Theses and Dissertations

Formable Advanced High-Strength Steels (AHSS) have a unique combination of strength and ductility, making them ideal in the effort to lightweight vehicles. The AHSS in this study, Quenched and Partitioned 1180, rely on the Transformation Induced Plasticity (TRIP) effect, in which retained austenite (RA) grains transform to martensite during plastic deformation, providing extra ductility via the transformation event. Understanding the factors involved in RA transformation, such as local strain and grain attributes, is therefore key to optimizing the microstructure of these steels. This research seeks to increase understanding of those attributes and the correlations between microstructure and RA transformation in …

Three-Dimensional Hydrodynamic Focusing For Integrated Optofluidic Detection Enhancement, Erik Scott Hamilton Apr 2020

Three-Dimensional Hydrodynamic Focusing For Integrated Optofluidic Detection Enhancement, Erik Scott Hamilton

Theses and Dissertations

The rise of superbugs, including antibiotic-resistant bacteria, and virus outbreaks, such as the recent coronavirus scare, illustrate the need for rapid detection of disease pathogens. Widespread availability of rapid disease identification would facilitate outbreak prevention and specific treatment. The ARROW biosensor microchip can directly detect single molecules through fluorescence-based optofluidic interrogation. The nature of the microfluidic channels found on optofluidic sensor platforms sets some of the ultimate sensitivity and accuracy limits and can result in false negative test results. Yet higher sensitivity and specificity is desired through hydrodynamic focusing. Novel 3D hydrodynamic focusing designs were developed and implemented on the …

Loading Orientation Dependence On The Compressive Response Of Ice-Templated Ceramic-Polymer Composites, Sashanka Akurati, Justine Marin, Dipankar Ghosh

College of Engineering & Technology (Batten) Posters

Natural materials are made from weak constituents, yet exhibit an excellent synergy of high stiffness, strength, and damage-tolerance. They consist of alternate layers of the hard and soft phases with a complex hierarchical structural organization. The ice-templating technique provides an approach to fabricate multilayered architectures for engineering applications. In this technique, an aqueous ceramic suspension is solidified unidirectionally leading to phase separation into alternating layers of ice-crystals and ceramic particles. Ice-crystals are sublimated by freeze-drying process and resultant ceramic foams are sintered to impart strength. The fabricated sintered ceramic foams contain alternate layers of oriented ceramic lamella walls and pores. …

Power and Energy Institute of Kentucky Faculty Publications

Axial flux PM (AFPM) machines are used particularly in applications requiring a compact structure. Their disc shape topology and axial air-gap have led to a variety of configurations including two popular ones: the yokeless and segmented armature (YASA), and the single-stator single rotor or single sided machine. In this study, a comprehensive comparative analysis of these configurations is conducted at different magnetic and electric loadings. It is found that at lower loadings, typically employed for air-cooled machines, the torque/ampere characteristics of the YASA machine are almost identical to those of a single sided machine constructed with half the magnet volume. …

Design And Analysis Of A Piezoelectric Pulse Wave Velocity Sensor, Haley Richards, Andrew Closson, John Xj Zhang

ENGS 88 Honors Thesis (AB Students)

Pulse Wave Velocity (PWV) is strongly correlated with blood pressure and a valuable biomarker in assessing hypertension. This report evaluates the feasibility of noninvasive blood pressure determination from piezoelectric PWV measurements. A wearable piezoelectric prototype was developed to detect PWV in the radial artery, and a computational model of the sensor was designed to simulate a piezoelectric sensor response under ideal conditions. These methods provide insight into the relationship between PWV and blood pressure, and the ability of piezoelectric materials to detect blood flow characteristics. This work lays the groundwork for future device development in this field.

Engineering A Switchable Nanosystem For Customizable Therapeutics, David Diebold, Hanzhe Chen Apr 2020

Engineering A Switchable Nanosystem For Customizable Therapeutics, David Diebold, Hanzhe Chen

Bioengineering Senior Theses

Exosomes are nanovesicles that are naturally secreted by mammalian cells in vivo for intercellular communications. Due to their inherent targeting ability, exosomes have a potential for therapeutic applications. However, due to their physiological derivation, the isolation of engineered exosomes has been a major obstacle to their therapeutic application, and successful disease-targeting has been difficult to control. Recently, we have developed an exosome technology that borrows from switchable Chimeric Antigen Receptor (sCAR) T-Cell Therapy and Strep-tag engineering to overcome these obstacles. Here, we describe the development of a de novo method to produce genetically modified exosomes with switchable targeting ability and …

Predicting Depression Progression Rates In Radiotherapy Patients, Ardella Phoa, Joshua Vincent, Shani Williams Apr 2020

Predicting Depression Progression Rates In Radiotherapy Patients, Ardella Phoa, Joshua Vincent, Shani Williams

Bioengineering Senior Theses

Cancer is a highly prevalent disease that affects millions of people worldwide. In addition to the physiological effects of the disease, cancer patients are more likely to be diagnosed with Major Depressive Disorder (MDD). Unfortunately, prior research has shown that MDD can also decrease the efficacy of radiotherapy cancer treatments. Currently, there is no way to predict, prevent, or mitigate this comorbidity, preventing physicians from administering supplemental therapies. In this paper, we propose a low-cost and efficient computational tool that can be utilized to quantify a patient’s likelihood of developing depression. To do so, we used PET images and a …
