Gli proteins are involved in cell fate determination, proliferation, and patterning in many species and are major effectors of Hedgehog (Hh) signaling. There are three Gli proteins in humans, and mutations or errors in their regulation lead to a variety of developmental disorders or cancer. However, the mechanisms by which they interact with co-factors are poorly understood. We are analyzing co-factors of Gli proteins using TRA-1 in Caenorhabditis nematodes. Native plants are of great value in landscape maintenance. Despite their importance in the landscape, the salt tolerance of most native plants has received little attention. The present research was designed to assess morphological, physiological, and biochemical responses of four Utah-native plants [Arctostaphylos uva-ursi (kinnikinnick), Cercocarpus ledifolius (curl-leaf mountain mahogany), Cercocarpus montanus ‘Coy’ (alder-leaf mountain mahogany), and Shepherdia ×utahensis ‘Torrey’ (hybrid buffaloberry)] at different salinity levels. Apoptosis is a type of regulated cell death important for normal embryonic development and maintenance of adult tissues by removing excess or dysfunctional cells to ensure proper functioning of organs. The Bcl-2 family of proteins determines whether apoptosis remains suppressed or becomes activated through the balance of interactions among pro-survival and pro-death members. OBJECTIVES: To establish a framework by which experts define disease subsets in systemic sclerosis associated interstitial lung disease (SSc-ILD).
METHODS: A conceptual framework for subclinical, clinical and progressive ILD was provided to 83 experts, asking them to use the framework and classify actual SSc-ILD patients. Each patient profile was designed to be classified by at least four experts in terms of severity and risk of progression at baseline; progression was based on 1-year follow-up data. A consensus was reached if ≥75% of experts agreed. Experts provided information on which items were important in determining classification.

To date, it has invaded and has now established its existence in Australia, New Zealand, and several of the Pacific Islands. The integration of the human fat mass and obesity associated (FTO) gene into turfgrass is a novel approach at improving cell proliferation and abiotic stress resistance. The FTO protein is an RNA demethylase responsible for epigenetic regulation of the genome. In related rice, the gene is associated with increased crop yield, tiller number, and aerial biomass. It is proposed to work via demethylation of repeat RNA associated with chromatin remodeling, causing widespread transcriptional activation. Dysregulated hyperinflammatory host immune response to underlying bacterial infections is a characteristic of sepsis. In sepsis, bacteria often trigger abnormal hyperinflammatory responses which can cause multiple organ failure and if sustained can lead to an immunosuppressive phase where the host is susceptible to secondary infections caused by opportunistic bacteria like Pseudomonas aeruginosa (P. aeruginosa). TDG, which is a member of the uracil DNA glycosylase superfamily, plays a critical role in the active demethylation process in mammals by removing oxidized derivatives of 5-methylcytosine (mC). While human TDG has been extensively studied and found to possess 5-formylcytosine and 5-carboxylcytosine DNA glycosylase activity, little is known regarding this demethylation process in vertebrates other than the human species. Previous research proposed a catalytic sequence in motif 1 of zebrafish TDG1 and TDG2 that modulates DNA glycosylase activity based on mutational, kinetic, and modeling analyses of these enzymes. Cryptococcus neoformans is a pathogenic basidiomycetous yeast that causes meningitis in immunocompromised patients. This lethal fungus is sometimes referred to as the “sugar coated killer” due to its polysaccharide capsule. It is estimated that 152,000 cases of cryptococcal infection occur each year and result in 112,000 deaths. The ability of C. neoformans to adapt to host temperature is a main factor responsible for virulence. Cryptococcus neoformans is a fungus that primarily infects humans who have weakened immune systems. An azole drug, Fluconazole, is commonly administered against C. neoformans in regions were cryptococcosis is most prevalent, most notably Sub-Saharan Africa. However, C. neoformans can gain resistance to Fluconazole through becoming an aneuploid. Supplementation with nicotinamide riboside (NR), a form of vitamin B3 and a precursor of nicotinamide adenine dinucleotide (NAD+) extends lifespan in the nematode C. elegans and delays aging-related pathologies in mammals. During aging, levels of NAD+ decline causing metabolic dysfunction and oxidative damage. Studies in C. elegans found that when NR was administered during larval development it induced the mitochondrial unfolded protein response (UPRmt), which is frequently associated with lifespan extension. The free-living amoeba, Naegleria fowleri, can cause a rare yet usually lethal infection of the brain called primary amebic meningoencephalitis. Because of poor diagnostics and limited treatment options, the mortality rate associated with the disease is >97%. Due to our finding that glucose is critical for trophozoite growth in culture, we have been interested in exploiting amoebae glucose metabolism to identify new potential drug targets. Kinetoplastid parasite infections remain a global health burden. Here, we have characterized inhibitors of an essential Trypanosoma brucei glycolytic enzyme, enolase (TbENO). We anticipate TbENO inhibitors will be potent anti-trypanosomals, as T. brucei relies on glycolysis for ATP production in the blood of infected mammals. Additionally, the phosphonate enolase inhibitors being considered are well-tolerated in mammals, suggesting their potential in developing novel therapeutics for kinetoplastid infections. TbENO was cloned into the bacterial expression vector, pQE-30, and the heterologously-expressed protein was purified by nickel affinity and assayed in a coupled enzyme assay. Reactions between Rongalite (Na+O-SOCH2OH) and epoxides were studied with the aim of developing new synthetic routes for the procurement of sulfone diols. This class of compounds contains essential building blocks in the construction of sulfur heterocycles, which have been implied in the development of several medicinal compounds. Thus far, the desired diols have not been isolated. During control experiments with bromo alcohols, an unknown product was isolated but remains to be identified.

Tuberculosis (TB) is an infectious disease caused by the bacteria Mycobacterium tuberculosis that primarily attacks the lungs. Drug resistance for M. tuberculosis is emerging and therefore new drug targets need to be identified. One way to target TB is through the inhibition of the tryptophan biosynthetic pathway of M. tuberculosis since production of tryptophan is essential for the replication of M. tuberculosis.
