Emerging evidence indicates that around 80-100% of children with autism spectrum disorder (ASD) have motor impairments in gross motor skills, strength, and endurance. In the current study, which is a multisite collaboration with the physical therapy programs at the University of Connecticut and the University of Delaware, we are assessing the effects of two types of movement-based interventions compared to a standard-of-care seated play control intervention on motor performance in children with ASD. Depression is a debilitating disorder that can cause motivational deficits such as psychomotor retardation, anergia, apathy, and fatigue. Recent research indicates that these motivational deficits, and potential pathways of therapeutic intervention, can be studied in animal models involving rats and mice. Treatments with the VMAT-2 inhibitor tetrabenazine (TBZ) and cytokine interleukin-1ß (IL-1ß) can create a low-effort bias and impair effort-related motivation (Nunes et al. 2013, 2014). A number of high-affinity DA transport inhibitors such as d-amphetamine, methylphenidate, and cocaine can restore extracellular DA, albeit with the cost of undesirable effects such as high abuse liability. Self-regulation is the cognitive process of controlling our thoughts and behaviors to be aligned with our goals. This process is used in many different contexts and has been associated with contributions from several brain regions. This research aimed to investigate differences in four prefrontal areas of the brain while participants applied four different self-regulation strategies. We recorded EEG while participants (N = 132) performed three tasks which engaged each of the four self-regulation strategies: the AX-CPT task engaged proactive and reactive control, the Go/Nogo task engaged inhibitory control, and the hybrid Flanker Global/Local task engaged the resolution of response conflict. …

Ongoing (apparently spontaneous) pain at rest is a major complaint of patients suffering from many forms of acute and chronic pain, including acute and persistent postsurgical pain. Accumulating evidence suggests ongoing activity in nociceptors is a major driver of ongoing pain. Ongoing activity can be generated in sensory neurons in the absence of sensory generator potentials if one or more of three neurophysiological alterations occur – prolonged depolarization of resting membrane potential (RMP), hyperpolarization of action potential (AP) threshold, and/or increased amplitude of depolarizing spontaneous fluctuations of membrane potential (DSFs) to bridge the gap between RMP and AP threshold. The human capability for fluent speech profoundly directs inter-personal communication and, by extension, self-expression. Language is lost in millions of people each year due to trauma, stroke, neurodegeneration, and neoplasms with devastating impact to social interaction and quality of life. The following investigations were designed to elucidate the neurobiological foundation of speech production, building towards a universal cognitive model of language in the brain. Understanding the dynamical mechanisms supporting cortical network behavior will significantly advance the understanding of how both focal and disconnection injuries yield neurological deficits, informing the development of therapeutic approaches.

As cancer research continues to move towards more personalized methods, targeted inhibitors of oncogenic drivers of cancer are found to be an innovative and promising therapeutic approach. Previously thought to be undruggable regions of the genome, RNA has become an area of increasing interest in cancer due to the identification of more targetable regions and their relationship to tumor growth and progression. This work reports on the development of IACS-13743, a novel small molecule found to directly bind and inhibit microRNA-10b. Automaticity --- the ability to perform a task with directing attentional resources to its completion --- is commonly reduced among individuals with neurodegenerative diseases. These automaticity deficits result in impaired functional and daily activities and are sensitive to subtle, subclinical impairments. However, current measurement of automaticity by dual task paradigms is methodologically limited. In order to gain insight into the current state of the literature regarding cognitive-motor interference in symptomatic and prodromal neurodegenerative disease, the author of this dissertation conducted a scoping review (Chapter 1). Migraine is a neurological disease marked by recurrent headache and migraine attacks. Migraine is one of the most common diseases in the world, and as a result of high prevalence and symptoms, migraine is also quite disabling. One-third of people who have migraine experience aura, and these individuals have a greater risk for stroke, psychiatric comorbidities, and suicide attempts. Depressed short-term memory (STM) abilities during non-adaptive times of the day can significantly impact those who work occupations that require peak levels of cognitive functioning around the clock. Retrieval Induced Forgetting (RIF) is a type of active forgetting that may play beneficial and detrimental roles in long-term memory. The benefit of the retrieval of certain information is that information will become more readily available following subsequent retrieval; a concept termed the retrieval practice effect (RP). The detrimental effect of RIF may be that, upon the subsequent recall of certain information, related information may be inhibited from recall.
