An assay procedure for in vitro enzymatic methylation of mannnalian ribosomal RNA has been developed in this study. The assay procedure, utilized for the comparison of normal and neoplastic methylase activities (using mouse liver and Ehrlich ascites cells as sources of enzyme), is a modification of previously published methods (52,53). A 100,000 x g supernatant (SlOO) enzyme preparation was incubated with 28S-5.8S rRNA and tritium-labeled S-adenosyl-L-methionine. The RNA was extracted, applied to DEAE cellulose paper, washed, and the radioactivity counted. The neoplastic cell methylase preparation was more active in methylating both exogenous neoplastic and normal 28S-5.8S rRNA than the normal …

Crude extracts of triple-cloned, purified cultures of 22 species of Mycoplasma and Acholeplasma were examined for expression of 21 isozyme systems routinely used to type mammalian cells. Nine previously described enzymes (purine nucleoside phosphorylase, adenylate kinase, dipeptidase, esterase, glyceraldehyde-3-phosphate dehydrogenase, glucose phosphate isomerase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and superoxide dismutase) and three enzymes not previously reported in mycoplasma (triose phosphate isomerase, inorganic pyrophosphatase, and acid phosphatase) were detected in some or all of the species examined. These findings provide new information on the enzymatic expressions of these organisms. A transmission electron microscope analysis of the changes that occur in the nuclear chromatin and the Feulgen-positive areas of the oocyte cytoplasm as the female develops, feeds and mates is described. The description includes analysis of oocytes from three types of females: unfed, unmated; fed, unmated; and fed, mated. In all types, dense material passes through the extremely porous nuclear membrane where it accumulates in the cytoplasm.

Nuclei of previtellogenic oocytes appear the same in all females studied. Fibrillar bodies and up to four large nucleoli are seen. No typical chromatin is seen, but structures resembling nucleosomes appear throughout the …

Interpopulation Variation In Calcareous And Proteinaceous Shell Components In The Stream Limpet, Ferrissia Rivularis, Carl M. Way, Daniel J. Hornbach, Albert J. Burky

Natural populations of the North American stream limpet, F. rivularis, were studied in upstate New York [USA], in a set of localities whose waters have a 15-fold range of dissolved Ca (4.6-67.6 mg/liter) and also range from oligotrophy to eutrophy. Shell component analyses (CaCO3, total organic C and total N) are reported as component mass-fractions (mg/g or .mu.g/g dry weight) and as values for a standard limpet shell of 35 mm aperture length (AL). Seasonal metabolic rates were examined for the sphaeriid clam, Musculium partumeium (Say) from a permanent and a temporary pond. For the fall-born generation from the permanent pond and for the single generation from the temporary pond, metabolic rates peak during periods of greatest growth and reproduction. Metabolic rates were consistently higher for the permanent pond population. These rates for Musculium partumeium extend the available information on the family Sphaeriidae to the genus Musculium.

Clarke (1973) states that Pisidium casertanum (Poli) is perhaps the most common species of Pisidium and is truly cosmopolitan, occurring throughout Eurasia, Africa, Australia, and South, Central and North America. Populations of these clams are reported in habitats ranging from ephemeral ponds to benthic zones of deep lakes. Sodium Flux Ratio In Voltage-Clamped Squid Giant Axons, David D. Busath, Ted Begenisich

The sodium flux ratio across the axolemma of internally perfused, voltage-clamped giant axons of Loligo pealei has been measured at various membrane potentials. The flux ratio exponent obtained from these measurements was about unity and independent of membrane voltage over the 50 mV range from about -20 to +30 mV. These results, combined with previous measurements of ion permeation through sodium channels, show that the sodium channel behaves like a multi-ion pore with two ion binding sites that are rarely simultaneously occupied by sodium.

Chromosomal and morphometric studies were conducted on a sample from an assemblage of Akodon spp. occurring in various patterns of sympatry from the provinces of Catamarca, Jujuy, Salta, and Tucumán, Argentina. Results showed three distinct morphometric groups based upon size. Size also varied with age, but there were no significant differences in measurements of males and females. The three morphometric groups have distinct karyotypes. Akodon caenosus Thomas is the smallest of the three, and has a karyotype of 2n = 34, FN = 40. A. boliviensis tucumanensis J. A. After vasectomy, the passage of sperm antigen through the epithelium of the efferent duct in the rhesus macaque probably results in immune complex deposition. Immune complexes can be visualized in the thickened basement by fluorescence microscopy. Subsequent electron microscopic evaluation, with the appearance of electron-dense deposits in the basement membrane of the efferent ducts, substantiates these findings.

Five-week-old male BALB/c mice were anesthetized and incised mid-dorsally to expose the fascia over the right kidney. The kidney was then palpated up and an inoculum of 0.05 ml containing approximately 100,000 viable Asperaillus flavus spores in phosphate-buffered saline was injected into the cortex. A second group of mice underwent similar surgical procedures, but were injected with phosphate-buffered saline only into the right kidney. This group was designated the sham-operated control mice. All mice, including controls, were bled for differential blood counts 24 and 48 h after surgical procedures and subsequently on an every-other-day basis for the duration of the …

It has been previously demonstrated that 7,12-dimethylbenz(a) anthracene (DMBA) is an initiator of ovarian granulosa cell tumors. It is also known that DMBA leads to destruction of primordial oocytes, and that tumor induction may involve the interaction of pituitary gonadotropins. However, the effects of DMBA on ovarian steroidogenesis and uterine growth in relation to the tumorigenic process have not been investigated. Therefore, the present study was conducted to examine these parameters using adult C57Bl/6N mice treated in proestrus and diestrus II with 0.1 ml corn oil with or without 1.5 mg DMBA and killed 44 hours later. Altered patterns of methylation have been shown in both ribosomal and transfer RNA isolated from cancer cells. In contrast to the elevated activity of the methyltransferases responsible for this modification, hypomethylation appears as a general characteristic. The endogenous levels of S-adenosylmethionine and S-adenosylhomocysteine are important in relationship to their role as substrate and product of the transmethylation reaction. The levels of S-adenosylmethionine and S-adenosylhomocysteine were determined using phosphocellulose cation exchange or high pressure liquid chromatography. Damage to primary photosynthetic reactions caused by environmental stress can be assessed by measurement of chlorophyll fluorescence induction in intact leaves. This approach was applied in studies of ultraviolet-B photoinhibition of photosynthesis in Pisum sativum L. and Rumex patientia L. leaves. At ultraviolet-B dose rates insufficient to cause inhibition of net photosynthesis, changes in the magnitude of fluorescence transients did occur, which suggested direct effects on chloroplast pigments in Pisum and inhibition of photosynthetic electron transport between the photosystems in both species. FTIR was used in conjunction with a surface selective technique, known as attenuated total reflectance (ATR), to study the infrared spectra of preimplantation rabbit embryos. The rabbit embryos ranged in aged from 3 to 6 days. Since an embryo is mostly water, deuteration of the embryo was necessary to eliminate the intense water background present in the mid-infrared region. A set of spectral standards were tested, in order to identify the bands seen in embryo spectra. By analyzing and comparing the spectra from 3, 4, 5 and 6 day embryos, differences could be noted. 5.8S rRNA is a low molecular weight ribosomal RNA which is found hydrogen bonded to 28S rRNA in the eucaryotic cell. There are two nucleotides which have 2'-0-methylations; a guanine residue at position 77 is fully methylated and a uridine residue at position 14 which is partially methylated. This partial 2'-0-methylation of the uridine residue has been found to vary with the tissue source, with the highest level in normal tissue and the lowest level in neoplastic tissue. In order to study the significance of this site-specific methylation, a reliable and convenient method of analysis was needed.

A two-dimensional polyacrylamide gel electrophoretic technique was developed for the detection of unique tumor-associated proteins in urine of patients with transitional cell carcinoma (TCC) of the urinary bladder. This technique yielded high resolution of polypeptides in the urine of cancer patients. Urine was chosen as a source of tumor-associated components because the location of the tum or should release already solublized tum or substances in to the urine.

Urinary proteins were concentrated and then analyzed by the two - dimensional electrophoretic technique using isoelectric focusing in the first dimension and sodium dodecylsulfate - polyacrylamide gel electrophoresis in the second dimension. …

Regenerating sabellid polychaetes were histochemically studied with regard to the synthesis and deposition of glycosaminoglycans (GAG) and collagen. Both of these types of macromolecules are known to be actively involved in cell migrations and differentiations of several vertebrate systems. The purpose of this study was to investigate whether or not these extracellular materials might be similarly involved in invertebrate systems. Results demonstrate for the first time that these same extracellular materials, GAG and collagen, actively participate during invertebrate development. Sand G2 phase cells were exposed to 150 ergs mm⁻² UV and their ability to photoreactivate the induced cell killing (loss of colony forming ability) and chromosomal aberrations was determined as a function of time following the UV exposure. In S phase cells, the lesions leading to cell death and those leading to aberrations were both converted to a non-photoreactivable state shortly after the UV exposure. A significant fraction of the lesions induced in G2 cells, that led to cell death, were converted to a non-photoreactivable state before the progeny of the exposed cells reached the next succeeding S phase. …

Neurospora produces a phase specific, cationic mucopolysaccharide composed primarily of galactosamine (GAG-MP) which becomes part of the cell wall and is later secreted into the media. Its appearance coincides with the onset of the restricted phase of growth and causes efflux of small molecular weight metabolites when incubated with conidial cells. This activity may be a product of electrostatic interactions of the GAG-MP with the conidial plasma membrane. The activity is blocked by acetylation of the primary amines on the molecule, digestion by enzymes that hydrolyze carbohydrate linkages and the inclusion of NaCl (lM) in the GAG-MP/conidia reaction mixture. The morphology of Parascaris equorum of horses was studied; light and scanning electron microscopy were used. Late fourth-stage larvae have narrow, rectangular lips bearing few, large. triangular, irregularly spaced denticles and the lips are not markedly set off from the body. The cutick of the late fourth stage is completely transversely striated, with incomplete longitudinal ridges creating a brickwork pattern; longitudinal alae are present. Total body lengths of fourth-stage larvae range from 10 to 32 mm. A single molting fourth-stage male was found to be 33 mm long. Male Callagur borneoensis exihibit sexual dichromatism and seasonal coloration which is rare among turtles. In the breeding season male's heads are colored white with a scarlet stripe from the occiput to the tip of the snout. After the breeding season they change to drab charcoal-gray to black with a dull orange-yellow stripe. Females and juveniles are a drab brown throughout the year.

This study was performed to determine the mechanisms of this color change at the histological level. Histological preparation of the head revealed a significant increase in vascular tissue just below the epidermis of the stripe area with increased …

Acute inflammation is associated with injury to tissues, whether it be from mechanical destruction, or from the introduction of a noxious substance into the tissue. Inflammation is a homeostatic process, adapted to neutralize harmful substances and restore injured tissue. Celsus (c. A.D. 178) provided the first documented characterization of inflammation. He recognized redness, heat, swelling and pain as the cardinal signs of the inflammatory reaction. Loss of function was later added to this list of symptons (Movat. 1971).
