Screening And Characterization Of A Pan-Gtpase Inhibitor, Leyla Akhadov May 2020

Screening And Characterization Of A Pan-Gtpase Inhibitor, Leyla Akhadov

Biomedical Engineering ETDs

Abnormal functioning of small GTPases is implicated in a variety of diseases, ranging from neurological and developmental diseases to cancer. In fact, mutant GTPases are found in up to 30% of cancers. Thus, small GTPases are a highly relevant target in drug discovery and development. High-throughput targeted screening of small molecules is the most productive method of discovering compounds that can give insights into drug development. This thesis describes improvements made to a high-throughput GTPase-targeted screening method to minimize confounding systematic error. It also describes the follow-up characterization of a compound that was identified in a high-throughput screen. The compound …

Orthostatic Blood Pressure And Arterial Stiffness In Persons With Spinal Cord Injury: The Effect Of The Renin Angiotensin Aldosterone System, Caitlyn G. Katzelnick May 2020

Orthostatic Blood Pressure And Arterial Stiffness In Persons With Spinal Cord Injury: The Effect Of The Renin Angiotensin Aldosterone System, Caitlyn G. Katzelnick

Seton Hall University Dissertations and Theses (ETDs)

With advances in acute medical care, longevity has increased in persons with SCI; however, morbidity due to cardiovascular disease (CVD) occurs at an earlier age compared to the general population. Arterial stiffness (AS) is recognized as an independent risk factor for CVD and, specifically, pulse wave velocity (PWV) has been proven to be a valid tool to predict and track structural arterial changes that reflect arteriosclerosis. Evidence has shown that persons with SCI have increased AS compared to uninjured able-bodied controls; however, possible contributors to this increase are not yet fully understood. After a SCI, sympathetic control in the regions …

Allosteric Inhibitor Of Β-Catenin Selectively Targets Oncogenic Wnt Signaling In Colon Cancer., Anton Cheltsov, Natsuko Nomura, Venkata Mahidhar Yenugonda, Jatin Roper, Rajesh Mukthavaram, Pengfei Jiang, Nam-Gu Her, Ivan Babic, Santosh Kesari, Elmar Nurmemmedov

Articles, Abstracts, and Reports

Abnormal regulation of β-catenin initiates an oncogenic program that serves as a main driver of many cancers. Albeit challenging, β-catenin is an attractive drug target due to its role in maintenance of cancer stem cells and potential to eliminate cancer relapse. We have identified C2, a novel β-catenin inhibitor, which is a small molecule that binds to a novel allosteric site on the surface of β-catenin. C2 selectively inhibits β-catenin, lowers its cellular load and significantly reduces viability of β-catenin-driven cancer cells. Through direct binding to β-catenin, C2 renders the target inactive that eventually activates proteasome system for its removal. …

Black Pregnancy Matters: Racial Reproductive Bias In African American Maternal Mortality, Juhi Khemani May 2020

Black Pregnancy Matters: Racial Reproductive Bias In African American Maternal Mortality, Juhi Khemani

Master's Projects and Capstones

This semester-long research project uncovers how racial biases demonstrated in the reproductive health field, and specifically before, during, and after childbirth, affect the rate of maternal mortality and morbidity for African American mothers in the United States. The rate of maternal mortality for this specific racial group is four times greater as compared to Caucasian women and this discrepancy will be investigated and analyzed throughout this capstone thesis. Interviews were conducted with key figures in the obstetric and gynecological field as well as with Black mothers themselves in an effort to uncover what factors, aside from medical anomalies, are leading …

Anticipatory Threat Responding: Associations With Anxiety, Development, And Brain Structure, Rany Abend, Andrea L. Gold, Jennifer C. Britton, Kalina J. Michalska, Tomer Shechner, Jessica F. Sachs, Anderson M. Winkler, Ellen Leibenluft

School of Medicine Publications

BACKGROUND: While translational theories link neurodevelopmental changes in threat learning to pathological anxiety, findings from studies in patients inconsistently support these theories. This inconsistency may reflect difficulties in studying large patient samples with wide age ranges using consistent methods. A dearth of imaging data in patients further limits translational advances. We address these gaps through a psychophysiology and structural brain imaging study in a large sample of patients across the lifespan.

METHODS: A total of 351 participants (8-50 years of age; 209 female subjects; 195 healthy participants and 156 medication-free, treatment-seeking patients with anxiety) completed a differential threat conditioning and …

Transdiagnostic Multimodal Neural Correlates Of Psychosis Dimensions, Dov Bernard Lerman-Sinkoff May 2020

Transdiagnostic Multimodal Neural Correlates Of Psychosis Dimensions, Dov Bernard Lerman-Sinkoff

McKelvey School of Engineering Graduate Student Theses & Dissertations

Psychosis refers to a debilitating set of symptoms that impacts individuals, their communities, and society at large. Current psychiatric nosology treats psychosis as a categorical construct. However, recent evidence suggests that a dimensional approach that cuts across extant nosological boundaries may more accurately represent the underlying phenomena contributing to dysfunction in psychosis. One putative domain of transdiagnostic variation is cognitive control, a construct that refers to the set of functions that enable and support goal-directed behavior and regulation of one’s thoughts and actions. Previous analyses in both healthy individuals and individuals with psychosis have led to a number of findings …

A Public Health Examination Of The Discipline Gap, Karishma Furtado May 2020

A Public Health Examination Of The Discipline Gap, Karishma Furtado

Arts & Sciences Graduate Student Theses and Dissertations

Education is often positioned as the great equalizer in the United Statesѡ cure all for many social ills, from poverty, to family instability, to exposure to violence. However, disadvantaged students tend, on average, to get a lower quality education. One example of a barrier that impedes educational equity is the discipline gap, or the disproportionate rate of exclusionary discipline like suspension and expulsion experienced by students with disabilities and those from historically disadvantaged racial, ethnic, and gender subgroups. Removing a student from class for punitive reasons puts them at greater risk of academic disengagement, a diminished sense of belonging and …

Pillars Of The Nutrition Transition: The Global Impacts Of Ultra-Processed Foods And Beverages On Overweight And Obesity And National Nutrient Supplies, Jacob Eaton May 2020

Pillars Of The Nutrition Transition: The Global Impacts Of Ultra-Processed Foods And Beverages On Overweight And Obesity And National Nutrient Supplies, Jacob Eaton

Arts & Sciences Graduate Student Theses and Dissertations

Malnutrition in some form impacts nearly one-third of the global population. Across the world, countries are undergoing the Үutrition transitionӠfrom traditional and largely unprocessed diets to Western-style, energy-dense diets. At the same time, rates of overweight and obesity and diet-related chronic diseases continue to climb. Ultra-processed foods (UPF), sugar-sweetened beverages (SSB), and vegetable oils are three of the foods driving the nutrition transition. This dissertation calculates changes in the global food supply between 1961 and 2013 and quantifies the influence of UPF and SSB (as measured through sales) on national nutrient supplies between 2005 and 2013 and trends in adult …

Broken With Nature, Meghan Bowler May 2020

Broken With Nature, Meghan Bowler

Akesis

This is a short reflection on two separate periods of intense mental and physical self-healing. Nature removed me from the imprisonment of my own mind around 8 years ago, only to fall back into its grips after a recent hip surgery. Hiking/physical activity was the only form of therapy that I was vulnerable enough to let heal me. It would break me down mentally and physically while rebuilding me at the same time. Some information was derived from personal journal entries but all are very relevant to each and every hike I take today.

This photo is an overhead shot of the waves breaking at Pine Point beach. This represents so much for me. In these times of uncertainty it gives me hope that the tides will eventually turn. And just like the tides, this too will take a turn for the better. It also represents the healing and cleansing qualities of the sea.

Motus Habitus: An Anatomical Ethnography, Michael Cyr, Jordan G. Lamb May 2020

Motus Habitus: An Anatomical Ethnography, Michael Cyr, Jordan G. Lamb

Akesis

Although this project began long before the release of Akesis' Spring theme, it perfectly encapsulates adaptation in many ways. The response to the recent pandemic and the disruption to people's daily routines has caused us to reflect on the ways we experience our bodies and minds in the absence of work, recreation, sociality, and movement. Movement, a key to human health and happiness, has been greatly restrained as most of us are confined to our living quarters. Now more than ever it is important to acknowledge the effects of lifestyle on body and health and find creative ways to move …

Photographic Validation Of Target Versus Nontarget Take Of Brown Treesnake Baits, Shane R. Siers, Aaron B. Shiels, Cynthia G. Payne, Francinem M. Chlarson, Craig S. Clark, Stephen M. Mosher May 2020

Photographic Validation Of Target Versus Nontarget Take Of Brown Treesnake Baits, Shane R. Siers, Aaron B. Shiels, Cynthia G. Payne, Francinem M. Chlarson, Craig S. Clark, Stephen M. Mosher

United States Department of Agriculture Wildlife Services: Staff Publications

Use of toxic baits or other tools for managing nuisance species must ensure that the species of interest is adequately targeted while exposure to nontarget species is minimized. Nontarget takes of acetaminophen‐laced baits for control of invasive brown treesnakes (Boiga irregularis) on Guam may put those animals at risk of lethal intoxication and render the bait unavailable to the intended target species. We used wildlife cameras to identify species removing toxic and nontoxic baits from brown treesnake bait stations designed to exclude nontarget taxa in 2015 and 2016. Throughout various sites and habitat types, and balanced by season (wet vs. …

Time Allocation To Resources By Three Species Of Rats (Rattus Spp.) In A Radial Arm Maze, Gary Witmer, Nathan P. Snow, Rachael S. Moulton May 2020

Time Allocation To Resources By Three Species Of Rats (Rattus Spp.) In A Radial Arm Maze, Gary Witmer, Nathan P. Snow, Rachael S. Moulton

United States Department of Agriculture Wildlife Services: Staff Publications

Context. Introduced rats (Rattus spp.) can pose a serious threat to native flora and fauna, especially on islands where most species have evolved in the absence of terrestrial predators. Effective detection and eradication methods for introduced rats are essential to the maintenance of insular ecosystem integrity. Thus, it is important to better understand the behaviour of rats when they first arrive in a new setting.

Aims. To determine whether rats would find some novel stimuli to be significantly more attractive than other novel stimuli.

Methods. An eight-arm radial maze was used to study the behaviour of three species of Rattus …

Interactions With Humans Shape Coyote Responses To Hazing, Julie K. Young, Edd Hammill, Stewart W. Breck May 2020

Interactions With Humans Shape Coyote Responses To Hazing, Julie K. Young, Edd Hammill, Stewart W. Breck

United States Department of Agriculture Wildlife Services: Staff Publications

Medium and large carnivores coexist with people in urban areas globally, occasionally resulting in

negative interactions that prompt questions about how to reduce human-wildlife conflict. Hazing,

i.e., scaring wildlife, is frequently promoted as an important non-lethal means for urbanites to reduce

conflict but there is limited scientific evidence for its efficacy. We used a population of captive coyotes (Canis latrans) to simulate urban human-coyote interactions and subsequent effects of hazing on coyote behavior. Past experiences with humans significantly affected the number of times a coyoteapproached a human to necessitate hazing. coyotes that had been hand fed by adults had to …

United States Department of Agriculture Wildlife Services: Staff Publications

Invasive species are a leading driver of global change, with consequences for biodiversity and society. Because of extraordinary rates of endemism, introduction, and extinction, Hawaii offers a rich platform for exploring the cross-disciplinary challenges of managing invasive species in a dynamic world. We highlight key successes and shortcomings to share lessons learned and inspire innovation and action in and beyond the archipelago. We then discuss thematic challenges and opportunities of broad relevance to invaded ecosystems and human communities. Important research needs and possible actions include eradicating mammals from mainland island sanctuaries, assessing hidden threats from poorly known introduced species, harnessing …

Deciphering Interactions Between White-Tailed Deer And Approaching Vehicle, Morgan Pfeiffer, Raymond B. Iglay, Thomas W. Seamans, Bradley F. Blackwell, Travis L. Devault

United States Department of Agriculture Wildlife Services: Staff Publications

Deer-vehicle collisions are a major transportation hazard, but factors affecting deer escape decision-making in response to vehicle approach remain poorly characterized. We made opportunistic observations of deer response to vehicle approach during daylight hours on a restricted- access facility in Ohio, USA (vehicle speeds were ≤64 km/h). We hypothesized that animal proximity to the road, group size, vehicle approach, and ambient conditions would affect perceived risk by white-tailed deer (Odocoileus virginianus) to vehicle approach, as measured by flight-initiation distance (FID). We constructed a priori models for FID, as well as road-crossing behavior. Deer responses were variable and did not demonstrate …
