Cyclic And Linear Cell-Penetarating Peptides Composed Of Tryptophan (Ww) And Arginine (Rr) Residues As Molecular Transporters, Lois Kim Aug 2022

Cyclic And Linear Cell-Penetarating Peptides Composed Of Tryptophan (Ww) And Arginine (Rr) Residues As Molecular Transporters, Lois Kim

Pharmaceutical Sciences (MS) Theses

The cell membrane has polar and negative charged characteristics and separates the inside of cells from the outside environment, and only allows small molecules to go into the cell to protect them from toxic substances. However, cell membrane characteristics make it difficult for polar and negatively-charged molecules to interact and penetrate the membrane. The delivery of cell impermeable compounds, such as phosphopeptides and siRNA, across the cell membrane remains a challenge. Previous studies in Dr. Parang’s laboratory have shown that cyclic peptides composed of tryptophan (W) and arginine (R) amino acids [WR]5 have higher cellular uptake than the corresponding …

Identifying A Glucocorticoid-Activated Gpcr That Rapidly And Non-Genomically Increases Camp Levels In Mammalian Cells, Francisco Nunez Aug 2022

Identifying A Glucocorticoid-Activated Gpcr That Rapidly And Non-Genomically Increases Camp Levels In Mammalian Cells, Francisco Nunez

Pharmaceutical Sciences (PhD) Dissertations

Glucocorticoids (GCs) are steroid hormones that regulate diverse physiological processes. Synthetic versions of GCs are commonly used to treat inflammatory diseases such as asthma by modulating gene expression to suppressing several inflammatory activities. However, it is estimated that 5-10% of asthmatics are unresponsive to GCs, which may be explained by receptor desensitization and/or the presence of a neutrophilic endotype. One understudied phenomenon of GCs is their ability to induce rapid, non-genomic actions. For example, GCs can acutely modulate calcium concentrations levels, induce smooth muscle relaxation and modulate nitric oxide synthase activity, within minutes and sometimes seconds, which is too rapid …

Prevalence And Factors Associated With Undernutrition Among Children With Congenital Heart Disease At Three Tertiary Hospitals In Nairobi, Kenya., Shamsa Yahya Aug 2022

Prevalence And Factors Associated With Undernutrition Among Children With Congenital Heart Disease At Three Tertiary Hospitals In Nairobi, Kenya., Shamsa Yahya

Master of Medicine in Paediatrics and Child Health

Background: Congenital heart disease (CHD) is the commonest birth defect. Children with CHD have a high burden of undernutrition and its consequences. Some of the factors that are associated with undernutrition in children with CHD include the type and complexity of the heart defect, associated pulmonary hypertension, late diagnosis, timing of surgical correction, low socioeconomic status, lack of adequate nutritional support, associated genetic conditions and the presence of other clinical comorbidities. Justification: Although these factors are well defined, they vary geographically in their distribution and extent to which they influence the level of undernutrition. This limits the extrapolation of findings …

Translating Particulate Hexavalent Chromium-Induced Chromosome Instability From Human Lung Cells To Experimental Animals, Human Lung Tumors And Whale Cells., Haiyan Lu Aug 2022

Translating Particulate Hexavalent Chromium-Induced Chromosome Instability From Human Lung Cells To Experimental Animals, Human Lung Tumors And Whale Cells., Haiyan Lu

Electronic Theses and Dissertations

Lung cancer is a major human health problem. While smoking is the most well-known cause of lung cancer, people who never smoked develop the disease. Understanding how non-tobacco environmental carcinogens cause lung cancer is key to combating this disease. Hexavalent chromium [Cr(VI)] is a well-established human lung carcinogen, but its carcinogenic mechanism is uncertain. Chromosome instability (CIN) is a hallmark of lung cancer and a major factor in Cr(VI)-induced lung cancer. Studies in human lung cells show Cr(VI) induces DNA double strand breaks and suppresses homologous recombination (HR) repair by targeting RAD51, resulting in CIN. We translated these outcomes to …

“…You Can't Unknow It, It's A New Reality": A Case Study Exploring A Social Justice Public Health Intervention's Impact On Critical Consciousness Development In Youth., Monique Williams

Electronic Theses and Dissertations

Even though public health purports to be rooted in social justice, it is not clear how social justice frameworks are integrated in public health research and intervention. The discipline focuses on groups/populations on the margins, without integrating the social and political factors that cause marginalization into the intervention. Seeing communities, and individuals, as assets and experts in their own experiences is key to population-level health improvement. Particularly for public health youth intervention, how youth are labeled and engaged in social change is critical to their healthy development. While public health lags in practically applying asset-based approaches to youth intervention, research …

Molecular Mechanisms Of Hexavalent Chromium-Induced Centrosome Amplification., Jennifer Haruka Toyoda

Electronic Theses and Dissertations

Lung cancer is the deadliest form of cancer and resulted in 1.8 million deaths worldwide in 2020. While cigarette smoking is the most familiar cause of lung cancer, up to 25% of cases occur in non-smokers, thus other environmental agents are also causative. Hexavalent chromium [Cr(VI)] is a known lung carcinogen and poses occupational and environmental exposure risks relevant to humans, wildlife, and ecosystems. This dissertation considers the carcinogenic mechanisms of a highly potent, particulate, hexavalent chromium compound, zinc chromate. The molecular mechanism of carcinogenesis induced by Cr(VI) is not fully understood, but it is known that chromosome instability is …

Laboratory-Reported Normal Value Ranges Should Not Be Used To Diagnose Periprosthetic Joint Infection., Salvador A. Forte, Joseph A. D'Alonzo, Zachary Wells, Brett Levine, Stephen Sizer, Carl Deirmengian

Orthopedic Surgery Resident Research

INTRODUCTION: Clinical laboratories offer several multipurpose tests, such as the erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP), which are not intended to diagnose any specific disease but are used by clinicians in multiple fields. The results and laboratory interpretation (normal/abnormal) of these multipurpose tests are based on laboratory-reported normal thresholds, which vary across clinical laboratories. In 2018, the International Consensus Meeting on Musculoskeletal Infection (2018 ICM) provided a gold-standard definition to diagnose periprosthetic joint infection (PJI) which included many multipurpose laboratory tests, along with thresholds optimized to diagnose PJI. The discrepancy between laboratory-reported normal thresholds and 2018 ICM-recommended PJI-optimized …

Somatostatin receptors (SSTRs) play versatile roles in inhibiting the secretion of multiple hormones such as growth hormone and thyroid-stimulating hormone, and thus are considered as targets for treating multiple tumors. Despite great progress made in therapeutic development against this diverse receptor family, drugs that target SSTRs still show limited efficacy with preferential binding affinity and conspicuous side-effects. Here, we report five structures of SSTR2 and SSTR4 in different states, including two crystal structures of SSTR2 in complex with a selective peptide antagonist and a non-peptide agonist, respectively, a cryo-electron microscopy (cryo-EM) structure of Gi1-bound SSTR2 in the presence of the …

Utah has a rich history related to air pollution; however, it is not widely known or documented. This is despite air quality being a top issue of public concern for the state’s urban residents and acute episodes that feature some of the world’s worst short-term particulate matter exposure. As we discuss in this narrative review, the relationship between air pollution and the state’s residents has changed over time, as fuel sources shifted from wood to coal to petroleum and natural gas. Air pollution rose in prominence as a public issue in the 1880s as Utah’s urban areas grew. Since then, …

The Media Discourses On Organ Donation And Transplantation In Spain (1954-2020) And Their Implications For Spanish Nationalism, Rebeca Herrero Sáenz Aug 2022

The Media Discourses On Organ Donation And Transplantation In Spain (1954-2020) And Their Implications For Spanish Nationalism, Rebeca Herrero Sáenz

Legacy Theses & Dissertations (2009 - 2024)

Spain has been the global leader in organ donation and transplantation since 1992, an achievement that has become a source of national pride, in a country where national symbols are heavily contested. In this dissertation I examine the changing meanings that organ donation and transplantation have acquired in contemporary Spain, focusing specifically on their implications for different aspects of Spanish nationalism. To do so, I employ a modified version computational grounded theory, a mixed-methods approach that combines topic modeling with interpretive analysis, to identify and interpret the narratives around organ donation and transplantation circulated by the Spanish press between 1954 …

Intolerance Of Uncertainty Specific To Compulsive Exercise : Development And Preliminary Validation Of The Exercise-Specific Intolerance Of Uncertainty Scale, Christina Scharmer Aug 2022

Intolerance Of Uncertainty Specific To Compulsive Exercise : Development And Preliminary Validation Of The Exercise-Specific Intolerance Of Uncertainty Scale, Christina Scharmer

Legacy Theses & Dissertations (2009 - 2024)

Compulsive exercise is poorly understood and difficult to treat. Overlap between definitions of compulsive exercise, eating disorders and OCD suggests that conceptualizations of these disorders may include shared underlying mechanisms, including intolerance of uncertainty. Although intolerance of uncertainty is traditionally conceptualized as a non-specific construct representing an individual’s dispositional distress in the face of general uncertainty, identifying specific forms of intolerance of uncertainty may further assist in understanding the specific factors that contribute to the development and maintenance of problematic behaviors, including compulsive exercise. The current study aimed to develop an assessment instrument that captures exercise-specific variations in intolerance of …
