Variously aged (C57BL X C3H)F1 hybrid mice were used as recipients in experiments to determine if spleen or bone marrow cells from normal C57BL or C3H donors, spleen or bone marrow cells from C57BL donors previously treated with cortisone, or C57BL bone marrow cells previously incubated with serum from X-irradiated C57BL mice could induce a graft-versus-host reaction (GVHR).
The mice were assayed for GVHR by mortality or splenomegaly as detected by the discriminant spleen assay developed by Simonsen. Results show that spleen cells from normal C57BL, normal C3H, or cortisone-treated C57BL mice are capable of inducing a GVHR in …

The purpose of this research was to investigate the effect of selenium upon methyl mercury poisoning in Medaka fish (Oryzias latipes) embryos. Selenium has been shown to provide a protective mechanism against mercury poisoning in some adult animals. Three experiments were done. In the first, selenium and methyl mercury were put directly in the growing medium of the eggs. In the second experiment, selenium was included in the diet of the parent fish. In the third part, methyl mercury exposure was delayed three days. The results of the first experiment were quite dramatic and showed a significant difference when selenium …

Two factors which affect leaf ontogeny and ultimate leaf size: (1) the rate and duration of cell expansion, and (2) the rate and duration of cell division, were examined for their role in the slowed early growth rate and smaller ultimate leaf size when plants are exposed to ultraviolet-B (UV-B) radiation. Rumex patientia L. was grown in controlled environment chambers under enhanced UV-B radiation (equivalent to daily solar UV-B irradiation at 40°N latitude in mid-May with an atmospheric ozone concentration of 0.20 atm-cm) and control treatments. Lehman's polychrome stain was used to follow the fate of germinal plasm in early development of Bombina orientalis embryos. Islets of germinal plasm approximately 6 um in diameter were observed in the vegetal pole region of the 2-cell embryo. By the 4-cell stage, some of these islets had coalesced to form larger ones so that two size classes of islets were present (approximately 7.5 and 19.5 um in diameter).
As cleavage continued to the 16-cell stage, the islets were moved further up the cleavage furrow and were always observed to remain in close association with the cleavage membrane. The islets …

The normal morphological development of the Chinese hamster (Cricetulus griseus) during the last week of gestation was studied. A review of the literature reveals no distinct study of tooth development in this species. An identical gestation period is found in the mouse (Mus musculus) making direct comparison between the embryos of the same age of the two species feasible. During this investigation, it was found that morphological tooth development in the Chinese hamster lagged behind that of the mouse by about 2 days. The pleomorphism of Coprinus radians was studied. The three distinct growth forms--sporophore, oidial stage, and ozonium stage--were examined in relation to ultrastructure, morphology and development. Scanning and transmission electron microscopy are correlated with photomicrographs of developmental stages. Development of the oidia and of the sporophore are of particular interest. The multinucleate oidia develop quite differently from oidia as generally described for Basidiomycete imperfect stages. Development of the oidia of this Coprinus is holoblastic and sympodial. The mushroom primordia are differentiated internally into gill, cap, and stipe hyphae while only l-2mm in diameter. At this stage they look superficially like sclerotial …

The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level. They may achieve overall specific gravities of <1 to > 1 after a ventilation. They may remain submerged (s.g. > 1) or float (s.g.

Eremosphaera viridis de Bary was studied with light microscope techniques to determine the method of cytokinesis and to explain the ontogeny of the membranous ring separating the mature daughter cells in the intact sporangium.
It is proposed that cytokinesis in this organism is phylogenetically advanced and that the separation of mature daughter cells involves the sequential deposition of two cell walls. The first wall develops centripetally by the addition of membrane subunits. The resulting cleavage furrow is oriented by transverse microtubules found along the cell division plane. Nemerteans are common invertebrates along coastlines in both temperate and arctic climates throughout the world. Amphiphorus imparispinosus is a carnivorous member of this phylum that is found abundantly in the intertidal areas of the Oregon coast. As in many other invertebrate phyla, endocrine integration within the nemerteans is thought to be accomplished by neurosecretory mechanisms. In Amphiphorus the presence of these neurosecretory systems has not been previously shown. The dispersal and dispersion patterns of Hydra fusca were examined. Hydra were placed in petri dishes at various densities. The water in the dishes was swirled forcing the animals to the center. The location of each animal was marked at time intervals thereafter. Analyses of the dispersal rates and dispersion patterns were made. Hydra dispersed from a central release point at a non-random rate. There was rapid movement from the center followed by a minimal daily movement. This eventually resulted in a uniform dispersion pattern at high densities in a limited environment. There was a relationship between the ratio of …

The establishment of normal chromosomal patterns for a wide range of plants and animals has led to the recognition of abnormalities as well as their relationship to phenotypic irregularities. Research dealing with the expression of human karyotype is of prime interest; but, closely rivaling that is the genetic study of domestic animals which are of economic importance to man.

Blastospore and mycelial cytoplasmic extracts from four strains of Candida albicans, varying in virulence, were compared by sodium dodecyl sulfate (SDS) polyacrylamide disc gel electrophoresis Cytoplasmic extracts were made by disrupting the cultures in a Braun homogenizer followed by ultracentrifugation. The supernatant fractions were then resolved on polyacrylamide gels containing SDS. Each of the preparations yielded a distinct pattern. There were mycelial specific and blastospore specific protein bands. There were more components resolved from the blastospore phase cytoplasmic extracts than from the mycelial phase cytoplasmic extracts. There were bands common to both phases. The time-course of changes in the pattern of newly synthesized proteins in the R15 neuron of the parietovisceral ganglion of Aplysia californica has been studied at 14 degrees C. 5% polyacrylamide gels containing sodium dodecyl sulfate (SDS) have been used to separate newly synthesized (leucine-labeled) proteins from the neuron. We have demonstrated that the pattern of newly synthesized proteins from the R15 neuron does not change significantly if 5-h pulses of labeled leucine are given during the first 72 h of in vitro incubation of the excised ganglion. However, the level of leucine incorporation begins to decline somewhere between 17 …

The external surfaces of pear sclereids commonly are illustrated as covered with apertures. This SEAA investigation of the surface features has shown the surface to have few or no apertures. When the primary wall layer was removed the typical ramiform canal system was obvious. This observation confirms the often-ignored fact that the pitapertures of the secondary wall are not continuous with the primary wall. Hence, they do not show on the surfaces of the intact cell.

The upper optimum temperature for embryonic development of mountain whitefish is 6 C, and for the post yolk sac stage is 9 to 12 C. The need to know effects on all stages in the life cycle in cases of thermal pollution is demonstrated. Abnormalities caused by thermal pollution in hatched larvae were agape jaws, coloboma or fissure of the eye, monophthalmia or the presence of only one eye, monomicrophthalmia or the presence of one small eye and one case of twinning. High mortalities of eggs occurred at 9 C and higher.

The ultrastructure of abnormal meiotic cytokinesis in an autosomal male sterile mutant of Drosophila melanogaster is described. In the mutant ms(2)3R mitotic cytokinesis proceeds normally, but abnormal meiotic divisions give rise to four spermatids developing in one cytoplasmic mass. The contractile ring which is responsible for cell constriction during cytokinesis appears to form and function normally during meiosis. Ring canals which form intercellular bridges between synchronously developing spermatids are also apparently normal. However, there is an apparent adhesion and subsequent disintegration of the furrow membranes after division is completed. In addition to double membrane fragments in areas of open …

Light And Electron Microscope Study Of The Mitotic Apparatus Of The Ring-Legged Earwig, Euborellia Annulipes (Lucas), Vonnie R. Prentice, William L. Evans

The mitotic apparatus in dividing cells of the ring-legged earwig is composed of the cell center or pole, chromosomal fibers, continuous fibers, and chromosomal and background structure. Parts from cellular elements such as Golgi membranes, endoplasmic reticulum, and nuclear envelope as well as whole mitochondria associate with the spindle microtubules to produce the diffraction pattern of the spindle fibers as seen with the light microscope. The microtubules of the chromosomal fibers attach independently and demonstrate the diffuse nature of the centromere or polycentric condition of the chromosomes.

Jackson and Patton stated that the key to understanding mechanisms of chromosomal evolution lies in studies of chromosomal variation within populations that can hybridize. Hybrids between cytotypes contain the answer to problems of meiotic pairing and reduced fertility, as well as information concerning the nature of zones of contact. However, before detailed studies can be initiated, chromosomal races and hybrid zones must be located. Few chromosomal races have been described for bats. Within the family Vespertilionidae, five species are known to have such races and races have been reported in two species of the family Phyllostomatidae.

The ry+ gene of Drosophila melanogaster and its associated enzyme, xanthine dehydrogenase, were employed in an analysis of the relationship between regulation and the location of a gene in the genome. Enzyme assays as an indication of gene activity were performed on genotypes containing zero, one and two doses of ry+ genes. Xanthine dehydrogenase activity of rosy genes in normal and relocated positions was determined during development.
The results indicate: 1) the gene ry+ is differentially active during ontogeny; 2) the activity during ontogeny is proportional to the number of ry+ genes present in the genome; …
