Regulation Of P53 Stability And Apoptosis By A Ror Agonist, Yongjun Wang, Laura A. Solt, Douglas J. Kojetin, Thomas P. Burris

Activation of p53 function leading to cell-cycle arrest and/or apoptosis is a promising strategy for development of anticancer therapeutic agents. Here, we describe a novel mechanism for stabilization of p53 protein expression via activation of the orphan nuclear receptor, RORa. We demonstrate that treatment of cancer cells with a newly described synthetic ROR agonist, SR1078, leads to p53 stabilization and induction of apoptosis. These data suggest that synthetic ROR agonists may hold utility in the treatment of cancer.

Mutations of comparative gene identification 58 (CGI-58) in humans cause Chanarin-Dorfman syndrome, a rare autosomal recessive disease in which excess triacylglycerol (TAG) accumulates in multiple tissues. CGI-58 recently has been ascribed two distinct biochemical activities, including coactivation of adipose triglyceride lipase and acylation of lysophosphatidic acid (LPA). It is noteworthy that both the substrate (LPA) and the product (phosphatidic acid) of the LPA acyltransferase reaction are wellknown signaling lipids. Therefore, we hypothesized that CGI-58 is involved in generating lipid mediators that regulate TAG metabolism and insulin sensitivity. The objective of this study was to determine the effects of muscadine grape or wine (cv. Noble) phytochemicals on obesity and associated metabolic complications. Muscadine grape or wine phytochemicals were extracted using Amberlite FPX66 resin. Male C57BL/6J mice were given a low-fat diet (LF, 10% kcal fat), high-fat diet (HF, 60% kcal fat), HF + 0.4% muscadine grape phytochemicals (HF+MGP), or HF + 0.4% muscadine wine phytochemicals (HF+MWP) for 15 weeks. At 7 weeks, mice fed HF+MGP had significantly decreased body weights by 12% compared to HF controls. Dietary MGP or MWP supplementation reduced plasma content of free fatty acids, …

Objectives.Using data from the first two phases of the Georgia Centenarian Study, we proposed a latent factor structure for the Duke OARS domains: Economic Resources, Mental Health, Activities of Daily Living, Physical Health, and Social Resources. Methods. Exploratory and confirmatory factor analyses were conducted on two waves of the Georgia Centenarian Study to test a latent variable measurement model of the five resources; nested model testing was employed to assess the final measurement model for equivalency of factor structure over time. Background: Given the documented physical activity disparities that exist among low-income minority communities and the increased focused on socio-ecological approaches to address physical inactivity, efforts aimed at understanding the built environment to support physical activity are needed. This community-based participatory research (CBPR) project investigates walking trails perceptions in a high minority southern community and objectively examines walking trails. Objective: There is little evidence of how fruit and vegetable (FV) household food availability changes over a one-month period among low-income households. The objective of this study was to analyze how FV variety and sustainability changes over a 4-week period.
Design: Inventories were conducted in low-income family households (n=49) once a week over a 4-week period. Trained researchers gathered the weights of all FV, including legumes, within the home. Previously determined mean container weights were subtracted to obtain the estimated weight of the FV. All weights were then converted to edible cups of FV, taking into account the weight that …

Interest in consuming unprocessed foods has led to assumptions that raw milk has health benefits over pasteurized milk. This study was designed to evaluate the belief that raw milk is nutritionally superior. Eighteen mice were randomly assigned to one of two groups: raw milk or pasteurized milk. Mice were grouped into breeding trios and given a fresh supply of milk every four hours between 7 a.m. and 11 p.m. for 55 days. Milk consumption was tracked at each feeding by measuring the amount of milk that was provided and the amount of milk that was left from the previous feeding. …

Nutritional and energetic needs of female mammals depend upon size or reproductive investment and shape individual activity budgets and behavioral patterns. To maximize nutritional and energetic intake females may increase time allocated to foraging or access resources through risk-prone behaviors, represented as aggression or leadership. Conversely, to minimize nutritional and energetic spending females may engage in risk-averse activities such as resting or nonaggressive social interactions. Objective: This study was aimed at assessing the effect of maternal smoking during pregnancy on metabolic cardiovascular risk factors in early adulthood in a Brazilian birth cohort, after controlling for possible confounding variables and health behaviors in early adulthood.

Methods: In 1982, the maternity hospitals in Pelotas, southern Brazil, were visited and all births were identified. Those livebirths whose family lived in the urban area of the city were studied prospectively. The agency directors of the 332 member agencies of the LCFB are in direct contact with clients, therefore they could provide nutrition education rather than the LCFB staff alone. The purpose of this study was to examine the effects of smoking on risk factors associated with cardiovascular disease (CVD) and determine the efficacy of antioxidant-rich berries in ameliorating these risk factors in postmenopausal smokers. Menopause and cigarette smoking have been identified as major risk factors for CVD due to a decrease in antioxidant protection, increase in inflammation and oxidative stress, and adverse changes in serum lipids. Antioxidants, specifically flavonoid compounds found in blackberries and blueberries, have been shown to prevent low-density lipoprotein cholesterol oxidation and thus prevent downstream inflammation and oxidative stress. The diets of Mongolia infants are deficient in vitamins and minerals and complementary foods are introduced too early. Breastfeeding is the best source of nutrition for infants until 6 months of age. A survey of 50 Mongolian mothers with children under the age of 5 in the town of Sainshand, Dornogobi, was conducted in July 2011. Mongolian infants are among the highest percent of infants in the world who are exclusively breastfed for the first 6 months of their lives, thus providing them with excellent nutrition. Understanding the adaptive significance of variation in female mating behaviour is important because variation may often be favored by selection instead of a change in mean mating behaviour, particularly in variable environments. Females are known to adjust their mating behaviour to a variety of intrinsic and extrinsic factors including nutrition. There are multiple reasons why female behaviours might vary with nutrition; we tested two of these hypotheses: the search cost hypothesis and the direct benefits hypothesis. Hydrolyzing food by-products is a unique approach to potentially increase by-product value and reduce waste. An abundant by-product of cheese production, whey, contains all essential amino acids and some distinctive peptides with functional and nutraceutical properties. Typically, proteins from whey are tailored for specific uses by chemical or enzymatic hydrolysis; however, subcritical water hydrolysis is a novel alternative used successfully to hydrolyze various substrates. Nevertheless, minimal research exists on: (1) the hydrolysis of whey protein; (2) the incorporation of additives; (3) the hydrolysis of whole whey; and (4) the production of volatiles when using subcritical water hydrolysis.
