Modulation Of Tlr4/Nf-Κb, Nrf2/Ho-1 And Pi3k/Akt Signaling By Cilostazol Mitigates Lipopolysaccharide-Induced Septic Acute Kidney Injury, Ahmed F. Mohamed, Helmy M. Sayed, Hala F. Zaki, Marwa M. Safar

Bulletin of Faculty of Pharmacy Cairo University

Cilostazol was investigated as a protective agent against lipopolysaccharide (LPS)-associated acute kidney damage in mice. Cilostazol (50 mg/kg/day; p.o) administered for 7 consecutive days before a single LPS dose (2 mg/kg; i.p) hampered serum creatinine and cystatin C as well as renal KIM-1 and NGAL, repressed TLR4 and MYD88 transcription as well as NF-κB p65, IL-1β and MDA content, boosted Nrf-2 mRNA expression, HO-1 activity and GSH content. This was synchronous with an upregulation of p-PI3K and p-Akt expressions. Collectively, cilostazol prevented LPS renal injury which might correspond to modulation of TLR4/NF-κB, Nrf2/HO-1 and PI3K/Akt pathways.

Comparing The Toxicant Responses And Culturing Characteristics Of Long-Term Laboratory-Reared And Field Populations Of Ceriodaphnia Dubia, Victoria Rose Lydy

Ceriodaphnia dubia is an EPA-recommended organism used in regulatory toxicity testing of surface waters and commercial chemicals. They are often cultured for extended periods in the laboratory with little knowledge of how it impacts subsequent generations. This thesis investigated if C. dubia cultured for an extended period were representative of three recently-collected field populations by comparing their culturing characteristics and sensitivities to toxicants. Comparative chronic toxicity tests with sodium chloride, the neonicotinoid insecticide thiamethoxam, and mixtures of the two toxicants measured differences in sensitivities among the laboratory and field cultures, specifically in the variability of responses. A variety of molecules can be radiolabeled and delivered to a cancer site for the purposes of diagnostics and therapy. Among the most promising of tumor targeting molecules are peptides and antibodies. These bio-inspired molecules can be designed and synthesized to target and respond to cancer cells based on the properties of those cells. Matrix metalloproteinase (MMP) enzymes are over-expressed by some metastatic cancers, in which they are responsible for the degradation and remodeling of the extracellular matrix. Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is an organophosphate flame retardant. The primary TDCPP metabolite, bis(1,3-dichloro-2-propyl) phosphate (BDCPP), is detectable in the urine of over 90 % of Americans. Epidemiological studies show sex-specific associations between urinary BDCPP levels and metabolic syndrome, which is an established risk factor for type 2 diabetes, heart disease, and stroke. We used a mouse model to determine whether TDCPP exposure disrupts glucose homeostasis. Successful invasive plant management—where invaders are sufficiently reduced and diverse native plant communities recover—remains an elusive goal for land managers. The site- and landscape-scale drivers of variable management outcomes and vegetation recovery are poorly understood due to a lack of rigorous experiments that characterize longer term vegetation trends across contexts. We present the results of a five-year experiment across eight subestuaries of Chesapeake Bay, representing a gradient of watersheds with differing dominant land-use types and anthropogenic impacts, to evaluate invasive and native plant response to herbicide management. Pharmaceuticals are increasingly detected in water bodies, and their presence can negatively impact aquatic organisms. This effect can be amplified when combined with increasing salinity in freshwater ecosystems. Acetaminophen is a widely used analgesic that is commonly found in river, streams, and waters where it is discharged directly. Therefore, organisms present in these locations (e.g., insects, snails, amphibians, and fish) are likely to be affected by acetaminophen. West Nile virus (WNV) causes annual outbreaks globally and is the leading cause of mosquito-borne disease in Unite States. In the absence of licensed therapeutics, there is an urgent need to develop effective and safe human vaccines against WNV. Honey bee colonies have perished at an alarming rate for over 15 years. Among the multitude of threats to bee health, virus infections and agrochemical exposure appear to be major drivers of colony loss. As virus infection is among the most damaging influences on bee health, we first investigated a putative target for antiviral immunity, termed KATP channels. The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a highly transmissible virus that causes acute respiratory disease with possible long-term complications, known as coronavirus disease 2019 (COVID-19). Griffithsin (GRFT; including the oxidation-resistant variant Q-GRFT used in this study) is an algal carbohydrate-binding protein exhibiting antiviral effects against many enveloped viruses, including all major variants of SARS- CoV-2 reported to date. Q-GRFT has been recombinantly manufactured in Nicotiana benthamiana plants, thoroughly purified, and formulated as an intranasal spray designed to be developed as a non-vaccine broad-spectrum prophylactic product for acute use. The fetus is deprived of vital blood, oxygen, and nutrients due to faulty maternofetal circulation, leading to a severe lack of fetal growth. Since current prenatal testing is highly ineffective at diagnosing the condition, many babies with IUGR are negatively impacted before, during, and after birth. Physicians can put patients at risk of infections if bacteria contaminate their ties during their normal hospital routines.
