Comparative Whole-Genome And Phenotype Analyses Of Virulence Factors And Lethality In Nematode Challenge Assay Among Aeromonas Isolates, Pyeongsug Kim
Master of Science in Integrative Biology Theses
Aeromonads are bacteria found globally in fresh and brackish waters, with some species causing water-borne diseases vertebrate hosts such as diarrhea and wound-infections in humans and motile Aeromonas septicemia in fish. Although many virulence factors have been described, determining which factors are most important in pathogenesis is ongoing. To examine this further, the genomes of five Aeromonas strains isolated from various animals were sequenced and annotated in this study. Each isolate was further characterized using comparative genomic analyses and testing for production of selected virulence factors. Although previously identified as Aeromonas hydrophila, modern genotypic approaches including multilocus sequence typing …

Purpose: Bupropion is an antidepressant drug that facilitates weight loss. It is a highly water-soluble drug that needs multiple dosing, so it is considered a potential candidate for oral controlled-release dosage form. The aim of this research was to formulate and evaluate satiety-inducing swellable floating bupropion tablets by direct compression targeting depression associated with eating disorders. Various combinations of natural and semi-synthetic hydrogels were selected to achieve maximum swelling and remaining buoyant in the stomach. This synergistically enhances weight loss by increasing satiety. Methods: An I-optimal mixture design was conducted to establish the optimal quantitative composition of tablets. Friability, floating …

Background: Asthma is one of the most prevalent chronic respiratory diseases. Despite increased investment in treatment, little progress has been made in the early recognition and treatment of asthma exacerbations over the last decade. Nocturnal cough monitoring may provide an opportunity to identify patients at risk for imminent exacerbations. Recently developed approaches enable smartphone-based cough monitoring. These approaches, however, have not undergone longitudinal overnight testing nor have they been specifically evaluated in the context of asthma. Also, the problem of distinguishing partner coughs from patient coughs when two or more people are sleeping in the same room using contact-free audio …

Global Challenges Of Being A Strength Athlete During A Pandemic: Impacts And Sports-Specific Training Considerations And Recommendations, Christopher Latella, G. Gregory Haff
Research outputs 2014 to 2021
The ongoing global pandemic brought about by Coronavirus II (SARS-Cov-2 or COVID-19) has caused an ongoing cessation of sporting competitions and training facility closures. This is a fundamental challenge for amateur and elite sporting professionals. Although recommendations have been provided for team-sport athletes to maintain general and sport-specific conditioning, these methods are often not optimal for strength athletes (i.e., powerlifting (PL) and weightlifting (WL)) due to the unique and narrow set of performance requirements posed by these sports. The purpose of this review is to provide evidence-based information and recommendations and highlight potential strategies and approaches that may be used …

There is a lack of large comparative study on the outcomes of reduced intensity conditioning (RIC) in acute myeloid leukemia (AML) transplantation using fludarabine/busulfan (FB) and fludarabine/melphalan (FM) regimens. Adult AML patients from Center for International Blood and Marrow Transplant Research who received first RIC allo-transplant between 2001 and 2015 were studied. Patients were excluded if they received cord blood or identical twin transplant, total body irradiation in conditioning, or graft-versus-host disease (GVHD) prophylaxis with in vitro T-cell depletion. Primary outcome was overall survival (OS), secondary end points were leukemia-free survival (LFS), nonrelapse mortality (NRM), relapse, and GVHD. Multivariate survival …

Factors In The Probability Of Covid-19 Transmission In University Classrooms, Charles Connor
Numeracy
University students and faculty members need an effective strategy to evaluate and reduce the probability that an individual will become infected with COVID-19 as a result of classroom interactions. Models are developed here that consider the probability an individual will become infected as a function of: prevalence of the disease in the university community, number of students in class, number of class meetings, and transmission rate in the classroom given the presence of an infected individual. Absolute probabilities that an individual will become infected in a classroom environment cannot be calculated because some of these factors have unknown values. Nevertheless, …

Journal of the Saudi Heart Association
Current generation LVADs provide continuous blood flow which has been shown to increase sympathetic tone. Pheochromocytoma is a rare neuroendocrine tumor which classically presents with episodic hypertension, palpitations and headaches. Careful perioperative management with alpha blockade is required to prevent cardiovascular complications, specifically hypertensive crisis. Here we present a description of perioperative management of pheochromocytoma in a patient with a continuous flow LVAD. Traditional assessment of sympathetic tone is challenging in LVAD patients due to the presence of continuous circulatory flow. Invasive management via an arterial line was useful in perioperative management, allowing for accurate circulatory monitoring and hemodynamic control. …

Hba-Deals: Accurate And Simultaneous Identification Of Differential Expression And Splicing Using Hierarchical Bayesian Analysis., Guy Karlebach, Peter Hansen, Diogo Ft Veiga, Robin Steinhaus, Daniel Danis, Sheng Li, Olga Anczuków, Peter N Robinson
Faculty Research 2020
We present Hierarchical Bayesian Analysis of Differential Expression and ALternative Splicing (HBA-DEALS), which simultaneously characterizes differential expression and splicing in cohorts. HBA-DEALS attains state of the art or better performance for both expression and splicing and allows genes to be characterized as having differential gene expression, differential alternative splicing, both, or neither. HBA-DEALS analysis of GTEx data demonstrated sets of genes that show predominant DGE or DAST across multiple tissue types. These sets have pervasive differences with respect to gene structure, function, membership in protein complexes, and promoter architecture.

The Experience Of Being A Parent Of A Child Who Stutters And Subsequent Involvement In Support Groups: A Narrative Study, Katlyn A. Ferguson, Richard Arenas
Speech and Hearing Sciences ETDs
INTRODUCTION: Developmental stuttering generally begins after a period of typical fluency and is highly variable in its presentation and persistence. This variability along with the lack of a definitive cause and social stigma often negatively impact both children who stutter (CWS) and their caregivers. However, research on the specific effects of stuttering on the caregiver is quite sparse compared to research into the caregiver experience in other disorders. Additionally, although social support has been identified as a primary protective factor for other caregivers, little evidence exists to show how support groups benefit caregivers of CWS.
METHOD: Five parents participated in …

Background: While optimal cardiovascular risk factor (CRF) profile is associated with lower mortality, morbidity, and healthcare expenditures among individuals with atherosclerotic cardiovascular disease (ASCVD), less is known regarding its impact on financial hardship from medical bills. Therefore, we assessed whether an optimal CRF profile is associated with a lower burden of financial hardship from medical bills and a reduction in cost-related barriers to health.
Methods: We used a nationally representative sample of adults between 18 and 64 years from the National Health Interview Survey between 2013 and 2017. We assessed ASCVD status and the number of risk factors to categorize …

Granulocyte-monocyte colony stimulating factor (GM-CSF) is used as an adjuvant in various clinical and preclinical studies with contradictory results. These were attributed to opposing effects of GM-CSF on the immune or myeloid systems of the treated patients or to lack of optimal dosing regimens. The results of the present study point to inter-tumor heterogeneity as a possible mechanism accounting for the contrasting responses to GM-CSF incorporating therapies. Employing xenograft models of human melanomas in nude mice developed in our lab, we detected differential functional responses of melanomas from different patients to GM-CSF both in vitro as well as in vivo. …

Anthropometry is important for predicting sport performance. While 3-dimensional (3D) body scanners increase the feasibility of anthropometric assessment, reliability data on athletes are lacking. The aim of this study was to determine the test-retest reliability of a portable, single-camera 3D body scanning system (Styku S100) to assess circumferences, and whole-body and segmental surface areas and volumes of athletes. Forty-nine (19 male) athletes were scanned six times (two sessions of three scans). The Styku scanner demonstrated nearly perfect reliability. Systematic errors were negligible (mean standardized bias [95%CI]: scan 1 vs. 4, 0.04 [0.02, 0.06]), random errors were negligible (mean standardized typical …

Proximity To Comprehensive Emergency Obstetric Care And The Utilization Of Facility-Based Delivery Services Across Six Low-And-Middle Income Countries, Chioma T. Amadi
Dissertations and Theses
Background: More than 90% of maternal deaths occur in low-and-middle-income countries (LMICs) and are largely attributed to preventable pregnancy-related causes. Comprehensive emergency obstetric care (EmOC), also known as signal functions, is the most effective life-saving intervention for managing obstetric and newborn emergencies. Health facilities offering delivery services are generally classified as having either comprehensive, basic or less than basic EmOC capacity based on their obstetric resources. Multiple EmOC methods are regularly utilized in characterizing facility obstetric capacity and this introduces inconsistencies that pose significant public health and policy implications for access to delivery care among women within a service environment. …

Background: Inflammatory arthritis in children with Down syndrome (DS) was first described in 1984 and is now termed Down syndrome-associated arthritis (DA). Studies have shown that DA is under-recognized with a 19-month average delay in diagnosis. Additionally, most patients present with polyarticular, rheumatoid factor (RF) and anti-nuclear antibody (ANA) negative disease. Current therapies for juvenile idiopathic arthritis (JIA) have been used, but appear to be poorly tolerated, more toxic and less effective in patients with DA. There is currently no standardized approach to the assessment or management of DA. The objective of this study was to describe provider perspectives toward …

Sickle cell disease (SCD) is a recessive genetic blood disorder exhibiting abnormal blood rheology. Polymerization of sickle hemoglobin, due to a point mutation in the β-globin gene of hemoglobin, results in aberrantly adhesive and stiff red blood cells (RBCs). Hemolysis, abnormal RBC adhesion, and abnormal blood rheology together impair endothelial health in people with SCD, which leads to cumulative systemic complications. Here, we describe a microfluidic assay combined with a micro particle image velocimetry technique for the integrated in vitro assessment of whole blood viscosity (WBV) and RBC adhesion.
