Most Veterans who use the Veterans Health Administration (VHA) also utilize private-sector health care providers. To better inform local and regional health care planning, we assessed the association between reliance on VHA ambulatory care and total and system-specific preventable hospitalization rates (PHRs) at the state level. We conducted a retrospective dynamic cohort study using Veterans with diabetes mellitus, aged 66 years or older, and dually enrolled in VHA and Medicare parts A and B from 2004 to 2010. While controlling for median age and proportion of males, we measured the association between reliance on VHA ambulatory care and PHRs at …

Heteroatom-doped carbon dots (CDs) with excellent optical characteristics and negligible toxicity have emerged in many applications including bioimaging, biosensing, photocatalysis, and photothermal therapy. The metal-doping of CDs using various heteroatoms results in an enhancement of the photophysics but also imparts them with multifunctionality. However, unlike nonmetal doping, typical metal doping results in low fluorescence quantum yields (QYs), and an unclear photoluminescence mechanism. In this contribution, we detail results concerning zinc doped CDs (Zn-CDs) with QYs of up to 35%. The zinc ion charges serve as a surface passivating agent and prevent the aggregation of graphene p–p stacking, leading to an …

Accumulation of senescent cells over time contributes to aging and age-related diseases. However, what drives senescence in vivo is not clear. Here we used a genetic approach to determine if spontaneous nuclear DNA damage is sufficient to initiate senescence in mammals. Ercc1-/Δ mice with reduced expression of ERCC1-XPF endonuclease have impaired capacity to repair the nuclear genome. Ercc1-/Δ mice accumulated spontaneous, oxidative DNA damage more rapidly than wild-type (WT) mice. As a consequence, senescent cells accumulated more rapidly in Ercc1-/Δ mice compared to repair-competent animals. However, the levels of DNA damage and senescent cells in Ercc1-/Δ mice never exceeded that …

Background: Patients with penicillin allergies who require broad-spectrum antimicrobial therapy for the treatment of sepsis often receive a carbapenem instead of a cephalosporin due to concerns of penicillin-cephalosporin cross-reactivity. • As rates of antibiotic resistance continue to increase, institutions should prioritize efforts to preserve the efficacy of carbapenems. • Reduction of carbapenem use in institutions has been shown to lower incidence rates of carbapenem-resistant organisms . • Cephalosporins have previously demonstrated low rates of cross-reactivity in penicillin allergic patients. • As of May 29th, 2018, updates to our …

Anecdotal evidence indicates that naloxone prices have risen in recent years, but limited research has examined the magnitude of these increases and potential causes. We contribute nationally representative evidence to help answer each of these questions, including wholesale pricing data from a proprietary drug sales database span- ning January 2006 to February 2017. We find that all formulations of naloxone increased in price since 2006 except for Narcan Nasal Spray. These cumulative increases totaled 2281% for the 0.4 MG single-dose products, 244% for the 2 MG single-dose products, 3797% for the 4 MG multi-dose products, and 469% for the 0.4 …

Giant salvinia (Salvinia molesta) is one of the most noxious invasive species in the world. The fern is known to have primary, secondary, and tertiary growth forms, which are also commonly hypothesized as growth stages. The identification of these forms is primarily based on the size and folding status of the floating leaves. However, we identified 12 forms in the greenhouse and the field. Our experiments showed that the folding of floating leaves is a reversible trait dependent on water access. The floating leaves quickly fold in response to water shortage, reducing water loss and needs, decreasing growth, …

Abstract
Background: Brain tumor vasculature can be significantly compromised and leakier than that of normal brain blood vessels. Little is known if there are vascular permeability alterations in the brain adjacent to tumor (BAT). Changes in BAT permeability may also lead to increased drug permeation in the BAT, which may exert toxicity on cells of the central nervous system. Herein, we studied permeation changes in BAT using quantitative fluorescent microscopy and autoradiography, while the effect of chemotherapy within the BAT region was determined by staining for activated astrocytes.
Methods: Human metastatic breast cancer cells (MDA-MB-231Br) were injected into left ventricle …

APOBEC3G (A3G) belongs to the family of cytosine deaminases that play an important role in the innate immune response. Similar to other, two-domain members of the APOBEC family, A3G is prone to concentration-dependent oligomerization, which is an integral for its function in the cell. It is shown that oligomerization of A3G is related to the packing mechanism into virus particle and, is critical for the so-called roadblock model during reverse transcription of proviral ssDNA. The role of oligomerization for deaminase activity of A3G is widely discussed in the literature; however, its relevance to deaminase activity for different oligomeric forms of …

Precocious puberty, measured by thelarche or menarche, is a well-established breast cancer risk factor. Using a unique and patented tissue engineering system we established cell lines from 48 primary cultures of normal breast tissue from reduction mammoplasties and used them as an in vitro model of thelarche, quantifying episphere formation and ductal differentiation. Differential ductal formation was observed based on ancestry. In light of African American (AA) precocious thelarche both in vivo and as we observed in vitro, we hypothesized that AA women would exhibit an increased proportion of breast stem cells, with greater potency for differentiation. Five AA cell …

NER plays an important role in remediating DNA damage. Latimer et al., have previously shown nucleotideexcision repair (NER) to be intrinsically deficient in stage I sporadic BC by function and gene expression. Previous work performed on 12 isogenically matched stage I tumors and non-tumor adjacent (NTA) samples identified 2 types of NTA tissue with regard to functional NER. One type that represented 75% ofthe samples, had lower NER capacity, similar to the tumor (Low: Low pair). The other type had high NER relative to the tumor (High: Low pair). Two isogenic NTA/tumor cell line pairs representing these 2 types ofNTA …

G protein-coupled receptor (GPCR) heteromerization is rapidly evolving field. Traditionally, GPCRs were thought to behave has independent entities or protomers. However, that line of reasoning has since been brought into question. Currently, it appears that many, if not all, GPCRs signal and function as heteromeric receptor complexes. The roles of GPCR heteromerization are vast and can include: maturation and trafficking; dynamic regulation; allostery and cooperativity; signal transduction; and receptor internalization.
The majority of studies identifying and characterizing GPCR heteromers were performed using recombinant receptors. Only four receptor pairs have been identified and validated in native tissues. The overall objective of …
