Equine arteritis virus (EAV) is the causative agent of equine viral arteritis (EVA), a respiratory, systemic, and reproductive disease of horses and other equid species. Following natural infection, 10–70% of the infected stallions can become persistently infected and continue to shed EAV in their semen for periods ranging from several months to life. Recently, we reported that some stallions possess a subpopulation(s) of CD3+ T lymphocytes that are susceptible to in vitro EAV infection and that this phenotypic trait is associated with long-term carrier status following exposure to the virus. In contrast, stallions not possessing the CD3+ T …

Assessing Readiness For Clinical Practice: Students’ Perspectives Of Their Veterinary Curriculum, L. Chris Sanchez, Alison Kwiatkowski, Jeff Abbott, Dana N. Zimmel, Linda S. Behar-Horenstein
The Qualitative Report
Studies describing the effectiveness of a veterinary curriculum from the student perspective are currently sparse. The overall purpose of this investigation was to describe students’ perceived preparedness for clinical practice. Three focus group meetings with fourth year veterinary students were conducted. Data were open-coded and categorized to identify themes. Four main themes emerged: Challenging communications, Un/appreciating curricular experiences, Documenting demands impede case involvement, and Hungering for timely, effective feedback. Overall students felt comfortable talking to clients about medicine but less comfortable discussing euthanasia or money; they appreciated the split clinical curriculum but questioned the value of the 1st/2nd year courses; …

Evaluation Of Several Pre-Clinical Tools For Identifying Characteristics Associated With Limb Bone Fracture In Thoroughbred Racehorses, Anthony Nicholas Corsten
Open Access Theses
Catastrophic skeletal fractures in racehorses are devastating not only to the animals, owners and trainers, but also to the perception of the sport in the public eye. The majority of these fatal accidents are unlikely to be due to chance, but are rather an end result failure from stress fractures. Stress fractures are overuse injuries resulting from an accumulation of bone tissue damage over time. Because stress fractures are pathological, it is possible that overt fractures can be predicted and prevented. In this study, third metacarpals (MC3) from 33 thoroughbred racehorse comprised of 8 non-fractured controls and 25 horses that …

Chikungunya virus (CHIKV) infection is an emerging or re-emerging infectious disease found in several countries. The ecology of this virus involves humans, reservoir animals, and mosquito vectors. Two experiments were conducted in this study. Blood samples were collected and tested for CHIKV infection by using immunocytochemistry (ICC) staining and reverse transcription polymerase chain reaction (RT-PCR). For the first experiment, BALB/c mice were inoculated with 106.3 CID50/mouse. Mean CHIKV titers in the BALB/c mice were 103.3, 101.5, 102.3, and 102.3 CID50/ml of serum on day 1 to day 4 post inoculation (PI), respectively. For the second experiment, there were two groups …

Regulation Of Alpha-Herpesvirus Reactivation From Latency By Stress, Insun Kook
School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research
Bovine herpes virus 1 (BHV-1) and Herpes simplex virus 1 (HSV-1) are crucial etiological viral agent of clinical diseases. HSV-1 and BHV-1 establish latent infection in sensory neurons. Periodically, reactivation from latency occurs resulting in virus excretion and transmission. Stress increases corticosteroid levels and the incidence of HSV-1 and BHV-1 reactivation from latency. The synthetic corticosteroid, dexamethasone (DEX) mimics stress and induces BHV-1 and HSV-1 reactivation. However, molecular mechanisms by which corticosteroid mediates viral reactivation are not well understood. My dissertation has focused on elucidating events that induce BHV- 1 or HSV-1 reactivation during the early stages of stress-induced escape …

The Impact Of Tickling Rats On Human-Animal Interactions And Rat Welfare, Megan Renee Lafollette
Open Access Theses
Rats initially fear humans which can lead to negative affect, poor welfare, and difficult handling. Also, modeling and measuring positive affect states in rats can pose an additional challenge. Heterospecific play, or “tickling,” is a handling habituation technique that mimics rat rough-and-tumble play that is being used to study positive affect. It can also be used to reduce fear of human, improve welfare, and elicit a positive affect state. However, current studies implementing the technique in laboratory rats use a wide variety of protocols to achieve differential results. Unlike in laboratory environment, pet store rats experience high levels of novelty …

Challenges And Prospects Of The Embryo Transfer Industry From A European Standpoint, Michel Thibier
The Thai Journal of Veterinary Medicine
In vivo derived (IVD) and in vitro produced (IVP) embryo transfers are respectively the 2nd and 3rd generations of the reproductive biotechnologies. Commercial application of these technologies began in the seventies for the former and in the eighties for the latter. After 40 years, they have clearly proved their sustainability to the livestock industry. The present article deals with bovine embryos only. The numbers of embryos collected or produced and transferred worldwide are reported in the first part. About one million bovine embryos are transferred each year. This clearly shows the significant impact they have on the cattle industry. The …

Advanced Biotechnologies For Wildlife Fertility Preservation, Pierre Comizzoli
The Thai Journal of Veterinary Medicine
Reproductive biotechnologies are critical tools for saving and maintaining endangered species. Some successes have been reported with the use and integration of artificial insemination (with fresh or frozen-thawed semen) in conservation programs. However, not a single species is currently managed through oocyte freezing or embryo-based technologies. This is primarily due to the lack of knowledge of species biology, as well as inadequate facilities, space, expertise, and funding needed for their successful application. More fundamental studies of animal reproductive biology as well as more fertility preservation options are needed with all parties involved (reproductive technologists, zoo biologists and conservationists) adopting parallel …

Encephalitozoon cuniculi (E. cuniculi) is an intracellular zoonotic parasite that is recognized as a common parasite of rabbits. Infected rabbits show neurological and renal disorder. The polar tube protein 1 of the spore is revealed to be antigenicity. The present study aimed to produce a recombinant polar tube protein 1 (rPTP1) for which the codon usage of a native ptp1 gene was optimized and synthesized for protein production in Escherichia coli (E. coli). Both a native ptp1 gene amplified by PCR technique and an optimized ptp1 gene were ligated into a pQE30 expression vector and transformed into an E. coli …

Canine atopic dermatitis (AD) is a common, chronic relapsing inflammatory skin disease. An aqueous extract, PG102, derived from Actinidia arguta fruit, has been investigated as an anti-inflammatory agent in humans and atopy-like animal models, but its underlying mechanism for canine AD treatment has not been fully understood. This study evaluated the in vitro anti-inflammatory effect of PG102 with respect to the representative allergy-related cytokine panel from dust mite allergen extract (DME)-stimulated peripheral blood mononuclear cells (PBMCs) of canine AD patients using quantitative real-time PCR. PBMCs were isolated from eight dogs with definite diagnoses of AD and stimulated with DME for …

Stem cell-based therapy employing bone marrow-derived mesenchymal stem cells has been proposed as a promising approach for bone regenerative treatment. Despite numerous studies supporting the application in human medicine, such information in veterinary practice is still lacking. Thus, this study aimed to investigate in vitro osteogenic differentiation potential of canine bone marrow-derived mesenchymal stem cells under different doses of β-glycerophosphate supplement. The isolated cells showed self-renewing ability and expressed pluripotent marker genes, zinc finger protein 42 and octamer-binding transcription factor 4, suggesting their potentiality in vitro. In vitro osteogenic differentiation was performed by using a regular osteogenic induction medium containing …

The Thai Journal of Veterinary Medicine
Enterococcus gallinarum and E. casseliflavus possess intrinsic vancomycin resistance, which is mediated by vanC-1 and vanC-2 gene clusters, respectively. The VanC resistance phenotype is expressed inducibly or constitutively. This study characterized the expression types for vancomycin resistance in 23 porcine E. gallinarum and 19 porcine E. casseliflavus isolates with vancomycin minimum inhibitory concentrations between 4 and 16 μg/ml. The vanSC and vanSC-2 genes were sequenced and the deduced amino acid sequences were compared to those of the inducible and constitutive resistance reference strains. Analysis of growth curves revealed that the resistance phenotype in all isolates was induced by vancomycin. In …
