DNA damage, such as single base lesions and mismatches, is highly prevalent within cells. If these DNA damage events are not repaired, they could lead to mutations and thus disease and cancer. Intricate repair mechanisms are in place to fix these damage events, one such being Base Excision Repair (BER) and associated enzyme: Thymine DNA Glycosylase (TDG). The goal of this study was to investigate changes in hearing sensitivity and neural changes in the brainstem by measuring neural activity of the auditory brainstem in an established preclinical model of chronic migraine. Thermostable fluorescent proteins, such as thermal green protein (TGP) and yellow thermal protein (YTP), could be used as biosensors to monitor cellular activity and as a fusion tag to monitor a protein of interest. The use of fluorescent proteins can sometimes be limited in certain organelles with low pH and in thermophilic organisms. This research aims to improve the thermal stability of TGP and YTP. This thesis uses quantitative and qualitative research methods to: (1) explore the base causes of insurgency in Ukraine, (2) examine the historical basis for Ukrainian insurgency, (3) provide historical examples of successful and unsuccessful insurgencies to contrast against Ukraine, and (4) provide recommendations for NATO and Ukrainian policy. Collectively, this project demonstrates that current Russian counterinsurgency tactics will not be successful without significant adjustment. This Ukrainian insurgency may also derail the possibility of peace in the region.

NOD-like receptors (NLRs) are intracellular proteins that play an important role in the regulation of the innate immune response to pathogens. Since being identified, various functions for NOD-like receptor pyrin domain containing 12 (NLRP12) have been suggested. It has been shown to negatively regulate the inflammatory response through canonical and noncanonical NF-kB signaling pathways, control tumorigenesis and gut homeostasis and exacerbate inflammation through the formation of a multi-protein complex called an inflammasome. The clinical construct of psychopathy has been researched extensively relative to incarceration. Cognitive insight has shown to be related to prognosis and treatment outcomes with severe psychopathologies. There has been, however, limited research on both psychopathy and cognitive insight in relation to incarceration. The purpose of the present study was to examine the moderation of cognitive insight levels on rates of incarceration (the number of times someone has been incarcerated in a government detention facility) based on their level of self-reported psychopathic traits. This study explored students’ mathematical reasoning skills and offered supports to advance them through a collaborative argumentation framework in a college intermediate algebra class. The goals of this study were to make observations about student reasoning, identify specific actions to address those observations, and document student growth in reasoning as a result of those actions. An iterative analysis, mixed method study was conducted in which the researcher engaged students in responding to questions that required conceptual understandings using a collaborative argumentation framework as a tool to identify and code components of their responses—claim, evidence, and reasoning. This current study advanced understanding of the career goal-setting and relevant goal-related attitudes to the career planning process. Specifically, this study evaluated how career goal commitment, career goal self-efficacy and grit affect career goal structures and effort. A multidimensional career goal commitment scale was developed to differentiate intrinsic (affective) from rational types of commitments. Findings supported affective career goal commitment as consequential to the completeness of career goal structures and in career-relevant effort. Also, career goal self-efficacy was found to predict career planners’ self-efficacy for shorter term performance.

In this study, I developed Deep Learning interatomic potentials to model a multi-phase and multi-component system of Ni-based Superalloys. The system has up to three major phase constituents, namely Gamma, Gamma Prime, and Transition-metal rich Carbide. I utilized invariant scalar-based and/or equivariant, tensor-based neural network (NN) approach as implemented in DEEPMD, NEQUIP/ALLEGRO codes, respectively, and Moment Tensor Potential (MTP). The Black-capped chickadee (Poecile atricapillus) and Carolina chickadee (P. carolinensis) are North American songbird species that hybridize in a narrow contact zone stretching latitudinally from New Jersey to Kansas, USA. The association between genetic ancestry and song type in this hybrid zone has been studied independently several times and found to be minimal or absent, likely due to the influence of cultural transmission on learned song in the oscine passerine clade to which the chickadees belong. The North American grapes species Vitis rupestris Scheele and Vitis riparia Michx have been the pillars of rootstock breeding for many decades. Though a large body of viticultural knowledge has been accumulated on their impact on grafted scions, the genetic basis of their root system architecture (RSA) has received limited scientific attention. In this study, I generated and analyzed adventitious root systems from dormant cuttings of 22 V. riparia and 19 V. rupestris accessions, as well as 162 interspecific F1 hybrid progeny from a cross between V. rupestris (♀) and V. riparia (♂). This thesis integrates and discusses two collaborative manuscripts that revolve around the shared subject matter of multi-level research on systemic issues. All the signaling pathways and inflammatory reactions are maintained in a healthy mode if cell cycle checkpoints are precisely regulated and kept in homeostasis. AIM2 inflammasome is a cytoplasmic double-stranded DNA sensor. Upon activation, it elicits an inflammatory reaction with the release of pro-inflammatory cytokines such as IL-1β and IL-18 and they are cleaved by the proteases, Caspase-1 from their inactive form. Metal oxide nanoparticles (MONPs) are becoming more popular in today’s environment. They contribute significantly to the technologies in agriculture and food development but there is little understanding to how MONPs, including ZnO, CuO, TiO2, and SnO2, impact human health and the environment. Our growth assay revealed that none of these negatively affects viability in the budding yeast, Saccharomyces cerevisiae. In contrast, both human thyroid cancer cells (ML-1) and rat medullary thyroid cancer cells (CA77) displayed a significant reduction in viability with the treatment of CuO and ZnO. First found experimentally in 2015, topological Weyl materials are desirable compounds that have garnered much interest due to their ability to conduct electricity via their surface states even though the bulk material is a semimetal. Such a candidate, Sm3HfBi5, was discovered with a flux crystal growth method, following an extensive amount of reaction syntheses. This thesis reports on the discovery, growth, structural characterization via x-ray diffraction, and magnetization measurements on Sm3HfBi5.

Closeness And Conflict In The Parent-Child Relationship And Parental Self-Efficacy During Childhood, Stephanie Boekweg

The present study examined the relationship between closeness in the parent-child relationship, conflict in the parent-child relationship, and parental self-efficacy during childhood. Quantitative data from an electronic survey on one hundred eighty-eight participants was collected. Results from a simple linear correlation indicated that parental self-efficacy during childhood and the parent-child relationship quality measures of closeness and conflict were moderately correlated. A simple regression showed that parental self-efficacy during childhood did not predict closeness nor conflict in the parent-child relationship when controlling for current parental self-efficacy. However, parental self-efficacy during childhood significantly predicted current-parental self-efficacy.

In this research, I examine the concept of the Other in horror films. I use Kenneth Burke’s identification, Jean-Francois Lyotard’s metanarrative concept, and Lennard Davis’s bell curve of normalcy to describe the Other and how otherness relates to disability. First, I discuss how horror films have portrayed the Other historically in a negative context and slowly transition to the virtuous Other, the final girl.
