The present work studied the optimization of aeration rate, agitation rate and oxygen transfer and the use of various batch fermentation strategies for xylanase production from a recombinant Aspergillus nidulans strain in a 3 L stirred tank reactor. Maximum xylanase production of 1250 U/mL with productivity of 313 U/mL/day was obtained under an aeration rate of 2 vvm and an agitation rate of 400 rpm using batch fermentation. The optimum volumetric oxygen transfer coefficient (kLa) for efficient xylanase production was found to be 38.6 h1. Fed batch mode and repeated batch fermentation was also performed with kLa was 38.6 h1. …

Land application of manure introduces gastrointestinal microbes into the environment, including bacteria carrying antibiotic resistance genes (ARGs). Measuring soil ARGs is important for active stewardship efforts to minimize gene flow from agricultural production systems; however, the variety of sampling protocols and target genes makes it difficult to compare ARG results between studies. We used polymerase chain reaction (PCR) methods to characterize and/or quantify 27 ARG targets in soils from 20 replicate, long-term no-till plots, before and after swine manure application and simulated rainfall and runoff. The disclosure is directed to intrapleural air leak detection and monitoring. According to various embodiments of the disclosure, an air leak may be detected utilizing at least one sensor to determine whether fluid extracted from a pleural cavity of a patient includes carbon dioxide and/or a second substance. The second substance may be a foreign substance inhaled by the patient to confirm presence of the air leak. Hydrogen peroxide (H2O2) is an important molecule within the human body, but many of its roles in physiology and pathophysiology are not well understood. To better understand the importance of H2O2 in biological systems, it is essential that researchers are able to quantify this reactive species in various settings, including in vitro, ex vivo and in vivo systems. Grain yield, crop evapotranspiration (ETc), basal evapotranspiration (ETb), ETc-yield production functions (ETYPF), and crop water use efficiency (CWUE) response of three drought-tolerant (DT) and one non-drought-tolerant (NDT) maize (Zea mays L.) hybrids to two plant population densities (PPDs) [84,000 plants ha-1 (high PPD) and 59,300 plants ha-1 (low PPD)] and three irrigation levels were researched at two semi-arid locations: North Platte (WCREC) and Scottsbluff (MAL), Nebraska, in 2010, 2011, and 2012. The 2017 M7.1 Central Mexico Earthquake caused significant infrastructural damage in the Mexico City area. The earthquake contained a significant pulse in the long period, resulting in numerous buildings severely damaged or collapsed. This paper discusses a reinforced concrete building which was still partially occupied post-earthquake. The building’s interior walls were examined to have substantial damage, including some extensive cracking. In January 2018, the authors visited the structure and collected detailed assessment data. Magnetic Co-doped Fe3O4@FeOOH nanocomposites were prepared in one step using the hydrothermal synthesis process for catalyzing peroxymonosulfate (PMS) to degrade refractory methylene blue (MB) at a wide pH range (3.0–10.0). The catalysts' physiochemical properties were characterized by different equipment; Fe3+/Fe2+ and Co3+/Co2+ were confirmed to coexist in the nanocomposite by X-ray photoelectron spectroscopy. The nanocomposite effectively catalyzed PMS's decoloration (99.2%) and mineralization (64.7%) of MB. In this study, the hydroxyapatite powder is investigated for both of methylene blue and thymol blue in aqueous solution. The physical and chemical properties of the adsorbent were evaluated systematically using the different techniques including Microsoft Excel programming, linear regression model and also the coefficient of determination. Batch adsorption experiments were conducted to determine the effect of contact time, solution pH, initial dye concentrations, and also the adsorbent dosage on adsorption. Crop residue is an abundant resource for the potential production of biofuels, but a better understanding of its use on net carbon emissions must be developed to mitigate climate change. Small RNAs (sRNAs) remain an understudied class of regulatory molecules in bacteria in general and in Gram-positive bacteria in particular. In the major human pathogen Staphylococcus aureus, hundreds of sRNAs have been identified; however, only a few have been characterized in detail. In this study, we investigate the role of the sRNA Teg41 in S. aureus virulence. We demonstrate that Teg41, an sRNA divergently transcribed from the locus that encodes the cytolytic alpha phenolsoluble modulin (αPSM) peptides, plays a critical role in αPSM production. Hyperinsulinemia affects 72% of Fanconi anemia (FA) patients and an additional 25% experience lowered glucose tolerance or frank diabetes. The underlying molecular mechanisms contributing to the dysfunction of FA pancreas β cells is unknown. Therefore, we sought to evaluate the functional role of FANCA, the most commonly mutated gene in FA, in glucosestimulated insulin secretion (GSIS). This study reveals that FANCA or FANCB knockdown impairs GSIS in human pancreas β cell line EndoC-βH3. The objective of this study is to characterize the micromechanical properties of poly-L-lactic acid (PLLA) composites reinforced by grade 420 stainless steel (SS) particles with a specific focus on the interphase properties. The specimens were manufactured using 3D printing techniques due to its many benefits, including high accuracy, cost effectiveness and customized geometry. Mg/Nb nanolaminates synthesized through vapor deposition techniques exhibit high flow strength without conventional twinning in Mg. In this work, we investigated the influence of laminated microstructures on deformation mechanisms of Mg nanolayers. Using molecular dynamics simulations, we explored that (0001)-oriented Mg layers transform or re-orient to {10¯10}-oriented Mg layers through nucleation and growth of {10¯12} twins by atomic shuffling, instead of conventional {10¯12} twinning shear. Such a reorientation accommodates in-plane compressive strain and out-of-plane tensile strain when Mg/Nb laminates are subjected to compression parallel to the Mg/Nb interfaces. In this work, the peridynamic corrosion model is used for 3D simulation of pitting corrosion in stainless steel. Models for passivation and salt layer formation are employed to predict detailed characteristics of pit growth kinetic in stainless steels, such as lacy cover formation on top of the pit, and the diffusion-controlled regime at the pit bottom. The model is validated against an experimentally grown pit on 316L stainless steel in NaCl solution. Lacy covers in this model are formed autonomously during the simulation process. They are remarkably similar to the covers observed on top of the real pits.

In this work, the peridynamic corrosion model is used for 3D simulation of pitting corrosion in stainless steel. Models for passivation and salt layer formation are employed to predict detailed characteristics of pit growth kinetic in stainless steels, such as lacy cover formation on top of the pit, and the diffusion-controlled regime at the pit bottom. The model is validated against an experimentally grown pit on 316L stainless steel in NaCl solution. Lacy covers in this model are formed autonomously during the simulation process. They are remarkably similar to the covers observed on top of the real pits.

The study of propagating, evanescent and complex wavenumbers of guided waves (GWs) in high-performance composites using a stable and robust semi-analytical finite element (SAFE) method is presented. To facilitate understanding of the wavenumber trajectories, an incremental material change study is performed moving gradually from isotropic aluminum alloy to carbon fiber reinforced polymer (CFRP) composites. The SAFE results for an isotropic aluminum alloy plate are compared with the exact analytical solutions, which shows that N = 20 SAFE elements across the thickness provides <0.5% error in the highest evanescent wavenumber for the given frequency-wavenumber range. The material change study reveals that reducing the transverse and shear moduli moves the wavenumber solution towards one similar to composite material. The comparison of the propagating, evanescent and complex wavenumber trajectories between composites and aluminum alloy show that antisymmetric imaginary Lamb wave modes always exist in composites although they may not exist in isotropic aluminum alloy at some frequencies. The wavenumber trajectories for a unidirectional CFRP plate show that the range of real wavenumber is much smaller than in the isotropic aluminum alloy. For laminated CFRP composite plates (e.g., unidirectional, off-axis, transverse, cross-ply and quasi-isotropic laminates), the quasi Lamb wave and shear horizontal (SH) wave trajectories are also identified and discussed. The imaginary SH wave trajectories in laminated composites are distorted due to the presence of ±45 plies. The convergence study of the SAFE method in various CFRP laminates indicates that sufficient accuracy can always be achieved by increasing the number of SAFE elements. Future work will address the stress-continuity between composite layers.

This longitudinal study investigated development in expectancy for success (perceived competence), 3 types of task value (utility, interest, attainment), and 3 types of perceived cost (opportunity, effort, psychological) for engineering students during their first 2 years of college. Latent growth curve models indicated declines in expectancy and values, with attainment value declining more slowly than expectancy, interest value, and utility value. Costs increased over time, with effort cost increasing more rapidly than psychological cost. Demographic differences were observed in initial levels of motivation, but not in rates of change over time. Students with slower declines in expectancy and value and …

Subsurface volatile organic compounds (VOCs) can pose risks to human and environmental health and mediate biological processes. Volatile organic compounds have both anthropogenic and biogenic origins, but the relative importance of these sources has not been explored in subsurface environments. This study synthesized 17 yr of VOC data from the Amargosa Desert Research Site in Nevada with the goal of improving understanding of spatial and temporal variations that distinguish sources of VOCs from a landfill and from ambient sources including biogenic VOCs (bVOCs). Scanning Kelvin probe force microscopy (SKPFM) is used in corrosion studies to quantify the relative nobility of different microstructural features present within complex metallic systems and thereby elucidate possible corrosion initiation sites. However, Volta potential differences (VPDs) measured via SKPFM in the literature for metal alloys exhibit large variability, making interpretation and application for corrosion studies difficult. We have developed an improved method for referencing SKPFM VPDs by quantifying the closely related work function of the probe relative to an inert gold standard whose modified work function is calculated via density functional theory (DFT).
