In recent decades, cases of community-acquired MRSA (CA-MRSA) and healthcare-associated MRSA (HA-MRSA) have been on the rise. The ability of antibiotic resistance to transfer between microorganisms greatly expands its threat to the effective treatment and containment of bacterial infections. The mecA gene is found on a mobile cassette within the Staphylococcal cell and encodes for PBP2a, an alternate version of penicillin-binding protein (PBP) with less binding affinity for β-lactam antibiotics, increasing resistance to the effects of antibiotics. การคลอดก่อนกำหนด (ก่อน 37 สัปดาห์) เป็นปัญหาสาธารณสุขสำคัญทั่วโลก และในไทย ซึ่งเป็นสาเหตุหลักของการเสียชีวิตและภาวะแทรกซ้อนในทารกแรกเกิด การคลอดก่อนกำหนดส่วนใหญ่เกิดขึ้นเองโดย ไม่ทราบสาเหตุ จากหลักฐานก่อนหน้านี้พบว่า anti-HLA antibody อาจมีส่วนเกี่ยวข้องกับการคลอดก่อน กำหนดในประชากรชาวชิลี วัตถุประสงค์ของงานวิจัยจึงต้องการศึกษาความถี่และความสัมพันธ์ของแอนติบอดีต่อ Human Leukocyte Antigen class I ในหญิงตั้งครรภ์ชาวไทยที่คลอดก่อนกำหนด และสร้างโมเดลทำนายภาวะ คลอดก่อนกำหนดโดยใช้ปัจจัยเสี่ยง และผล anti-HLA class I กลุ่มตัวอย่างประกอบด้วยหญิงตั้งครรภ์ชาวไทยที่ คลอดที่โรงพยาบาลเจริญกรุงประชารักษ์จำนวน 299 คน แบ่งเป็นกลุ่มคลอดก่อนกำหนด 135 คน และกลุ่ม คลอดตามกำหนด 164 คน ได้รับการตรวจ anti-HLA class I ด้วยเทคนิค Multiplex Fluorescent Immunoassay และรวบรวมข้อมูลทางคลินิก ผลการศึกษาพบว่าความชุกของการตรวจพบ anti-HLA class I antibody โดยรวมอยู่ที่ 33.11% และสูงกว่าอย่างมีนัยสำคัญทางสถิติในกลุ่มที่คลอดก่อนกำหนด (42.96%) เมื่อ เทียบกับกลุ่มที่คลอดตามกำหนด (25.00%) การวิเคราะห์ความสัมพันธ์ด้วยสถิติ Chi-square test และ Logistic Regression พบว่าการตรวจพบ anti-HLA class I มีความสัมพันธ์อย่างมีนัยสำคัญกับการคลอดก่อนกำหนด (P < 0.001) โดยเพิ่มโอกาสในการคลอดก่อนกำหนดสูงขึ้น 2.260 เท่า นอกจากนี้งานวิจัยได้สร้างแบบจำลองการ ทำนายการคลอดก่อนกำหนดโดยใช้อัลกอริทึม Machine Learning หลายประเภท และใช้วิธีการคัดเลือกตัวแปร ที่แตกต่างกัน พบว่าแบบจำลอง LightGBM ที่ใช้การคัดเลือกตัวแปรแบบ Backward มีประสิทธิภาพสูงสุดในแง่ ของความแม่นยำ (Accuracy = 0.6018 ± 0.0482) และความสามารถในการตรวจจับการคลอดก่อนกำหนด (Sensitivity = 0.6444 ± 0.0726) ซึ่งแสดงให้เห็นว่า anti-HLA class I antibody และปัจจัยเสี่ยงบางประการ สามารถนำมาใช้เป็นตัวบ่งชี้เพื่อทำนายความเสี่ยงของการคลอดก่อนกำหนดได้ ผลการวิจัยนี้เป็นองค์ความรู้ พื้นฐานที่สำคัญในการพัฒนา biomarker เพื่อช่วยแพทย์ในการวางแผนการดูแลรักษาหญิงตั้งครรภ์ในอนาคต

วัณโรค (TB) ซึ่งมีสาเหตุเกิดจากเชื้อ Mycobacterium tuberculosis (MTB) และโรคติดเชื้อกลุ่ม Non-tuberculous mycobacteria (NTM) เป็นโรคติดเชื้อที่มีความสำคัญซึ่งเกิดจากเชื้อกลุ่ม Mycobacteria ส่งผลกระทบต่อปอดและอวัยวะอื่นๆ ของร่างกาย แม้ว่าสาเหตุของการก่อโรคต่างกัน แต่อย่างไรก็ตามอาการแสดงมีความคล้ายคลึงกัน เช่น ไอ มีไข้ น้ำหนักลด และอ่อนเพลีย แนวทางในการรักษาวัณโรคและโรคติดเชื้อกลุ่ม NTM นั้นเป็นการรักษาโดยการใช้ยาต้านเชื้อ Mycobacteria หลายชนิดผสมผสานกัน แต่อย่างไรก็ตามแนวทางในการรักษาและระยะเวลาที่ใช้ในการรักษาแตกต่างกันระหว่างโรคดังกล่าว ดังนั้นวิธีการวินิจฉัยที่มีความรวดเร็วและจำเพาะในการจำแนกการติดเชื้อระหว่างสองกลุ่มจึงมีความจำเป็น เพื่อที่ผู้ป่วยจะได้รับการรักษาและการจัดการกับผู้ป่วยที่เหมาะสมต่อไป การศึกษาในครั้งนี้มีวัตถุประสงค์ในการพัฒนา Primer ในการจำแนกเชื้อกลุ่ม NTM จาก MTB และพัฒนาชุดตรวจแถบตรวจสารพันธุกรรมต้นแบบที่สามารถอ่านผลการทดสอบได้ด้วยตาเปล่าจากการเพิ่มประมาณสารพันธุกรรมที่อุณหภูมิเดียวซึ่งจำเพาะต่อยีน IS1081 และ ku ของเชื้อ MTB และ NTM ตามลำดับ แถบตรวจที่ผลิตพัฒนาขึ้นซึ่งผลิตจาก Nitrocellulose MTB/NTM strip สามารถตรวจจับผลผลิต Multiplex-recombinase polymerase amplification (M-RPA) ได้ โดยผลการทดสอบเบื้องต้นพบว่า Primer ที่ออกแบบขึ้นมานั้นสามารถเพิ่มปริมาณสารพันธุกรรมและจำแนก DNA ที่สกัดได้จากโคโลนีของเชื้อ MTB และ NTM ได้อย่างถูกต้อง โดยปฏิกิริยาบ่มที่อุณหภูมิ 37 ˚C เป็นระยะเวลา 30 นาที และเวลาในการอ่านผลของแถบตรวจ MTB/NTM strip คือ 15 นาที โดยตรวจสอบความเข้มข้นของเชื้อที่น้อยที่สุดที่ตรวจวัดได้ด้วยเทคนิค M-RPA ร่วมกับแถบตรวจ MTB/NTM strip สำหรับโคโลนีของเชื้อ MTB และ NTM คือ 1.97 x 104 CFU/mL และไม่พบปฏิกิริยาข้ามกลุ่มสำหรับเชื้อแบคทีเรียก่อโรคทั่วไปสายพันธุ์อื่น รวมถึงเชื้อกลุ่ม Higher bacteria เทคนิค M-RPA ร่วมกับแถบตรวจ MTB/NTM strip ได้นำมาประเมินกับ DNA …

การจัดการประมงอย่างยั่งยืนจำเป็นต้องอาศัยองค์ความรู้เกี่ยวกับการวางไข่ของปลาที่มีความสำคัญทางเศรษฐกิจหลายชนิด ซึ่งประเทศไทยในปัจจุบันยังขาดข้อมูลที่ครอบคลุมในด้านนี้ การศึกษานี้จึงสำรวจการวางไข่ของปลาหลากหลายสปีชีส์โดยใช้เทคนิคดีเอ็นเอบาร์โค้ด เพื่อระบุสปีชีส์ของไข่ปลา ตัวอย่างไข่ปลาถูกเก็บทุก ๆ 3 เดือน เป็นระยะเวลา 1 ปีจากสามพื้นที่ในอ่าวไทย ได้แก่ อ่าวไทยตอนใน จังหวัดจันทบุรี และจังหวัดชุมพร จากนั้นจึงทำการเพิ่มจำนวนชิ้นส่วนยีน COI (710 คู่เบส) และ 16S rRNA (570 คู่เบส) ด้วยเทคนิค PCR และนำไปหาลำดับนิวคลีโอไทด์เปรียบเทียบกับฐานข้อมูล GenBank เพื่อระบุสปีชีส์ของปลา โดยสามารถระบุสปีชีส์ได้เมื่อลำดับนิวคลีโอไทด์ตรงกับฐานข้อมูลมากกว่า 97% ขึ้นไป ผลการศึกษาพบรูปแบบการวางไข่ของปลาที่หลากหลาย และสามารถระบุสปีชีส์ของปลาได้มากกว่า 34 วงศ์ ซึ่งแสดงให้เห็นถึงความหลากหลายของสปีชีส์ปลาในอ่าวไทย นอกจากนี้ ยังพบว่าปลาบางชนิดวางไข่ตลอดทั้งปี ในขณะที่บางชนิดวางไข่เฉพาะช่วงฤดูกาล สะท้อนให้เห็นถึงความแตกต่างในกลยุทธ์การสืบพันธุ์ของปลาแต่ละชนิด อีกทั้งยังพบว่าปลาหลายชนิดมีแหล่งวางไข่เฉพาะพื้นที่ ผลการศึกษานี้แสดงให้เห็นถึงความหลากหลายทางชีวภาพที่สูงในอ่าวไทย และให้ข้อมูลเชิงลึกเกี่ยวกับแหล่งวางไข่ของปลา ซึ่งอาจเป็นประโยชน์ต่อการหารือเกี่ยวกับแนวทางการจัดการประมงในอนาคต

During infection, Listeria adhesion protein (LAP), a housekeeping enzyme, acts as a tight junction modulator (TJM) through interaction with Hsp60 to facilitate Listeria monocytogenes translocation across the intestinal epithelial barrier. Here, we used purified LAP as a potential TJM to overcome the limiting and variable effects observed by other agents in the class. We structurally determined the LAP interaction alone and in complex with Hsp60 utilizing cryo-EM and computational analysis. LAP structure resolved at 2.83 Å, forms multimeric interlocking dimers and tetramers, and the N-domain interacts with Hsp60, while the C-domain bridges the bacterial surface receptor InlB. Mobulas (manta and devil rays) are large-scale ram filter feeders that separate planktonic food particles from large volumes of water with minimal clogging. This contrasts with most human-made filters that can suffer from problematic clogging requiring additional mechanisms for clearing blocked surfaces and maintaining performance. Prior studies have shown that mobulas employ a unique mechanism referred to as ricochet separation to filter feed, whereby captive vortices in filter pores cause particles to bounce off the filter surfaces and away from the filter pores. Soybean (Glycine max) peptide lunasin exhibits significant cancer-preventive, antioxidant, and hypocholesterolemic effects. This study aimed to identify quantitative trait nucleotides (QTNs) associated with lunasin content and to annotate the candidate genes in the soybean genome. The mapping panel of 144 accessions was gathered from the USDA Soybean Germplasm Collection, encompassing diverse geographical origins and genetic backgrounds, and was genotyped using SoySNP50K iSelect Beadchips. Alcohol-induced dysregulation of microglial activity is associated with neuroinflammation, cognitive decline, heightened risk for neurodegenerative diseases, alcohol dependence, and escalation of alcohol drinking. Given the challenge of longitudinally sampling primary microglia, we optimized an in vitro method to differentiate peripheral blood mononuclear cells (PBMC) from rhesus macaque (RM) into induced microglia-like cells (RM-iMGLs). The RM-iMGLs displayed transcriptional profiles distinct from monocyte progenitors and closely resembling primary microglia. Notably, morphological features showed that differentiated RM-iMGLs derived from subjects with chronic alcohol consumption (CAC), while bigger, exhibited a bipolar-like morphology. Fusarium Head Blight (FHB) of wheat, caused by the fungus Fusarium graminearum, is a devastating disease that led to epidemics in the U.S. during the 1990s, resulting in economic losses exceeding $2.6 billion. Typical symptoms include bleached spikelets and shriveled grains, often contaminated with trichothecene mycotoxins that make them unsafe for food or feed. FHB severity and mycotoxin production are influenced by a complex interplay between host genotype, environmental factors, and pathogen genotype. Dysbiosis and impaired gut barrier integrity contribute to chronic immune activation associated with both obesity and HIV infection. Given the increased incidence of obesity in people living with HIV, we explored the impact of obesity on the gut microbiome and microbial translocation (MT) biomarkers during HIV infection and antiretroviral therapy (ART). Lean and obese rhesus macaques were infected with simian immunodeficiency virus (SIV) and subsequently treated with ART. Obese animals exhibited higher initial MT and inflammation biomarkers that remained constant throughout the study, while lean animals exhibited significant increases in these biomarkers that approached levels observed in obese animals. In the early 1980s Diadema antillarum, or the long-spined urchin, was hit by a catastrophic mass mortality event across the western Atlantic and Gulf of Mexico, inclusive of the Florida Keys. The cause of this die-off event was likely triggered by a pathogen or environmental stressor that decimated native populations by up to 98%. The species has not yet fully recovered and is struggling with recruitment success which is why it may benefit from an ex-situ recovery program. Cells exposed to proteotoxic stress invoke adaptive responses aimed at restoring proteostasis. Our previous studies have established a firm role for the transcription factor Nuclear factor-erythroid derived-2-related factor-1 (Nrf1) in responding to proteotoxic stress elicited by inhibition of cellular proteasome. Following proteasome inhibition, Nrf1 mediates the synthesis of new proteasomes, thus enabling cells to mitigate proteotoxic stress. Here, we report that under similar circumstances, multiple components of the autophagy-lysosomal pathway (ALP) were transcriptionally upregulated in an Nrf1-dependent fashion, thus providing the cells with an additional route to cope with proteasome insufficiency. Staphylococcus aureus, traditionally viewed as an extracellular pathogen, is increasingly recognized for its ability to persist intracellularly, particularly within macrophages. This intracellular lifestyle is central to osteomyelitis, a chronic bone infection characterized by persistent inflammation, bone destruction, and impaired repair. Within bone, S. aureus exploits macrophage plasticity by driving a shift from pro-inflammatory, bactericidal M1-like states to anti-inflammatory, tissue-reparative M2-like phenotypes. This polarization suppresses immune clearance and promotes an environment conducive to bacterial survival and dissemination. Additional strategies – including biofilm formation, small colony variants, and inhibition of phagolysosomal killing – further enhance persistence and immune evasion. While these …

Tricolored bats (Perimyotis subflavus) have experienced substantial population losses from white-nose syndrome (WNS), a disease caused by the pathogenic fungus Pseudogymnoascus destructans (Pd) which infects hibernating bats. Determining population genetic structure helps assess genetic diversity and the impacts of wildlife threats to populations. Pre-WNS data shows weak-to-negligible structure and panmictic P. subflavus populations at core portions of their range. However, current genetic data from their southeastern U.S. range is lacking. The Enneacanthus genus consists of three species of freshwater fish endemic to eastern North America: E. obesus (Banded Sunfish), E. gloriosus (Blue Spotted Sunfish), and E. chaetodon (Black Banded Sunfish). Previous studies using molecular data suggest that E. obesus and E. gloriosus are more closely related, with E. chaetodon as the most distantly related species in the genus. This study investigates the phylogenetic relationship among the Enneacanthus species, using karyotype characteristics - chromosome number, morphology and fundamental number - as comparative metrics. Background: Glioblastoma (GBM) is the most common malignant brain tumor with an abysmal prognosis. Since complete tumor cell removal is impossible due to the infiltrative nature of GBM, accurate measurement is paramount for GBM assessment. Preoperative magnetic resonance images (MRIs) are crucial for initial diagnosis and surgical planning, while follow-up MRIs are vital for evaluating treatment response. The structural changes in the brain caused by surgical and therapeutic measures create significant differences between preoperative and follow-up MRIs.
