Bariatric surgery, in addition to the benefit of sustained weight loss, can also reduce cardiometabolic risk and mortality. Lifelong vessel maintenance is integral to the prevention of cardiovascular disease. Using aldehyde dehydrogenase activity, an intracellular detoxifying enzyme present at high levels within pro-vascular progenitor cells, we observed an association between chronic obesity and "regenerative cell exhaustion" (RCE), a pathology whereby chronic assault on circulating regenerative cell types can result in adverse inflammation and diminished vessel repair. We also describe that, at 3 months following bariatric surgery, systemic inflammatory burden was reduced and pro-angiogenic macrophage precursor content was improved in subjects …

© 2020 The Author(s). Background: Osteoarthritis (OA) is the most common form of arthritis and characterized by degeneration of the articular cartilage. Mitogen-inducible gene 6 (Mig-6) has been identified as a negative regulator of the epidermal growth factor receptor (EGFR). Cartilage-specific Mig-6 knockout (KO) mice display increased EGFR signaling, an anabolic buildup of the articular cartilage, and formation of chondro-osseous nodules. Since our understanding of the EGFR/Mig-6 network in the cartilage remains incomplete, we characterized mice with cartilage-specific overexpression of Mig-6 in this study. Methods: Utilizing knee joints from cartilage-specific Mig-6-overexpressing (Mig-6 over/over ) mice (at multiple time points), we …

Endothelial Progenitor Cell-Breast Cancer Interaction, Ghada Ben Rahoma
NYMC Student Theses and Dissertations
Breast cancer remains the second leading cause of cancer death among women in the U.S. Although organ-confined disease is curable, metastatic disease remains incurable and an unmet clinical need. Hence, the development of new therapeutic targets and further understanding of the genesis of malignant progression is needed and to this end analysis of the tumor microenvironment is imperative for novel therapeutic targets in breast cancer. We and others have demonstrated that bone marrow derived endothelial progenitor cells (EPCs) incorporate in the neovasculature around implanted tumors supporting their growth and metastasis. The incorporation of EPCs from the marrow to the tumor …

Department of Public Health Scholarship and Creative Works
COVID-19 has imposed unprecedented challenges to society. As the pandemic evolves, the social distancing measures that have been globally enforced, while essential, are having undesirable socioeconomic side effects particularly among vulnerable populations. In Mexico, families who depend upon informal employment face increased threats to their wellbeing, and households who in addition have young children may face long-term consequences. The Mexican government has not yet taken actions, but a coalition of non-governmental organizations is advocating in partnership with academic institutions for social protection actions such as a cash transfer and basic services subsidies for families with young children, subsisting from the …

Fall Prevention Assessment in the Intermediate Care Patient Population
Falls cause greater than 600,000 deaths per year and are the most common injury in people over 65 years of age. Fall prevention in the acute care setting has been identified as key goal in the most recent JCAHO review process. Falls on an intermediary care unit lead to an increased rate of morbidity, delayed discharge, and excess cost expenditure.
Even with appropriate evidence-based tools in place, Hospital A’s intermediate care unit, recorded an increased rate of falls; the highest rate in its health care system. An investigation identified factors contributing …

Spectrum Of Imaging Findings In Aidsrelated Diffuse Large B Cell Lymphoma, Edward Nganga, Samuel Gitau
Imaging & Diagnostic Radiology, East Africa
Lymphoma in HIV-infected patients is AIDS defining. This is the second most common AIDS defining malignancy after Kaposi’s sarcoma. Development of lymphoma in HIV patients is related to immunosuppression and high viral load. Co-infection with other lymphotrophic viruses especially EBV is also strongly associated with development of lymphoma in HIV patients. Despite advances in HAART therapy, incidence of diffuse large B cell lymphoma in HIV-infected patients remains significantly higher than in the general population.
Early diagnosis is challenging due to presence of opportunistic infections and atypical presentation of the lymphoma in this subset of patients. Atypical imaging findings are not …

Challenges and special aspects related to the management and prognosis of pulmonary hypertension (PH) in middle- to low-income regions (MLIRs) range from late presentation to comorbidities, lack of resources and expertise, cost, and rare options of lung transplantation. Expert consensus recommendations addressing the specific challenges for prevention and therapy of PH in MLIRs with limited resources have been lacking. To date, 6 MLIR-PH registries containing mostly adult patients with PH exist. Importantly, the global prevalence of PH is much higher in MLIRs compared with high-income regions: group 2 PH (left heart disease), pulmonary arterial hypertension associated with unrepaired congenital heart …

Patients' Disclosure Of Sexual And Gender Minority Status: A Comparative, Multiple Case Study, Damon B. Cottrell
Nursing Theses and Dissertations
This work explores the healthcare and related experiences of sexual and gender minority (SGM) people. This population faces unique challenges in addressing their health needs. In the context of these challenges, the health outcomes are often poorer than those of heterosexuals. SGM subpopulations experience unique disparities making healthcare needs more complex and necessitating an understanding of SGMs’ being. Essential to addressing SGMs’ healthcare needs is the healthcare provider’s (HCP’s) knowledge of sexual and gender minority status. The first manuscript in this portfolio dissertation, Fostering Sexual and Gender Minority Status in Patients, provides an overview of the issues surrounding disclosure to …

Friend Or Foe? Recent Strategies To Target Myeloid Cells In Cancer, Mehdi Chaib, Subhash C. Chauhan, Liza Makowski
School of Medicine Publications
The tumor microenvironment (TME) is a complex network of epithelial and stromal cells, wherein stromal components provide support to tumor cells during all stages of tumorigenesis. Among these stromal cell populations are myeloid cells, which are comprised mainly of tumor-associated macrophages (TAM), dendritic cells (DC), myeloid-derived suppressor cells (MDSC), and tumor-associated neutrophils (TAN). Myeloid cells play a major role in tumor growth through nurturing cancer stem cells by providing growth factors and metabolites, increasing angiogenesis, as well as promoting immune evasion through the creation of an immune-suppressive microenvironment. Immunosuppression in the TME is achieved by preventing critical anti-tumor immune responses …

A Role For Retinoids In The Treatment Of Covid-19?, Steven E. Trasino
Publications and Research
The 2020 global outbreak of the novel coronavirus (SARS-CoV-2 or COVID-19) is a serious threat to international health, and thus, there is an urgent need for discoverynof novel therapies or use of repurposed drugs that can make a significant impact on slowing the spread of the virus. Type 1 interferons (IFN-I) are a family cytokines of the early innate immune response to viruses that are being tested against SARSCoV- 2. However, coronaviruses similar to SARS-CoV-2 can suppress host IFN-I antiviral responses. Retinoids are a family molecules related to vitamin A that possess robust immune-modulating properties, including the ability to increase …

We are summarizing the recommendations for the use of Echocardiography in patients during COVID-19 pandemic. The patient risk for COVID-19 should be assessed according to the Saudi CDC guidelines. Echocardiography should only be performed of considered appropriate and will likely alter the clinical decision. In COVID-19 suspected/ confirmed patients, echocardiography study should be performed bedside and in infection control approved area with airborne precaution. Limited focused imaging is recommended to minimize contact time. A dedicated machine for COVID-19 suspected/confirmed cases is recommended. Transesophageal echocardiography is considered an aerosol generating procedure; therefore, an alternative modality should be strongly considered. In COVID-19 …

Department of Neurology Faculty Papers
Adenosine-to-inosine RNA editing, catalyzed by adenosine deaminase acting on RNA (ADAR) enzymes, alters RNA sequences from those encoded by DNA. These editing events are dynamically regulated, but few trans regulators of ADARs are known in vivo. Here, we screen RNA-binding proteins for roles in editing regulation with knockdown experiments in the Drosophila brain. We identify zinc-finger protein at 72D (Zn72D) as a regulator of editing levels at a majority of editing sites in the brain. Zn72D both regulates ADAR protein levels and interacts with ADAR in an RNA-dependent fashion, and similar to ADAR, Zn72D is necessary to maintain proper neuromuscular …

The Apicomplexan-Specific Fikk Kinase Is Crucial For Cyst Formation In Toxoplasma Gondii, Corey Gaylets
NYMC Student Theses and Dissertations
Toxoplasma gondii is an obligate intracellular pathogen. It is considered the second deadliest foodborne pathogen in the world with up to 1/3 of the human population estimated to be currently infected. The majority of immune competent people may exhibit mild symptoms following T gondii infection. The acute stage of infection is often well controlled by cellular immunity in healthy individuals. However sterilizing immunity does not occur as the tachyzoite stage of the parasite responsible for acute infection differentiates to a slow growing bradyzoite stage contained in tissues cysts that can persist for the life of the host. However, if the …

Cell Surface Regulatory Immune Targets In Anaplastic Thyroid Cancer, Sanjukta Chakraborty
NYMC Student Theses and Dissertations
Thyroid cancer incidence is increasing at an alarming rate almost trebling every decade. 52,070 new cases of thyroid cancer (14,260 in men and 37,810 in women) were diagnosed in 2019 with an estimated death toll of 2170. Although most thyroid tumors are treatable and has good prognosis, anaplastic thyroid cancer (ATC) is extremely aggressive with a grim poor prognosis of 6-9 months post diagnosis. ATC is completely refractory to mainstream therapies. In our study, immunohistochemical analyses of ATC tissues confirmed a T cell inflamed “hot” tumor immune microenvironment (TIME) as evidenced by presence of CD3+ and CD8+ T cells. This …

BACKGROUND: Risk of stroke-related morbidity and mortality increases significantly with age. Aging is associated with chronic, low-grade inflammation, which is thought to contribute to the poorer outcomes after stroke seen in the elderly. Histamine (HA) is a major molecular mediator of inflammation, and mast cells residing in the gut are a primary source of histamine.
METHODS: Stroke was induced in male C57BL/6 J mice at 3 months (young) and 20 months (aged) of age. Role of histamine after stroke was examined using young (Yg) and aged (Ag) mice; mice underwent MCAO surgery and were euthanized at 6 h, 24 h, …

Huntington’s disease (HD) is a rare and incurable autosomal neurodegenerative disease affecting 1-10 in every 100,000 people in the world. There is no cure for HD and treatments available alleviate certain symptoms for short periods of time. Evidence suggests that neuropathology of HD begins with the proteolysis of the mutated Huntingtin (mHTT) protein. A variety of proteases, like the matrix metalloproteases, cleave mHTT creating proteinaceous fragments that are thought to be neurotoxic. As these fragments increase in the brain, the damage to neurons also increases, leading to chronic inflammation due to hyper reactive microglia and astrocytes attempting to minimize and …
