Considering the disadvantages of the costly, laborious, and time-consuming traditional clinical analyses, there is a tendency of developing non-hospital point-of-care which allows patients to monitor their health conditions conveniently at home. To optimize the convenience of the point-of-care instruments, a wearable device with the ability of detecting different biological components in different biological conditions is preferer. The device should also have the qualities of rapid analyses, high accuracy, and high sensitivity. Training methods for both shallow and deep neural nets are dominated by first order algorithms related to back propagation and conjugate gradient. However, these methods lack affine invariance so performance is damaged by nonzero input means, dependent inputs, dependent hidden units and the use of only one learning factor. This dissertation reviews affine invariance and shows how MLP training can be made partially affine invariant when Newton's method is used to train small numbers of MLP parameters. Several novel methods are proposed for scalable partially affine invariant MLP training. The potential application of the algorithm to deep learning is discussed. …

The research focuses on the mathematical modeling and control of an unmanned aerial vehicle with cable suspended payload. A comprehensive mathematical model is derived for a quadcopter with a cable suspended payload using the Newton-Euler method and the Euler-Lagrange formulation. These methods assume that the cable is massless and always taut and cannot be used to simulate the cases when the cable is flexible. Hence, an alternative approach to model the flexibility of the cable is presented using Lagrangian mechanics by approximating the cable to be a chain of serially connected links. Cavitation appears when local static pressure drops below the water vapor pressure and causes significant impacts on marine propeller blades. The Volume of Fluid (VOF) method, which is also referred to as the Transport Equation-based Model (TEM), has been extensively adopted in the cavitating flow simulations. Piecewise Linear Interface Calculation (PLIC) schemes have been extensively employed in the VOF method for interface capturing in numerical simulations of multiphase flows. This study investigates the feasibility of using continuous wave lasers and high-speed CMOS cameras together in 2D particle image velocimetry (PIV) which is a widely-used non-intrusive method in experimental aerodynamics. In order to validate the setup, laminar and turbulent boundary layers over a flat plate with elliptical leading edge are examined in a low-speed wind tunnel. Before the experiments, the suitability of the elliptical-nose flat plate is determined (i.e. pressure gradient over the surface and separation), and the free-stream conditions (flow velocity and test section turbulence) are measured. CCL2 is an inflammatory mediator that is released by tumor cells to activate and direct immune cell species, especially macrophages, to inflammatory sites within the body. The goal of this project was to successfully generate knockout the CCL2 ligand gene using a CRISPR/Cas9 complex delivered via lipid mediated transfection. The sgRNA and Cas9 mRNA were introduced into the cells via lipid-mediated transfection. The cells were incubated for 4 days, before being analyzed using PCR and gel electrophoresis. High performance cooling is a critical requirement for modern microprocessors that generate large amounts of non-uniformly distributed heat. Metal heat spreaders and heat sinks have been traditionally used for thermal management. While Li-ion cells show outstanding electrochemical performance, their poor thermal transport characteristics results in reduced performance as well as significant safety concerns. The heterogeneous interface between cathode and separator plays a vital role in the dissipation of heat within a Li-ion cell. Previous studies showed that the cathode-separator interfacial thermal resistance contributes around 88% of total thermal resistance within the cell. In this research, thermal conductance across the cathode-separator interface is calculated using molecular dynamics simulations. Ultrasound switchable fluorescence (USF) imaging has been proposed as a novel imaging modality which provides microscopic fluorescence imaging in centimeter-deep tissue. USF is possible to reveal many interesting phenomena such as microcirculation, tumor angiogenesis, and cancer metastasis and diagnose early-stage cancer. USF adopts a focused ultrasound (FU) to repetitively switch “on” fluorescent agents in tissue, and the USF photons scatter out and are captured by a detector. Despite breakthroughs in the field of neuroimaging, there remain many unanswered questions about brain functions and activities and their underlying physiological processes. In my dissertation research, I examined brain functions while human subjects experienced four vigilance states: (1) resting state with eyes open, (2) resting awake state with eyes closed, (3) sleep stage 1 and (4) sleep stage 2. Simultaneous dual modality measurements using electroencephalography (EEG) and functional near infrared spectroscopy (fNIRS) were acquired during the aforementioned vigilance states. My research focused on applying biomedical engineering techniques to diagnose different diseases and conditions. The results of these work led to one published paper and two submitted manuscripts which are summarized in Chapters 2, 3 &4. Briefly, in Chapter 2, my work laid emphasis on developing a method for early detection of cartilage injury. An apoptotic cell-detecting probe was fabricated by the conjugation of the apoptotic cell-binding peptide (CQRPPR) and a sulfo-cyanine7 N-hydroxysuccinimide ester (Cy7 dye). Blast-induced traumatic brain injury (bTBI) is a serious concern among military personnel and their families. Although the mechanisms responsible for disruption to the brain are not well understood, the development of reliable diagnosis along with effective therapeutic treatment is urgently warranted. Recent findings suggest that shockwaves produced by a blast can generate micron-size bubbles that subsequently collapse in the brain tissue. The collapse of microbubbles (referred to as microcavitation) may compromise the integrity of the blood-brain barrier (BBB). Utility managers and owners have challenges when addressing appropriate intervals for inspection of gravity sanitary sewer pipelines and other underground pipeline systems. Frequent inspection of sewer network is not cost-effective due to large inventory of pipes and high cost of inspections, such as using closed-circuit television (CCTV) surveys. The effects of a temporally modulated, distributed cathode in a linear format crossed-field amplifier (CFA) are simulated in VSim and analyzed. A linear format, 150 MHz, low power (100 W), moderate gain (7 dB), meander line CFA is used as the basis for the simulation model. This paper describes simulations with different time-varying distributed cathodes in which electron injection is modulated at the RF frequency both in and out of phase with the RF input. At low RF input power the modulated electron injection dominates the operation. Injecting in phase with the RF input shows gain increases from 23 dB …
