Michigan Bovine Tuberculosis Bibliography and Database
Bovine tuberculosis (bTB) is a contagious disease of livestock, wildlife, and humans. Typically, it is transmitted through inhalation of aerosolized bacilli and direct or indirect contact between animals. In northeastern Michigan, bTB is endemic in white-tailed deer, and evidence suggests deer have spread the disease to domestic cattle. Previous research indicates bTB transmission likely occurs through contamination of cattle feeding sources by infected deer and subsequent use by cattle. We are investigating deer movements in relation to farm management practices such as feeding schedules as well as locations of cattle feeding areas, hay storage sites, barns, and water sources. All …

Mitogen- and antigen-induced interferon-γ (IFN-γ) responses of peripheral blood leucocytes from cervids were evaluated by a commercial whole-blood assay. The assay was applied to Mycobacterium bovis-infected white-tailed deer and reindeer, M bovis BCG-vaccinated white-tailed deer and elk, and unvaccinated, uninfected white-tailed deer, fallow deer, elk and reindeer. The responses of the M bovis-infected white-tailed deer to pokeweed mitogen (PWM) varied with time and between individuals. The responses of the M bovis-infected reindeer to PWM and M bovis purified protein derivative (PPD) were positively associated. Samples from tuberculosis-free captive herds in various parts of the USA were also …

Bovine tuberculosis (TB) is endemic in white-tailed deer (Odocoileus virginianus) in the northeastern portion of Michigan’s Lower Peninsula. Bovine TB in deer and cattle has created immense fi nancial consequences for the livestock industry and hunting public. Surveillance identified coyotes (Canis latrans) as potential bio-accumulators of Mycobacterium bovis, a finding that generated interest in their potential to serve as sentinels for monitoring disease risk. We sampled 175 coyotes in the bovine TB–endemic area. Fifty-eight tested positive, and infection prevalence by county ranged from 19% to 52% (statistical mean 33%, SE 0.07). By contrast, prevalence in …

Computational models have been successfully applied to a wide variety of research areas including infectious disease epidemiology. Especially for questions that are difficult to examine in other ways, computational models have been used to extend the range of epidemiological issues that can be addressed, advance theoretical understanding of disease processes and help identify specific intervention strategies. We explore each of these contributions to epidemiology research through discussion and examples. We also describe in detail models for raccoon rabies and methicillin-resis-tant Staphylococcus aureus, drawn from our own research, to further illustrate the role of computation in epidemiological modeling.

Molecular Determinants Of Kaposi's Sarcoma-Associated Herpesvirus Tumorigenicity, Haixia Kong
LSU Master's Theses
A conditional silencing system using anti-gB siRNAs was devised to investigate the structure and function of the KSHV gB. Transient co-transfection of plasmids constitutively expressing gB and anti-gB siRNAs in 293 cells substantially inhibited gB mRNA levels and protein production. Similarly, transient expression of siRNAs into the basal cavity-based lymphoma cells (BCBL-1) caused substantial reduction of gB transcription and protein synthesis. TaqMan real-time PCR and infectivity assays showed that gB was essential for virion egress and infectivity. Transfection of a codon-optimized gB not recognized by the anti-gB siRNAs, efficiently rescued virion egress and infectivity in BCBL-1 cells in the presence …

Comparative Roles Of Herpes Simplex Virus Type 1 (Hsv-1) Glycoproteins In Cytoplasmic Virion Egress, Hyuncheol Lee
LSU Master's Theses
HSV-1 acquires its final envelope by budding into cytoplasmic vesicles thought to be derived from Trans-Golgi Network (TGN) membranes. This process is facilitated by interactions among the carboxyl termini of viral glycoproteins and tegument proteins. To investigate the relative importance of different viral glycoproteins in cytoplasmic virion morphogenesis, a set of recombinant viruses were constructed silencing expression of the glycoprotein E (ΔgE), the carboxyl terminus of glycoprotein D (gDΔcp), and the membrane protein UL20 (ΔUL20). In addition, recombinant viruses were constructed having the ΔgE+gDΔcp, and the ΔgE+ΔgM (glycoprotein M) deletions. These recombinant viruses were constructed using the double-red, site-directed mutagenesis …

Effects Of Fescue Herbicides Plateau® And Cimarron® On Pregnancy Maintenance In Broodmares And On Alkaloid Concentrations In Endophyte Infected Tall Fescue, Kathleen Scarlett Black
University of Kentucky Master's Theses
Ingestion of endophyte infected (E+) fescue by pregnant mares can cause significant reproductive problems. Plateau® and Cimarron® herbicides suppress fescue while leaving desired forages unharmed. To determine if these herbicides are harmful to pregnant mares, they were allowed to graze pastures treated with Plateau®, Cimarron®, or vehicle carrier. Pregnancies were monitored via ultrasonography, blood chemistry, and hematology. Of the components measured only creatinine differed among treatments over time (P=0.0003) and that increase was only significant in one of four studies.
Two additional experiments were conducted to determine the effect of the herbicides on alkaloids within E+ fescue. A greenhouse experiment …
The Zinc Induction Of Heme Oxygenase-1 Gene Promoter In Mouse Hepa Cells, Salim M. Alawneh
The Zinc Induction Of Heme Oxygenase-1 Gene Promoter In Mouse Hepa Cells, Salim M. Alawneh
LSU Doctoral Dissertations
Heme oxygenase-1 (HO-1) catalyzes the initial and rate-limiting reaction in heme catabolism (yielding equimolar quantities of iron, carbon monoxide, and biliverdin), which is further metabolized to bilirubin by biliverdin reductase. HO-1, an important enzyme with antioxidant and cytoprotective properties, functions against oxidative stress generated in response to heme and to various stress stimuli, such as zinc. The cytoprotective function of HO-1 is dependent on both the activities of the reaction products and on the inducibility of the heme oxygenase gene (hmox-1), which is regulated primarily at the level of gene transcription by two distal enhancers, E1 and E2. Characterization of …

Kaposi’s sarcoma-associated herpesvirus (KSHV) replication and transcription activator (RTA) is necessary and sufficient for the switch from KSHV latency to lytic replication. RTA activates promoters by several mechanisms. RTA can bind to sequences in viral promoters and activate transcription. In addition, RTA interacts with the cellular recombination signal sequence-binding protein-J kappa (RBP- Jκ), a transcriptional repressor, converts the repressor into an activator and activates viral promoters via RBP- Jκ. Because RBP- Jκ is required for RTA to activate lytic replication, it is important to understand how RTA cooperates with RBP- Jκ protein to activate KSHV lytic replication program. Previously, we …

Bovine herpesvirus type 1 (BHV-1) is an important pathogen that can initiate bovine respiratory disease complex. Like other members of the subfamily Alphaherpesvirinae, BHV-1 establishes latency in sensory neurons. The latency-related (LR) gene expresses a family of alternatively spliced transcripts in infected sensory neurons that have the potential to encode several LR proteins. An LR mutant virus that contains three stop codons near the 5’ terminus of the first open reading frame in the LR gene does not express two LR proteins or reactivate from latency. In addition, the LR mutant virus induces higher levels of apoptosis in trigeminal …

Sheep scrapie is the prototypical transmissible spongiform encephalopathy (prion disease), which has a fundamental pathogenesis involving conversion of normal cellular prion protein (PrPC [C superscript stands for cellular]) to disease-associated prion protein (PrPSc [Sc superscript stands for sheep scrapie]). Sheep microglial cell cultures, derived from a prnp 136VV/171QQ near-term fetal brain, were developed to study sheep scrapie in the natural host and to investigate potential cofactors in the prion conversion process. Two culture systems, a primary cell culture and a cell line transformed with the large T antigen of simian virus 40, were developed, and both were identified …

Bovine Herpesvirus Type 1 Induces Cell Death By A Cell Type Dependent Fashion, Vicki Geiser, Suzanne Rose, Clinton J. Jones
School of Veterinary and Biomedical Sciences: Faculty Publications
Bovine herpesvirus 1 (BHV-1), a member of the alpha-herpesvirinae subfamily, causes significant losses to the cattle industry. BHV-1 establishes latency in trigeminal ganglionic sensory neurons, but periodically reactivates from latency. Previous studies suggested that infection with BHV-1 induced novel morphological changes in rabbit skin cells (RS) versus bovine kidney cells (MDBK). Consequently, we hypothesized that viral infection led to a novel form of cell death in RS cells compared to MDBK cells. To test this hypothesis, we examined the levels of apoptosis in these cell types following infection with BHV-1. Infection of RS, but not MDBK, cells leads to high …

Infection of cattle by bovine herpesvirus type 1 (BHV-1) can lead to upper respiratory tract disorders, conjunctivitis, genital disorders and immune suppression. BHV-1-induced immune suppression initiates bovine respiratory disease complex (BRDC), which costs the US cattle industry approximately 3 billion dollars annually. BHV-1 encodes at least three proteins that can inhibit specific arms of the immune system: (i) bICPO inhibits interferon-dependent transcription, (ii) the UL41.5 protein inhibits CD8+ T-cell recognition of infected cells by preventing trafficking of viral peptides to the surface of the cells and (iii) glycoprotein G is a chemokine-binding protein that prevents homing of lymphocytes to …

In this study Mentofin^® was used in commercial broiler chickens naturally exposed to infectious bursal disease virus (IBDV) and administered live Newcastle disease virus (NDV) vaccine to evaluate the ability of Mentofin^® to reduce Escherichia coli-related respiratory lesions, immunomodulate Newcastle disease (ND) vaccine response, change pharyngeal aerobic bacterial counts, and have certain impacts on specific production parameters. Mentofin^® was added to the drinking water (100 ml/500 l water) of broiler chickens aged between 3 and 5 days. ND and IBD vaccinations were administered via drinking water to 15-day-old chickens. NDV vaccination was given again when the chickens were 20 days …

Presence of the chicken infectious anemia virus (CIAV) infection in chicken flocks in some provinces of Turkey was investigated by ELISA and PCR in this study. From 38 flocks (16 commercial layer, 10 commercial broilers, 4 breeder layers and 8 breeder broilers), 922 serum samples were tested for CIAV-specific antibodies using a commercially available competitive ELISA. CIAV antibodies were positive in 609 (66.0%) sera from 34 flocks (89.5%). A total of 95 thymus samples from 25 flocks, 57 samples from 15 commercial layers, and 38 samples from 10 commercial broiler flocks were tested by PCR. In 53 (55.8%) thymus samples …
