The work on the active substance of Eremocarpus setigerus Benth has been previously performed by Naito and Noller. However, the investigation was not complete. Therefore we isolated and tested this substance by steam distillation and thin layer chromatography technique. A yellow sticky toxic substance was isolated with Rf values of 0.15 in benzene system and 0.57 in benzene-absolute ethanol (95:5 v/v) system.
The toxic substance which is free of halogen, nitrogen, and sulfur, is soluble in ethyl ether, ethanol, and concentrated sulfuric acid. It has a maximum absorption in the ultraviolet region that indicates the presence of unsaturation. Its infrared …

Synthesis of piperidine-2-carboxylic acid (pipecolic acid) in soybean was studied by infiltration of radioactive carbon labeled precursor and by experiments using enzyme extracts from control and fluoride-fumigated plants.
Pipecolic acid was synthesized from lysine in both control and treated plants. Normal plants, in which pipecolic acid had not been detected by conventional procedures, were capable of synthesizing pipecolic acid. Greater pipecolic acid synthesis was shown in treated plants than in controls, and was attributed to increased enzyme concentration induced by fluoride treatment.

Guinea pig brain cortex slices incubated in media containing U-14-aspartate or glutamate (GA) (J. Neurochem. in press) form glutamine ( GM) of relative specific activity (RSA)> 1 (GA=l). In the presence of 10-3 fluoroacetate (FA) the RSA of GM decreased to values considerably < 1 without any changes in levels of amjno acids; the % of label in GA doubled whilA that in GM decreased correspondingly. Similar results were obtained with 1-14 C-acetate as tracer in vivo (mice) and in vitro. RSA' s of Grvl > 1 have been explained by the compartmentation of GA in cerebral cortex. Consequently the effect of GA may be explained by an inhibition of the flow of metabolites through that pool of GA used preferentially for GM synthesis. Evidence suggested there might exist in brain a contractile protein: 1) actomyosin contains Nacetyl aspartic acid; brain contains a high concentration of the free compound, 2) actomyosin functions as a Mg dependent ATPase; brain contains a Mg dependent ATPase, 3) actomyosin-like contractile proteins have been described in cells, membranes, and tissues other than muscle. Therefore whole rat brains were extracted with 0.6M KCl and the extract diluted to O.lM KCl. The precipitate exhibited superprecipitation (SP) properties which required both Mg and ATP. SP was inhibited by p-chloro-mercuribenzene sulfonic acid (10- M) but not by ouabain (10 M). The protein purified …

(1) Compartmentation of glutamate metabolism in brain cortex previously observed only in vivo, has now been demonstrated in vitro. This was shown by using [U-14C]aspartate and [U-14C]glutamate as tracer substrates. (2) Preparation and maintenance of the slices at 0° resulted in reversible inhibition of glutamine synthesis. Preincubation at 37° for 10 min or preparation of the slices at room temperature partially overcame this inhibition. (3) Transfer to fresh medium after preincubation had an added stimulatory effect on glutamine synthesis. (4) Incubation in high K+ medium (27 mM) altered the relative specific activity of glutamine. THE judicious use of sulphur dioxide is perhaps the most important of the fundamentals of winemaking in a hot climate.
Western Australia's vintage, centred in the Swan Valley just a few miles north-east of Perth, is subjected to some of the highest summer temperatures experienced by a quality wine producing district.
In this environment, it is important to make good use of sulphur dioxide to avoid wine spoilage.

The survey of plant species for aminolevalinic acid synthetase (ALAS) source was conducted. Under the conditions of this study, spinach leaves were found to be a good source of ALAS activity. The amount of amino levulinic acid (ALA) synthesized by the plant extracts was very small and was rapidly metabolized to porphyrins. ALA and porphyrin synthesis was found to be confined to the particulate fractions. Rupturing of the particles with ultrasonifier enhanced the penetration of succinyl-CoA and increased the activity. A method has been described to suspend seedling wheat roots in a saturated atmosphere. Wheat (Triticum vulgate, L.) seeds were germinated in aluminum trays and the seedlings transferred to seedling chambers. After a day's growth, the roots were treated with various concentrations of solutions containing CaSO4, KCl, KF, NaCL, NaF, CH3COONa, FCH2COONa, K2C2O4 and Na2EDTA and observations were recorded 24 and 48 hours after treatment. A study was made of the urinary excretion of estrogens of healthy university students consuming self-selected diets living under home conditions (4 men and 5 women). Two 31-day test periods were used. During the two test periods, two meal patterns were used; three meals a day or two meals a day with no lunch. Weight was kept constant. Calories, protein, carbohydrate, and fat of their diets were calculated.
Urine specimens were collected twice a week, and additional samples were also collected on specified days for female subjects. Urinary estrogens were determined by using Brown's method (1955, 1957) with some modifications. …

Soybean plants were fumigated in HF and the organic acid-fractions of the leaf extracts were compared to those of the leaf extracts from soybean plants grown in nutrient solution containing fluoroacetate. The extracts from both treatments contained fluoroorganic compounds that were chromatographically similar to fluoroacetate and fluorocitrate.
The fluorocitrate separated from HF fumigated leaves was found to inhibit aconitase prepared from pig-heart. This inhibition was similar to that caused by commercial fluorocitrate and the fluorocitrate synthesized using guinea-pig kidney. The aconitase from soybean leaves was found to be sensitive to fluorocitrate as the aconitase that isolated from pig-heart. Inorganic fluoride …

Fourteen compounds were isolated from an alkaline hydrolyzed extract of alfalfa leaves by using cellulose thin layer plates. The Rf values, fluorescent characteristics, and color reactions were given for all the compounds isolated.
Eight of these were identified as trans-ferulic acid, trans-p-coumaric acid, trans-caffeic acid, o-coumaric acid, tricin, syringic acid, cis-ferulic acid and cis-p-coumaric acid. The identity of the compounds was confirmed by comparison of the Rf values on thin layer chromatograms, fluorescent characteristics under ultraviolet light, and color reactions of spray reagents with those of authentic samples. The purpose of this paper is to summarize the results of my research on the enzymic steps of cholesterol biosynthesis (Dempsey, 1962; 1964; 1965; Dempsey, Seaton, and Trockmon,1963; Dempsey, Seaton, Sanford, and Trockmon,19640; Dempsey, Seaton, Schroepfer, and Trackman, 19646.2 These data were obtained by use of an enzyme system isolated from rat liver homogenates. The findings have elucidated the pathways of and cofactor requirements for cholesterol synthesis from naturally occurring sterol precursors.

Guinea pig brain cortex slices incubated in media containing U-C14 aspartic acid form glutamine of higher specific activity than glutamic acid (Fed. Proc. -25, 713 (1966) ). Glutamic acid and glutamine from such experiments were enzymatically decarboxylated and the o/o of label in C-1 ranged from 37 to 42o/o. These values exceed the maximum predicted from the known operation of the citric acid cycle viz. 33o/o. When l-C14 acetate was injected into mice or incubated with brain slices the aspartate isolated from brain had more than 50o/o of the label in C-4. From the known operation of the citric acid …

The determination of urinary 3-methoxy-4-hydroxyphenylethylene glycol (HMPG), a major metabolite of epinephrine and norepinephrine by gas-liquid chromatography and electron capture detection is described. A purified extract of HMPG was treated with trifluoroacetic anhydride to form the highly sensitive HMPG-trifluoroacetate derivative. Chromatography was performed on a 6% QF-I- 0065 column at 158°. Recovery was found to be 55·4% using [3H]HMPG. Two sets of eight identical samples demonstrated a standard deviation from the mean of 6.9% and 5.7%. Thirty-five normal human urine specimens assayed by this procedure gave a mean HMPG excretion of 0.86 #gfmg creatinine (range 0.25-1.49). Six urine samples from …

Uredospores of Puccinia gramintis (Pers.) tritici (Eriks. and Henn.) were uniformly labeled with 14C by permitting the host (Triticuml aestivum L.) to carry out photosynthesis in 14CO2 during the process of spore production by the obligate parasite. The use of 14C labeled spores provided advantages in a study of the utilization of endogenous substrates at frequent intervals with small amounts of spores under conditions conducive to germination.

On The Mechanism Of The Thiamine Catalyzed Reductive Dimerization Of Farnesyl Pyrophosphate To Squalene., Horatio D. Durst

The mechanism of the dimerization of farnesyl pyrophosphate to squalene was investigated in detail. Thiamine was found to catalyze this dimerization in yeast-soluble enzyme preparations. Removal of thiamine was found to cause the buildup of farnesyl pyrophosphate in the enzyme system. Derivatives of thiamine were tested to examine their effect on the dimerization reaction and were found to be inactive. This study was undertaken to provide a circumscription of the genus Ficus in Venezuela. The material studied was representative of the collections that have been made throughout the country. A great portion of Venezuela is apparently unexplored botanically. With this in mind I have attempted to do as complete a study as possible with the material available with fully realizing the many limitations. this circumscription is intended as a starting point for future work,not a stopping place.

The size of explants in organ culture is limited by their ability to feed, respire, and excrete by diffusion. The perfusion procedure described in this report is not limited to a particular explant size. The test tissue was the adult human tooth.
In order to maintain the pulp tissue of the intact human tooth in organ culture, special precautions were taken to prevent internal necrosis and to allow diffusion of nutrients and waste products.
