The REPRIEVE Trial recently reported high rates of sudden cardiac death (SCD) middle-aged people living with HIV-1 infection (PWH) using the WHO/NIH-recommended two nucleoside reverse transcriptase inhibitors (NRTIs)/one integrase strand inhibitor (INSTI) regimen to manage HIV-1 viremia. To date, clinically relevant animal models to delineate underlying causes for this remain limited. Hepatocyte peroxisomes are required for metabolism of phytol-derived branched-chain fatty acids through the ACOX2-SCPx beta-oxidation pathway. This study tests whether hepatocyte-specific deletion of PEX16 (Pex16Alb-Cre), which eliminates peroxisome biogenesis, sensitizes the liver to phytol-induced injury. The Pex16Alb-Cre mice and their normal control Pex16fl/fl mice were fed the Lieber-DeCarli liquid with or without 0.1 percent phytol for eight weeks. Serum alanine aminotransferase (ALT) and liver index were measured. Liver sections were stained with hematoxylin and eosin and Masson’s trichrome. Osteoporosis and vertebral degeneration are major contributors to musculoskeletal morbidity, yet the molecular mechanisms regulating vertebral bone and intervertebral disc (IVD) homeostasis remain incompletely understood. GPNMB (osteoactivin) has been identified as a regulator of bone remodeling, though its role in spine health is unclear. This study evaluated the effects of GPNMB deficiency on lumbar vertebral bone microarchitecture and IVD integrity in a sex-dependent context.

Predicting PFAS adsorption across diverse adsorbents and environmental matrices remains challenging because adsorbent physicochemical properties, PFAS molecular descriptors, and operational conditions simultaneously influence adsorption. This study develops and evaluates a unified hybrid modeling framework that integrates Response Surface Model (RSM) with machine-learning algorithms to quantify how six key variables, surface area, Log Kow, pHpzc, pKa, log dose, and log-initial concentration, affect PFAS distribution coefficients (Log Kd). Background: This case features the surgical reconstruction of the coracohumeral ligament (CHL) in conjunction with open capsulorrhaphy and capsular shift for the management of chronic, recurrent, multidirectional shoulder instability (MDI). While there is a paucity of existing literature examining the clinical outcomes of this technique, biomechanical studies have implied its potential to restore stability. This procedure may be of particular benefit to patients who have failed previous conservative management and/or attempts at surgical stabilization, as well as those with severe joint hyperlaxity or connective tissue disorders.

While excess adiposity is a known risk factor for incident heart failure ( HF), once the condition is established, observational data suggest that increased body mass index (BMI) may confer a survival advantage. This paradox has emphasized the underlying roles of cardiorespiratory fitness (CRF), and body composition, particularly lean mass (LM), in influencing clinical outcomes.

Pancreatic ductal adenocarcinoma (PDAC) is the fourth leading cause of cancer-related mortality, characterized by intrinsic resistance to conventional therapies and limited effective treatment options. In this study, we investigated the role of the CXCR2 axis in PDAC therapy resistance. CXCR2, a chemokine receptor, is actively involved in inflammation, tumor angiogenesis, and metastasis. Our working hypothesis is that CXCR2 contributes to PDAC chemotherapy resistance. To test this, we generated gemcitabine-resistant (GemR) lines using T3M4 and CD18/HPAF (CD18) cell lines. Midkine (MDK) is an oncofetal, heparin-binding cytokine that is re-expressed across diverse cancers and correlates with aggressive disease and treatment resistance. This review synthesizes current evidence on MDK as a coordinator of tumor-intrinsic signaling and microenvironmental remodeling. We summarize MDK structural features, extracellular matrix interactions, and receptor systems that mediate MDK signaling, highlighting LRP1 and PTPRZ1 with context-dependent participation of ALK, nucleolin and integrins. Downstream, MDK engages MAPK, PI3K-AKT, STAT3 and NF-κB pathways to promote tumor cell survival, epithelial-mesenchymal plasticity, and therapeutic stress tolerance. Pancreatic cancer (PC) is characterized by extensive desmoplasia, with heterogeneous cancer-associated fibroblasts (CAFs) as a major component. However, the contribution of distinct precursor cells to CAF heterogeneity remains poorly defined. This study investigated the role of Muc5ac in modulating CAF heterogeneity by maturing precursor cells, including adipose-derived mesenchymal stem cells (AD-MSCs), bone marrow-derived MSCs (BM-MSCs), and pancreatic stellate cells (PSCs), into different CAF subsets. RNA sequencing of precursor cells treated with conditioned media from Muc5ac-proficient or -deficient cancer cells revealed distinct transcriptional profiles. Objective: This study aimed to establish cephalometric norms for the Tanzanian population, assess sexual dimorphism in cephalometric measurements, and compare these findings with established Caucasian and West African norms to enhance orthodontic diagnosis and treatment planning. Materials and Methods: A crosssectional study was conducted on 309 subjects (142 males, 167 females) at Muhimbili University Dental Clinic. Cephalometric radiographs were digitally traced using Quick Ceph Studio. Reliability was assessed using the inter-class correlation coefficient. The Kolmogorov-Smirnov test checked normality, while one-sample t-tests and Student’s ttests (p < 0.05) were used for comparisons. Data were analyzed with RStudio. Results: Tanzanians showed significantly smaller craniofacial measurements than Caucasians. Males had larger skeletal dimensions than females. Compared to West Africans, Tanzanians had generally higher values, with a few exceptions. Conclusion: Tanzanians display distinct craniofacial characteristics, differing from both Caucasians and West Africans. These findings support the need for population-specific cephalometric norms to guide accurate orthodontic diagnosis and treatment.

Objective: This study intended to evaluate changes, clinically and radiographically, associated with the immediate placement of dental implants in the maxillary anterior esthetic zone, comparing vestibular socket therapy with the ice cream cone technique. Purpose. This study aimed to evaluate the clinical performance of CAD-CAM-milled PEKK frameworks versus PEEK frameworks in the fabrication of clasp-retained maxillary obturators, assessing retention outcomes and patient satisfaction over a sixmonth follow-up period. Materials and Methods. Ten patients with maxillary defects (Armany class I) were randomly assigned to two equal groups (n=5). Objective: To investigate the impact of serum vitamin D levels on marginal bone loss (MBL) in patients restored with unsplinted-mandibular-implant overdentures using LOCATOR attachments. Materials and Methods: Thirty participants rehabilitated with three-implant-mandibular overdentures were included. Based on serum vitamin D at the time of prosthetic loading, participants were categorized into two equal groups: a test group with deficient vitamin D (< 20 ng/mL) and a control group with sufficient levels (≥30 ng/mL). Forty-eight sound human maxillary central incisors were collected and prepared for anterior laminate veneer restorations following standardized preparation protocol. Objective: The present study aimed to assess the dose-dependent effect of Biodentine (BD) and human-treated dentin matrix (hTDM) on the proliferation of human dental pulp stem cells (hDPSCs).
Materials and methods: hTDM was prepared and characterized. Objectives: This study was conducted to prepare and characterize root-shaped treated dentin matrix (TDM) scaffolds, and to evaluate their efficacy together with seeded dental pulp stem cells (DPSCs) as potential bio-roots in extraction sockets of rat models.
Materials and Methods: Root-shaped TDM specimens were prepared and characterized. Following extraction of maxillary incisors, the alveolar sockets were implanted with TDM segments measuring 8 mm in length, either alone (TDM group) or seeded with dental pulp stem cells (TDM/DPSCs group). Purpose: To evaluate, both clinically and radiographically, the use of extended platelet-rich fibrin versus a pericardium membrane in mandibular ridge splitting accompanied by immediate implant placement.
Materials and Methods: A total of sixteen implants were placed in eight patients with horizontally atrophic posterior mandibular ridges measuring 3–6 mm in width were randomly assigned to two groups with equal numbers. Nutrition epidemiological models involve many analytic decisions, such as defining exposures, selecting which covariates to include, or configuring variables in different ways. We explored the impact of analytical decisions on conclusions in nutrition epidemiology using self-reported beef intake and incident coronary heart disease as a case study. Chronic alcohol consumption (CAC) is associated with an enhanced risk of bone fracture, reduced bone density, and osteoporosis. In a rhesus macaque model of voluntary ethanol consumption, CAC induces functional, transcriptomic, and epigenomic changes in hematopoietic stem and progenitor cells (HSPCs) and their resultant monocytes/macrophages, skewing them toward a hyper-inflammatory response. In the present study, those studies were extended to investigate alterations in osteoclast development, which, in postnatal life, differentiate from HSPCs and play a critical role in maintaining bone homeostasis. Chronic alcohol consumption increases the risk of osteoporosis and fracture by disrupting bone remodeling, in part by enhancing osteoclastogenesis. However, the cellular mechanisms underlying this process remain incompletely defined. We analyzed scRNA-seq data from osteoclasts differentiated in vitro from bone marrow mononuclear cells obtained from macaques following 12 months of chronic ethanol or isocaloric control solution consumption. Module scoring, trajectory inference with generalized additive modeling (tradeSeq), and CellChat-based analyses of intercellular communication were applied to uncover ethanol-induced changes in metabolic reprogramming, lineage progression, and signaling network dynamics. Objective: Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) and Metabolic Dysfunction-Associated Alcohol-related Liver Disease (MetALD) exhibit systemic immune abnormalities. Given that such immune dysregulation is closely linked to the skeletal complications frequently observed in MASLD and MetALD, we sought to comprehensively characterize the bone marrow hematopoietic compartment and its link to osteoclastogenesis.
Methods: We utilized bone marrow from mouse models of MASLD (high-fat diet, HFD) and MetALD (high-fat diet plus ethanol, HFD+EtOH), followed by flow cytometric analysis to phenotype hematopoietic stem and progenitor cells (HSPCs), as well as in vitro and in vivo assays to evaluate osteoclastogenesis.

Hospice care is intended to prioritize comfort, dignity, symptom management, and quality of life for individuals with life-limiting illness. However, when hospice-eligible patients are managed within inpatient hospital settings, significant ethical challenges frequently emerge due to the tension between comfort-focused care and the curative, intervention-driven structure of acute care medicine. Synchronous coexistence of plasma cell neoplasms (PCNs) and metastatic carcinoma is not a common phenomenon and poses a significant diagnostic challenge requiring meticulous histopathological and immunophenotypic analysis for accurate lineage assignment. We report two cases of collision tumors comprising PCN and metastatic lung carcinoma. Case 1 involved a 68-year-old male with metastatic non-small cell lung adenocarcinoma who developed intracranial lesions; resection revealed metastatic adenocarcinoma intermingled with clonal plasma cells. Case 2 was a 73-year-old woman with a lytic iliac crest lesion whose bone marrow and lesion biopsies showed diffuse infiltration by both carcinoma and atypical plasma cells.
