Mechanisms Of Thai Medicinal Plant Extracts On The Attenuation Of Glutamate-Mediated Neurotoxicity, Anchalee Prasansuklab

Chulalongkorn University Theses and Dissertations (Chula ETD)

As the world population ages, neurodegenerative diseases, particularly Alzheimer's disease, are becoming a major public health concern with increasing incidence and prevalence worldwide while no cure currently exists. To challenge this situation, herbal medicine may provide a potential alternative treatment as use of natural-derived substances has been proven to be effective in the prevention and/or treatment of several diseases. During the past decades, increasing evidence has implicated excessive glutamate levels in the pathway of neuronal cell death. Thailand is a place known for cultivating a variety of tropical plants and herbs, so far many of them have never been examined …

Chulalongkorn University Theses and Dissertations (Chula ETD)

Ultraviolet radiation from sunlight is a significant environmental factor in skin damage and can induce hyperpigmentation disorder and aesthetic problem. Development of novel whitening phytochemical compounds from natural products has been become trends recently. The purpose of this study was to find some plant extracts that reduce melanin synthesis and melanogenetic gene expression in alpha-MSH-induced B16F10 mouse melanoma cells. To screen total phenolics and flavonoids, antioxidant activity, and anti-mushroom tyrosinase activity, we used 13 plants which were extracted with petroleum ether, dichloromethane and ethanol solvents, subsequently. We found that total phenolic content of 13 plants extracts was found in the …

Role Of Progesterone Receptor A And B Isoforms In Lung Neuroendocrine Tumors, Teeranut Asavasupreechar 2017 Faculty of Allied Health Sciences

Role Of Progesterone Receptor A And B Isoforms In Lung Neuroendocrine Tumors, Teeranut Asavasupreechar

Chulalongkorn University Theses and Dissertations (Chula ETD)

Lung neuroendocrine tumors (NETs) incidences are increasing in recent years. Women with lung NETs have significantly better survival rates as compared to men suggesting the involvement of sex steroids and their receptors in the progression of lung NETs. Recent data suggested that progesterone receptor (PR) may play a role in the survival of lung NET patients. PR exists as two major isoforms, PRA and PRB. How expression of PR isoforms affects proliferation of lung NETs and patient's survival is not known. To determine the role of PR isoforms in lung NETs, we constructed H727 lung NET cell models expressing PRB …

Navigating Human Cytomegalovirus (Hcmv) Envelopment And Egress, William Longeway Close 2017 Wayne State University

Navigating Human Cytomegalovirus (Hcmv) Envelopment And Egress, William Longeway Close

Wayne State University Dissertations

Human cytomegalovirus (HCMV) is a ubiquitous viral pathogen. In individuals with fully functioning and mature immune systems, HCMV is associated with mild symptoms prior to establishing latency. In individuals with naïve or compromised immune systems, HCMV is capable of causing severe organ damage. HCMV is the leading infectious cause of congenital birth defects and a major non-genetic cause of hearing loss. Unfortunately, antiviral treatment options lack diversity due to limited knowledge of virion replication. If HCMV replication were better understood, new antiviral treatments could be developed.

In this work, we describe the development and implementation of new tools to study …

Expression, Purification And Characterization Of Lysine Methyltransferase Smyd5, Wen Xue 2017 Wayne State University

Expression, Purification And Characterization Of Lysine Methyltransferase Smyd5, Wen Xue

Wayne State University Theses

Methylation of histones and non-histone proteins play vital roles in numerous cellular processes including gene expression regulation and DNA damage response. The identifications of methyltransferase SMYD protein family are not well characterized. SMYD5 is a unique and critical member of SMYD family involved in immune response, stem cell renew, hematopoiesis regulation and cancer metastasis. Understanding its function and structure is monumental to human disease. With the achievement of SMYD5 expression and purification, the association between SMYD5 and its substrate histone H4 has been investigated. While possessing a poly-E tail instead of the TPR domain included in other members, SMYD5 is …

The rapid proliferation of publications employing chemical assays of fish hard parts, and otoliths in particular, has led to many novel insights into the migration patterns, life history strategies and mixed stock dynamics of fishes across the globe (Campana et al., 2000; Elsdon et al., 2008; Walther & Limburg, 2012). These insights include uncovering diverse migratory strategies within species and populations (Kerr et al., 2009; Hogan et al., 2014; Schoen et al., 2016), quantifying rates of mixing among stocks across management boundaries (Rooker et al., 2008; Walther & Thorrold, 2010) and estimating the relative …

The Effect Of Chronic Nutrient Addition From Wastewater On Forest Ecosystems At The Rice Rivers Center, Michael Beck 2017 Virginia Commonwealth University

The Effect Of Chronic Nutrient Addition From Wastewater On Forest Ecosystems At The Rice Rivers Center, Michael Beck

Theses and Dissertations

Wastewater application to land can be a useful tool for mitigating impacts of nutrient enrichment on aquatic systems. A land application treatment system at VCU’s Rice Rivers Center in Charles City County, VA provided an opportunity to study the impact of wastewater addition on the biogeochemistry of forests representative of the Mid-Atlantic Coastal Plain. Nutrient concentrations in throughfall and leachate were measured at Treatment and Control sites to assess differences in nutrient deposition and retention. Wastewater amended plots from the Walter L. Rice education building received 20-fold (N) and 6-fold (P) higher inputs relative to Control plots and plots located …

Biochemical Analysis Of Putative Single-Stranded Nucleic Acid Binding Proteins In Porphyromonas Gingivalis, Steve H. Kokorelis 2017 Virginia Commonwealth University

Biochemical Analysis Of Putative Single-Stranded Nucleic Acid Binding Proteins In Porphyromonas Gingivalis, Steve H. Kokorelis

Theses and Dissertations

Proteins that bind to both DNA and RNA embody the ability to perform multiple functions by a single gene product. These nucleic acid binding proteins in prokaryotes can play a vital role in many cellular processes, including replication, transcription, gene expression, recombination, and repair, to name a few. Nucleic acid binding proteins have unique functional characteristics that stem from their structural attributes that have evolved in a widely-conserved manner. In Escherichia coli (E. coli), the highly-conserved histone-like protein, HU, which predominates as a heterodimer of HUα and HUβ, has been found to bind to both dsDNA and ssDNA. …

The Localization And Function Of Novel Tetrahymena Thermophila Cytoskeletal Genes Bbc29 And Bbc39, Emily K. Moore, Nicole C. Zanolli

Undergraduate Research Symposium Posters

Ciliary function is known to play an important role in many human conditions, including chronic sinus and pulmonary diseases and problems with infertility. Cilia are cytoskeletal structures that protrude from the cell body to facilitate movement. Ciliary structure is conserved throughout eukaryotes, from unicellular to multicellular organisms, including humans. A clear understanding the proteins that compose cilia and how they interact with one another will increase our knowledge about important cytoskeletal elements. Because cilia are difficult to study in multicellular organisms, the unicellular eukaryote Tetrahymena thermophila serves as a useful model for the study of cytoskeletal genes, due to their …

Evolution Of Protein Complexes In Bacterial Species, Shwetha Hara Sridhar, Wedad Albalawi, Peter Uetz 2017 Virginia Commonwealth University

Evolution Of Protein Complexes In Bacterial Species, Shwetha Hara Sridhar, Wedad Albalawi, Peter Uetz

UROP Posters

Protein complexes are composed of two or more associated polypeptide chains that may have different functions. Protein complexes play a critical role for all processes in life and are considered as highly conserved in evolution. In previous studies, protein complexes from E. coli or Mycoplasma pneumoniae have been characterized experimentally, revealing that a typical bacterial cell has on the order of 500 protein complexes. Using gene homology (orthology), these experimentally-observed complexes can be used to predict protein complexes across many species of bacteria. Surprisingly, the majority of protein complexes is not conserved, demonstrating an unexpected evolutionary flexibility.

Identifying and analyzing biological mixture samples at a crime scene are of paramount concern for forensic scientists, especially if that type of evidence contains only one cell type. The presence of multiple contributors in a biological evidence sample reduces the probative value of DNA evidence and can sometimes lead to its eventual loss of value. As such, this study was performed in an attempt to examine and evaluate flow cytometry analysis as a means to separate blood mixture samples labeled with fluorescent antibodies. Fluorescein Isothiocyanate (FITC) antibodies were specifically targeted and bound to HLA (Human Leukocyte Antigens) markers present on …

Manipulating Lipid Spreading Domain Formation With Compositional Gradients And Plasmonic Nanoparticles, Chen-min Hung 2017 Claremont McKenna College

Manipulating Lipid Spreading Domain Formation With Compositional Gradients And Plasmonic Nanoparticles, Chen-Min Hung

CMC Senior Theses

After colliding two solid-supported spreading bilayers of different compositions, we produce a dynamic gradient as the newly-healed bilayer equilibrates. We apply this approach to study the formation of galactosyl ceramide (GalCer) domains. With a single experiment, we are able to explore the effects of varying cholesterol concentration on GalCer domains. To further control domain formation, we aimed to locally heat membranes with embedded plasmonic nanoparticles. However, we discovered a percolation threshold of spreading lipids over the array of nanoparticles. We found that surfaces with gold nanoparticles deposited at an average inter-particle distance of 40 nm inhibited lipid bilayer spreading, but …

Phage Display To Identify Functional Resistance Mutations To Rigosertib, Nedim Filipovic 2017 Claremont McKenna College

In vitro protein selection has had major impacts in the field of protein engineering. Traditional screens assay individual proteins for specific function. Selection, however, analyzes a pool of mutants and yields the best variants. Phage display, a successful selection technique, also provides a reliable link between variant phenotype and genotype. It can also be coupled with high throughput sequencing to map protein mutations; potentially highlighting vital mutations in variants. We propose to apply this technique to cancer therapy. RAF, a serine/threonine kinase, is critical for cell regulation in mammals. RAF can be activated by oncogenic RAS, found in over 30% …

Molecular Modeling Of Novel Tryptamine Analogs With Antibiotic Potential Through Their Inhibition Of Tryptophan Synthase, Jared Schattenkerk 2017 Claremont McKenna College

Molecular Modeling Of Novel Tryptamine Analogs With Antibiotic Potential Through Their Inhibition Of Tryptophan Synthase, Jared Schattenkerk

CMC Senior Theses

The growing prevalence of antibiotic-resistant bacteria is a global health crisis that threatens the effectiveness of antibiotics in medical treatment. Increases in the number of antibiotic-resistant bacteria and a drop in the pharmaceutical development of novel antibiotics have combined to form a situation that is rapidly increasing the likelihood of a post-antibiotic era. The development of antibiotics with novel enzymatic targets is critical to stall this growing crisis. In silico methods of molecular modeling and drug design were utilized in the development of novel tryptamine analogs as potential antibiotics through their inhibition of the bacterial enzyme tryptophan synthase. Following the …
