Pharmacogenomics (PGx) is an emerging field that merges genetics with pharmacology. It focuses on how an individual's genetic makeup influences their response to specific medications. This relatively new area of medicine seeks to understand the genetic factors that affect drug efficacy and safety. One practical application of PGx is in the context of treatment with angiotensin-converting enzyme inhibitors (ACEIs), which are widely used medications for managing hypertension. In this study, we evaluated the suitability of elutriation, a method successfully employed in the extraction of microplastics from marine sediments, for the extraction of microplastics from freshwater and terrestrial soils. Five soils were sampled throughout Oklahoma, USA in order to capture a range of sand, silt, clay, and organic matter composition. Each soil was subjected to microplastic extraction with and without elutriation, followed by digestion in 7.5% NaOCl, and then flotation in 6 M ZnCl2. Hair external contamination poses significant challenges to the interpretation of hair testing results. Previous studies have shown that cosmetic hair treatments can reduce the concentration of drugs already present in the hair shaft. However, the impact of these treatments on externally deposited drugs remains largely unexplored. Lewy body disorders are a family of neurodegenerative conditions characterized by proteostatic and redox disequilibrium, leading to deposition of the abundant protein α-synuclein in hallmark inclusions and oxidative damage to DNA. Sublethal oxidative DNA damage in neurons is repaired predominantly by the base excision repair pathway, which involves the coordinated activity of several enzymes, including apurinic/apyrimidinic endonuclease 1 (APE1). Despite emerging connections between α-synuclein, oxidative stress, and DNA damage/repair, along with evidence demonstrating the multifunctional nature of APE1, the link between APE1 and α-synuclein has not yet been explored.

In Lewy body disorders, α-synuclein aggregates are believed to destabilize the microtubular framework. However, microtubules are a challenging therapeutic target, as excessive rigidity or flexibility afforded by microtubule modulators may be neurotoxic. Thus, the objective of the present study was to evaluate dual-acting microtubule- stabilizing/destabilizing agents in cellular and animal models of early-stage Lewy body disease.
In Aim 1 we tested the dual-acting tricylic pyrimido[4,5-b]indole analogs AG161-47 and AG161-41 in an in vitro preformed fibril induced model of Lewy body disease. We found that both compounds reduced preformed fibril-seeded α-synucleinopathy without toxicity in primary hippocampal cultures. AG161-47 subtly impacted microtubule …

In August 2017, nearly 1 million Rohingya people fled to Bangladesh to save their lives from the genocide inflicted by the military of Myanmar. At present, over 1.3 million Rohingyas are staying at Cox’s Bazar, Bangladesh, in 34 overcrowded refugee camps. This research examined the environmental impacts especially the change in vegetation cover and land surface temperature as well as the socioeconomic alteration of the host country after the refugee influx. In compliance with 902 KRS 10:010 Kentucky Swimming and Bathing Facility regulation, all bathing and swimming pools for public access are subject to inspection and monitoring by local and state health agencies. It is established that Δ9-THC exposure early in life causes adverse effects in children’s development including behavioral disorders. Despite the opioid crisis taking headlines, the abuse of cocaine and amphetamines remains rampant and laboratory instruments must be able to effectively and reliably handle the overwhelming case load. In this study, an Agilent 6475 Triple Quadrupole (QQQ) LCMS, recently acquired by the Tarrant County Medical Examiner’s Office (TCMEO) in Fort Worth, Texas, was validated for identifying and quantifying CocAmps. Results show that the instrument was fit-for-purpose and that the instrument passed all validation tests. Reproducibility was ≤ 20% RSD for 12 QC replicates, 6 of blood and 6 of urine. Chronic inflammation, when left untreated, may result in significant morbidity and in severe cases, mortality. While glucocorticoids (GCs) are still being extensively used clinically as a class of very potent anti-inflammatory agents, they are also notorious for diverse adverse side effects. To mitigate these side effects, we have developed a polyethylene glycol (PEG)-based prodrug (ZSJ-0228) of dexamethasone (Dex). When tested on rodent models of Lupus Nephritis and Focal Segmental Glomerulosclerosis (FSGS), ZSJ-0228 was found to provide potent and sustained therapeutic efficacy without any detectable typical GC-associated side effects.
