Somatostatin (SST) is a neuropeptide hormone that regulates the release of secondary hormones. Evidence suggests SST plays a neuromodulatory role due to its distribution throughout the central nervous system. Interestingly, SST has been suggested to affect the carotid body, the small peripheral chemoreceptors that regulate breathing. It has been shown that the peripheral chemoreflex sensitivity to CO2 and hypoxia is reduced by SST in humans (Pedersen et al., 1999; Pandit et al., 2014). Hypoxic events encountered during the perinatal period leading to pathologic decreases in fetal cerebral oxygen availability are detrimental resulting in neonatal hypoxic-ischemic encephalopathy (HIE). This work focused on the characterization of a rat model of neonatal HIE for use in the preclinical evaluation of infrared light therapy (IRL). Consistent with results from previous IRL efficacy studies using a rat model of adult global brain ischemia, IRL treatment attenuated cerebral injury resulting from hypoxia-ischemia in the neonatal rat. The purpose of this paper is to investigate the adaptations that occur in human skeletal muscle in response to endurance and resistance exercises. The advances made in science over the past several decades increased the number of methods available for the classification of muscle fibers, resulting in many fiber type classes and their corresponding characteristics. This allows for the tracking of changes that occur within the muscle fibers. The heterogeneous collection of fiber types found within a muscle allows for its dynamic nature. Prostate cancer development is uniquely dependent on the androgen receptorâ??s (ARâ??s) transcriptional regulation in response to hormone binding. Current therapies directly target AR acting as an antagonist at androgen binding sites. However, once androgen-dependence is lost, meaning the patient has progressed into a late-stage hormone resistant phenotype, all current treatments are essentially ineffective. Utilizing the yeast strain Saccharomyces cerevisiae we are capable of creating a model system that allows for the exogenous expression of AR while still retaining the chaperone components needed for this steroid receptor complex. Direct-developing frogs lack, wholly or in part, a wide range of larval features found in metamorphosing species and form adult-specific features precociously, during embryogenesis. Most information on thyroid regulation of direct development relies on hormone manipulations; the ontogeny of many thyroid axis components has not been fully described. This analysis examines differentiation of the median eminence of the hypothalamus and production of thyroid-stimulating hormone (TSH) by the pituitary of the direct-developing frog Eleutherodactylus coqui. The mammalian/mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway appears to be the primary regulator of protein synthesis and growth. While resistance exercise paradigms are often divided into hypertrophy (HYP) and strength (STR) protocols, it is unknown whether these protocols differentially stimulate mTORC1 signaling. The purpose of this study was to examine mTORC1 signaling in conjunction with circulating hormone concentrations following a typical lower-body HYP and STR resistance exercise protocol. Ten resistance-trained men (24.7±3.4y; 90.1±11.3kg; 176.0±4.9cm) performed each resistance exercise protocol in a random, counterbalanced order. The successful regeneration of muscle tissue is dependent upon the infiltration of phagocytic CD14++CD16- monocytes that support the proliferation and differentiation of myogenic precursor cells. Physiologically, the magnitude of the cellular response following resistance exercise is dictated by the level of receptor expression on the plasma membrane of the monocyte, as well as the secretion of their cognate ligands from tissue resident cells. Spinal manipulative therapy (SMT) is a therapeutic procedure employed by various healthcare practitioners for alleviating acute and chronic musculoskeletal complaints. This form of treatment is also delivered to enhance the performance and augment the rehabilitation of athletes. However, despite research findings alleging the strength-modulating effects of SMT alongside numerous professional athletes’ positive anecdotal claims concerning its results, the physiological processes to explain its effects remain largely unexplained. To investigate further into the mechanism in which these defects are obtained, local brain insulin-like growth factor 1 (Igf1) was investigated in relation to the hypothyroid brain. There are many similarities between hypothyroidism and the Igf1 deficient brain. Additionally, TH (thyroid hormone) is known to interact with Igf1. Many mammalian species live in complex social groups that profoundly affect the behavior and ecology of their members. In ring-tailed lemurs (Lemur catta) these groups are characterized by female dominance, where females have priority over males for food, grooming, and space. Among females there is also a linear dominance hierarchy. L. catta have a strong matrilineal social core due to females staying within their natal group and males dispersing at sexual maturity. Daughters “inherit” their mother’s social rank. Vagal tone, a measure of parasympathetic activity via the vagus nerve, is known to be associated with positive emotion because it promotes social engagement and self-soothing behavior. Heart rate variability (HRV), especially high frequency oscillation, is a direct measure of vagal tone, and has been used in previous studies to test the correlation between vagal tone and positive emotion. The purpose of this study was to determine physiological differences in drought response among Garry oak (Quercus garryana Douglas ex Hook. [Fagaceae]) seedlings that grow in distinct populations in Washington state, in order to aid in restoration efforts by informing growers of the potential differences between the acorn collection sites. Acorns from six Washington populations east of the Cascades, as well as one population from Whidbey Island, were collected and grown in containers under controlled conditions. The impact of restraint stress on changes in cytokine expression and oxidative stress markers in hippocampal (Hipp) and hypothalamic (Hyp) regions of the brain, were assessed in a rat model. In addition, immune and inflammatory variables were assessed in peripheral blood. Male Wistar rats were acutely (30 min) or repeatedly (30 min/d for 14 consecutive days) exposed to a restraint stress, or maintained as non-stressed controls. In order to determine if mutations made to phospholemman (PLM) could increase PLM binding to the Na/K-ATPase (NKA) and cause positive inotropy we performed scanning mutagenesis of the transmembrane domain of PLM and measured FRET between each mutant and NKA. We observed increased binding to NKA for several PLM mutants compared to WT, including L27A, L30A, and I32A. In isolated cardiomyocytes, overexpression of WT PLM increased the amplitude of the Ca2+ transient compared to GFP control. Ca2+ transient amplitude was further increased by L30A PLM overexpression. L30A mutation also delayed Ca2+ extrusion and increased the duration of the cardiomyocyte contraction. …

As the central pacemaker, the suprachiasmatic nucleus (SCN) has long been considered the primary regulator of blood pressure circadian rhythm; however, this dogma has been challenged by the discovery that each of the clock genes present in the SCN is also expressed and functions in peripheral tissues. The involvement and contribution of these peripheral clock genes in the circadian rhythm of blood pressure remains uncertain. There is currently no direct evidence that chemotherapy induced peripheral neuropathy (CIPN) necessarily explains the sensorimotor deficits seen in 90% patients treated with oxaliplatin (OX). Some patients develop sensory symptoms without CIPN. Our laboratory reported abnormal signaling from IA afferents in OX treated rats with no evidence of neuropathy. We hypothesized that in the absence of CIPN, the behavioral disability is associated with impaired sensory encoding in OX treated rats. Malignant transformation is the process by which cells develop cancer properties. While many causes for malignant transformation are known (i.e. common genetic mutations and/or exposure to toxins or viruses), the basic requirements that allow a cell to stay alive with altered nutrient and energy requirements are just now being studied. In some tumor types malignant cells undergo changes that result in metabolic differences compared to normal cells. Circadian rhythms play crucial roles in orchestrating diverse physiological processes that are critical for health and disease. Cyclic AMP responsive element binding protein 3-like 3 (CREB3L3, also known as CREBH) is a liver-enriched, endoplasmic reticulum (ER)-tethered transcription factor known to regulate hepatic acute-phase response and energy homeostasis under stress conditions. Here, we demonstrate that CREBH is regulated by the circadian clock and functions as a diurnal regulator of hepatic lipid and glucose metabolism. CREBH is required to maintain circadian profiles of blood triglycerides, fatty acids, and glucose as well as hepatic glycogen storage. CREBH rhythmically regulates expression levels and amplitudes …

The placenta is vital for the short- and long-term health of the fetus, and significantly impacts the health of the mother. During the first and second trimesters of pregnancy, extravillous trophoblast (EVT) cells invade the uterus and remodel the maternal spiral arteries, which, if inadequate, leads to pregnancy complications, including early pregnancy loss (EPL), preeclampsia (PE), and intra-uterine growth restriction (IUGR). EVT migration into the uterine wall is dependent on growth factors and cytokines that signal between maternal and fetal tissues. We investigated the acute and chronic effects of resveratrol on renal function and blood pressure. We hypothesized that resveratrol would act as a renal vasodilator through a nitric oxide-dependent mechanism. In our acute studies, we found an intravenous bolus of resveratrol influenced changes in renal hemodynamics by increasing renal blood flow and decreasing renal vascular resistance. The mechanism of renal vasodilation was nitric oxide dependent and through a reduction of endogenous reactive oxygen species. Resveratrol-induced renal vasodilation was not influenced by COX metabolism and vasodilatory prostanoids. We found with continuous intravenous resveratrol infusion induced significant renal vasodilation while not altering …

The purpose of this study was to determine if pilots are at risk for computer vision syndrome (CVS) as computer display technology becomes commonplace in modern cockpits and simulators. A review of computer vision syndrome respect to causes and effects is provided. Display technology used in aircraft and simulators are outlined. Ergonomics and human factors for pilots in aircraft and in simulators are discussed in relation to video displays. Paddlefish often frequent oxbows and backwater habitats between periods of migration. Speculation as to the use and energetic savings associated with this seasonal residence has not been substantiated. This study used radio telemetry to track the seasonal movements and swim tunnel respirometry to estimate metabolic rates and swimming efficiency of paddlefish within an oxbow located just north of Clarksdale, Mississippi (Moon Lake). Paddlefish overwintered in the deepest portions of Moon Lake with little movement into shallower areas. Paddlefish swimming performance was measured at four swimming speeds (76.04, 91.27, 106.50, and 121.72 cm/s) to determine the cost of locomotion over season …

Dogs admitted to animal shelters experience psychological stressors resulting in elevated plasma cortisol. We previously found 30 min of human interaction reduced this response. The present study further characterized this effect, with the aim of developing a practical means of reducing stress of shelter dogs. We found that a second day of 30 min of petting reduced cortisol levels as effectively as the first. Further, 15 min of this interaction was as effective as 30 min. During petting, signs of excitation (vocalizations) and anxiety (panting) as well as escaped attempts were reduced, and social solicitation (tail-wagging) increased. However, cortisol levels …

Transcranial Direct Current Stimulation (tDCS) has been used to treat various mental and neurological illnesses. Rodent models have been used to examine physiological changes in the brain after tDCS, as well as to develop safety standards. However, most animal tDCS studies implant an electrode on the brain, potentially altering the path of current during stimulation. Sleep deprivation causes many adverse effects on work performance. Many experiments in both human and rodent models reveal detriments that sleep deprivation has on learning and memory, including performance in a water radial arm maze (WRAM) task. This study utilizes the modified multiple platform method (MMPM) of sleep deprivation; rats were sleep deprived in order to study memory errors they may make during the WRAM task. The findings indicate that 6 hours of sleep deprivation for 2 five-day week periods did not affect performance in the WRAM task except on the initial day compared to the large platform group. The …

Available data suggest that snake hemoglobins (Hbs) are characterized by a combination of unusual structural and functional properties relative to the Hbs of other amniote vertebrates, including oxygenation-linked tetramer-dimer dissociation. However, standardized comparative data are lacking for snake Hbs, and the Hb isoform composition of snake red blood cells has not been systematically characterized. Here we present the results of an integrated analysis of snake Hbs and the underlying α- and β-type globin genes to characterize 1) Hb isoform composition of definitive erythrocytes, and 2) the oxygenation properties of isolated isoforms as well as composite hemolysates. This project study addressed the problem of alcohol and binge drinking at a local rural college campus in the Northeast United States and the lack of an effective long-term academic intervention program to address the problem. The purpose of this research study was to determine the prevalence of the problem of the alcohol abuse problem and to develop a long-term program that would respond to the problem of repeat alcohol offenders.
