West Nile (WN) disease is one of the encephalitic diseases in humans and mammals. This disease has beenreported in several areas of the world. Thailand is located in the area of high risk of WN virus (WNv) infectionconcerning a natural transmission cycle. However, the WNv infection has not been previously reported. Therefore,the aim of this study was to detect WNv in mosquitoes in some areas in Nakhon-pathom and Phetchaburi provinces,central Thailand. To determine the seroprevalence of caprine arthritis-encephalitis virus infection (CAEV) in goats in theWestern part of Thailand, a cross-sectional serological survey was conducted in three provinces, Ratchaburi,Petchaburi and Kanchanaburi, situated in the western part of the country along the border area. A total of 1,129serum samples from 74 randomly selected goat farms containing different breeds of goats were collected during theperiod from November 2009 to January 2011. Seroprevalence of CAEV antibodies was determined using competitiveenzyme-linked immunosorbent assay (cELISA) test. Eleven multiparous crossbred lactating sows were used to determine the influence of deficient protein dietcontaining 8.2% crude protein and normal protein diet containing 18.2% crude protein on plasma amino acidconcentrations of sows and their piglets, physical mammary gland and histological changes of mammary cells. Theywere provided sufficient water and feed access to one of two isocaloric diets, either deficient (6 sows) or normalprotein diets (5 sows). The experimental period started from farrowing (day 0) until weaning day (day 28). The prevalence of Enterococcus faecalis was investigated in diseased swine in Hunan Province, China betweenFebruary 2007 and February 2011. A total of 385 diseased swine dissected in prosectorium from 10 representativeadministrative regions in Hunan Province were examined for the presence of E. faecalis. Forty-two out of 385 (10.91%)diseased swine were found to be infected with E. faecalis by bacteria isolations. E. faecalis was mainly recovered fromthe lung and joint cavity. Identification of the organisms was based on biochemical assays and sequencing of thepartial 16S rRNA. The origin of dogs came from the limited number of founders leading to inbreeding or genetic diseasewithin or among breeds. Thai Ridgeback (TR) and Bangkaew (BK) dogs were the only native dog breeds that webelieved to originate in Thailand. The study of genetic variability in Thai dogs (39 Thai Ridgeback dogs, 82 Bangkaewdogs) was defined by using 12 microsatellite markers. These DNA markers were highly informative and distributedthrough the canine genome. Immunoprecipitation and subsequent mass spectrometry analysis of the cellular proteins from cells expressing the vesicular stomatitis virus (VSV) P protein identified the poly(C) binding protein 2 (PCBP2) as one of the P protein-interacting proteins. To investigate the role of PCBP2 in the viral life cycle, we examined the effects of depletion or overexpression of this protein on VSV growth. Small interfering RNA-mediated silencing of PCBP2 promoted VSV replication. Conversely, overexpression of PCBP2 in transfected cells suppressed VSV growth. Further studies revealed that PCBP2 negatively regulates overall viral mRNA accumulation and subsequent genome replication. Acute fatty liver of pregnancy (AFLP), characterized by hepatic microvesicular steatosis, is a sudden catastrophic illness occurring almost exclusively in the third trimester of pregnancy. Defective fatty acid oxidation in the fetus has been shown to be associated with this disease. Since the placenta has the same genetic makeup as the fetus and as AFLP patients generally recover following delivery, we hypothesized that the placenta might be involved in pathogenesis of this disease. In an animal model of hepatic microvesicular steatosis (using sodium valproate), we found that microvesicular steatosis results in mitochondrial structural alterations and oxidative stress in subcellular organelles …

Microarray containing 13319 probes was used to construct gene expression profiles in order to screendifferentially expressed genes of muscle tissues in Rugao chicken and investigate the molecular mechanism relatedwith muscle tissue traits at 2 and 12 weeks old of chicken. Two hundred and eight differentially expressed genes, ofwhich 108 were known genes, 94 up-regulated, 114 down-regulated were involved in growth, molecular mechanism,fat metabolism, cell proliferation, transcription and splicing factor, protein synthesis and degradation. We studied the prevalence of swine salmonellosis and PFGE genotype of isolates in Gunma Prefecture,Japan. Between 2005 and 2008, swine salmonellosis was confirmed in 430 of 2,707,402 (0.02%) swine atslaughterhouses. All isolates were identified as deriving from Salmonella Choleraesuis, biotype Choleraesuis (negativefor H2S production). We used 30 bacterial strains from 15 farms that had experienced outbreaks in 2006 and 2007. Allstrains were susceptible to various antibiotics such as cephems (cefotaxime), fluoroquinolones (norfloxacin andciprofloxacin), and fosfomycin. On the other hand, all strains were resistant to tetracycline (TC), and 29 of 30 (97%)strains were resistant to streptomycin (SM). The most predominant profiles …

An experiment was conducted to investigate the effect of Roselle calyx in two preparation forms in layerdiets on egg production performance, egg quality and TBARS value in plasma and yolk. Two hundred and seventy,33-week-old, CP Brown laying hens were randomly allocated into 6 treatments with 5 replications of 9 hens each.The dietary treatments were control diet, control diet supplemented with 250 mg/kg α-tocopheryl acetate, dietcontaining 1 and 2% of Roselle calyx crude extracts, and diet containing 2% and 4% Roselle calyx powder. Background: Recurrent airway obstruction (RAO) is a severe chronic respiratory disease affecting horses worldwide, though mostly in the Northern hemisphere. Environmental as well as genetic factors strongly influence the course and prognosis of the disease. Research has been focused on characterization of immunologic factors contributing to inflammatory responses, on genetic linkage analysis, and, more recently, on proteomic analysis of airway secretions from affected horses. Porcine reproductive and respiratory syndrome virus (PRRSV) causes late-term abortion in sows and pneumonia in growing piglets. PRRSV evades the host immune response by several mechanisms, including the modulation of cytokine secretions in infected pigs, which is the subject of this dissertation. Particularly, PRRSV reduces the secretion of the pro-inflammatory cytokine, tumor necrosis factor-α (TNF-α) but increases the secretion of the anti-inflammatory cytokine interleukin-10 (IL-10). The latter effect, however, is PRRSV strain-specific. In this dissertation, we have examined mechanisms by which PRRSV regulates TNF-α and IL-10 expressions. Viruses usurp host cell pathways for different stages of their infection. Understanding virus-host interaction will be invaluable to elucidate molecular mechanisms of virus infection and to identify drug targets. In order to identify such critical cellular genes in vesicular stomatitis virus (VSV, a model non-segmented negative strand RNA virus) infection, we developed a stable cell line constitutively expressing replication proteins of VSV. Glutaredoxin 2 (Grx2) belongs to the oxidoreductase family and is an isozyme of glutaredoxin 1 (Grx1) present in the mitochondria, however its function is not well understood. The purpose of this study is to evaluate the potential anti-apoptotic function of Grx2 by examining its ability to protect complex I in the mitochondrial electron transport system using human lens epithelial cells as a model. We found that cells treated with 200 μM hydrogen peroxide (H2O2) for 24 h exhibited decreased viability and became apoptotic with corresponding Bax up-regulation, Bcl-2 down-regulation, caspase 3 activation and mitochondrial cytochrome c leakage. PURPOSE. To investigate the molecular mechanism for cytosolic phospholipase A2 (cPLA2) regulation and its association to platelet-derived growth factor (PDGF)-induced cell proliferation.

METHODS. cPLA2 was examined using human lens epithelial (HLE) B3 cells. Reactive oxygen species (ROS) generation induced by PDGF was analyzed by luminescence assay. Cell proliferation was measured by cell counting and by BrdU assay. Human cPLA2 gene was cloned via RT-PCR followed by sitedirected mutagenesis to construct HLE B3 cells expressing either inactive cPLA2 enzyme with S228A mutation (S228A), or cPLA2 truncated at the calcium-binding C2 domain (C2D). Canine genetic disorders inherited through genes or chromosomes. Mutations can also occur spontaneouslyor influenced by environmental factors. Such disorders can be transmitted to offspring. However, the disease may notbe developed owing to other influenced factors. Dominant and recessive genetic mechanisms also affect the traitoutcome. The study aimed to apply flow cytometry (FC) for early detection and monitor of treated response cats withIMHA by measuring immunoglobulin-G (IgG) and immunoglobulin-M (IgM) on red blood cells. Coombs' test and FCmethod were performed in twenty-nine cats including: ten clinically normal client-owned cats and nineteen anemiccats with packed cell volume of less than 27%. Five anemic cats were positive for Coombs' test and had significantlyhigh levels of IgG and IgM on the red blood cells with high level of mean fluorescene intensity (MFI) measured by theFC method. piRNAs are a testis-specific class of small RNAs in mammal. Recently, we reported the preferential infection of a novel glial cell type from adult canine brain, termedSchwann cell-like brain glia (SCBG) with canine distemper virus (CDV). To further characterize virus-cell and cell-cellinteractions, SCBG in purified and mixed brain cell cultures was infected with different CDV strains. Cell cultureswere infected with the attenuated CDV-Ond, CDV-2544, CDV-OndeGFP, CDV-R252 and the mustelid virulent CDV-5804PeGFP strain. Cytopathic effects (CPEs) and the percentage of infection were monitored daily at 3 and 10 dayspost-infection (dpi), respectively. Derivation of embryonic stem (ES) cells from parthenogenetic embryos represents a possible alternativeapproach to create histocompatible cells for regenerative medicine. The objectives of this study were to establishmouse parthenogenetic ES (pES) cell lines from parthenogenetically-derived blastocysts as a model system for humanand animal research and to examine pluripotency differences among the pES cell lines. We are able to report thesuccessful establishment of four pluripotent pES cell lines from blastocysts of parthenogenetic origin (22% efficiencyof pES cell line establishment). This study was conducted from 2004 to 2009 to investigate the prevalence of Listeria monocytogenes in thechicken production chain in Thailand from primary production stages to processing plants. A total of 14,670 sampleswere taken from 43 breeder farms, 32 hatcheries, 1331 broiler farms, 22 slaughterhouses and 22 ready-to-eat (RTE)chicken products from processing plants. Various types of samples were collected: soiled litter, water, and chickenfeed from breeder and broiler farms; swabs and mecomium from hatcheries; cloacal swabs and fresh frozen chickenmeat from slaughterhouses; and RTE chicken products from processing plants.
