Preclinical work indicates that exposure to traditional antidepressant medications, in adolescent and adult female subjects, alters reward-related behavior later in life. In recent years, the anesthetic ketamine (KET), now used as a fast-acting antidepressant, has shown promising therapeutic efficacy for the management of depression. However, the potential long-term behavioral consequences of KET exposure across development have not been thoroughly assessed. Thus, to address this issue, we examined if KET exposure, during adolescence or early adulthood, results in enduring alterations in responsivity to the rewarding properties of sucrose and cocaine later in life. Purpose of review: Impaired reward processing and amotivation are well documented in schizophrenia. We aim to review the current state of neuroimaging and behavioral research addressing components of motivational deficits in this complex and impairing syndrome. Evidence will be integrated to inform the ongoing development of effective strategies for behavioral rehabilitation. Recent findings: While striatal dopamine and aberrant reward prediction errors have been connected to amotivation in schizophrenia, they are not sufficiently full explanations of reward processing impairments. Frontal dysfunction and associated cognitive control deficits also have evidenced involvement in atypical reward prediction, learning, and valuation. Ongoing work supports the …

The increasing use of quantitative real-time polymerase chain reaction (qPCR) as a method for quantifying gene expression has led to an increased demand for standardization of data analysis methods to ensure accurate reporting and robust, reproducible results. The exponential nature of qPCR amplification results in the potential magnification of what are usually very small sources of error. Relative gene expression calculations circumvent this issue by normalizing target gene expression data to within-sample expression of a previously validated, stably expressed reference gene or genes. Alcohol use disorder (AUD) is a prevalent neuropsychiatric disease with profound health, social, and economic consequences. With an estimated 50% heritability, identifying genes that engender risk and contribute to the underlying neurobiological mechanisms represents an important first step in developing effective treatments. Gene expression studies are an important source of candidate genes for studying AUD, providing windows into the molecular machinery engaged by the brain in response to ethanol. Social support is associated with various outcomes such as depression and anxiety. Less work has investigated how support received from various sources cumulatively is associated with internalizing symptoms. Research suggests that above other sources, parents and peers are the most strongly associated with internalizing outcomes. Looking at female adolescents specifically, the current study explored 1) what patterns of social support across sources emerge in a female adolescent sample, and 2) how do depressive and anxiety symptoms vary based on those patterns of social support? Autism spectrum disorder (ASD) and intellectual disability (ID) are highly prevalent neurodevelopmental disorders characterized by social and communication deficits, stereotyped behaviors, cognitive dysfunction, and deficits in adaptive behaviors. The pathogenesis underlying these disorders remains unknown, and thus no pharmacologic or genetic therapies are currently available. Recent progress in the field has shown that haploinsufficiency of the AT-rich interactive domain-containing 1B (ARID1B) gene is a genetic cause of ASD and ID. The common marmoset is a good model for research because they are easy to house and have complex social relationships (French et al., 2019). Marmosets are sensitive to social isolation, and when introduced to a stressor, the HPA axis is activated (Saltzman & Abbott, 2011). The purpose of this experiment is to validate marmosets as a translational model for stress due to social relationships in humans. This is done by validating biomarker concentration levels at baseline, then comparing the concentration when introduced to a stressor. The results from these experiments have begun to provide new insights into the effects of spaceflight on mammalian physiological systems. This study assessed the role of stress systems in the nucleus accumbens (NAc) in promoting sex differences in the reinforcing effects of nicotine. Intravenous self-administration (IVSA) of various doses of nicotine was compared following overexpression of corticotropin-releasing factor (CRF) in the NAc of female and male rats. Ovariectomized (OVX) females were also included to assess the role of ovarian hormones in promoting nicotine reinforcement. Rats received intra-NAc administration of an adeno-associated vector that overexpressed CRF (AAV2/5-CRF) or green fluorescent protein (AAV2/5-GFP). All rats were then given extended access (23 h/day) to an inactive and an active lever that delivered nicotine. …

The hindlimb unloading (HU) model has been used extensively to simulate the cephalad fluid shift and musculoskeletal disuse observed in spaceflight with its application expanding to study immune, cardiovascular and central nervous system responses, among others. Most HU studies are performed with singly housed animals, although social isolation also can substantially impact behavior and physiology, and therefore may confound HU experimental results. Psychopathy is a multifaceted disorder characterized by a lack of cognitive and emotional empathy. The traditional model of psychopathy divides the disorder into two factors: Factor 1 consists of the interpersonal and affective traits of psychopathy while Factor 2 measures antisocial behaviors and lifestyle choices. The attention-to-the-eyes hypothesis argues that psychopathic individuals have impaired emotion recognition (specifically for fear) due to deficits in orienting attention to salient facial features like the eyes. Psychopathic individuals also display blunted autonomic responding to emotional stimuli, though whether this is due to attention-orienting deficits remains to be clarified. Adult neurogenesis is the addition of new neurons in the adult brain. In mammals, neurogenesis is largely restricted to the hippocampus and olfactory bulb, which underlie learning, memory, and olfaction. In songbirds, new neurons are incorporated into telencephalic regions that subserve the production, perception, and maintenance of song. Studies of the regulation of neurogenesis have exclusively focused on a single brain region and within a single hemisphere. Interestingly, it has yet to be investigated whether there are correlations between numbers of new neurons across regions and hemispheres within the same animal. Animals engage in intricately woven action sequences that are constructed from trial-and-error learning, but the mechanisms by which the brain links together individual actions which are later recalled as fluid chains of behavior are not fully understood. The aim of this dissertation is to investigate the learning and goal-directed control of action sequences in rats. Experiment 1 addresses a question that comes out of a reinforcement learning model of action sequencing: how does the extent of training change how the performance of an action sequence is impacted by reward devaluation. Daily rhythms in light exposure influence the expression of behavior by entraining circadian rhythms and through its acute effects on behavior (i.e., masking). Importantly, these effects of light are dependent on the temporal niche of the organism; for diurnal organisms, light increases activity, whereas for nocturnal organisms, the opposite is true. Here we examined the functional and morphological differences between diurnal and nocturnal rodents in retinorecipient brain regions using Nile grass rats (Arvicanthis niloticus) and Sprague-Dawley (SD) rats (Rattus norvegicus), respectively. Objective: The present study examined forgiveness of others, self-forgiveness, sleep, and health in a nationally representative sample of United States adults. It was hypothesised that sleep would mediate the associations of forgiveness of others and self-forgiveness with health.
Design: A nationally representative survey of 1,423 United States adults.
Main Outcome Measures: Measures included forgiveness of others, self-forgiveness, sleep quantity, sleep quality, psychological distress, life satisfaction, and self-rated physical health.
Results: Forgiveness of others (β = .20, p < .001) and self-forgiveness (β = .11, p < .01) were associated with sleep and forgiveness of others (β = .24, p < .001) and self-forgiveness (β = .27, p < .001) were associated with health. Sleep was associated with health (β = .45, p < .001) and also acted as a mediator of the associations of forgiveness of others (β = .09, p < .01) and self-forgiveness (β = .05, p < .01) with health.
Conclusions: Forgiveness of others and self-forgiveness may attenuate emotions such as anger, regret, and rumination and provide …

Defects in the nervous system can impact the normal function of an organism. By examining mutations during neuronal development, we can better understand the significance of specific pathways. The EphR/ephrin pathway is crucial for axonal guidance during development. The simple organism, C. elegans, allow us to study the impact of EphR/ephrin genes on neuronal morphology, synapse connections, and basic behavior. In specific EphR/ephrin mutants, we see change in morphology and loss of synapses in a thermosensory and also a chemosensory circuit. The translational gap between animal models and clinical trials is a longstanding, yet largely unresolved, limitation in the study of cognition. This discrepancy is largely due to the differences in how cognition is assessed in animal models compared to those in clinical populations. In the stimulus-response (S-R) learning literature, for example, the techniques used to assess the acquisition of habitual behaviour differ greatly across species, leading to poor cross-species translation and often conflicting results.
