Salmonella enterica is, globally, an important cause of human illness with beef being a significant attributable source. In the human patient, systemic Salmonella infection requires antibiotic therapy, and when strains are multidrug resistant (MDR), no effective treatment may be available. MDR in bacteria is often associated with the presence of mobile genetic elements (MGE) that mediate horizontal spread of antimicrobial resistance (AMR) genes. In this study, we sought to determine the potential relationship of MDR in bovine Salmonella isolates with MGE. The present study involved 111 bovine Salmonella isolates obtained collectively from specimens derived from healthy cattle or their environments …

Lactogenic immunity is important for the protection of piglets against many pathogens including porcine epidemic diarrhea virus. Circulating neutralizing antibodies levels in sow sera may help determine if a detectable immune response could confer protection to piglets. Neutralizing antibodies can be detected through various diagnostic assays. This study evaluated the diagnostic characteristics of two neutralizing antibody assays for porcine epidemic diarrhea virus neutralizing antibodies in serum of challenged gilts. Four treatment groups, control, non-vaccinated, vaccinated prior to challenge, and vaccinated following challenge, were comprised of 20 gilts. Serum sample were collected from each gilt prior to and following challenge with …

An Evaluation Of Avian Influenza Virus Whole-Genome Sequencing Approaches Using Nanopore Technology, Hon S. Ip, Sarah Uhm, Mary Lea Killian, Mia K. Torchetti

United States Department of Agriculture Wildlife Services: Staff Publications

As exemplified by the global response to the SARS-CoV-2 pandemic, whole-genome sequencing played an important role in monitoring the evolution of novel viral variants and provided guidance on potential antiviral treatments. The recent rapid and extensive introduction and spread of highly pathogenic avian influenza virus in Europe, North America, and elsewhere raises the need for similarly rapid sequencing to aid in appropriate response and mitigation activities. To facilitate this objective, we investigate a next-generation sequencing platform that uses a portable nanopore sequencing device to generate and present data in real time. This platform offers the potential to extend in-house sequencing …

Red blood cell (RBC) transfusion is associated with recipient inflammation and infection, which may be triggered by excessive circulating iron. Iron chelation following transfusion may reduce these risks. The aim of this study was to evaluate the effect of deferoxamine on circulating iron and inflammation biomarkers over time and in vitro growth of Escherichia coli (E. coli) following RBC transfusion in dogs with atraumatic hemorrhage. Anesthetized dogs were subject to atraumatic hemorrhage and transfusion of RBCs, then randomized to receive either deferoxamine or saline placebo of equivalent volume (n = 10 per group) in a blinded fashion. Blood was sampled …

Consumption of the total Western diet (TWD) in mice has been shown to increase gut inflammation, promote colon tumorigenesis, and alter the fecal microbiome composition in mice as com-pared to mice fed a healthy diet, AIN93G (AIN). However, it is unclear whether the gut micro-biome contributes directly to colitis-associated CRC in this model. The objective of this study was to determine whether dynamic fecal microbiota transfer (FMT) from host mice fed either AIN or TWD basal diets would alter colitis symptoms or colitis-associated CRC in recipient mice, which were fed either AIN or TWD directly using a 2x2 factorial experiment …

Displaying And Delivering Viral Membrane Antigens Via Ww Domain–Activated Extracellular Vesicles, Sengjin Choi, Zhiping Yang, Qiyu Wang, Zhi Qiao, Maoyun Sun, Joshua Wiggins, Shi-Hua Xiang, Quan Lu

School of Veterinary and Biomedical Sciences: Faculty Publications

Membrane proteins expressed on the surface of enveloped viruses are conformational antigens readily recognized by B cells of the immune system. An effective vaccine would require the synthesis and delivery of these native conformational antigens in lipid membranes that preserve specific epitope structures. We have created an extracellular vesicle–based technology that allows viral membrane antigens to be selectively recruited onto the surface of WW domain–activated extracellular vesicles (WAEVs). Budding of WAEVs requires secretory carrier-associated membrane protein 3, which through its proline-proline-alanine-tyrosine motif interacts with WW domains to recruit fused viral membrane antigens onto WAEVs. Immunization with influenza and HIV viral …

Vaccines Against Group B Coxsackieviruses And Their Importance, Kiruthiga Mone, Ninaad Lasrado, Meghna Sur, Jay Reddy

School of Veterinary and Biomedical Sciences: Faculty Publications

The group B coxsackieviruses (CVBs) exist in six serotypes (CVB1 to CVB6). Disease associations have been reported for most serotypes, and multiple serotypes can cause similar diseases. For example, CVB1, CVB3, and CVB5 are generally implicated in the causation of myocarditis, whereas CVB1 and CVB4 could accelerate the development of type 1 diabetes (T1D). Yet, no vaccines against these viruses are currently available. In this review, we have analyzed the attributes of experimentally tested vaccines and discussed their merits and demerits or limitations, as well as their impact in preventing infections, most importantly myocarditis and T1D.

Despite progress, 2-year pancreatic cancer survival remains dismal. We evaluated a biomarker-driven, combination/N-of-one strategy in 18 patients (advanced/metastatic pancreatic cancer) (from Molecular Tumor Board). Targeted agents administered/ patient = 2.5 (median) (range, 1–4); first-line therapy (N = 5); second line, (N = 13). Comparing patients (high versus low degrees of matching) (matching score ≥50% versus <50%; reflecting number of alterations matched to targeted agents divided by number of pathogenic alterations), survival was significantly longer (hazard ratio [HR] 0.24 (95% confidence interval [CI], 0.078–0.76, P = 0.016); clinical benefit rates (CBR) (stable disease ≥6 months/partial/complete response) trended higher (45.5 vs 0.0%, P = 0.10); progression-free survival, HR, 95% CI, 0.36 (0.12–1.10) (p = 0.075). First versus ≥2nd-line therapy had higher CBRs (80.0 vs 7.7%, P = …

Whole Genome Sequencing For Identification Of Putative Causative Alleles For Hemochromatosis In Horses, Anna M. Rauber-Ramos 2023 South Dakota State University

Whole Genome Sequencing For Identification Of Putative Causative Alleles For Hemochromatosis In Horses, Anna M. Rauber-Ramos

Schultz-Werth Award Papers

Hemochromatosis is liver disorder in which excess iron is stored in the liver, leading to iron overload and extensive liver damage that can prove fatal. Based on human precedence, a genetic cause for idiopathic hemochromatosis in horses is suspected. Two horses with hemochromatosis were whole genome sequenced, with variants in candidate genes prioritized in analyses of potential pathogenicity. No clear putative variants were found to be present in both horses, suggesting an etiology more complex than the initially suspected Mendelian inheritance. A heterozygous nonsense mutation in STEAP3 metalloreductase (STEAP3) was identified as a potential contributor to hemochromatosis in one of …

Animal Shelter Management Of Feline Leukemia Virus And Feline Immunodeficiency Virus Infections In Cats, Julie Levy, Cynda Crawford, Keegan Spera, E. Susan Amirian, Paola Dezubiria

Diseases and Health

Approximately 5% of cats in animal shelters in the United States test positive for either feline leukemia virus (FeLV) or feline immunodeficiency virus (FIV), which translates to more than 100,000 positive cats managed by shelters each year. Little is known about the current status of retroviral management in animal shelters, particularly in regions burdened by chronic pet overpopulation and high shelter admissions, such as the southern United States. The purpose of this study was to describe feline retroviral management in Florida shelters. Shelters were surveyed on practices including selection of cats for testing, diagnostic techniques, and outcome options for cats …

Environmental Transmission Of Influenza A Virus In Mallards, Kim M. Pepin, Clinton B. Leach, Nicole L. Barrett, Jeremy W. Ellis, Kaci K. Vandalen, Colleen T. Webb, Susan A. Shriner

United States Department of Agriculture Wildlife Services: Staff Publications

Abstract

Influenza A viruses present a major challenge for animal and human health. They circulate widely in wild waterfowl and frequently spillover into poultry, emphasizing the need for risk-based surveillance in wild birds and an understanding of the relative importance of different transmission mechanisms. We addressed this objective with a replicated (N = 6) experimental infection study in which we serially exposed eight cohorts of four naïve contact mallards to an experimentally infected mallard and a shared water pool. Viral concentration in the water was a better predictor of transmission than several direct measures of viral shedding in the …

Changes In Wild Pig (Sus Scrofa) Relative Abundance, Crop Damage, And Environmental Impacts In Response To Control Efforts, Joseph W. Treichler, Kurt C. Vercauteren, Charles R. Taylor, James C. Beasley

United States Department of Agriculture Wildlife Services: Staff Publications

Background

As the population and range of wild pigs (Sus scrofa) continue to grow across North America, there has been an increase in environmental and economic damages caused by this invasive species, and control efforts to reduce damages have increased concomitantly. Despite the expanding impacts and costs associated with population control of wild pigs, the extent to which wild pig control reduces populations and diminishes environmental and agricultural damages are rarely quantified. The goal of this study is to quantify changes in wild pig relative abundance and subsequent changes in damages caused by invasive wild pigs in response to …

Individual‑Level Patterns Of Resource Selection Do Not Predict Hotspots Of Contact, Anni Yang, Raoul Boughton, Ryan S. Miller, Nathan P. Snow, Kurt C. Vercauteren, Kim C. Pepin, George Wittemyer

United States Department of Agriculture Wildlife Services: Staff Publications

Contact among animals is crucial for various ecological processes, including social behaviors, disease transmission, and predator–prey interactions. However, the distribution of contact events across time and space is heterogeneous, influenced by environmental factors and biological purposes. Previous studies have assumed that areas with abundant resources and preferred habitats attract more individuals and, therefore, lead to more contact. To examine the accuracy of this assumption, we used a use-available framework to compare landscape factors influencing the location of contacts between wild pigs (Sus scrofa) in two study areas in Florida and Texas (USA) from those influencing non-contact space use. …
