Diabetes mellitus (DM) is a group of metabolic disorders in which high blood glucose levels over a prolonged period, caused by deficiency of insulin production from the pancreas, or by the insulin resistance. Several epidemiological studies suggest that diabetic population is not only at increased risk of cardiovascular complications, but also at substantially higher risk of many types of malignancies. Hyperglycemia induced free radical reactive oxygen species (ROS) production, which is a major cause of cell injury and organ damage, especially affects pancreatic beta cells. หลังคาเขียวเพิ่มพื้นที่สีเขียวในเมืองและอาจรองรับสังคมนกโดยให้ที่พักพิง อาหารและแหล่งน้ำ งานวิจัยนี้ศึกษาความหลากชนิดของนกและรูปแบบการใช้ประโยชน์บนหลังคาเขียว 7 แห่งในกรุงเทพมหานครที่มีขนาดพื้นที่และสัดส่วนพื้นที่สีเขียวแตกต่างกัน โดยสำรวจนกบนหลังคาเขียวทุกเดือนระหว่างเมษายน 2559-พฤษภาคม 2560 พบนกรวม 26 ชนิดและนกส่วนใหญ่เป็นนกประจำถิ่น พบนก 7-18 ชนิดบนหลังคาเขียว นกกินปลีอกเหลือง Cinnyris jugularis เป็นนกที่พบบนทุกหลังคาเขียว นกกระจอกบ้าน Passer montanus เป็นนกที่มีความชุกชุมสูงสุดโดยมักปรากฏเป็นฝูงใหญ่ พบนกอพยพบางชนิด เช่น นกอัญชันป่าขาเทา Rallina eurizonoides นกแซงแซวสีเทา Edolius leucophaeus นกขมิ้นท้ายทอยดำ Oriolus chinensis นกกระเบื้องผา Monticola solitarius และนกอีเสือสีน้ำตาล Lanius cristatus การหาอาหารและการเกาะพักเป็นรูปแบบการใช้ประโยชน์ที่มีความถี่สูงสุด (ร้อยละ 47 และ 31 ตามลำดับ) นกใช้ไม้ยืนต้นสำหรับการเกาะพักและการเคลื่อนที่ ลักษณะทางกายภาพและชีวภาพบางประการส่งผลต่อสังคมนกที่ใช้ประโยชน์บนหลังคาเขียว ดังนั้นหลังคาเขียวจึงมีศักยภาพในการอนุรักษ์ความหลากหลายทางชีวภาพในระบบนิเวศเมือง

Micro- and nano-motors have attracted numerous attentions from various scientific areas due to their potential applications. Most studies on self-propelled colloidal engines have exploited catalytic decomposition of hydrogen peroxide to drive the motor. Since the hydrogen peroxide is caustic, it is not suitable to use in biological applications, encouraging people to develop “greener” fuels. The aim of this research is to study a new transduction mechanism for self-propulsion not tied to hydrogen peroxide, and which can in particular be used with biological molecules as fuels. In vitro protein selection has had major impacts in the field of protein engineering. Traditional screens assay individual proteins for specific function. Selection, however, analyzes a pool of mutants and yields the best variants. Phage display, a successful selection technique, also provides a reliable link between variant phenotype and genotype. It can also be coupled with high throughput sequencing to map protein mutations; potentially highlighting vital mutations in variants. We propose to apply this technique to cancer therapy. A diet consisting of a high intake of saturated fat and refined sugars is characteristic of a Western-diet and has been shown to have a substantial negative effect on human health. Expression proteomics were used to investigate changes to the parietal lobe proteome of rhesus monkeys consuming either a high fat and sugar (HFS) diet, a HFS diet supplemented with resveratrol (HFS+RSV), or a healthy control diet for 2 years. Here we discuss the modifications in the levels of 12 specific proteins involved in various cellular systems including metabolism, neurotransmission, structural integrity, and general cellular signaling following a nutritional intervention. …

Biologically, costal cartilage is an understudied tissue type and much is yet to be learned regarding underlying mechanisms related to form and function, and how these relate to disease states, specifically chest wall deformity. Chest wall deformities have a component of inheritance, implying underlying genetic causes; however the complexity of inheritance suggests multiple genetic components. Objective: p120 catenin (p120ctn) has been reported to play a critical role in maintenance of the stability of adherens junctions. It also has potential anti-inflammatory effects in epithelial and endothelial cells. This research was designed to evaluate the effects of p120ctn on inflammatory responses in human macrophages upon LPS stimulation, as well as the possible mechanism by which p120ctn regulates LPS-induced proinflammatory response in macrophages. he multisubunit SIN3 complex is a global transcriptional regulator. In Drosophila, a single Sin3A gene encodes different isoforms of SIN3, of which SIN3 187 and SIN3 220 are the major isoforms. Previous studies have demonstrated functional non-redundancy of SIN3 isoforms. The role of SIN3 isoforms in regulating distinct biological processes, however, is not well characterized. In addition, how the components of the SIN3 complex modulate the gene regulatory activity of the complex is not well understood. SH3 and multiple ankyrin repeat domains 3 (SHANK3) is a multidomain scaffold protein that is highly augmented in the postsynaptic density (PSD) of excitatory glutamatergic synapses within the central and peripheral nervous systems. SHANK3 links neurotransmitter receptors, ion channels, and other critical membrane proteins to intracellular cytoskeleton and signal transduction pathways. Mutations in SHANK3 are linked with a number neuropsychiatric disorders including autism spectrum disorders (ASDs). Intellectual disability, impaired memory and learning, and epilepsy are some of the deficits commonly associated with ASDs that result from mutations in SHANK3. Niemann Pick Type C (NPC) disease is a fatal childhood neurological disease caused by mutations in the NPC-1 protein, resulting in cholesterol buildup in the late endosomes. StarD4 and StarD5 are cholesterol binding proteins that play a role in the intracellular cholesterol transport. In this study we overexpress StarD4 and StarD5 in in vitro and in vivo models, and find evidence of amelioration of the NPC phenotype. This study demonstrates that the overexpression of these proteins has the potential to be a therapeutic treatment for NPC disease.

The objective of this project was to identify important sources of bacterial loading in Plaster Creek (Kent County). The following goals were set to reach this objective: 1) the relative contributions of bacterial loading for 10 major tributaries will be assessed and the five most egregious contributors will be prioritized; 2) molecular markers will be used to try to identify the primary taxonomic sources, and 3) to pinpoint bacterial loading locations using molecular markers and scent-trained canines in sub-watersheds.

It serves as a hydrogen bond donor to both the solvent molecules at the active site and Tyr 34, the conserved “gateway” amino acid residue. This dissertation develops our understanding of the effect of amino acid Gln146 at the second shell of the active site of metalloprotein MnSOD in facilitating metal binding, the modulation of redox potential, and the optimization of catalytic activity and structure stability. Escherichia coli transporter protein AcrB and its homologues are the inner membrane components of the Resistance-Nodulation-Division (RND) family efflux pumps in Gram-negative bacteria. It is well accepted that soluble proteins are only marginally stable, but such insight is missing for membrane proteins. The lack of stability data, including thermodynamic stability and oligomer association affinity is a result of intrinsic difficulties in working with membrane proteins. Toluene / o-xylene monooxyenase (ToMO) from Pseudomonas sp. OX1 is a multimeric metalloenzyme enzyme that efficiently catalyzes the hydroxylation of aromatic hydrocarbons with high specificity. Though included in a larger group of conserved bacterial multicomponent monooxygenases (BMMs) studied as potential biocatalysts for industrial hydrocarbon chemistry, the substrate specificity and oxygenated intermediates of ToMO differ greatly from its well-characterized, alkane-hydroxylating analog sMMO. Despite a shared global topology and near identical active sites, sMMO can cleave inert C-H bonds in alkanes while ToMO cannot - two seemingly similar structures give rise to vastly different chemistries. Accurate maps and DNA sequences for human subtelomere regions, along with detailed knowledge of subtelomere variation and long-range telomereterminal haplotypes in individuals, are critical for understanding telomere function and its roles in human biology. Here, we use a highly automated whole genome mapping technology in nano-channel arrays to analyze large terminal human chromosome segments extending from chromosome-specific subtelomere sequences through subtelomeric repeat regions to terminal (TTAGGG) n repeat tracts. We establish detailed maps for subtelomere gap regions in the human reference sequence, detect many new large subtelomeric variants and demonstrate the feasibility of long-range haplotyping through segmentally duplicated subtelomere regions. …

The accumulated evidence points to the microenvironment as the primary mediator of cellular fate determination. Comprised of parenchymal cells, stromal cells, structural extracellular matrix proteins, and signaling molecules, the microenvironment is a complex and synergistic edifice that varies tissue to tissue. Furthermore, it has become increasingly clear that the microenvironment plays crucial roles in the establishment and progression of diseases such as cardiovascular disease, neurodegeneration, cancer, and ageing. "Age-related macular degeneration (AMD) is a leading cause of irreversible blindness among adults, age 60 and older, in developed countries. While oxidative stress is implicated in the pathogenesis of AMD, clinical studies have shown that dietary antioxidants can delay progression of AMD. Currently, there is no FDA-approved treatment for AMD. Therefore, we hypothesized that N-acetylcysteine amide (NACA), a thiol antioxidant, would protect retinal pigment epithelium and impede the progression of retinal degeneration. Alternative splicing occurs throughout the human genome, leading to multiple proteins from a single gene. The resulting proteins can be nearly identical or vastly different in how they function. Alternative splicing within the perilipin family has been observed in perilipins 1 and 3, giving rise to proteins with varying functions. In the course of our studies on perilipin 5, an immunoblot signal was observed that corresponded to an approximately 35 kDa protein. The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. Alpha-glucosidase [EC 3.2.1.20; α-D-glucoside glucohydrolase] is an exohydrolase which catalyzes non-reducing end of substrates to release D-glucose. In addition, alpha-glucosidase also displays a transferase activity, which bring about the formation of α-glucosylated compounds such as soluble starch and glycogen. This research aims to express and characterize α-glucosidase from Weissella confusa BBK-1 (WcAG). WcAG gene was expressed in modified pET28b vector. Fisetin is a flavonoid which was widely exerted in pharmacological industries. However, fisetin has low water solubility and unstable in aqueous solution. So the uses of fisetin have been limited. The purpose of this study was to increase water solubility of fisetin by transglycosylation reaction using cyclodextrin glycosyltransferase (CGTase) from Paenibacillus sp. RB01. Women with lung NETs have significantly better survival rates as compared to men suggesting the involvement of sex steroids and their receptors in the progression of lung NETs. ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …
