A significant fraction of Earth consists of mountainous terrain. However, the question of how to monitor the surface–atmosphere carbon exchange over complex terrain has not been fully explored. This article reports on studies by a team of investigators from U.S. universities and research institutes who carried out a multiscale and multidisciplinary field and modeling investigation of the CO2 exchange between ecosystems and the atmosphere and of CO2 transport over complex mountainous terrain in the Rocky Mountain region of Colorado. The goals of the field campaign, which included ground and airborne in situ and remote-sensing measurements, were to characterize unique features …

Chapter 1

DNA barcoding based on the mitochondrial cytochrome c oxidase subunit I (COI) gene sequence is emerging as a useful tool for identifying unknown, whole or partial organisms to species level. However, the application of only a single mitochondrial marker for robust species identification has also come under some criticism due to the possibility of erroneous identifications resulting from species hybridizations and/or the potential presence of nuclear-mitochondrial psuedogenes. The addition of a complementary nuclear DNA barcode has therefore been widely recommended to overcome these potential COI gene limitations, especially in wildlife law enforcement applications where greater confidence in the …

The measurement of trace analytes in aqueous systems has become increasingly important for understanding ocean primary productivity. In oceanography, iron (Fe) is a key element in regulating ocean productivity, microplankton assemblages and has been identified as a causative element in the development of some harmful algal blooms. The chemosenor developed in this study is based on an indicator displacement approach that utilizes time-resolved fluorescence and fluorescence resonance energy transfer as the sensing mechanism to achieve detection of Fe3+ ions as low as 5 nM. This novel approach holds promise for the development of photoactive chemosensors for ocean deployment.

Characterization Of Winds Through The Rotor Plane Using A Phased Array Sodar And Recommendations For Future Work, Regina Anne Deola

Publications (E)

Portable remote sensing devices are increasingly needed to cost effectively characterize the meteorology at a potential wind energy site as the size of modern wind turbines increase. A short term project co-locating a Sound Detection and Ranging System (SODAR) with a 200 meter instrumented meteorological tower at the Texas Tech Wind Technology Field Site was performed to collect and summarize wind information through an atmospheric layer typical of utility scale rotor plane depths. Data collected identified large speed shears and directional shears that may lead to unbalanced loads on the rotors. This report identifies suggestions for incorporation of additional data …

Sudden Surface Warming/Drying Events Caused By Typhoon Passages Across Taiwan, T. C. Chen, Shih-Yu (Simon) Wang, M. C. Yen, A. J. Clark, J. D. Tsay

Plants, Soils, and Climate Faculty Publications

Typhoon passages across Taiwan can generate sudden surface warming in downslope regions. Special characteristics and mechanisms for 54 such warming events that were identified during the 1961–2007 period are examined. Preferred warming regions were identified in northwest Taiwan, where warming is generated by downslope flow from east or northeast winds in westward-moving typhoons, and in southeast Taiwan, where it is generated by downslope flow from west or northwest winds in northwestward-moving typhoons. In addition to the orographic effect, warmings occurred exclusively within nonprecipitation zones of typhoons. Most northwest (southeast) warmings occur during the day (night) with an average lifetime of …

Study Of Secondary Metabolite Gene Expression In Marine Microbial Co-Cultures Using Quantitative Real-Time Pcr, Crystal A. Conway 2010 Nova Southeastern University

Study Of Secondary Metabolite Gene Expression In Marine Microbial Co-Cultures Using Quantitative Real-Time Pcr, Crystal A. Conway

HCNSO Student Theses and Dissertations

Interactions among microbial organisms often cannot be observed directly, but they can be inferred genetically using new molecular techniques. The analysis of secondary metabolite gene expression produced by co-cultured marine microbial species allows us to see how these organisms interact with one another when kept in the same environment. Co-cultures of three different strains of marine bacteria, P. aeruginosa PAO1, Roseobacter denitrificans OCH114, and Salinispora arenicola CNS-205 were grown in a laboratory setting, and using the Real-Time qPCR method gene expression levels of two different secondary metabolite producing genes from each organism was accessed across three time points. P. aeruginosa …

Analysis Of Causes Of Icing Conditions Which Contributed To The Crash Of Continental Flight 3407, Frederick R. Mosher, Debbie Schaum, Chris Herbster, Tom Guinn

Applied Aviation Sciences - Daytona Beach

On February 12, 2009, at 10:20 p.m. EST, Continental Connection Flight 3407 from Newark to Buffalo crashed 5 miles short of the runway at Buffalo, killing all 49 people on board and one person on the ground. The National Transportation Safety Board (NTSB) is still investigating the crash, but preliminary reports show the airplane was experiencing icing conditions before the crash. Preliminary reports indicate that the pilot did not respond properly to the icing conditions, which contributed to the crash. However, the presence of the icing conditions which were significant enough to cause aircraft problems needs further investigation to determine …

Characterization of phytoplankton community composition is critical to understanding the ecology and biogeochemistry of the oceans. One approach to taxonomic characterization takes advantage of differing pigmentation between algal taxa and thus differences in fluorescence excitation spectra. Analyses of bulk water samples, however, may be confounded by interference from chromophoric dissolved organic matter or suspended particulate matter. Here, we describe an instrument that uses a laser trap based on a Nikon TE2000-U microscope to position individual phytoplankton cells for confocal fluorescence excitation spectroscopy, thus avoiding interference from the surrounding medium. Quantitative measurements of optical power give data in the form of …

A Transition-Phase Teleconnection Of The Pacific Quasi-Decadal Oscillation, Shih-Yu (Simon) Wang, R. R. Gillies, L. E. Hipps, J. Jin

Plants, Soils, and Climate Faculty Publications

The atmospheric circulation patterns associated with the Pacific quasi-decadal oscillation (QDO) are investigated using available observational data from 1948 to 2007. Previous studies indicate that the Pacific QDO is characterized by a distinct lifecycle in the form of sea surface temperature (SST) patterns. In the warm and cool phases of the Pacific QDO, the SST patterns resemble those associated with the El Niño-Southern Oscillation (ENSO). During the warm–cool and cool–warm transitions of the Pacific QDO, recurrent SST patterns are also clearly visible. The rotated empirical orthogonal function analysis on the 10–15 year filtered data shows that the evolutions of SST …

Diurnal Movements And Site Fidelity Of The Florida Manatee, Trichechus Manatus Latirostris, In Fort Lauderdale, Fl., Kym Walsh

HCNSO Student Theses and Dissertations

The diurnal movements of Florida manatees (Trichechus manatus latirostris) in warmwater refugia are poorly understood and these may prove critical in accurately estimating populations of these animals. Previous studies indicate that manatee populations in such refuges in Tampa Bay, FL, peak mid-day and decline towards evening when the animals leave to forage. This trend suggests that variation in the timing of aerial manatee population surveys may hamper accurate estimation of the true population size. Aerial survey counts are known to underestimate population size, yet the extent of the underestimation may be greater than expected. In this study data …
