Differential Expression Patterns Of Proteins Involved In Epidermal Proliferation And Differentiation In Canine Atopic Dermatitis, Sirin Theerawatanasirikul, Achariya Sailasuta, Roongroje Thanawongnuwech, Tossaporn Nakbed, Komkrid Charngkaew, Gunnaporn Suriyaphol

The Thai Journal of Veterinary Medicine

The pathological alterations in skin keratinocyte proliferation and differentiation in canine atopic dermatitis(CAD) were investigated in 20 small breed dogs with CAD (10 of which displayed lesional CAD skin and 10 nonlesionalCAD skin) and 11 healthy control animals. Biopsy of lesional CAD skin showed orthokeratotichyperkeratosis. Immunohistochemistry (IHC) of Ki-67, a cellular marker for proliferation, showed a higher epidermalcell proliferation rate in the CAD dogs (p< 0.05) which positively correlated with Canine Atopic Dermatitis Extent andSeverity Index, CADESI-03 (p< 0.01). IHC labeling indicated reduced expression of cornified envelope proteins, involucrin (IVL) and filaggrin (FLG) in both lesional and non-lesional skin (p< 0.05). In contrast, expression oflympho-epithelial Kazal-type inhibitor (LEKTI), which has been reported to inhibit proteases that cleave proFLG intoFLG, was increased in lesional skin compared to the normal controls (p< 0.05). The IHC results showed a positivecorrelation between CADESI-03 and LEKTI expression and a negative correlation between CADESI-03 and IVLexpression. In conclusion, the present study demonstrates that epidermal hyperkeratosis in CAD is related todisturbance of both epidermal proliferation and cornified envelope differentiation. This is the first report of acorrelation of LEKTI with CAD.

Department of Animal Science: Faculty Publications

Locomotion in mammals relies on a central pattern-generating circuitry of spinal interneurons established during development that coordinates limb movement. These networks produce left–right alternation of limbs as well as coordinated activation of flexor and extensor muscles. Here we show that a premature stop codon in the DMRT3 gene has a major effect on the pattern of locomotion in horses. The mutation is permissive for the ability to perform alternate gaits and has a favorable effect on harness racing performance. Examination of wild-type and Dmrt3-null mice demonstrates that Dmrt3 is expressed in the dI6 subdivision of spinal cord neurons, takes …

Development Of Repressible Systems To Control Gene Expression In Vaccinia Virus, Allison Titong Aug 2012

Development Of Repressible Systems To Control Gene Expression In Vaccinia Virus, Allison Titong

Master's Theses

Two vaccinia virus (VACV) expression systems that contain elements from the lactose (lac) and the tetracycline (tet) operons of E. coli were developed to repress the expression of a reporter gene, enhanced green fluorescent protein (EGFP), in the presence of tet operon inducers. In the first system, lac and tet operon elements were arranged in a gene circuit, and in the presence of increasing concentrations of a lac operon inducer (isopropyl-β-D-thiogalactoside, IPTG), EGFP expression increased in a dose dependent manner and at high IPTG concentrations, expression reached the same levels as a positive control virus. Importantly, in the …

Expression And Distribution Of Thiol-Regulating Enzyme, Glutaredoxin 2 In Porcine Ocular Tissues, Bijaya Prasad Upadhyaya

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Glutaredoxin 2 (Grx2), a thiol-regulating enzyme of oxidoreductase family and a mitochondrial isozyme of glutaredoxin 1, was discovered 11 years ago in our laboratory. Grx2 is present in the lens where it shows dethiolase, peroxidase, and ascorbate recycling activities. Recently, Grx2 has also been identified to protect the mitochondrial electron transport system with anti-apoptotic function. Since other eye tissues besides the lens are rich in mitochondria and are very sensitive to oxidative stress, we speculate the presence of Grx2 therein as an important redox regulator. This study is to investigate the expression and distribution of Grx2 in ocular tissues using …

Melanopsin Mediates Retrograde Visual Signaling In The Retina, Dao-Qi Zhang, Michael A. Belenky, Patricia J. Sollars, Gary E. Pickard, Douglas G. Mcmahon Aug 2012

Melanopsin Mediates Retrograde Visual Signaling In The Retina, Dao-Qi Zhang, Michael A. Belenky, Patricia J. Sollars, Gary E. Pickard, Douglas G. Mcmahon

School of Veterinary and Biomedical Sciences: Faculty Publications

The canonical flow of visual signals proceeds from outer to inner retina (photoreceptors→bipolar cells→ganglion cells). However, melanopsin-expressing ganglion cells are photosensitive and functional sustained light signaling to retinal dopaminergic interneurons persists in the absence of rods and cones. Here we show that the sustained-type light response of retinal dopamine neurons requires melanopsin and that the response is mediated by AMPA-type glutamate receptors, defining a retrograde retinal visual signaling pathway that fully reverses the usual flow of light signals in retinal circuits.

Development Of One-Step Taqman® Real-Time Reverse Transcription-Pcr And Conventional Reverse Transcription-Pcr Assays For The Detection Of Equine Rhinitis A And B Viruses, Zhengchun Lu, Peter J. Timoney, Jena White, Udeni B R Balasuriya

Veterinary Science Faculty Publications

BACKGROUND: Equine rhinitis viruses A and B (ERAV and ERBV) are common equine respiratory viruses belonging to the family Picornaviridae. Sero-surveillance studies have shown that these two viral infections are prevalent in many countries. Currently, the diagnosis of ERAV and ERBV infections in horses is mainly based on virus isolation (VI). However, the sensitivity of VI testing varies between laboratories due to inefficient viral growth in cell culture and lack of cytopathic effect. Therefore, the objective of this study was to develop molecular diagnostic assays (real-time RT-PCR [rRT-PCR] and conventional RT-PCR [cRT-PCR] assays) to detect and distinguish ERAV from ERBV …

Duplex Pcr For Simultaneous And Unambiguous Detection Of Streptococcus Iniae And Streptococcus Agalactiae Associated With Streptococcosis Of Cultured Tilapia In Thailand, Channarong Rodkhum, Pattanapon Kayansamruaj, Nopadon Pirarat, Janenuj Wongtawatchai

The Thai Journal of Veterinary Medicine

Warm-water streptococcosis outbreaks in Thai cultured tilapia are caused by Streptococcus iniae and S.agalactiae. However, distinguishing between these two species of streptococcus are very difficult due to their similarmicrobiological appearance and clinical signs of infected fish. In this study, we proposed a new duplex-PCR basedmethod for simultaneous detection of these pathogens. The duplex-PCR amplified partial lctO and 16s rRNA gene ofS. iniae and S. agalactiae at 870 bp and 220 bp, respectively. This technique gave 100% specificity while sensitivity ofreaction was 100 fg of each bacterial genomic DNA. Detection limit of duplex-PCR applicable to clinical specimens isalso evaluated as 106 …
