The isolation of Rhizobium japonicum cell walls was performed by ultrasonication and the enzymatic digestion with DNase, RNase and pronase separately. Crude cell walls were washed with M NaCl and 5% SDS and were isolated by 20,000 g centrifugation. Muropeptide amino acids were freed by hydrolysis at 121C for 10 minutes in 6N HCl.
Detection of Rhizobium japonicum murein layer amino acids was done by 2-dimensional instant thin layer chromatography on silica gel plates. The solvents found most favorable, were n-butanol: acetic acid: water and n-propanol: 34% ammonium hydroxide. Amino acid spots were developed by spraying ninhydrin on the chromatograms, …

Isolation And Characterization Of Two Bacteriophages Infecting The Myxobacterium Cytophaga Succinicans, Kenneth Wayne Lee

Two bacteriophages which infect the myxobacterium Cytophaga succinicans, strains 14 and 16, were isolated from fish hatchery effluent. Each was submitted to a detailed study. These phages have a limited host range, even within the genus Cytophaga. The turbid plaques formed by these phages are similar in appearance to those produced by coliphage Lambda, and average 0.85 mm in diameter. Both phages have a latent period of 195 minutes and a rise period of an additional 165 minutes. Phage 0CS-16 has a burst size of 137 and phage 0CS-14 has a burst of 45. This thesis was designed to provide a histological and histochemical survey of the small intestine of the mink, Mustela vison. These two objectives were achieved and it was found that the histology and enzyme activity in this animal is in most respects similar to other mammals. Exceptions to this are the absence of a cecum and an ileocecal valve and the absence of lactase activity in the small intestine.

Induction And Initial Characterization Of Male-Sterile Mutants On The Second Chromosome Of Drosophila Melanogaster, Susan M. Graves

Fourteen recessive, male-sterile, autosomal mutants of Drosophila melanogaster were induced using ethyl methanesulfonate. The techniques employed restricted the recovery of mutants to those located on the second chromosome. Eleven of the mutants were male-sterile at 23°C, and the remaining three, though fertile at 23°C, were male-sterile at 28°C. Complementation tests involving all possible pairs of mutations showed each to be located in a unique cistron.
Spermiogenesis in each mutant was studied by light microscopy to determine the stage at which cellular differentiation deviates from normal and to characterize the associated gross abnormalities. Phase contrast microscopy showed that each mutant sterile …

Spermiogenesis In Two Male-Sterile Third-Chromosome Mutants Of Drosophila Melanogaster, Richard F. Wilkinson

An ultrastructural study of sperm differentiation was made on two male-sterile mutants of Drosophila melanogaster. The mutations were induced on the third chromosome with the monoalkylating agent ethyl methanesulfonate and were maintained in stock cultures over a balancer chromosome using a system of multiple inversions. The stages of sperm differentiation, a most dramatic form of cellular differentiation, were then studied at the electron microscope level to determine the effect of the mutations on sperm structure and to correlate those changes with normal sperm development.

The Effects Of Moist-Heat Treatments And Stratification On Germination Of Prairie Plant Seeds, John E. Stuurwold

Germination of Silphium terebinthenaceum was enhanced by a post-stratification most-heat treatment at 80°C. Tested seeds of four other perennial herbs common to midwestern prairies (Andropogon scoparius, Sorghastrum nutans, Eryngium yuccifolium, and Solidago rigida) showed little adverse or beneficial effects of the heat treatment. Heat treatments alone had little effect on the seeds; the length of stratification periods necessary to effect maximal germination varied. The significance of these findings is examined in the light of related literature on grassland fire ecology.

Lipid is associated with all of the major serum protein fractions of both the mallard duck and man. however, whereas the ρ lipoproteins predominate in man with little lipo-albumin evident, according to available literature, in ducks the lipo-albumin constitutes more than half of the serum lipoproteins, and a substantial lipo-prealbumin fraction is evident. Most studies of lipo-proteins are carried out on human serum primarily for the purpose of diagnosis of abnormalities. In order to obtain proper muscle development in tissue culture, certain conditions must prevail. There is a delicate balance of factors, and elimination of even one of these factors leads to poor and improper development. Muscle cells grown in the absence of fibroblasts usually fail to differentiate. Therefore, proper development depends upon the presence of two cell types: myoblasts and fibroblasts. Investigators have demonstrated that the fibroblasts are necessary in that they produce collagen which is necessary for proper muscle development. The Role Of D-Gluconic Acid In The Regulation Of The Synthesis Of The Enzymes Of The Entner-Doudoroff Pathway In Pseudomonas Fluorescens, Steven C. Quay

Induction of Entner-Duodoroff pathway enzymes in Pseudomonas fluorescens was investigated to study the role of gluconate as a possible inducer. Glucose oxidase-deficient mutants were isolated and characterized. One of these mutants, gox-7, was deficient in particulate glucose oxidase; another mutant, gox-17, was deficient in particulate glucose and gluconate oxidase activities. Gluconate, but not glucose, induced synthesis of gluconokinase and 6-phosphogluconate dehydratase in both mutants. High constitute levels of 2-keto-3-deoxy-6-phosphogluconate aldolase were found when both mutants were grown on glucose. Growth of parent and both mutant strains on glycerol also resulted in high levels of Entner-Doudoroff pathway enzymes. It was concluded …

Parietovisceral ganglia from Aplysia californica were incubated in medium containing leucine-3H. Single, identified nerve cell somas were isolated from the ganglia, and their proteins extracted and separated by electrophoresis on 5% SDS-polyacrylamide gels. The distribution of total or newly synthesized proteins from the single neurons was determined by staining or slicing and liquid scintillation counting of the gels. Experiments showed that: (a) a number of proteins were being synthesized in abundance in the nerve cells; (b) different, identified neurons showed reproducibly different labeling patterns in the gels; (c) cells R2 and R15, …

ALAR* is a growth retardant which has been die subject of intensive research in other countries. It tends to slow both fruit growth and ripening and to reduce shoot extension.
Overseas and local investigations with apples have shown that Alar will reduce preharvest drop, restrict shoot growth and increase flower bud formation for the following season.

A cell has been designed and built that allows for rapid measurement of volume moved in a definite time by electroosmosis. The cell is simple to use and is not very elaborate. Using a water jacket, the cell temperature can be controlled to ± 0.1° C. Measurements are presented for acetonitrile, dimethylformamide, and nitrobenzene at 25° C for applied voltages of 25, 50, 75, and 100 volts.

The effects of urethan on RTG-2 and FHM cells were studied in vitro, by using the mitotic index, it was determined that 0.3 percent urethan caused an increase in the rate of cell division while higher concentrations (0.6, 0.9, 1.2, and 1.5 percent) caused either a decrease in the rate or a cessation of cell division. Concentrations of urethan higher than 1.5 percent killed the cells. The mitotic index data also indicated that epithelial cells continued to divide at a higher concentration of urethan than did the fibroblast cells. Showers of muons which result from interactions of primary cosmic rays high in the atmosphere appear deep underground as bundles of nearly parallel muons. For threshold energies of 1 TeV and arrival zenith angles of 45 degrees, those events having a small mean number of muons are described by a power law distribution in shower size having an exponent of ≅ -3.7 and a radial density distribution that decreases with distance R from the shower axis a little less rapidly than exp(-R/5.4 m). The following members of the Rhizobiaceae were tested for their lytic response to various treatments: Agrobacterium tumefaciens, Chromobacterium violaceum, Rhizobium phaseoli, Rhizobium trifolii, Rhizobium lupini, and Rhizobium japonicum. Lysis was observed in spent growth media and as a function of saline concentration, pH, buffer composition, disodium (ethylenedinitrilo) tetraacetate (EDTA) concentration, and lysozyme.
Cells were harvested by centrifugation, washed, and resuspended in various treatment solutions. The lytic response was followed spectrophotometrically.

The chemical composition of algae grown in batch culture depends mainly on environmental conditions, nutrient availability, presence of predators, cell age, and species. The effects of nutrient availability and cell age on the composition of three unialgal cultures (algae + bacteria) and one hetergeneous culture (algae + bacteria + microscopic animals) were evaluated. The cultures were grown in batch culture under both nutrient-abundant and nutrient deficient conditions and the changes in compositions were observed. Luxurious uptake where nutrients are incorporated into cellular protoplasm at levels greater than those necessary for growth, and super-luxurious uptake, where some nutrients are stored rather …

Analysis of lambing trials with Merino ewes has shown a highly significant association between pre-mating liveweight and lambing performance.
With May-June lambing ewes there was a 2.9 per cent, lambing increase for every extra 10 lb. liveweight; for July-August- September lambing ewes there was a 6.9 per cent, increase for every 10 lb. liveweight increase.
With the May-June lambing there was a critical weight below which twinning was negligible.

Ovulation studies in Western Australia have indicated that increased lambing percentages in crossbred and Merino ewes could be expected from mating in February-April rather than December- January.
Such increases have been demonstrated in time of lambing trials with crossbred ewes, but had not been seen in Merino flocks until recently.
This report summarises the results of time of lambing trials with Merinos carried out at the Merredin and Wongan Hills Research Stations between 1963 and 1969.

Department of Agriculture experiments have shown that biennial bearing of apples can be controlled by chemical thinning, but both winter and spring weather conditions must be taken into account when the need for spraying is assessed.
BIENNIAL BEARING is a regular cycle in which each heavy crop of fruit is followed by a light crop, or, in extreme cases, no crop at all.

The action of a chemotherapeutic agent as a tool in studying its effect upon a primary growth center, the mandibular condyle, was investigated. So far as can be ascertained, no studies have been reported where these growth sites have been eliminated or modified by the use of chemotherapeutic growth inhibitors.
Observation of mandibular and alveolar growth following the infusion into the left external carotid artery of a cancer chemotherapeutic agent, 5-Fluorouricil (Roach-Hoffman), was noted. Comparisons were made between the infused and the uninfused sides of the mandible observing gross growth changes.
