Studies Of The Dutpase Of The Western Corn Rootworm, Carlos A. Riera-Ruiz 2021 University of Nebraska-Lincoln

Studies Of The Dutpase Of The Western Corn Rootworm, Carlos A. Riera-Ruiz

School of Biological Sciences: Dissertations, Theses, and Student Research

The western corn rootworm (WCR), Diabrotica virgifera virgifera, is a major corn pest in the United States and Europe. WCR has developed resistance to multiple management strategies, including Cry proteins. Even though the biology and ecology have been thoroughly studied in WCR, their genome and molecular mechanisms are understudied. This work focuses on the ubiquitous enzyme deoxyuridine triphosphatase (dUTPase) encoded by the DUT gene. dUTPase hydrolyzes dUTP into dUMP and pyrophosphate. It contributes to genome stability by keeping the uracil-to-thymine ratio at a certain level. In WCR, two dUTPase isoforms were predicted using transcriptome analyses. These two potential isoforms …

Biochemical Characterization Of Small Molecule Inhibitor Binding On A Ras Related Gtpase And Its Effector Interactions, Djamali Muhoza 2021 University of Arkansas, Fayetteville

Biochemical Characterization Of Small Molecule Inhibitor Binding On A Ras Related Gtpase And Its Effector Interactions, Djamali Muhoza

Graduate Theses and Dissertations

The Ras superfamily of GTPases has 167 proteins that are involved in various cellular processes such as proliferation, transformation, migration, and inhibition of cell death. Mutations, abnormal expression, and function of these proteins are observed in many diseases, including several forms of cancer. Even though these GTPases were among the first discovered oncogenes, no successful Ras drug candidate has successfully passed clinical trials. Drugs targeting these proteins have failed mainly because of the complexity of their regulation, their high affinity to GTP, and their structure’s dynamic nature. Recently, novel promising targeting approaches have renewed interest in the Ras drug discovery …

Validation Of Anti-Oxidative Stress Genes From Genome-Wide Screening Of Escherichia Coli, Carson Ercanbrack 2021 University of Arkansas, Fayetteville

Validation Of Anti-Oxidative Stress Genes From Genome-Wide Screening Of Escherichia Coli, Carson Ercanbrack

Chemistry & Biochemistry Undergraduate Honors Theses

The primary purpose of this project is to evaluate the genes that play a role in the oxidative stress response in Escherichia coli. In doing so, the entire genome of E. coli was subject to throughput in which individual genes were determined to have a role in the bacteria’s oxidative stress response. Moreover, this project focused on the validation of the genes that were able to pass the initial throughput stage. The genes were subject to two forms of validation. In the first validation technique, candidate genes were overexpressed and minimum inhibitory concentrations of hypochlorous acid were taken. Following, a …

Understanding The Non-Classical Secretion Mechanism Of Interleukin-1 Alpha, Clay Martin 2021 University of Arkansas, Fayetteville

Understanding The Non-Classical Secretion Mechanism Of Interleukin-1 Alpha, Clay Martin

Chemistry & Biochemistry Undergraduate Honors Theses

The Interleukin 1 family is a unique collection of cytokines in that they are primarily associated with both acute and chronic inflammation, which is associated with many diseases. In classical protein secretion, a protein, typically with an N-terminal signal peptide, is synthesized on a ribosome of a cell either in the cytoplasm or on the rough endoplasmic reticulum, transported to and merges with the Golgi apparatus, and is then sent to bind with either a lysosome or the cell membrane or is released from the cell via exocytosis. Thus, a non-classical secretion mechanism is any mechanism that differs from this. …

Reaction Pathways Initiated By One-Electron Oxidation Of Guanine: Reaction Pathways Leading To Stable End Products, Zach Cutright 2021 East Tennessee State University

Reaction Pathways Initiated By One-Electron Oxidation Of Guanine: Reaction Pathways Leading To Stable End Products, Zach Cutright

Undergraduate Honors Theses

8-hydroxyguanine (8-oxoG) is one of the most important products resulting from the oxidation of guanine in DNA. 8-oxoG is known as a biomarker of oxidative stress such as lung cancer. 2,5-diamino-4H-imidazol-4-one (Iz) and its hydrolysis product 2,2-diamino-4-[(2-deoxy-β-D-erythro-pentofuranosyl)-amino]-2,5-dihydrooxazol-5-one (Oz) are also important products of oxidation of guanine in DNA. Recently, novel types of dimer intermediates related to 8-oxoG and Iz have been identified in our research lab. Using high performance liquid chromatography (HPLC), LC-MS, and NMR coupled with educated hypotheses, it is possible to identify the products, X1 and X2, resulting from the oxidation of guanine and decipher the mechanisms leading …

Analyzing The Effects Of E-Hook Peptides On Kinesin-1, Ashton Ward Murrah, Baylee Hope Howard 2021 University of Mississippi

Analyzing The Effects Of E-Hook Peptides On Kinesin-1, Ashton Ward Murrah, Baylee Hope Howard

Honors Theses

Cancer is the second leading cause of death in the United States. Cancerous growth is a result of oncogenes, or mutated genes that increase the rate of cell division in an uncontrolled manner. Cell division, which consists of mitosis and cytokinesis phases, is dependent upon the active movement of kinesin motor proteins along microtubules to rearrange the cytoskeleton for equitable distribution of genetic material to daughter cells. As kinesins are vital to this process, if we could prevent kinesin from binding to the microtubules, cell division would cease.

The goal of this study is to develop a method to prevent …

Detailed Analysis Of The Role Of Mtr4 Arch Domain In Rna Binding And Helicase Activity, Keith J. Olsen 2021 Utah State University

Detailed Analysis Of The Role Of Mtr4 Arch Domain In Rna Binding And Helicase Activity, Keith J. Olsen

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The central dogma of biology states that DNA is transcribed into RNA, which is translated into protein. However, this concept portrays RNA purely as an intermediary step from DNA to protein. RNA plays an essential and vital role in a cell, including; regulating protein and RNA levels, act as a template for protein synthesis, cell differentiation, regulatory processes, and signaling molecules. If cells do not adequately monitor and regulate RNA levels, it can lead to disease states and even cell death. The main protein complex that regulates the maturation and degradation of RNA in the nucleus is the RNA nuclear …

Chemical Engineering Faculty Publications and Presentations

Ultrafiltration membranes, that respond to an external magnetic field and local temperature have been developed. Surface-initiated activator-generated electron transfer (AGET) atom transfer radical polymerization (ATRP) has been used to graft poly(N-isopropylacrylamide) (PNIPAm) from the surface of 300 kDa regenerated cellulose membranes. The polymerization initiator was selectively attached to the entire membrane surface, only the outer membrane surface or only the inner pore surface. A superparamagnetic nanoparticle was attached to the end of the polymer chain. The DI water flux as well as the flux and rejection of bovine serum albumin were investigated in the absence and presence of a 20 …

The Small Gtpase Rab-35 Facilitates The Initiation Of Phagosome Maturation And Acts As A Robustness Factor For Apoptotic Cell Clearance, Ryan Haley, Zheng Zhou 2021 The Texas Medical Center Library

The Small Gtpase Rab-35 Facilitates The Initiation Of Phagosome Maturation And Acts As A Robustness Factor For Apoptotic Cell Clearance, Ryan Haley, Zheng Zhou

Faculty, Staff and Students Publications

We recently identified the novel function of the small GTPase RAB-35 in apoptotic cell clearance in Caenorhabditis elegans, a process in which dying cells are engulfed and degraded inside phagosomes. We have found that RAB-35 functions in two separate steps of cell corpse clearance, cell corpse recognition and the initiation of phagosome maturation. During the latter process, RAB-35 facilitates the removal of phosphatidylinositol-4,5-bisphosphate (PI(4,5)P2) from the membranes of nascent phagosomes and the simultaneous production of phosphatidylinositol-3-P (PI(3)P) on these same membranes, a process that we have coined the PI(4,5)P2 to PI(3)P shift. RAB-35 also promotes the recruitment of the …

Membrane-bound angiotensin-converting enzyme 2 is important in regulation of the renin-angiotensin-aldosterone system, but the association of cleaved soluble ACE2 (sACE2) with cardiovascular disease (CVD) is unclear. We evaluated the association of sACE2 with cardiac biomarkers, structure, and function and cardiovascular events in the Atherosclerosis Risk in Communities Study. sACE2 was measured in a subset of 497 participants (mean age 78±5.4 years, 53% men, 27% black); Cox regression analyses assessed prospective associations of sACE2 with time to first CVD event at median 6.1-year follow-up. sACE2 was higher in men, blacks, and participants with prevalent CVD, diabetes, or hypertension. Higher sACE2 levels …

Pathological Mechanisms Of Vacuolar Aggregate Myopathy Arising From A Casq1 Mutation, Amy D Hanna, Chang Seok Lee, Lyle Babcock, Hui Wang, Joseph Recio, Susan L Hamilton

Faculty, Staff and Students Publications

Mice with a mutation (D244G, DG) in calsequestrin 1 (CASQ1), analogous to a human mutation in CASQ1 associated with a delayed onset human myopathy (vacuolar aggregate myopathy), display a progressive myopathy characterized by decreased activity, decreased ability of fast twitch muscles to generate force and low body weight after one year of age. The DG mutation causes CASQ1 to partially dissociate from the junctional sarcoplasmic reticulum (SR) and accumulate in the endoplasmic reticulum (ER). Decreased junctional CASQ1 reduces SR Ca

Molecular And Genetic Studies Of Robo2 Transcriptional Regulation In The Central Nervous System Of Drosophila Melanogaster, Muna Abdal Rahim Abdal Rhida 2021 University of Arkansas, Fayetteville

Molecular And Genetic Studies Of Robo2 Transcriptional Regulation In The Central Nervous System Of Drosophila Melanogaster, Muna Abdal Rahim Abdal Rhida

Graduate Theses and Dissertations

Drosophila Robo2 axon guidance receptor is a member of the evolutionarily conserved Roundabout (Robo) protein family that is involved in directing axons that cross the midline to the other side of the animal body. Robo2 roles mainly depend on two factors: The functional domains of the Robo2 protein, which is extensively studied, and the dynamic transcription of robo2 in various subsets of cells throughout embryogenesis which is not fully understood. Thus, knowing robo2 enhancers that transcriptionally regulate robo2 during embryogenesis is significant. To investigate robo2 potential enhancers, we screened 17 transgenic lines of Drosophila that were generated by Janelia Research …

Characterization Of Protein Aggregation Using A Solid-State Nanopore Device, Mitu Chandra Acharjee 2021 University of Arkansas, Fayetteville

Characterization Of Protein Aggregation Using A Solid-State Nanopore Device, Mitu Chandra Acharjee

Graduate Theses and Dissertations

Protein aggregation has been linked to many chronic and devastating neurodegenerative human diseases and is also strongly associated with aging. In the case of neurodegenerative diseases, α, β tubulins and tau proteins dissociate in a neuron cell and aggregate both intra and extra-cellularly. Tau and tubulin aggregations were found as one of the major causes of many neurodegenerative diseases, such as Parkinson’s, Picks, Alzheimer’s, Huntington, and Prion. Finding the state and mechanism of protein aggregation is significant. In this work, tau and tubulin aggregations were detected in ionic solutions using the solid-state nanopore technique. Besides tau and tubulin, aggregations of …

We developed High Resolution Melt Analysis (HERMA) as a rapid and reliable molecular diagnostic assay for the detection and identification of the main bacterial species recovered from the blood and lesions of bacterial chondronecrosis with osteomyelitis (BCO) from the lame broilers at the University of Arkansas poultry research farm. Also, the present study confirms that raising young birds on suspended wire flooring has been proved to successfully induce lameness attributable to BCO with birds being more susceptible to bacteraemia than those that were raised on litter flooring. The newly described pathogen, Staphylococcus agnetis, has been reported to be overrepresented in …

Effects Of Environmentally Relevant Concentrations Of 4-Methyl-1- Cyclohexanemethanol (Mchm) On General Esterase And Glutathione-S-Transferase Activity In Fathead Minnows (Pimephales Promela), Emily De Wind 2021 Southern Adventist University

Effects Of Environmentally Relevant Concentrations Of 4-Methyl-1- Cyclohexanemethanol (Mchm) On General Esterase And Glutathione-S-Transferase Activity In Fathead Minnows (Pimephales Promela), Emily De Wind

Research in Biology

Abstract

In 2014, several thousand gallons of coal-processing chemicals, which included 4-methyl-1- cyclohexanemethanol (MCHM), spilled into the Elk River in West Virginia. The location of this spill in West Virginia’s chemical valley is historically significant in defining the exploitations of people residing in this area. The concentration of crude MCHM in the river was approximated at 0.15 µg/L. Although some initial studies did consider the effects of MCHM exposure on humans, little attention has been given to aquatic wildlife. In this project, I will expose fathead minnows, Pimephales promelas, an environmentally relevant species, to concentrations of MCHM between zero …

Finite Element Analysis Of Impact Resistant Composites Inspired By Peacock Mantis Shrimp, Nandati Shrestha 2021 University of Southern Maine

Finite Element Analysis Of Impact Resistant Composites Inspired By Peacock Mantis Shrimp, Nandati Shrestha

Thinking Matters Symposium

The fist-like club of the peacock mantis shrimp, a 5-inch marine crustacean, can strike its prey with speed faster than a .22-caliber bullet with an impact force more than 1,000 times its own weight. Although these creatures punch so fast that it even boils the water, they don’t take any damage. This incredible insusceptibility is due to the arrangement of mineralized fiber layers in which each fibrous layer is laid at a slightly rotated angle to form a helicoidal structure that acts as a shock absorber for the club. The goal is to perform a finite element analysis to investigate …

Mems Directional Acoustic Sensors, Colby Damren 2021 University of Southern Maine

Mems Directional Acoustic Sensors, Colby Damren

Thinking Matters Symposium

The purpose of my project is to test and verify two Micro-Electro-Mechanical-System (MEMS) microphones. The two MEMS devices are biomimetic microphones that imitate the eardrums of the Ormia Ochracea, a parasitic fly that listens for host crickets to lay their eggs. The MEMS microphones mimic the eardrum spacing of the Ormia Ochracea with a spacing of 1000um. This is roughly twice the opening of the Ormia Ochracea. The microphones will be tested for directionality inside a wooden box. This box will be lined with beveled foam to prevent any echo or outside noise from interfering with the results. These results …

Design, Simulation And Testing Of Biomimetic Directional Acoustic Sensors, Brendan Francis 2021 University of Southern Maine

Design, Simulation And Testing Of Biomimetic Directional Acoustic Sensors, Brendan Francis

Thinking Matters Symposium

The Ormia ochracea, a species of parasitic fly, has become the focal point in sound localization research because of its finely tuned hearing abilities. The female of this species uses its super highly directional hearing to pinpoint the call of a host cricket, with hypersensitivity of frequency and phase difference, to reach and dispose of its eggs on the host. The goal of this study was to further the research of a previous Project in Professor Guvench’s group which implemented MEMS (Micro Electro-Mechanical System) technology on a chip to replicate these abilities. In this iteration, however, some commercially available …
