Elite human and animal athletes must acquire the fuels necessary for extreme feats, but also contend with the oxidative damage associated with peak metabolic performance. Here, we show that a migratory bird with fuel stores composed of more omega-6 polyunsaturated fats (PUFA) expended 11% less energy during long-duration (6 hr) flights with no change in oxidative costs; however, this short-term energy savings came at the long-term cost of higher oxidative damage in the omega-6 PUFA-fed birds. In species who consume folivorous diets, immature individuals must contend with the challenges of extracting nutrients from fibrous foods before dietary adaptations and strategies are fully developed. Additionally, immatures have distinct nutritional needs to support their stage-specific metabolic and biophysiological requirements. To meet these stage-specific needs, while constrained by underdeveloped feeding strategies and digestive capacities, immatures may adopt distinct diets better suited to their specific developmental context. Specifically, phytochemicals change human miRNA expression thereby impacting gut microbiome and/or increasing host immune functions. Cancer cachexia is a metabolic syndrome with alterations in gene regulatory networks that consequently lead to skeletal muscle wasting. Integrating microRNAs mRNAs omics profiles offers an opportunity to understand transcriptional and post-transcriptional regulatory networks underlying muscle wasting. Here, we used RNA sequencing to simultaneously integrate and explore microRNAs and mRNAs expression profiles in the tibialis anterior (TA) muscles of the Lewis Lung Carcinoma (LLC) model of cancer cachexia. We found 1,008 mRNAs and 18 microRNAs differentially expressed in cachectic mice compared with controls. The modern broiler is growing at a rapid rate generating tremendous amounts of heat. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The first two chapters present evaluation of the classic way to calculate NE versus a new methodology; the Arkansas NE (Ark NE) system, with birds from two genetic lines fed diets with different AA content or different ME content in two different environmental temperatures.

Fatty acid oxidation inhibition is one approach to reducing blood glucose levels in type II diabetes. Skeletal muscle specific Carnitine Palmitoyltransferase 1b knockout mice (Cpt1bm-/-) cannot transport long-chain fatty acids into the mitochondria to be oxidized in order to produce energy. Cpt1bm-/- mice have debilitated fat oxidation, less fat mass and improved glucose utilization compared to control C57BL/6 mice fed a 25% fat diet.

Purpose Weight loss can result in the loss of muscle mass and bone mineral density. Resistance exercise is commonly prescribed to attenuate these effects. However, the anabolic endocrine response to resistance exercise during caloric restriction has not been characterized. Methods Participants underwent 3-day conditions of caloric restriction (15 kcal kg FFM−1) with post-exercise carbohydrate (CRC) and with post-exercise protein (CRP), and an energy balance control (40 kcal kg FFM−1) with post-exercise carbohydrate (CON). Fulminant hepatic failure (FHF) is a rare, life-threatening liver disease with a poor prognosis. Administration of D-galactosamine (GalN) and lipopolysaccharide (LPS) triggers acute liver injury in mice, simulating many clinical features of FHF in humans; therefore, this disease model is often used to investigate potential therapeutic interventions to treat FHF. Background: Nut butter-based Ready to Use Supplemental Foods (RUSF) are an effective way to add nutrients and calories to diets of malnourished and food insecure populations. The RUSF formulations have been further modified to add micronutrients including iron and folic acid needed during pregnancy and lactation. Previous studies have demonstrated that ayurvedic ingredients exhibit ergogenic (performance enhancing) properties, however, no previous studies have examined the ergogenic potential of Asparagus racemosus. The purpose of the present study was to examine the ergogenic efficacy of supplementation with 500 mg·d−1 of A. racemosus during bench press training. Eighteen recreationally trained men (mean ± SD; age = 20.4 ± 0.5 yrs; height = 179.7 ± 1.5 cm; weight = 84.7 ± 5.7 kg) were randomly assigned either 500 mg·d−1 of A. racemosus (n = 10) or placebo (n = 8). CrAssphage (cross-assembly phage) is a bacteriophage that was first discovered in human gut metagenomic data. CrAssphage belongs to a diverse family of crAss-like bacteriophages thought to infect gut commensal bacteria belonging to Bacteroides species. However, not much is known about the biogeography of crAssphage and whether certain strains are associated with specific human populations. In this study, we screened publicly available human gut metagenomic data from 3,341 samples for the presence of crAssphage sensu stricto (NC_024711.1). Supplementation with n-3 long-chain (LC) polyunsaturated fatty acids (PUFA) is known to promote thermogenesis via the activation of brown adipose tissue (BAT). Agricultural products that are biofortified with α-linolenic acid (ALA), the precursor of n-3 LC PUFA, have been launched to the market, but their impact on BAT function is unknown. Background: The FUT2 gene (Se gene) encoding the enzyme α-1,2-L-fucosyltransferase 2 seems to have a significant effect on the number and type of bacteria colonizing the intestines. Methods: In a group of 19 patients after bariatric surgery, the polymorphism (rs601338) of FUT2 gene was analyzed in combination with body mass reduction, intestinal microbiome (16S RNA sequencing), and short chain fatty acids (SCFA) measurements in stools. Results: Among the secretors (Se/Se polymorphism of the FUT2 gene rs601338, carriers of GG variant), correlations between waist-hip ratio (WHR) and propionate content and an increase in Prevotella, Escherichia, Shigella, and Bacteroides were observed. Emerging evidence suggests intestinal microbiota as a central contributing factor to the pathogenesis of Relapsing-Remitting-Multiple-Sclerosis (RRMS). This novel RRMS study evaluated the impact of fecal-microbiota-transplantation (FMT) on a broad array of physiological/clinical outcomes using deep metagenome sequencing of fecal microbiome. FMT interventions were associated with increased abundances of putative beneficial stool bacteria and short-chain-fatty-acid metabolites, which were associated with increased/improved serum brain-derived-neurotrophic-factor levels and gait/walking metrics. This proof-of-concept single-subject longitudinal study provides evidence of potential importance of intestinal microbiota in the pathogenesis of MS, and scientific rationale to help design future randomized controlled trials assessing FMT in RRMS patients.

โรคออทิซึมสเปกตรัมเป็นความผิดปกติเกี่ยวกับพัฒนาการในช่วงเริ่มแรกของระบบประสาทและสมองซึ่งมีอาการทางคลินิกที่หลากหลายมาก ปัจจุบันยังไม่มีสารบ่งชี้เพื่อช่วยในการวินิจฉัยและแบ่งผู้ป่วยเป็นกลุ่มย่อย แต่การวินิจฉัยที่รวดเร็วเป็นสิ่งสำคัญที่จะช่วยให้ผู้ป่วยออทิซึมสเปกตรัมสามารถได้รับการเรียนรู้และพัฒนาพฤติกรรมที่จำเป็นในการดำรงชีวิตได้ การวินิจฉัยโรคออทิซึมสเปกตรัมตามมาตรฐานสากลใช้แบบสอบถามที่มีความซับซ้อนและใช้เวลามาก ซึ่งในปัจจุบันมีการนำเอาเทคโนโลยีปัญญาประดิษฐ์มาประยุกต์ใช้ในการวิเคราะห์ข้อมูลขนาดใหญ่ที่มีความซับซ้อน อย่างไรก็ตามการนำเอาปัญญาประดิษฐ์มาใช้วิเคราะห์อาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมยังไม่แพร่หลายนัก ในการศึกษานี้ผู้วิจัยต้องการนำเอาปัญญาประดิษฐ์มาใช้แบ่งผู้ป่วยออทิซึมสเปกตรัมจำนวน 85 ราย ออกเป็นกลุ่มย่อยตามอาการทางคลินิกจากข้อมูลแบบทดสอบในการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม ADI-R จำนวน 123 ข้อ จากนั้นได้ทำการสร้างโมเดลในการจัดกลุ่มและเปรียบเทียบประสิทธิภาพรวมถึงจำนวนข้อคำถามที่ใช้ในแต่ละโมเดล นอกจากนี้ผู้วิจัยได้ศึกษาหาสารบ่งชี้ทางชีวภาพที่เป็นตัวแทนของโรคออทิซึมสเปกตรัมกลุ่มย่อยจากลักษณะอาการทางคลินิกโดยอาศัยการบูรณาการโอมิกส์หลายระดับ จึงได้ดำเนินการวิเคราะห์รูปแบบทรานสคริปโตม และวิเคราะห์รูปแบบโปรตีโอมของผู้ป่วยออทิซึมสเปกตรัมโดยใช้เซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์เป็นต้นแบบ โดยใช้เทคนิคการแยกโปรตีนด้วยกระแสไฟฟ้า 2 มิติ (2D-GE) จากนั้นจะทำการระบุโปรตีนด้วยเทคนิค LC-MS/MS เพื่อหาโปรตีนที่ถูกรบกวนการทำงานอย่างมีนัยสำคัญ และบูรณาการข้อมูลเพื่อศึกษาความสอดคล้องและสัมพันธ์กันระหว่างการแสดงออกของโปรตีนและยีน และทำการยืนยันระดับการแสดงออกที่ผิดปกติไปของยีนด้วยเทคนิค Real-time qPCR และโปรตีนด้วยเทคนิค Western blot ในการศึกษาครั้งนี้จากผลการวิเคราะห์ข้อมูลลักษณะอาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมจากแบบทดสอบ ADI-R 123 ข้อ พบว่าวิธี K-means clustering ที่แบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อยนั้นให้ประสิทธิภาพในการแบ่งดีที่สุด และเมื่อทำการสร้างโมเดลในการจัดกลุ่มผู้ป่วยตามผลการแบ่งกลุ่มดังกล่าว พบว่าอัลกอริทึม Random forest ที่มีจำนวนต้นไม้ 10 ต้น โดยกระบวนการคัดเลือกข้อมูลแบบ Forward selection ให้ผลการทำนายถูกต้องมากที่สุด (100%) โดยใช้ข้อคำถาม ADI-R เพียง 5 ข้อ ในการแบ่งผู้ป่วยออกเป็นกลุ่มย่อย และจากการวิเคราะห์รูปแบบโปรตีโอมจากเซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์ของผู้ป่วยออทิซึมสเปกตรัมและออทิซึมสเปกตรัมกลุ่มย่อยด้วยเทคนิค 2D-GE ผู้วิจัยได้ทำการคัดเลือกจุดโปรตีนที่มีการแสดงออกผิดปกติอย่างมีนัยสำคัญและมีค่าสัดส่วนความเข้มแตกต่างกันอย่างน้อยสองเท่ามา 19 จุด เพื่อทำการระบุชนิดของโปรตีนด้วยเทคนิค LC-MS/MS โดยพบว่าจากโปรตีนทั้งหมด HNRNPA2B1 มีระดับการแสดงออกเพิ่มขึ้นในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ไม่มีความผิดปกติด้านภาษารุนแรง โดยที่โปรตีน HNRNPA2B1 MDH2 DLD และ MSN มีระดับการแสดงออกลดลงในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่มีความผิดปกติด้านภาษารุนแรง ดังนั้นจากการศึกษาวิจัยในครั้งนี้ได้แสดงให้เห็นว่าการใช้เทคโนโลยีปัญญาประดิษฐ์ในการแบ่งผู้ป่วยออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามลักษณะอาการทางคลินิกนั้น สามารถลดความหลากหลายตามลักษณะอาการที่แสดงออกของโรคออทิซึมสเปกตรัมในการวิเคราะห์รูปแบบทรานสคริปโตมและโปรตีโอม รวมถึงการค้นหาโปรตีนที่จำเพาะต่อผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อย ได้อย่างมีประสิทธิภาพมากขึ้น ซึ่งโปรตีนเหล่านี้อาจใช้เป็นโปรตีนบ่งชี้ในการศึกษาเชิงลึกในประชากรผู้ป่วย เพื่อพัฒนาเป็นสารบ่งชีทางชีวภาพ (Biomarker) หรือศึกษากลไกพยาธิสภาพในระดับชีวโมเลกุลของโรคออทิซึมสเปกตรัมได้ต่อไปในอนาคต

Carrier status of the E4 allele of the APOE gene is the strongest genetic risk factor for late-onset AD, and increasing evidence suggests that E4 carriers may be at an increased risk for neurodegeneration based on inherent metabolic impairments. A new appreciation is forming for the role of APOE in cerebral metabolism, and how nutritional factors may impact this role. Background: The appetite-regulating hormones may influence compensatory increases in energy intake with exercise, although this causal relationship has been difficult to prove in a longitudinal trial.

Methods: 37 participants (29 female) aged 18 to 40 years performed aerobic exercise 6 days (6d), 2 days (2d), or 0 days per week for 12-weeks. Concentrations of ghrelin, leptin, glucagon-like peptide 1 (GLP-1), and insulin were assessed before (fasting, minute 0) and after a standardized meal at minute 15, 30, 45, 60, 90, 120, 150, and 180.
