The overall objective of this dissertation was to examine the degree to which manipulation of the saturated fat content of a high-fat diet influenced obesity-related outcomes as well as to see if the naturally-occurring flavonoid, quercetin, could attenuate the resulting obesity and related metabolic and inflammatory side effects. Specifically, these studies examined 1) the influential role of saturated fat on macrophage function, inflammation, and other obesity-related comorbidities, including non-alcoholic fatty-liver disease, and 2) the potential benefits of quercetin supplementation. At the most basic level obesity is the result of a chronic imbalance between energy intake and energy expenditure. However, the exact etiology is considerably more complex and may involve a variety of physiological and behavioral factors. Metabolic disturbances, including reduced fat oxidation as measured by the respiratory quotient (RQ) and reduced resting metabolic rate (RMR), have been identified as possible predictors of changes in body weight and body composition. RMR represents the largest component (60-80%) of caloric expenditure that contributes to total daily energy expenditure in humans and has high inter-person variability (±25%) but not within individuals (<±5%). The cause of this variability between individuals and the exact role of RMR and RQ in determining body weight and body composition are uncertain.

Posterior shoulder tightness is a common physical impairment in overhand baseball athletes presenting with injury. The etiology of this physical impairment is poorly understood and theorized to be a combination of bony, muscular, and inert soft-tissue contributions occurring at the glenohumeral joint. The ability to discriminate between each tissue's influences on shoulder range of motion is often challenging to overcome within a clinical environment. Despite the current need for investigation of factors involved with soldiers' resilience to stressors in the Basic Combat Training (BCT) environment, and evidence in civilian populations that physical training is associated with psychological benefits, little is known about the relationship between physical fitness and psychological adjustment during BCT. Study one of this dissertation involved an extensive literature review of factors related to the relationship between physical fitness and psychological adjustment of soldiers during BCT. Gestational diabetes mellitus (GDM) is the most prevalent metabolic disorder during pregnancy. Women diagnosed with GDM have a substantially greater risk of developing type 2 diabetes within 5-10 years after delivery, and the risk is increased by excess body weight. Uncontrolled hyperglycemia during pregnancy is potentially harmful to both mother and fetus, resulting in a greater need for Caesarian-section deliveries, delivery of larger infants with more excess body fat, a greater risk of infant death and stillbirth, and an elevated risk of infant hypoglycemia immediately after birth. Essential tremor (ET) and Parkinson’s disease (PD) are two of the most common movement disorders. Tremors are the primary symptoms of ET and of some PD patients, the two are often mistaken for each other. Environmental temperature exhibits profound effects on the activity and ecology of ectotherms through its impact on muscle contractile physiology. While the performance of locomotor behaviors powered by muscle contraction directly decreases by at least 33% over a 10°C drop in body temperature, chameleons are known to feed, presumably with high performance, at body temperatures where sympatric lizard species remain inactive. I propose that ballistic movements that are powered by the recoil of preloaded elastic and collagenous tissues are less thermally dependent than movements that rely on direct muscular power. Pedicle screw-rod instrumentation is considered a standard treatment for spinal instability, and titanium is the most common material for this application. Cobalt-chromium has several advantages over titanium and is generating interest in orthopedic practice. The aim of this study was to compare titanium versus cobalt-chromium rods in posterior fusion, with and without transverse connectors, through in vitro biomechanical testing and determine the optimal configuration.

Quality physical education can positively influence students’ emotional development, specifically their perceptions of competence, self-esteem, and self-concept. Unfortunately, girls often become less engaged and involved in physical education as they grow older and consistently report lower physical self-concepts than males. Physical self-concept is associated with multiple positive outcomes, yet there is only speculation addressing why females report lower physical self-concepts than males in physical education. Gibson developed the affordance concept to complement his theory of direct perception that stands in sharp contrast with the prevalent inferential theories of perception. A comparison of the two approaches shows that the distinction between them also has an ontological aspect. We trace the history and newer formalizations of the notion of affordance and discuss some competing opinions on its scope. Next, empirical work on the affordance concept is reviewed in brief and the relevance of dynamical systems theory to affordance research is demonstrated. Context: Transdermal delivery of hydrocortisone by phonophoresis is used for the treatment of musculoskeletal conditions. Research shows hydrocortisone and other white or opaque topical preparations transmit ultrasound energy poorly. Effective transmission of ultrasound is important in phonophoresis. Main Outcome measured: Samples of subcutaneous interstitial fluid were collected during and for 20 minutes following phonophoresis treatment. Cortisol concentrations were analyzed by an enzyme linked immune-assay (ELISA) test. Objective: Determine the subcutaneous cortisol concentration after two different phonophoresis treatments using a 2.5% hydrocortisone preparation. To determine the relationship between overall psychological well-being and stress on diet quality among young adult women and to examine the potential confounding influence of season, physical activity and sleep on these relationships. Design/Participants. The study used a cross-sectional design. Three hundred and fifty-one women were recruited to participate in the study. All participants were university students (20.2 ± 1.6 y). Therefore the purpose of this study was to determine the steady state metabolic cost of repetitive jumping on the Digi-Jump machine, and to determine if exercise on this device is more or less strenuous than similar exercise with a jump rope. Rationale: Skeletal muscle blood flow is coupled with the oxygenation state of hemoglobin in young adults, whereby the erythrocyte functions as an oxygen sensor and releases ATP during deoxygenation to evoke vasodilation. Whether this function is impaired in humans of advanced age is unknown.
Objective: To test the hypothesis that older adults demonstrate impaired muscle blood flow and lower intravascular ATP during conditions of erythrocyte deoxygenation.
