Indications exist that paramagnetic calcium phosphates may be able to promote regeneration of bone faster than their regular, diamagnetic counterparts. In this study, analyzed was the influence of paramagnetic cobalt-substituted hydroxyapatite nanoparticles on osteoporotic alveolar bone regeneration in rats. Simultaneously, biocompatibility of the material was tested in vitro, on osteoblastic MC3T3-E1 and epithelial Caco-2 cells in culture. The material was shown to be biocompatible and nontoxic when added to epithelial monolayers in vitro, while it caused a substantial decrease in the cell viability as well as deformation of the cytoskeleton and cell morphology when incubated with the osteoblastic …

Menthol is a common compound in pharmaceutical and commercial products and a popular additive to cigarettes. The molecular targets of menthol remain poorly defined. In this study we show an effect of menthol on the α7 subunit of the nicotinic acetylcholine (nACh) receptor function. Using a two-electrode voltage-clamp technique, menthol was found to reversibly inhibit α7-nACh receptors heterologously expressed in Xenopus oocytes. Inhibition by menthol was not dependent on the membrane potential and did not involve endogenous Ca2+-dependent Cl− channels, since menthol inhibition remained unchanged by intracellular injection of the Ca2+ chelator BAPTA and perfusion with Ca2+-free bathing solution containing …

Metachromatic leukodystrophy (MLD) is a lysosomal storage disorder of the brain caused by mutations in the gene encoding the lysosomal sulfatase, arylsulfatase A (ASA). It is not possible to treat the brain in MLD with recombinant ASA, because the enzyme does not cross the blood-brain barrier (BBB). In the present investigation, a BBB-penetrating IgG-ASA fusion protein is engineered and expressed, where the ASA monomer is fused to the carboxyl terminus of each heavy chain of an engineered monoclonal antibody (MAb) against the human insulin receptor (HIR). The HIRMAb crosses the BBB via receptor-mediated transport on the endogenous BBB insulin receptor, …

alpha-Glucosidase inhibitor has known to be a therapeutic agent for diabetes mellitus (DM) treatment, especially type 2 DM. Based on previous studies. There are various plants that have the effect of inhibiting the activity of a-glucosidase, one of which is garu leaves (Antidesma montanum Blume). This research aimed to get the fraction which had the highest Il-glucosidase inhibiting activity from ethanol extract of garu leaves and identify the chemical compounds from the most active fraction. Simplisia powder was extracted by maseration using 80% ethanol then fractionated using n-hexane, ethyl acetate, and methanol. Inhibitory activity test was performed by measuring absorbance …

Over 4000 flavonoids have been identified so far and among these, many are known to have antitumor activities. The basis of the relationships between chemical structures, type and position of substituent groups and the effects these compounds exert specifically on cancer cells are not completely elucidated. Here we report the differential cytotoxic effects of two flavone isomers on human cancer cells from breast (MCF7, SK-BR-3), colon (Caco-2, HCT116), pancreas (MIA PaCa, Panc 28), and prostate (PC3, LNCaP) that vary in differentiation status and tumorigenic potential. These flavones are derived from plants of the family Asteraceae, genera Gnaphalium and Achyrocline reputed …

Selectivity Of The Ubiquitin-Binding Modules, Simin Rahighi, Ivan Dikic
Pharmacy Faculty Articles and Research
Ubiquitin-binding modules are constituents of cellular proteins that mediate the effects of ubiquitylation by making transient, non-covalent interactions with ubiquitin molecules. While some ubiquitin- binding modules bind single ubiquitin moieties, others are selective for specific ubiquitin chains of different linkage types and lengths. In recent years, functions of ubiquitin chains that are polymerized through their Lys or N-terminal Met (i.e. linear chains) residues have been linked to a variety of cellular processes. Selectivity of ubiquitin-binding modules for different ubiquitin chain types appears as a key to the distinct regulatory consequences during protein quality control pathways, receptor endocytosis, gene transcription, signaling …

Diabetes mellitus (DM) is a group of metabolic disorders characterized by hyperglicemia and associated with abnormalities in carbohydrate, fat, and protein metabolisms. Patients with DM in the world continue to increase along with population growths. Starting from this condition, searching for sources of DM treatment is always performed. One therapy of DM is a-glucosidase inhibitor. a-Glucosidase is an enzyme that can break down a complex carbohydrate into simple sugar. lhe inhibiton of this enzyme can retard the rate of carbohydrate digestion resulting in a delay in glucose absorption. The purpose of this studywas to identify the content of chemical compound …

Archaea have phytanyl ether lipids which is one of the characteristics that separates them from bacteria and eukaryotes. Some archaea have also unique membrane spanning tetrae ther lipids (T EL); in Sulfolobus and Ihermoplasma species these TELmake up the majority of total membrane lipids. Archaeal lipids are able to form stable liposomal structures, both from membrane factions, mainly the polar membrane fraction, i.e. archaeosomes, or from highly purified TEL (tetraether lipid liposomes). Liposomes of the main polar lipid (MPL) from thermoacidophilic archaeon Ihermoplasma acidophilum were thoroughly inevstigated. Archaeosomes and TEL liposomes exhibit extremely low proton permeability and high stability at …

Previous experiment reported that methanol extract from leaves of Antidesma neurocarpum Miq. has a strong inhibitory activity of a-glucosidase (IC50 = 2,18 mikrogram/mL). The aim of this experiment was to know about the most active extract and fraction of inhibitory activity of a-glycosidase and also the chemical compounds from those most active extract and fraction. Extraction is done by using multilevel maceration (n-hexane, ethyl acetate and methanol). Inhibitory activity of a-glucosidase tested with …

"Development of tumor resistance to chemotherapeutics is related to inherent tumor variations regarding sensitivity to chemotherapeutics and to sub-optimal dosing regimens, including variation in patient pharmacokinetics that result in suboptimal exposure of tumor cells to anti-neoplastic drugs [1, 2]. The rate and extent of drug efficacy depends on the extent of drug exposure at the tumor site and the time above the effective concentration [3]. In vitro models that incorporate these pharmacokinetic and pharmacodynamic (PK/PD) principles to optimize therapeutic response may be considered the method of choice for optimizing dosing schedules before translating data from static assays to animals and …
