The Level Of Lactic Dehydrogenase Activity As An Indicator Of The Growth Of Influenza Virus In The Embryonate Egg, Richard Kelly, Donald Greiff

The relation, between time and the levels of lactic dehydrogenase activity of chorioallantoic fluids from embryonate eggs infected with the PR8 strain of influenza virus were determined quantitatively. The mean values, based on 10 determinations for each time interval, followed a sigmoid curve, with the greatest rates of change occurring between 48 and 72 hours after the inoculation of virus. The activities of the fluids from infected eggs at the 72nd hour or later were approximately 18 times higher than those from non-infected eggs. Human oxalated plasma stored at 4° C. until the prothrombin time is increased beyond 60 sec. is a reliable medium for assaying labile factor (factor V) because its response to added labile factor corresponds quantitatively to that of plasma from patients with congenital deficiency of this factor. Such an agreement is not obtained with plasma stored at 37°C. The stability of labile factor is closely associated with ionized calcium. The addition of thrombin to fresh oxalated plasma causes an apparent hyperactivity of labile factor, but this is completely removed by adsorption with Ca3(PO)2. Oxalated plasma when …

The Effects Of X-Rays And Beta Rays (Tritium) On The Growth Of Rickettsia Mooseri And Rickettsia Akari In Embryonate Eggs, Donald Greiff, E. L. Powers, Walter E. Kisieleski, Henry Pinkerton

The growth of Rickettsia mooseri was accelerated and quantitatively increased in embryonate eggs containing tritium oxide at levels of 180, 90, and 45 mc./egg during the growth period. The eggs of a group containing 22.5 mc./egg showed only a slight increase in the rate of growth of organisms; the infections in the eggs of a group given 11.2 mc./egg did not differ significantly from those of the control group. 1. The widely accepted theory that calcium participates in the coagulation mechanism in the form of Ca++ and acts as a catalyst is not in accord with several important experimental findings:

(a) The anticoagulant action of sodium oxalate is much slower than the precipitation of ionized calcium as the oxalate salt.

(b) Sodium citrate begins to depress prothrombin activity at a concentration at which ionized calcium is still present. The inability of tricalcium phosphate to adsorb prothrombin from citrated plasma indicates that citrate forms a complex with prothrombin and it is postulated that prothrombin is …
