The fission yeast Schizosaccharomyces pombe provides a good model system to quickly study basic mechanisms underlying biological pathways conserved in higher eukaryotes. Here we utilized fission yeast to study the roles of the histone deacetylases (HDACs) Sir2 and Clr3 in heterochromatin formation and cancer associated mutations of the DEAD-box RNA helicase DDX3X, homolog of fission yeast Ded1, in translational control. Heterochromatin in fission yeast is characterized by hypoacetylation as well as methylation of histone H3 on lysine 9 (H3K9me). Heterochromatin assembly can now be separated into three distinct steps: heterochromatin establishment, spreading, and maintenance. These steps involve the actions of …

As part of the drug disposition process (absorption, distribution, metabolism, excretion), an often overlooked aspect is transport. In order for drugs to be metabolized and excreted from the body they go through the liver or other drug removal organs. For drugs that are polar or are large they must rely upon transport mechanisms to transport them across the biomembranes of the drug removal organs. OATP1B1 and OATP1B3 are transporters on the sinusoidal membrane of the liver which work in concert with the drug metabolizing enzymes as part of the drug removal process. Models of speech perception suggest a dorsal stream connecting the temporal and inferior parietal lobe with the inferior frontal gyrus. This stream is thought to involve an auditory-motor loop that translates acoustic information into motor/articulatory commands and is further influenced by decision making processes that involve maintenance of working memory or attention. Parsing out dorsal stream’s speech specific mechanisms from memory related ones in speech perception poses a complex problem. Here I argue that these processes may be disentangled from the viewpoint of the temporal dynamics of sensorimotor neural activation around a speech perception related event.

Rheumatoid arthritis (RA) is one of the autoimmune diseases, and is affecting 2.5 million Americans in total. Among the treatment options of RA, 1α,25-dihydroxyvitamin D3 [1,25(OH)2D3] is the only steroidal drug used clinically for anti-inflammatory and immune diseases. However, long-term use of 1,25(OH)2D3 (625 µg/day) in human would result in hypercalcemia (toxicity), and 1,25(OH)2D3 has substantial hypercalcemic effects (toxicity) in mice at a dose as low as only 2 µg/kg. Fortunately, during the investigation of novel metabolic pathway of vitamin D3 by cytochrome P450 enzymes, we found 20S-hydroxyvitamin D3 [20S(OH)D3] as a good lead compound. 20S(OH)D3 suppressed disease symptoms at …

Cellular protein homeostasis is achieved by a delicate network of molecular chaperones and various proteolytic processes such as ubiquitin–proteasome system (UPS) to avoid a build-up of misfolded protein aggregates. The latter is a common denominator of neurodegeneration. Neurons are found to be particularly vulnerable to toxic stress from aggregation-prone proteins such as α-synuclein. Induction of heat-shock proteins (HSPs), such as through activated heat shock transcription factor 1 (HSF1) via Hsp90 inhibition, is being investigated as a therapeutic option for proteinopathic diseases. HSF1 is a master stress-protective transcription factor which activates genes encoding protein chaperones (e.g. iHsp70) and anti-apoptotic proteins. However, …

Over the past two and a half years, I have studied fundamental aspects of astrocyte biology by investigating the existence and mechanism of astrocyte local translation peripherally around tripartite synapses consisting of pre- and post-synaptic neuron terminals and an associated astrocyte. Astrocytes are critical components of central nervous system synapses (which are predominately tripartite in nature); thus, it is important to consider how astrocyte dysregulation and dysfunction could contribute to the pathogenesis of diseases of synaptic connectivity such as autism spectrum disorders, Alzheimer’s disease, seizure disorders, and more. Putative cannabinoid receptor 2 (CB2)-selective agonists were identified from a library of commercially available compounds via inhibition of cAMP accumulation in high throughput screening. Binding affinity and receptor subtype selectivity were assessed using heterologous competition binding assays against the known cannabinoid orthosteric ligand CP55940. Test compounds ASX0152383 and CSC003141 preferentially bound to CB2, with no detection of binding to CB1 up to 1 mM. CMB038865 exhibited nearly 100-fold selectivity for CB1 over CB2, while CZ000026 bound non-preferentially to both receptors in the low micromolar range. To determine the extent of G protein …

Background: Many older adults suffer from chronic pain, a disease treated with opioids. Older adults are also highly susceptible to opioid ADEs. Research addressing older adults’ self-management of opioid ADEs is lacking.
Objectives: This study compared the opioid ADE profile found by the present research to those previously reported in the literature for older adults. It also described how older adults respond to opioid ADEs and the resulting outcomes.
Method: The study utilized a secondary analysis design to examine 15 opioid ADE cases from a pilot study that described older adults’ responses to analgesic ADEs.

West Nile virus (WNV) is a positive-sensed, single-stranded RNA flavivirus that can cause human neuroinvasive diseases, including encephalitis, meningitis, and flaccid paralysis. The mechanisms by which WNV enters the central nervous system and the host-factors that are involved in WNV-neuroinvasiveness are not completely understood. Osteopontin (OPN), a multifunctional glycoprotein, has been implicated as a bio-marker for a number of neuroinflammatory diseases. In particular, secreted (s)OPN has been implicated to participate in recruitment of polymorphonuclear neutrophils (PMN) to sites of its expression, while PMNs have been suggested to act as WNV reservoirs. The purpose of this study was to explore the relationship of adherence to evidence-based guidelines for pharmacologic management by select Mississippi hospitals and their hospital readmission rates. This was an exploratory study with a retrospective design. This design offered insight into the relationship between readmissions of heart failure patients and adherence to the national guidelines for the pharmacologic treatment of heart failure. The study took place in a hospital in the southeastern section of the United States. Data were collected from a database of patients with heart failure seen between January 2011 and June 2014.

Amblyomma maculatum (Gulf coast tick) ticks are prevalent across the Atlantic to Gulf Coast region of United States. These ticks are recognized vectors of Rickettsia parkeri, a spotted fever group of Rickettsia (SFGR) known to cause American boutonneuse fever associated with fever and eschar rashes localized to the site of bites. We hypothesized that Rickettsia parkeri colonization and proliferation in the tick vector involve pathogen-symbiont dynamics and tick-pathogen interactions, which influence rickettsial transmission to the victims after tick bites. The purpose of this cross-sectional, descriptive, correlational study was to examine ED staff nurses’ opinions of their ED nurse managers’ full range of leadership characteristics , the degree to which the characteristics correlate with specific outcomes of leadership behavior, and how the results of the measure and subscale compare to other nursing groups in literature. The need for nurses to pursue doctoral education is imperative to the professionalization of the discipline of nursing in light of the changing healthcare environment. Presently, < 1% of the nursing workforce possesses a doctoral degree (“Transforming Nursing Education,” 2016), albeit recommendations from the Institute of Medicine (IOM) (2010) admonish the need to increase the number of doctoral-prepared nurses. The need to understand characteristics of nurses who seek doctoral education is critical in planning long-term strategies for nursing education in the United States (US) (Kovner, Brewer, Katigbak, Djukic, & Fatehi, 2012). In conjunction with describing characteristics of nurses pursuing doctoral education, this research describes the motivational orientation and factors of registered nurses (RNs) pursuing doctoral education.
A descriptive correlational design was utilized to examine concepts relating to motivational orientation of RNs pursuing doctoral education. Participants were divided into two categories: (a) RNs seeking the Doctor of Philosophy (Ph.D.) degree and (b) RNs pursuing the Doctor of Nursing Practice (DNP) degree. A total of 173 RNs enrolled in either a Ph.D. or DNP program in the Gulf South region of the US comprised the final sample. Binary logistic regression was utilized …

The purpose of this study was to provide a better understanding of the goalkeeper position in order to prepare goalkeepers for competition. The objectives of this study were to characterize the technical and physical demands of an NCAA Division I collegiate goalkeeper over three seasons and examine trends from season to season. Count and frequency for each definition were analyzed. Incivility is rude or discourteous behavior that demonstrates a lack of respect for others. Some nurses ignore the dictates of professionalism and exhibit a total disregard for colleagues and peers by purposefully targeting each other with uncivil behaviors. Incivility has invaded the nursing educational environment with deleterious results. Uncivil behaviors perpetrated by nursing students against other nursing students cause psychological and physiological distress for victims and witnesses. Human papillomavirus (HPV) is the most prevalent sexually transmitted infection in the U.S., where one HPV-related cancer is diagnosed every 20 minutes. The most common HPV-related cancer is cervical cancer, with an estimated incidence of 12,000 cases annually, a third of which lead to death. Cervical cancer disparately affects women of ethnic minority groups and geographically isolated regions, such as Appalachia. Tennessee ranks third highest in cervical cancer incidence in the country. Lilium grayi (Gray’s Lily), a southern Appalachian endemic species, is threatened by a Lilium-specific fungal pathogen, Pseudocercosporella inconspicua. The disease is characterized by tan lesions that can cause early senescence, while also lowering seed production and viability. This project tested for P. inconspicua conidia and accessed health at nine locations. The disease was present and ubiquitous across the range of L. grayi. Through identification of P. inconspicua conidia in the field, L. superbum (Turk’s Cap Lily) was identified as an additional host, while L. michauxii (Michaux’s Lily) was disease-free. Fragile X Syndrome (FXS) is the most common form of inherited Intellectual Developmental Disorder and a genetic model for Autism Spectrum Disorder. Those living with FXS face emotional, social, intellectual, and physical challenges that impact engagement in occupations, yet to date, there has been limited qualitative research examining family occupations in FXS. In this research, twelve interviews were transcribed, coded, and analyzed for common themes anchored in the Person, Environment, Occupation (PEO) model. This critically appraised topic explores the functional task performance of adults with low vision utilizing tactile vision substitution systems, specifically tongue display units (TDUs). TDUs are a novel assistive device that functions to provide artificial vision to those with low vision. TDUs pixelate images captured on a camera the person wears and the images are translated via electronic stimulation on the tongue to paint a picture. Two studies that measured functional task performance utilizing a TDU with adults with low vision were explored. Therapeutic Listening® is an intervention increasingly used by occupational therapists despite the lack of supporting evidence in current literature. Therapeutic Listening® is a sound-based treatment developed by Sheila Frick, OTR/L, rooted in sensory integration. The purpose of this continuation study was to compare the quality of bilateral movement in typically developing children after either listening to Therapeutic Listening® Bilateral Coordination Quick Shift or listening to white noise. This study used a randomized control pretest-posttest experimental design to analyze posture, smooth and continuous movement, effort, precision, and arm/leg movements.
