To determine the role of Ca²⁺ signaling in activation of the Mitogen-Activated Protein Kinase (MAPK) pathway, we subjected MC3T3-E1 pre-osteoblastic cells to inhibitors of Ca²⁺ signaling during application of fluid shear stress (FSS). FSS only activated ERK1/2, rapidly inducing phosphorylation within 5 min of the onset of shear. Phosphorylation of ERK1/2 (pERK1/2) was significantly reduced when Ca_i²⁺ was chelated with BAPTA or when Ca²⁺ was removed from the flow media. Inhibition of both the L-type voltage-sensitive Ca²⁺ channel and the mechanosensitive cation-selective channel blocked FSS-induced pERK1/2. Introduction: Population-based studies in orthodontics have focused on differences between normative and perceived needs. However, information from national data on the prevalence of orthodontic visits and their associated factors in adults in the United States is scarce. We examined the demographic profile of likely adult users of orthodontic services and whether there is racial and ethnic disparity in orthodontic visits. Methods: We analyzed data from the Medical Expenditure Panel Survey, 2000-2004. Results: Overall, about 1% of the population reported an orthodontic visit. Subjects who made a general dental visit during the current year were significantly more likely to also have …

The clinical routine for ultrasound training is on real patients; therefore monitored and guided examinations involving medical students are quite time-constrained. Background: Maternal exposure to wood fuel smoke may lead to impaired fetal growth due to hypoxia and or oxidative stress from smoke constituents such as carbon monoxide and particulate matter. Objectives: We studied the risk of low birth weight (LBW) and reduced mean birth weight in relation to reported use of wood for cooking during the prenatal period, compared with natural gas (NG).

In this manuscript we give an overview of observational sleep research with a particular emphasis on sleep stage transitions. Sleep states represent a categorization of sleep electroencephalogram behavior over the night. We postulate that the rate of transitioning between sleep states is an important predictor of health. This claim is evaluated by comparing subjects with sleep disordered breathing to matched controls.

Blame the celebrity, chalk it up to reckless living, a self-destructive lifestyle, a pursuit of pleasure through recreational drugs. But the drugs that killed Ledger -- three types of benzodiazepines, an antihistamine, two pain relievers -- are all substances people take for sleep.

Born in El Paso in 1920, Gambrell entered this world in the family home due to his physician father’s decision to avoid local hospitals overflowing with Spanish Influenza patients. His father had recently returned from service in France during World War I; earlier he had served under General Jack “Black” Pershing during the latter’s 1916 punitive expedition into Mexico in Pershing’s ill-fated attempt to capture Pancho Villa. Serous cystadenomas (SC) of the pancreas are uncommon benign cystic neoplasms that were first differentiated from mucinous cystadenomas in 1978 by Compagno and Oertel (1-3). Serous cystadenomas characteristically have a spongy gross appearance and are composed of innumerable tiny cysts lined by flattened cuboidal or polygonal cells with clear to pale eosinophilic cytoplasm atop of thin fibrous septae. Since 1978, five unique SC subtypes have been identified (4-5). Here we report a case of a solid serous adenoma, a rare serous cystadenoma subtype.

Childhood obesity is a public health crisis in the United States. By the year 2015 an estimated 24 percent of all children will be obese. Children who are obese miss on average 20 percent more school than their healthy-weight classmates. To describe how frequently harm is reported in the abstract of high impact factor medical journals. We carried out a blinded structured review of a random sample of 363 Randomised Controlled Trials (RCTs) carried out on human beings, and published in high impact factor medical journals in 2003. Main endpoint: 1) Proportion of articles reporting harm in the abstract; and 2) Proportion of articles that reported harm in the abstract when harm was reported in the main body of the article. Analysis: Corrected Prevalence Ratio (cPR) and its exact confidence interval were calculated. Non-conditional logistic regression was used.

On a sunny fall day in 2007, a 50-year old semi truck driver hauling processed frozen chicken in a refrigerated trailer, died when his semi crashed into a rock wall. The driver was north bound on an interstate highway when witnesses saw the semi in the left lane contact the concrete median barrier then veer over to the right across one traffic lane and the shoulder, then strike a rock wall. More often than not biomarker studies analyze large quantities of variables with complicated and generally unknown correlation structure. There are numerous statistical methods which attempt to unravel these variables and determine the underlying mechanism through identification of causally related biomarkers. Results from these methods are generally difficult to interpret and nearly impossible to compare across studies. The FDA has currently called for a standardization of methods and protocol for biomarker detection. In response, we propose targeted variable importance (tVIM) as a standardized method for biomarker discovery. This research effort addressed the issue of long-term access to electronic medical records as technological generations become obsolete, thereby preventing the access to patient health information. Using the Delphi methodology, experts with experience in electronic medical records and applicable systems provided insight based on their years of hands-on experience managing and/or using records and these systems. This research developed and applied microlithography techniques to etch microscope slide platforms in order to measure thermal inactivation of Bacillus thuringiensis and Bacillus anthracis spores, two closely related Bacillus species. B.t. is widely used as a surrogate for B.a. in response studies and in some field studies. This work addressed the previously identified problem of measuring both spore growth and spore thermal kill threshold using the traditional method of spreading a diluted spore solution on a plate. The micro-etched platforms forced spore separation thereby preventing neighbor growth from obscuring germination and initial vegetative growth measurements using a microscope. The technique …

Lethal factor (LF), a component of anthrax toxin, is the primary virulence factor that allows Bacillus anthracis to evade the immune response by blocking the activation of mitogen-activated protein kinase (MAPK) enzymes. This research modifies three published MAPK models to reflect this signal inhibition and to estimate a first-order reaction rate by fitting the models to published viability data for two macrophage cell lines cultured with the LF-producing Bacillus anthracis-Vollum1B strain. One model appears to be ill-suited for this purpose because not all relevant MAPK components could be integrated into the inhibition equations. In the hippocampus, BK channels are preferentially localized in presynaptic glutamatergic terminals including mossy fibers where they are thought to play an important role regulating excessive glutamate release during hyperactive states. Large conductance calcium-activated potassium channels (BK, MaxiK, Slo) have recently been implicated in the pathogenesis of genetic epilepsy. However, the role of BK channels in acquired mesial temporal lobe epilepsy (MTLE) remains unknown. Telomerase is a ribonucleoprotein that is reactivated in cancer cells to allow for continuous cellular division and indefinite growth. With telomerase being expressed in more than 85% of all cancer, it is imperative that we understand how to selectively inactivate and degrade this unique DNA polymerase. In doing so, we can specifically target tumor cells to erode their telomeres so that they will undergo apoptosis or senescence. Through this research, we have learned that telomerase can be degraded in the nucleoplasm by Hsp90 chaperone inhibition and in the cytoplasm by the dominant negative mutant, D712A V713I. On a Fall day in 2005 at 3:00 PM, a 46 year-old male semi-tractor trailer owner-operator was hauling logs when he rounded a curve, drove off the right side of a two lane state highway, struck a tree and rolled over. Emergency medical services were called to the scene. When they arrived, they found the driver without vital signs and contacted the local coroner. Kentucky State Police arrived and called Kentucky Vehicle Enforcement to the scene. The driver had not been wearing his seatbelt. Toxicology detected methamphetamine and doxylamine in his system at the time of the crash.
