Marshes in Louisiana are under threat from numerous natural and anthropogenic sources. A consequence of these threats are sheared marsh margins, which result from the impact of storm surge on previously oiled, weakened marsh. These conditions occurred in Louisiana marshes after Hurricane Isaac in 2012 followed the Deepwater Horizon oil spill in 2010, particularly in the shorelines surrounding Bay Jimmy. The second and third chapters of this thesis focus on the differences in biodiversity between the sheared and intact marsh margins in impacted sites in Bay Jimmy. New technologies are vital for growing industries that require large amounts of enzymes at a cheaper production cost. Plant transformation technologies coupled with established cropping systems have allowed us to produce enzymes at more efficient rates than ever before. During breeding, offspring are selected for high production of the enzyme of interest and discontinues lines of low production. This continued range is an observed phenomenon in increasingly isogenic lines, but not fully understood. We hypothesize that distinct populations of non-coding RNAs in isogenic corn lines influence enzyme accumulation in seed. Understanding the structure of chromatin in chromosomes during normal and diseased state of cells is still one of the key challenges in structural biology. Using DAPI staining alone together with Fluorescence lifetime imaging (FLIM), the environment of chromatin in chromosomes can be explored. Fluorescence lifetime can be used to probe the environment of a fluorophore such as energy transfer, pH and viscosity. The overall goal of this thesis was to genetically improve agronomic traits of pennycress (Thlaspi arvense L.; Field Pennycress) and demonstrate the production of value-added designer seed oils to domesticate pennycress and enable its establishment as a new winter annual oilseed/meal/cover crop to be grown in temperate regions of the world. In the U.S. Midwest, pennycress can be double cropped on existing farmland during the time between corn harvest and subsequent planting of soybeans the following spring. Pennycress has the potential to produce 2,000 lbs/acre seeds, which at 33% by weight oil content and 20% protein, would yield 85 gallons/acre …

Glyphosate (N-(Phosphonomethyl) glycine) is the active ingredient in Round-Up Inc. Commonly known as a weed killer, glyphosate targets the enzyme 5-enolpyruvylshikimate-3- phosphate synthase (EPSPS) in the shikimate pathway. Ultimately, glyphosate selectively induces toxicity, targeting EPSPS present in plants and selected microorganisms. Therefore, glyphosate is anticipated to be harmless to animals and humans. Our laboratory studies Drosophila melanogaster - also known as the fruit fly, in an interest to understand its gut microbiota and immune system. Bone is a common site of metastasis for many solid malignancies. Bone-metastatic cancers pose a significant clinical problem worldwide and is among the main causes for cancer patient morbidity and mortality. Patients with advanced bone-metastatic diseases often present with either osteolytic or osteogenic bone diseases as their cancers progress. These bone pathologies are products of the cancer co-opting the local bone remodeling stroma to yield important growth nutrients and factors. Background: A Phase II proof of concept (POC) randomized clinical trial was conducted to evaluate the effects of rasagiline, a monoamine oxidase B (MAO-B) inhibitor approved for Parkinson disease, in mild to moderate Alzheimer's disease (AD). The primary objective was to determine if 1 mg of rasagiline daily for 24 weeks is associated with improved regional brain metabolism (fluorodeoxyglucose–positron emission tomography [FDG-PET]) compared to placebo. Secondary objectives included measurement of effects on tau PET and evaluation of directional consistency of clinical end points. During unfavorable cellular conditions (e.g., tumor hypoxia, viral infection, nutrient deprivation, etc.), the canonical, cap-dependent translation initiation pathway in human cells is suppressed by sequestration of the cap-binding protein, eukaryotic initiation factor(eIF) 4E, by 4E-binding proteins. Circumvention of cap-dependent translation shutdown has been linked to tumor development and cancer progression. The stress-induced repression of cap-dependent translation has also been correlated with increased eIF4GI and its homolog, Death Associated Protein 5 (DAP5) expression levels, suggesting these factors have a role in cap-independent translation. Duchenne muscular dystrophy (DMD) is a genetic disorder leading to progressive muscle degeneration and weakness due to mutation in dystrophin gene, which is very important for maintaining muscle membrane integrity. Dystrophin is the largest gene in the human genome therefore more prone to mutation. There is currently no cure for DMD. Our lab recently found that Lipin1 deficient myofibers showed upregulation of necroptosis correlated with the loss of muscle membrane integrity. Our primary approach for ameliorating dystrophic phenotype in DMD is through reduction of necroptosis using drugs which can potentially upregulate Lipin1 expression. Discovery studies have identified many metabolites contained in human sweat. However, quantitative analysis of the sweat metabolome content remains mostly unknown. Furthermore several attributes, including rate, have been defined to affect sweat metabolite content, while other effectors, like diet, remain unknown. This study works to quantitatively define the metabolite impact caused by nutritional supplementation. To better understand the effect diet plays, a LC-MS method was developed focusing on improving resolution and peak width. Among bony vertebrates, the melanocortin-2 receptor ortholog is unique among the family of five melanocortin receptors on the basis that it is dependent on its accessory protein, MRAP1, for trafficking and activation, and is selective for activation by ACTH alone. Previous studies on the MC2R orthologs of select cartilaginous fish, the elephant shark (Callorhinchus milii) and the red stingray (Dasyatis akajei), revealed divergent traits in a less obligatory relationship on MRAP1 and its ability to be activated by ACTH or the MSH-sized peptides. Molecular Dynamics (MD) is a computer simulation method to analyze and study the physical movement of atoms and molecules in an environment constrained by physical and quantum mechanics laws. The systems that were of interest during this summer were metal/ligand complexes, also known as a coordination complex. A coordination complex consists of a central atom or ion, which in this case is a metal, and an array of bound molecules that are called the ligands. The ligand of interest for this summer is ethylenediamine. Both copper(II) and nickel(II) coordinate to ethylenediamine.

Extracellular signal-regulated kinase 3 (ERK3) is a member of the atypical mitogen-activated protein kinase (MAPK) subfamily, whose members have been shown to play important roles in a number of cellular processes including proliferation, differentiation, migration, and apoptosis. While signals regulating ERK3 kinase activity remain unclear, ERK3 is known to be an unstable protein with function tightly regulated via ubiquitination and proteasomal turnover. The deubiquitinating enzyme USP20 has been shown to regulate ERK3 by stabilizing the kinase, but presently, no destabilizing ubiquitin ligases have been identified. Bilayers of lipid molecules form the basis of the cell membranes in living organisms. An ion channel is a type of molecule that can temporarily open a channel in a lipid bilayer in order to allow specific ions in and out. Problems with ion channels and their interactions with lipid bilayers are associated with several diseases, including osteoporosis. Force generation in epithelial tissues is often pulsatile, with actomyosin networks generating high-tension contractile forces at the cell cortex before cyclically disassembling. This pulsed nature of cytoskeletal forces implies that there must be cellular processes to extract unidirectional changes that drive processive transformations in cell shape. During Drosophila melanogastergastrulation, the invagination of the prospective mesoderm is driven by the pulsed constriction of apical surfaces. Here, we address the mechanisms by which the irreversibility of pulsed events is achieved while also permitting uniform epithelial behaviors to emerge.
