Development of new barley cultivars in Western Australia has substantially improved the grain yield and quality of the State's barley crop. This has increased returns to barley growers and enhanced Western Australia's reputation in export markets as a major supplier of barley for malting and feed.

The State exports more than 200, 000 t of malting barley, 60, 000 t of barley malt and 150, 000 t of feed barley each year.

The major challenge for the future is to develop new, high yielding cultivars with improved malting quality.

Sequences are given for nine complete genes and one partial mitochondrial tRNA gene of the black fly, Simulium vittatum (Zetterstedt). Sequenced tRNA genes were for alanine, arginine, asparagine, aspartic acid, glutamic acid, glycine, leucine(CUN), lysine, serine(AGN), and valine. Nucleotides were aligned with the same previously sequenced genes in Aedes albopictus Skuse and Drosophila yakuba Burla. A cluster of six tRNA genes, which differ in arrangement in Ae. albopictus and D. yakuba, was amplified by PCR and found to have the same position and orientation in S. vittatum as in D. yakuba. Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year‐on‐year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. The aim of the Department of Agriculture's long-term beef cattle selection experiment at Wokalup is to examine the effects of selection for faster-growing cattle on biological traits that may influence the productivity of a commercial beef herd. Selection lines were established from Hereford and multibreed cattle in 1977. Twelve years of selection have produced an increase in growth rate to weaning of about 120 g per day in both lines. About 40 per cent of this increase is the result of permanent genetic improvement.

A Temperature-Sensitive Mutant Of Escherichia Coli Affected In The Alpha Subunit Of Rna Polymerase, Majid Mehrpouyan

A temperature-sensitive mutant of Escherichia coli affected in the alpha subunit of RNA polymerase has been investigated. Gene mapping and complementation experiments placed the mutation to temperature-sensitivity within the alpha operon at 72 min on the bacterial chromosome. The rate of RNA synthesis in vivo and the accumulation of ribosomal RNA were significantly reduced in the mutant at 44$\sp\circ$C. The thermostability at 44$\sp\circ$C of the purified holoenzyme from mutant cells was about 20% of that of the normal enzyme. Assays with T7 DNA as a template showed that the fraction of active enzyme competent for transcription was reduced as a …

The present thesis work contributes to two areas of molecular genetics, namely the development of methods for identifying single nucleotides and the understanding of the mechanism by which antisense oligonucleotides inhibit eukaryotic gene expression. Three single nucleotide detection methods are presented. The first method utilizes hybridization with synthetic oligonucleotide probes to detect single or multiple nucleotide differences between alleles in genomic DNA without electrophoretic separation. Total genomic DNA (either digested or undigested with restriction endonucleases) was immobilized in depressions in an agarose gel (in situ dots) and hybridized with radiolabeled, allele-specific oligonucleotide probes. When commercial wool growers select Merino rams they often use the following pattern: the buyers first consider the main strains of Merinos in Western Australia - Bungaree, Collinsville and Peppin. They then study the 550 studs and numerous non-stud ram breeders in this State. Having chosen a breeder, they examine the rams on offer and buy their annual requirements from among these. I have studied 26 children who have a de novo rearrangement of the acrocentric chromosomes in order to understand the formation of these aberrations. The families include '25 probands ascertained for Robertsonian-type translocations, 13 between nonhomologous chromosomes and 12 between homologs, and one rea(21;21)(q22;q22). The parental origins of the de novo rearrangements were determined in 26/26 families using QFQ and NOR variants and/or RFLP analyses. While there was no overall difference in the sex distribution of the parents of origin, there were more maternally derived nonhomologous ("true" Robertsonian) translocations (8 mat: 5 pat) and more paternally derived homologous rearrangements (4 …

There are significant differences if the distribution of weedy characteristics among weeds, normal plants, and crops. The world’s most serious weeds possess on the average 10 or 11 of these characters, a random collection of British plants have an average seven of the traits, and crop plants only five. For the average crop to become as “weedy” as the average weed, it would need to acquire five weedy traits. Even using the unlikely assumption that those traits are single loci in which a dominant mutation would provide the weedy character, this would require the simultaneous acquisition of five gene substitutions. …

After mating, hypotrichous ciliated protozoa transform a set of their micronuclear chromosomes into thousands of short, linear DNA molecules that form the macronuclear genome. To examine micronuclear genome organization in the hypotrich Euplotes crassus, we have analyzed two cloned segments of micronuclear DNA as well as the macronuclear DNA molecules that are derived from them. E. crassus was found to display a number of features characteristic of other hypotrich genomes, including (i) clustering and close spacing of the precursors of macronuclear DNA molecules, (ii) the frequent occurrence of internal eliminated sequences within macronuclear precursors, (iii) overlapping macronuclear precursors, (iv) lack …

A novel 0.6 kb LINE family in human DNA, designated L2Hs, has been described (Musich and Dykes 1986). Studies employing clone N6.4, containing three 0.6 kb segments of this family, indicate that these sequences are interspersed and moderately repetitive. Two additional variant sequences of the L2Hs family, N6.1 and N6.3, have been identified. Restriction mapping of each cloned segment indicates similarities among N6.4, N6.3 and N6.1. When the cloned DNAs were cleaved with restriction enzymes and subjected to cross-hybridization, each cloned insert produced a pattern indicating that the sequences contained in N6.1 and N6.3 are represented in at least one …

The mitochondrial DNA restriction fragment patterns of killer whales (Orcinus orca) were investigated in order to determine the level of genetic differentiation that exists between killer whales from various geographic locations. Twenty one killer whales were examined, seventeen of which were captive killer whales that originated from the North Atlantic and Northeast Pacific Oceans. Two were captive-born animals and two were killer whales that stranded along the Northeast Pacific coast.

DNA was extracted from blood and/or tissue samples, cleaved with a variety of restriction endonucleases and the DNA fragments were separated by horizontal agarose gel electrophoresis. The DNA …

The total catch of about 12.1 million kilograms for the 1987/88 season, based on processor returns, was an increase of 44% on the 1986/87 season. This very high catch, the third highest on record, resulted from an above average perulus settlement in 1983/84 and a "flow-on" of fast growing animals stemming from the expected high recruitment in 1988/990 caught as "reds" in 1987/88.

The Committee has identified five major elements related to the state of the rock lobster fishery upon which management advice is considered.

Field peas have grown dramatically in popularity in western Australia in the past five years, with the planted area increasing from a mere few hundred hectares to about 70,000 ha in 1988. This growth may continue, as more than 700,000 ha of agricultural land is potentially suitable for growing field peas.

However, any such growth of the pea industry will require new cultivars with improved yield, adaptation and quality characteristics to meet local and export demands. The Department of agriculture therefore decided to start a selection and breeding programme to support the pea industry.

Following the sexual cycle, hypotrichous ciliated protozoans fragment a set of their micronuclear chromosomes to generate the thousands of short, linear DNA molecules present in the transcriptionally active macronucleus. We have used a hybrid selection procedure to examine macronuclear DNA molecules for subtelomeric length heterogeneity to determine whether chromosome fragmentation occurs at unique or multiple sites. The results suggest that multiple, but closely spaced, chromosome fragmentation sites are used by Oxytricha nova. In contrast, Euplotes crassus uses unique chromosome fragmentation sites in a reproducible manner to generate the ends of macronuclear DNA molecules. Additional studies compared DNA sequences in …

The interpretation of data on genetic variation with regard to the relative roles of different evolutionary factors that produce and maintain genetic variation depends critically on our assumptions concerning effective population size and the level of migration between neighboring populations. In humans, recent population growth and movements of specific ethnic groups across wide geographic areas mean that any theory based on assumptions of constant population size and absence of substructure is generally untenable. We have isolated ribosomal RNA gene (rDNA) chromatin from Physarum polycephalum using a nucleolar isolation procedure that minimizes protein loss from chromatin and, subsequently, either agarose gel electrophoresis or metrizamide gradient centrifugation to purify this chromatin fraction (Amero, S. A., Ogle, R. C., Keating, J. L., Montoya, V. L., Murdoch, W. L., and Grainger, R. M. (1988) J. Biol. Chem. 263, 10725-10733). Metrizamide-purified rDNA chromatin obtained from nucleoli isolated according to the new procedure has a core histone/DNA ratio of 0.77:1. The major core histone classes comigrate electrophoretically with their nuclear counterparts on Triton-acid-urea/sodium dodecyl sulfate two-dimensional gels, although they …

Recent studies indicate that polymorphic genetic markers are potentially helpful in resolving genealogical relationships among individuals in a natural population. Genetic data provide opportunities for paternity exclusion when genotypic incompatibilities are observed among individuals, and the present investigation examines the resolving power of genetic markers in unambiguous positive determination of paternity. Under the assumption that the mother for each offspring in a population is unambiguously known, an analytical expression for the fraction of males excluded from paternity is derived for the case where males and females may be derived from two different gene pools. The effects of density and food on remating were investigated using Drosophila melanogaster. The frequency of remating was unaffected by density for some combinations of fly strains but was reduced at low relative densities for other combinations. Until females had used most of their stored sperm, remating was less likely when food was absent or contact with food was prevented. Food availability had little effect on the incidence of remating once stored sperm were depleted and had no effect on initial virgin mating frequency. Bats of the genus Lasiurus present a number of interesting systematic problems that are difficult to resolve by traditional techniques. Members of the genus share a suite of derived morphological (Hall and Jones, 1961; Handley , 1960) and karyotypic (Bickham 1979, 1988) characteristics. However, until 1960 (Handley, 1960), members were placed in two genera- Lasiurus and Dasypterus-based primarily upon the presence or absence of the small, first upper premolar. We have undertaken the purification of ribosomal RNA gene (rDNA) chromatin from the slime mold Physarum polycephalum, in order to study its chromatin structure. In this organism rDNA exists in nucleoli as highly repeated minichromosomes, and one can obtain crude chromatin fractions highly enriched in rDNA from isolated nucleoli. We first developed a nucleolar isolation method utilizing polyamines as stabilization agents that results in a chromatin fraction containing far more protein than is obtained by the more commonly used divalent cation isolation methods. The Yellow-shafted Flicker (Colaptes auratus auratus) and Red-shafted Flicker (C. a. cafer) form a stable, narrow hybrid zone on the western Great Plains of North America. Allozyme data were obtained from 31 structural gene loci for 33 samples representing 246 Northern Flickers from throughout the Great Plains. Flickers were approximately equivalent to other birds in terms of proportion of polymorphic loci (P = 0.207) and average heterozygosity (H = 0.056). There was no concordant variation between plumage characters and allelic frequencies.
