Mycobacterium avium complex strains and Mycobacterium paratuberculosis are closely related intracellular pathogens affecting humans and animals. M. avium complex infections are a leading cause of morbidity and mortality in AIDS patients, and M. paratuberculosis is the agent of Johne's disease in ruminants. Genetic manipulation of these micro-organisms would facilitate the understanding of their pathogenesis, the construction of attenuated vaccine strains and the development of new drugs and treatment methods. This paper describes the replication of mycobacterial shuttle phasmids and plasmids, and the expression of the firefly luciferase reporter gene in M. avium complex and M. paratuberculosis. In a previous study, enterohemorrhagic Escherichia coli (EHEC) O157:H7 with a deletion and insertion in the eaeA gene encoding intimin was used to establish that intimin is required for the organism to attach to and efface microvilli in the piglet intestine (M. S. Donnenberg, S. Tzipori, M. L. McKee, A. D. O’Brien, J. Alroy, and J. B. Kaper, J. Clin. Invest. 92:1418–1424, 1993). However, in the same investigation, a role for intimin in EHEC adherence to HEp-2 cells could not be definitively demonstrated. Bovine herpesvirus 1 (BHV-1) establishes a latent infection in sensory ganglionic neurons of infected animals. Expression of latency-related (LR) gene products is controlled by a 980-bp fragment (LR promoter). DNA sequence analysis revealed that two major open reading frames (ORFs) are in the LR gene. Antibodies directed against both ORFs were generated in rabbits by using synthetic peptides. Antibody P2, which is directed to sequences near the amino terminus of ORF 2, recognized a 41-kDa protein in lytically infected cells, suggesting that ORF 2 encodes a protein. The Thai Journal of Veterinary Medicine
Thailand has one of the richest freshwater small fauna in the world; over 170 species are currently recognized. However, species that transmit animal diseases, or are suspected to, are relatively few. At present, only 23 freshwater snail species are implicated or suspected in animal disease transmission. These species are restricted to two orders and seven families. The parasites they transmit are blood flukes, liver flukes, lung flukes, intestinal flukes and some lung nematodes.(ประเทศไทยเป็นประเทศหนึ่งที่มีหอยน้ำจืดมากที่สุดในโลก ในปัจจุบันมีหอยน้ำจืดที่ได้ถูกค้นพบแล้วมากกว่า 170 ชนิด อย่างไรก็ตามชนิดของหอยที่นำโรคมาสู่สัตว์หรือสงสัยว่าอาจนำโรคมาสู่สัตว์นั้นค่อนข้างน้อย ในปัจจุบันมีหอยน้ำจืดเพียง 23 ชนิด ที่เกี่ยวข้องหรือ อาจเกี่ยวข้องในการนำโรคมาสู่สัตว์ พวกหอยเหล่านี้จัดอยู่ใน 2 อันดับ และ 7 วงศ์ พยาธิที่ใช้หอยน้ำจืดเป็นพาหะกลางคือ พยาธิใบไม้เลือด พยาธิใบไม้ตับ พยาธิใบไม้ลำไส้ และพยาธิตัวกลมในปอด)

B-mode, Real-Time Ultrasound with 5 MHz Linear Array Transducer were used for pregnancy examination. Twenty five female dogs with different gestation period were examined. During early gestation period (19-29 days), round or oval embryonic vesicles with embryo in embryonic fluid were seen. Embryonic heart activity was detected at about 21-22 days of gestation and was seen easily in the middle and late gestation. At 30-35 days of gestation, the embryo developed hypoechoic head and body with embryonic movement. After 36 days of gestation the hyperechoic skeleton, hypoechoic organs and embryonic activities can be clearly identified. The conventional colorimetric method of cholinesterase activity determination in blood and tissues of mammals was applied to investigate the enzyme activity in muscle and nervous cell of aquatic animals which are economically important, such as, tiger shrimp (Penaeus monodon fabricus) and giant perch (Lates calcarifer). Decreased enzyme activity in aquatic animals may indicate the contamination of insecticide in water resources. This study demonstrated that the method was suitable for detecting toxicity of organophosphate insecticide due to its convenience, simplicity and no sophisticated or expensive instruments needed. A PRRS virus specific antibody among swine breeding farms in Thailand was studied. Serum samples were collected from 15 breeder farms in central and north-eastern part of Thailand during January to March 1995. Of total 449 samples, 286 (64%) were revealed PRRS-antibody indicated the history of PRRS virus exposure. The exposure rate was varied from 20-90% from farm to farm which appeared among replacement gilts, primiparous sows, multiparous sows and boars. The highest prevalence (75%) was in replacement gilts. A continuing incidence of abortion record was investigated in a 1700 sow-breeding farm during 1992 through the first three quarters of 1995. The abortion rate per pregnant sows per year were 2.5, 3.7, 3.9 and 3.2% respectively compared to 0.3% recorded in 1991. Six months before, the farm has introduced the first batch of replacement gilts from a domestic breeding supplier into the farm. No obvious clinical signs were observed in sows. Laboratory evidences did not show any significant association with any conventional infection of the agents related the abortion. Prepubertal neutering in dogs has both advantages and disadvantages. The advantages include reduction of the incidence of mammary gland neoplasia and reduction of sexual behavior. The disadvantages are elongation of long bone and an inflammation of the external genitalia. Anesthesia of young dog may be more risky than in the adult. Effects of prepubertal neutering on urethral diameter and sex hormone imbalance dermatosis are not well documented. Acute Toxicity Of Carbaryl On Giant Pearch (Lates Calcarifer)(พิษเฉียบพลันของคาร์บาริลในปลากะพงขาว(Lates Calcarifer)), Supatra Srichairat, Supattra Jiamsak, Jirasak Tangtrongpiros

The acute toxicity of carbaryl was determined for giant pearch (Lates calcarifer) in static exposure systems. The mean 96-h LC was 2.95 mg/litre. All exposed fishes at first showed symptoms of carbaryl intoxication such as hyperexcitability, decrease in swimming performance, loss of coordination movement and reduction of food intake. Severity of intoxication and mortality rate were found to be dose dependent. Decreased cholinesterase activity in muscle and brain of all treated fishes was also observed withincreasing of carbaryl concentration. The reduction of cholinesterase in muscle and brain of all treated groups was statistically significant (p < 0.05) comparing with the control group.(ศึกษาความเป็นพิษเฉียบพลันของคาร์บาริลในปลากะพงขาวหลังแช่นาน 96 ชั่วโมง ค่า LC50 ภายใน 96 ชั่วโมงเท่ากับ 2.95 มก./ลิตร พบว่าปลาทุกกลุ่มที่ได้รับคาร์บาริลแสดง อาการเป็นพิษโดยมีอาการกระวนกระวาย ว่ายน้ำช้าลง สูญเสียการทรงตัว กินอาหารน้อยลง ความรุนแรงของอาการและเปอร์เซนต์การตายของปลาเพิ่มขึ้นตามขนาดความเข้มข้นของ สารฆ่าแมลงที่ได้รับ สมรรถนะของเอนไซม์โซลีนเอสเทอเรสในกล้ามเนื้อและสมองของปลาทุกกลุ่มลดลงตามขนาดความเข้มข้นของสารฆ่าแมลงที่เพิ่มขึ้นและต่ำกว่ากลุ่มควบคุมอย่างมีนัยสำคัญทางสถิติที่ p < 0.05)

Three hundred and fifty five neonatal piglets were used to determine the effect of piglet processing at two different ages on the survival rate from birth to weaning (28 days). Piglet processing procedures generally include teeth clipping, tail docking and injection of iron. Piglets were divided into two groups. The first group was processed at an age of 1 day (n 180). The second group was processed at an age of 3 days (n = 175). Neuraminidase-treated bovine serum amyloid P component (SAP) has chromatographic profile from DEAE-HPLC column of shorter retention time and less peak number than intact molecule does. SDS-PAGE analysis reveals a decrease in size of bovine SAP subunits after neuraminidase treatment, compared with an untreated molecule. Different freezing methods were performed to compare the viability of African catfish sperm. Cooling at 4°C can preserve the quality of spermatozoa up to three days. Freezing with the manual method using straw, cryotube, or pellet cryoprotected by methanol could not preserve the African catfish spermatozoa. Freezing by the programmable computerized freezing (PCF) method in cryotubes using 5 and 10% DMSO as cryoprotectant at -196°C for 5 minutes, 3, 7 and 15 days show a similar percentage motility of approximately 10, 5, <1 and <1% respectively. But freezing with 20% of DMSO showed lower percentage motility about 5, 5, <1 and <1% respectively. A freezing by PCF method using straw with DMSO and methanol fail to preserve the African catfish spermatozoa.(ใช้น้ำเชื้อที่เก็บจากพ่อพันธุ์ปลาดุกรัสเซียที่เจริญพันธุ์ เพื่อศึกษาวิธีการแช่เย็น (4 องศา เซลเซียส และการแช่แข็ง (-196 องศาเซลเซียส) ด้วยวิธี Manual Freezing (MF) และด้วยวิธี Programmable Computerized Freezing (PCF) การแช่เย็นพบว่าสามารถเก็บรักษาน้ำเชื้อปลาได้นานอย่างน้อย 3 วัน ส่วนการแช่แข็งโดยวิธี MF ทั้งในรูป straw, cryotube และ pellet โดยใช้ methanol เป็นสารป้องกันการแช่แข็งนั้นไม่สามารถเก็บรักษาคุณภาพน้ำเชื้อไว้ได้ ส่วนการแช่แข็งด้วยวิธี PCR ในรูป cryotube โดยใช้ DMSO ที่ 5 และ 10% เป็นสารป้องกันการแช่แข็ง พบว่าเปอร์เซ็นต์การเคลื่อนไหวของตัวอสุจิ นาน 5 นาที หรือ 3, 7 และ 15 วัน มีค่าใกล้เคียงกัน ประมาณ 10, 5, 11 และ <1% ตามลำดับ เมื่อใช้ DMSO ที่ 20% เปอร์เซ็นต์การเคลื่อนไหวจะน้อยกว่าประมาณ 5, 5, 41 และ 41% ตามลำดับ ส่วนการแช่แข็งโดยวิธี PCF ในรูปแบบ straw โดยใช้ methanol และ DMSO เป็นสารป้องกันการแช่แข็งไม่สามารถเก็บรักษาคุณภาพน้ำเชื้อไว้ได้)

This reports on 4 cases of canine anesthesia carried out for a thoracoscopy workshop. Since one lung needed to be collapsed for the surgical procedure, endo-bronchial tubes had to be used. For a 15-kg dog, a Portex left-sided double-lumen endobronchial tube, number 6, was easily passed without having to use a laryngoscope. The length from tip to angle of the mouth was 29 cm Auscultation was checked in the same way as is done in the human. Separate lung ventilation was successfully done on all dogs. We identified a semidominant, chemically induced, mouse use mutation with a complex array of abnormal behaviors including bidirectional circling and hyperactivity, abnormal circadian rhythmicity and abnormal responses to light. In this report, we genetically and phenotypically characterized the circling/ waltzing component of the abnormal behavior. We mapped the locus controlling this trait by heterozygosity mapping of partially congenic lines carrying the mutagenized chromosome outcrossed to different inbred strains for three generations. Canine Parvovirus serum antibody titres in puppies before the first parvovirus vaccination were studied in Thailand. The titres were measured by haemagglutination inhibition test (HI). The puppies had HI titers between 320-10,240 with a mean of 1,280. There was no correlation between titres and either age or previous history of CPV vaccination in the bitches that produced the puppies. There was also no apparent association between titres and either sex or breed. The suprachiasmatic nucleus (SCN) of the anterior hypothalamus (AH) is a circadian oscillator and an important component of the mammalian circadian system. To determine whether the SCN is the dominant circadian pacemaker responsible for generating a species-typical characteristic of circadian rhythms [i.e., period length (T)], neural transplantation was conducted using fetal AH donors of different species and SCN-lesioned (SCNx) hosts. The circadian behavior of each of the three donor species is clearly distinguishable by its species-typical T. Gonadotropin hormones in form of 400 iu of PMSG + 200 iu of HCG were given to anestrus or delayed oestrus sows after weaning and to anestrus replacement gilts (Landrace x Large White). The animals were divided into a treated group (sows, n = 20 and gilts, n = 17), and a control group (sows, n = 5 and gilts, n = 4). An experiment to stimulate follicular growth was performed in ten pre-pubertal swamp buffaloes (8-12 months old). They were divided into two groups according to the gonadotropin treatments. Five calves were injected intramuscularly with 24 mg of follicular stimulating hormone (FSH) and 2 months later with a 3000 IU of pregnant mare serum gonadotropin (PMSG), while the other 5 calves were first treated with PMSG which was followed by FSH. The ovarian responses were examined by laparotomy and scored according to the number of follicles (≥ 0.8 cm) and corpus hemorrhagicum. Ovaries had a more significant response to FSH than to …

The interaction between semen quality of Holstein Friesian, Jersey, Crossbred and Nili-Ravi buffaloes and the environment was studied. Semen was collected for one year at the Semen Production Unit, Surizai District, Peshawar from 10 Holstein Friesian, 4 Jersey and 4 Crossbred bulls and 3 buffaloes. Semen was collected early in the morning, twice weekly, using an artificial vagina. Environmental data regarding the monthly average temperature, humidity and daylight, was recorded. The correlation co-effi-cient between the semen volume, motility and sperm concentration and the three environmental parameters was calculated. An outbreak of concurrent Aspergillosis and Anatipestifer infection in Babary ducks was investigated. About 24.8 percent of 3,280 one-week-old babaries died, with lesions of focal pneumonia and multiple yellowish-white nodules, 1-4 mm in diameter, in the lung parenchyma. The lesions were also seen in the peritoneal cavity, brain tissue and bursa of Fabricius. Histological examination of lungs and brains revealed granulomas with giant cells. Mycelia elements were demonstrated in the granulomas by periodic acid schiff staining. Aspergillus fumigatus was cultured from the nodules and Pasteurella anatipestifer was isolated from lung tissue. Russell's viper venom 2 mg/kg bw was injected intramuscularly into rats. Renal functions after injection were studied at 1, 3, 6, 24, 72, 120 and 168 hours and compared with controls using normal saline. Within 6 hours of injection, all experimental rats produced oliguria, some anuria, some exhibited hemoglobinuria with severe bleeding from the nose and mouth and some were dead. After 6 hours all changes were less marked. At 24 hours, urine flow in the surviving rats increased without evidence of hemoglobinuria or severe bleeding. Two members of the genus Bartonella, Bartonella quintana (formerly Rochalimaea quintana) and Bartonella henselae (formerly Rochalimaea henselae), have recently been recognized as agents of severe or fatal disease in patients infected with human immunodeficiencyv irus (HIV). The development of infection with B. henselae in HIV-infected individuals has been associated with traumatic contact with cats (scratches or bites), and domestic cats have been identified as a major reservoir for this organism. Specific information regarding the transmission of B. henselae to humans is not yet available, but common-sense precautions that minimize exposure to cat-associated organisms are appropriate. Rabies is caused by a single-stranded, negative-sense RNA virus, maintained in nature by a variety of animal reservoirs. Rabies virus infects the central nervous system, resulting in progressive encephalopathy and ultimately death in an infected human. Globally, the risk of contracting rabies for humans is greatest in regions of the developing world where dog rabies is enzootic. Where rabies in dogs has been eliminated or otherwise controlled through vaccination programs, the disease can be maintained by wildlife. We review two groups of taxonomically unrelated viruses that share similarities in host preference and transmission routes to humans and pose a risk for mammalogists working with rodents. The rodent-borne hemorrhagic fever viruses in the Arenaviridae and Bunyaviridae are widely distributed on most continents where rodents occur. Their geographic distribution usually exceeds the distribution of the recognized human diseases they cause and has resulted from either natural coevolutionary events or the dissemination of viral passengers traveling with introduced mammalian hosts. Diseases of humans caused by these agents are among the most severe and most frequently fatal of zoonotic diseases. These …
