Latently human immunodeficiency virus (HIV)-infected cells are transcriptionally quiescent and invisible to clearance by the immune system. To demonstrate that the latency reversing agent vorinostat (VOR) induces a window of vulnerability in the latent HIV reservoir, defined as the triggering of viral antigen production sufficient in quantity and duration to allow for recognition and clearance of persisting infection, we developed a latency clearance assay (LCA). The LCA is a quantitative viral outgrowth assay (QVOA) that includes the addition of immune effectors capable of clearing cells expressing viral antigen. Here we show a reduction in the recovery of replication-competent virus from …

Climate change will increasingly affect the health of vulnerable populations, including maternal and fetal health. This systematic review aims to identify recent literature that investigates increasing heat and extreme temperatures on pregnancy outcomes globally. We identify common research findings in order to create a comprehensive understanding of how immediate effects will be sustained in the next generation. Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guide, we systematically reviewed articles from PubMed and Cochrane Reviews. We included articles that identify climate change-related exposures and adverse health effects for pregnant women. There is evidence that temperature extremes adversely …

Indoor environments can influence human environmental chemical exposures and, ultimately, public health. Furniture, electronics, personal care and cleaning products, floor coverings and other consumer products contain chemicals that can end up in the indoor air and settled dust. Consumer product chemicals such as phthalates, phenols, flame retardants and per- and polyfluorinated alkyl substances are widely detected in the US general population, including vulnerable populations, and are associated with adverse health effects such as reproductive and endocrine toxicity. We discuss the implications of our recent meta-analysis describing the patterns of chemical exposures and the ubiquity of multiple chemicals in indoor environments. …

Nursing Student Class Projects (Formerly MSN)
Chronic Obstructive Pulmonary Disease is the third leading cause of death in the United States and is affecting more than 32 million Americans (Mosenifar, 2017). The prevalence of the disease continues to rise and many people are unaware they even have the disease. Prevention is essential in decreasing the incidence of the disease and Advanced Practice Nurses (APN) play a vital role. Emphysema is abnormal permanent enlargement of air spaces distal to the terminal bronchioles, accompanied by the destruction of alveolar walls and without obvious fibrosis (Boka, 2016). Being able to understand the pathophysiology of emphysema will allow for providers …

Supporting children's self-regulation in eating through caregivers' practice of responsive feeding is paramount to obesity prevention, and while much attention has been given to supporting children's selfregulation in eating through parents' responsive feeding practices in the home setting, little attention has been given to this issue in childcare settings. This qualitative study examines childcare providers' perspectives on using responsive feeding practices with young children (2–5 years). Individual semistructured interviews were conducted with providers until saturation was reached. Data was analyzed using thematic analysis. The final sample included 18 providers who were employed full-time in Head Start or state-licensed center-based childcare …

Syk Inhibitors In Clinical Development For Hematological Malignancies, Delong Liu, Aleksandra Mamorska-Dyga
NYMC Faculty Publications
Spleen tyrosine kinase (Syk) is a cytosolic non-receptor protein tyrosine kinase (PTK) and is mainly expressed in hematopoietic cells. Syk was recognized as a critical element in the B-cell receptor signaling pathway. Syk is also a key component in signal transduction from other immune receptors like Fc receptors and adhesion receptors. Several oral Syk inhibitors including fostamatinib (R788), entospletinib (GS-9973), cerdulatinib (PRT062070), and TAK-659 are being assessed in clinical trials. The second generation compound, entospletinib, showed promising results in clinical trials against B-cell malignancies, mainly chronic lymphoid leukemia. Syk inhibitors are being evaluated in combination regimens in multiple malignancies.

Dysregulation of transforming growth factor-β1 (TGF-β1) and insulin-like growth factor (IGF) axis has been linked to reactive stroma dynamics in prostate cancer progression. IGF binding protein-3 (IGFBP3) induction is initiated by stroma remodeling and could represent a potential therapeutic target for prostate cancer. In previous studies a lead quinazoline-based Doxazosin® derivative, DZ-50, impaired prostate tumor growth by targeting proteins involved in focal adhesion, anoikis resistance and epithelial-mesenchymal-transition (EMT). This study demonstrates that DZ-50 increased expression of the epithelial marker E-cadherin, and decreased the mesenchymal marker N-cadherin in human prostate cancer cells. In DU-145 cells, the effect of DZ-50 on EMT …

Traumatic brain injury (TBI) produces profound and lasting neuroinflammation that has both beneficial and detrimental effects. Recent evidence has implicated microRNAs (miRNAs) in the regulation of inflammation both in the periphery and the CNS. We examined the expression of inflammation associated miRNAs in the context of TBI using a mouse controlled cortical impact (CCI) model and found increased levels of miR-21, miR-223 and miR-155 in the hippocampus after CCI. The expression of miR-155 was elevated 9-fold after CCI, an increase confirmed by in situ hybridization (ISH). Interestingly, expression of miR-155 was largely found in neuronal nuclei as evidenced by co-localization …

Spreading depolarization (SD) is a slow propagating wave of strong depolarization of neural cells, implicated in several neuropathological conditions. The breakdown of brain homeostasis promotes significant hemodynamic and metabolic alterations, which impacts on neuronal function. In this work we aimed to develop an innovative multimodal approach, encompassing metabolic, electric and hemodynamic measurements, tailored but not limited to study SD. This was based on a novel dual-biosensor based on microelectrode arrays designed to simultaneously monitor lactate and glucose fluctuations and ongoing neuronal activity with high spatial and temporal resolution. In vitroevaluation of dual lactate-glucose microbiosensor revealed an extended linear range, …

The genomes of cancers deficient in mismatch repair contain exceptionally high numbers of somatic mutations. In a proof-of-concept study, we previously showed that colorectal cancers with mismatch repair deficiency were sensitive to immune checkpoint blockade with antibodies to programmed death receptor-1 (PD-1). We have now expanded this study to evaluate the efficacy of PD-1 blockade in patients with advanced mismatch repair-deficient cancers across 12 different tumor types. Objective radiographic responses were observed in 53% of patients, and complete responses were achieved in 21% of patients. Responses were durable, with median progression-free survival and overall survival still not reached. Functional analysis …

Cahost: An Excel Workbook For Facilitating The Johnson-Neyman Technique For Two-Way Interactions In Multiple Regression, Stephen W. Carden, Nicholas S. Holtzman, Michael J. Strube
Psychology: Faculty Publications
When using multiple regression, researchers frequently wish to explore how the relationship between two variables is moderated by another variable; this is termed an interaction. Historically, two approaches have been used to probe interactions: the pick-a-point approach and the Johnson-Neyman (JN) technique. The pick-a-point approach has limitations that can be avoided using the JN technique. Currently, the software available for implementing the JN technique and creating corresponding figures lacks several desirable features–most notably, ease of use and figure quality. To fill this gap in the literature, we offer a free Microsoft Excel 2013 workbook, CAHOST (a concatenation of the first …
Enhancing The Well-Being Of Older Adults And Young Adults With Developmental Disabilities Through Participation In An Intergenerational Community Garden: Participatory Action Research, Diane L. Keen
Doctorate of Nursing Science Dissertations
The purpose of this Participatory Action Research (PAR) study was to discover ways to enhance well-being in young adults with DD and older adults in the Oak Grove community. This dissertation details how PAR was used in the Oak Grove community to identify ways to enhance purpose and meaningful activity for older adults while developing positive relationships with younger adults with DD through qualitative methods.
Ryff and Keyes (1995) discovered that although some elements of well-being remain stable as a person ages, purpose-in-life drops significantly in older adults. In addition, young adults with developmental disabilities (DD) lack meaningful relationships, which …

What You Know Makes A Difference: Physical Activity Maintenance And Adherence Of Collegiate Students, Vista Beasley, Brooke Thompson, Patrick R. Young, Itay Basevitch
Journal of Counseling and Psychology
For decades, exercise psychology researchers dismissed health/exercise knowledge as a determinant of physical activity (PA). We sought to overturn this misconception, showing that psychological theory may serve as a basis for informing physical education curriculum. Based on social cognitive and self-determined motivation theories, we examined health/exercise knowledge as a determinant of collegiate students’ PA maintenance (i.e., ≥ 6 months of regular PA involvement); adherence to United States Department of Health and Human Services (USDHHS) 2008 PA guidelines; and PA types (i.e., aerobic, weight training). Collegiate students (n = 231) provided data via online survey. ANOVA analyses revealed that knowledge …
