Laser Guide Star (LGS) systems are essential for adaptive optics in ground-based astronomical observation. This dissertation demonstrates the feasibility of semiconductor-based LGS systems using the membrane external-cavity surface-emitting laser (MECSEL) platform, employing multiple quantum wells. Various laser cavity configurations were analyzed through simulations and experiments. The in-well pumping method was explored to reduce the quantum defect and address thermal limitations. Multi-pass pumping schemes were designed with Zemax modeling and demonstrated experimentally. To simplify multi-pass pumping, the hybrid-MECSEL (H-MECSEL) design was introduced. COMSOL modeling studied thermal management and thermal lensing effect.

Many biophysical techniques, such as single-molecule fluorescence correlation spectroscopy, Förster resonance energy transfer, and fluorescence anisotropy, measure the translation and rotation of biomolecules to quantify molecular processes at the nanoscale. These methods often simplify data analysis by assuming isotropic rotational diffusion, e.g., that molecules wobble within a circular cone. This simplification ignores the anisotropy present in many biological contexts that may cause molecules to exhibit different degrees of diffusion in different directions. Here, we loosen this assumption and establish a theoretical framework for describing and measuring anisotropic rotational diffusion using fluorescence imaging. Radiosondes are balloon borne atmospheric instruments that are a critical tool for understanding dynamics in the lower layers of the atmosphere. The low-cost radiosondes developed in the Space and Atmospheric Instrumentation Lab have been further developed to improve the system's use as a science-quality atmospheric instrument that is unique in its ability to simultaneously track multiple sondes with a single ground station. Sensors to measure temperature and pressure were added to improve measurements of the atmospheric state. Amyloid-beta (Aβ42) aggregates are characteristic Alzheimer’s disease signatures, but probing how their nanoscale architectures influence their growth and decay remains challenging using current technologies. Here, we apply time-lapse single-molecule orientation-localization microscopy (SMOLM) to measure the orientations and rotational “wobble” of Nile blue (NB) molecules transiently binding to Aβ42 fibrils. We correlate fibril architectures measured by SMOLM with their growth and decay over the course of 5 to 20 min visualized by single-molecule localization microscopy (SMLM). We discover that stable Aβ42 fibrils tend to be well-ordered and signified by well-aligned NB orientations and small wobble. SMOLM also shows that increasing order …

Microscopy plays a crucial role across various scientific fields by enabling structural and functional imaging with microscopic resolution. In biomedicine, microscopy contributes to basic research and clinical diagnosis. Conventionally, optical microscopy derives its contrast from the amplitude of the optical wave and provides visualization of the physical structure of the sample qualitatively. In this dissertation, the problem of finding lower error bounds on the minimum mean-squared error (MMSE) and the maximum capacity achieving distribution for a specific channel is addressed. Presented are two parts, a new lower bound on the MMSE and upper and lower bounds on the capacity achieving distribution for a Binomial noise channel. The new lower bound on the MMSE is achieved via use of the Poincare inequality. It is compared to the performance of the well known Ziv-Zakai error bound. The second part considers a binomial noise channel and is concerned with the properties of the capacity-achieving distribution. …

Stabilization of topological spin textures in layered magnets has the potential to drive the development of advanced low-dimensional spintronics devices. However, achieving reliable and flexible manipulation of the topological spin textures beyond skyrmion in a two-dimensional magnet system remains challenging. Here, we demonstrate the introduction of magnetic iron atoms between the van der Waals gap of a layered magnet, Fe3GaTe2, to modify local anisotropic magnetic interactions. Consequently, we present direct observations of the order-disorder skyrmion lattices transition. In addition, non-trivial topological solitons, such as skyrmioniums and skyrmion bags, are realized at room temperature. Our work highlights …

This thesis presents the comprehensive design, fabrication, and demonstration of advanced high-power, high-efficiency single-mode semiconductor lasers operating at a wavelength of 9xxnm. We begin with the design of the laser epitaxial structure, serving as the cornerstone for achieving high-power high-efficiency lasers. Our methodology integrates a semi-analytical calculation model, which accounts for Longitudinal Spatial Hole Burning (LSHB) and Two-Photon Absorption (TPA) effects, facilitating a thorough exploration of how design parameters influence output power and conversion efficiency. This approach offers an effective and time-efficient epitaxial structure optimization strategy compared to conventional full 3D simulation models.

Biaxial-oriented polypropylene (BOPP) films are characterized by unfavorable aging behavior because of their poor susceptibility to high temperatures, humidity, and high electric fields. This makes them unqualified to withstand harsh operating conditions, such as in capacitor applications. This study investigates the impact of annealing BOPP samples at 100 °C for five hours after fluorination at different times (15, 30, and 60 minutes) on their electrical and mechanical performance under electro-thermal stresses. Scanning electron microscope (SEM) images confirm that there is an increase in surface roughness and the formation of a dense layer of fluorine-containing groups monotonically with fluorination time. A twelve-brick air insulated linear transformer driver (LTD) was characterized by charging to voltages ranging from 30 to 70 kV and delivering energy to two separate resistive loads. Various plasma diagnostics were built and fielded with an emphasis on the design, implementation and analysis of a Mach Zehnder interferometer, a moiré deflectometer and a spectroscopy system providing information on the temporal evolution of plasma electron density and atomic composition. A new method to nowcast spectral irradiance in extreme ultraviolet (EUV) and far ultraviolet (FUV) bands is presented here, utilizing only solar photospheric magnetograms and the Mg II index (i.e., the core-to-wing ratio). The EUV and FUV modeling outlined here is a direct extension of the SIFT (Solar Indices Forecasting Tool) model, based on Henney et al. (2015, https://doi.org/10.1002/2014sw001118). SIFT estimates solar activity indices using the earth-side solar photospheric magnetic field sums from global magnetic maps generated by the ADAPT (Air Force Data Assimilative Photospheric Flux Transport) model. Low-cost and low-size-weight-and-power (SWaP) magnetometers can provide greater accessibility for distributed simultaneous measurements in the ionosphere, either onboard sounding rockets or on CubeSats. The Space and Atmospheric Instrumentation Laboratory (SAIL) at Embry-Riddle Aeronautical University has launched a multitude of sounding rockets in recent history: one night-time mid-latitude rocket from Wallops Flight Facility in August 2022 and three mid-latitude rockets from White Sands Missile Range during the October 2023 annular solar eclipse. A suite of experiments measuring target-spin observables in electron-nucleon scattering (dubbed Run Group C) was conducted at Jefferson Lab’s Hall B in Newport News, VA with a new polarized nuclear target known as ‘APOLLO’ (Ammonia Polarized Longitudinally). This innovative target is engineered to seamlessly integrate with the advanced 12 GeV CEBAF (Continuous Electron Beam Accelerator Facility) accelerator and the Hall B CLAS12 (12 GeV CEBAF Large Acceptance Spectrometer) detector array. Ferroelectricity in hafnium zirconium oxide (Hf1−xZrxO2) and the factors that impact it have been a popular research topic since its discovery in 2011. Although the general trends are known, the interactions between fabrication parameters and their effect on the ferroelectricity of Hf1−xZrxO2 require further investigation. Reliable prediction of gas migration velocity, void fraction, and length of gas-affected region in water and oil-based muds is essential for effective planning, control, and optimization of drilling operations. Synthetic peptides that self-assemble into cross-β fibrils are versatile building blocks for engineered biomaterials due to their modularity and biocompatibility, but their structural and morphological similarities to amyloid species have been a long-standing concern for their translation. Further, their polymorphs are difficult to characterize using spectroscopic and imaging techniques that rely on ensemble averaging to achieve high resolution. Here, we utilize Nile red (NR), an amyloidophilic fluorogenic probe, and single-molecule orientation-localization microscopy (SMOLM) to characterize fibrils formed by the designed amphipathic enantiomers KFE8L and KFE8D and the pathological amyloid-beta peptide Aβ42. We develop six-dimensional single-molecule orientation-localization microscopy (SMOLM) to measure the 3D positions and 3D orientations simultaneously of single fluorophores. We show how careful optimization of phase and polarization modulation components can encode phase, polarization, and angular spectrum information from each fluorescence photon into a microscope’s dipole-spread function. We used the transient binding and blinking of Nile red (NR) to characterize the helical structure of fibrils formed by designed amphipathic peptides, KFE8L and KFE8D, and the pathological amyloid-beta peptide Aβ42. We also deployed merocyanine 540 to uncover the interfacial architectures of biomolecular condensates.

To help in building a quantum-based communication link, we experimentally designed a system to simulate atmospheric turbulence and characterize its effects on a polarization-entangled photon-pair source. The simulated turbulence is constructed using two afocal optical systems with a phase plate inserted in each to mimic both weak and strong atmospheric turbulence respectively. After propagation, quantum state tomography (QST) is performed on each pair to reconstruct the density matrix of the pair’s overall polarization state. In characterization of the simulated turbulence, we were able to reach strengths up to a D/r0 of 18.2, which begins to approach the strong turbulent regime. …

Telemedicine has the potential to improve access and delivery of healthcare to diverse and aging populations. Recent advances in technology allow for remote monitoring of physiological measures such as heart rate, oxygen saturation, blood glucose, and blood pressure. However, the ability to accurately detect falls and monitor physical activity remotely without invading privacy or remembering to wear a costly device remains an ongoing concern. Convolutional neural networks (CNNs) have become instrumental in advancing multi-frame image super-resolution (SR), a technique that merges multiple low-resolution images of the same scene into a high-resolution image. In this paper, a novel deep learning multi-frame SR algorithm is introduced. The proposed CNN model, named Exponential Fusion of Interpolated Frames Network (EFIF-Net), seamlessly integrates fusion and restoration within an end-to-end network. Key features of the new EFIF-Net include a custom exponentially weighted fusion (EWF) layer for image fusion and a modification of the Residual Channel Attention Network for restoration to deblur the fused image. Annealing is a critical process in modern GaN technology, essential for achieving p-type conductivity by activating the MgGa acceptor, as first demonstrated by Shuji Nakamura in the early 1990s. Despite the omnipresence of hydrogen as an impurity in GaN crystals, the precise mechanisms governing hydrogen diffusion and acceptor passivation remain only partially understood. The presented research investigates the effects of annealing-induced activation and hydrogen passivation on C and Be acceptors in GaN grown by MOCVD, HVPE, and MBE methods. Ocean and coastal waters are monitored by Ocean Color satellite sensors to determine concentrations of chlorophyll and water properties and identify areas of algal blooms and other events. The light radiance from the ocean is weak in comparison with the sky radiance, which requires very accurate atmospheric correction of the radiance measured at the top of the atmosphere (TOA) on the satellite and heavy validation of the derived water leaving radiance by field measurements from the ships and ocean platforms. We will describe spin-transparent storage rings that exhibit spin-coherence times of several hours and store a large number of particles and their use in novel applications. For example, these rings can be used to directly measure the electric dipole moment of the electron, relevant to CP violation and matter-antimatter asymmetry in the universe, and to search for dark energy and ultra-light dark matter*. These rings can also serve as a compelling platform for quantum computing. Photovoltaic (PV) systems are widely adopted for renewable energy generation, but their performance is influenced by complex interactions between longer-term trends and seasonal variations. This study aims to remove these factors and provide valuable insights for optimising PV system operation. We employ comprehensive datasets of measured PV system performance over five years, focusing on identifying the distinct contributions of longer-term trends and seasonal effects. To achieve this, we develop a novel analytical framework that combines time series and statistical analytical techniques. Niobium films are a key component in modern two-dimensional superconducting qubits, yet their contribution to the total qubit decay rate is not fully understood. The presence of different layers of materials and interfaces makes it difficult to identify the dominant loss channels in present two-dimensional qubit designs. In this paper, we present the study that directly correlates measurements of RF losses in such films to material parameters by investigating a high-power impulse magnetron sputtered (HiPIMS) film atop a three-dimensional niobium superconducting radio frequency (SRF) resonator.
