Volumetric Fluorescence Microscopy For High-Throughput And High-Sensitivity Imaging: From Single Molecules To Tissues, Le-Mei Wang

Fluorescence microscopy is an indispensable tool in the biological sciences, enabling researchers to investigate intricate subcellular structures, particularly for volumetric studies. However, conventional optical microscopy for volumetric imaging remains fundamentally constrained by imaging speed and throughput. To bypass traditional serial z-scanning, we introduce an axially scan-free method using a phase layer cake to modulate the system's point spread function. This approach projects volumetric information onto a 2D plane in a single shot, offering high flexibility in tuning axial depth alongside simultaneous multicolor imaging with high spatial resolution and sensitivity. Scalable Single-Erbium Telecom Qudits With Record Room-Temperature Quantum Coherence In Silicon-Based Nanostructures, Alexander Kaloyeros

Advancing quantum information science demands solid-state quantum systems that maintain long quantum coherence at elevated temperatures while supporting scalable, CMOS-compatible fabrication and telecom C-band operation. No existing platform has simultaneously achieved these requirements, as state-of-the-art demonstrations of coherent control of erbium ions, with an intrinsic telecom-band optical transition, have been confined to cryogenic temperatures below < 10 K under controlled vacuum conditions. This thesis introduces a new paradigm in which materials science and engineering provides the enabling pathway to quantum coherence.

A foundry-compatible nanofabrication approach, paired with targeted materials engineering, is developed to realize a new class of CMOS-scalable quantum system: arrays of spatially isolated single-erbium-ion qudits (five-level systems) embedded in silicon-based (e.g., silicon carbide (SiC) and SiCxOy) hollow nanopillars (HNPs). Gold nanoplates and nanostars were synthesized from a gold metal salt solution (HAuCl4) using a one-step synthesis method of ultraviolet C (UVC) light irradiation. The gold salt solution was mixed with ethanol in 6 mL with the following ratios: (1.5:4.5, 3:3, and 4.5:1.5), and subsequently irradiated with UVC light. Changes in localized surface plasmon resonance (LSPR) shifts were observed in real time at 30-min intervals for 2 h. The synthesis process produces gold nanoplates. In this paper, we systematically demonstrate the design and analysis of a new type of ultra-broadband tunable metamaterial perfect absorber (MPA) comprising a top vanadium dioxide (VO2) based patterned resonating patch, a continuous metallic film at the bottom, and an intermediate dielectric substrate having a thickness of only 0.18 at the center working frequency. The simulation results reveal that the absorber achieves a bandwidth of 7.26 THz, ranging from 5.40 THz to 12.66 THz, with more than 90% absorptance and an average absorption of 98.21% under normal incidence of the incoming THz wave. This work applies the Lyapunov method to identify instabilities and compute the growth rate of a linear time-varying system. The linear system studied describes cold fresh water on top of hot salty water with a periodically time-varying background shear flow. A time-dependent weighting matrix is employed to construct a Lyapunov function candidate. The resulting linear matrix inequalities are discretized in time using the forward Euler method. As the number of temporal discretization points increases, the growth rate predicted by the Lyapunov method or Floquet theory, used for comparison, will converge to the same value obtained from numerical simulations. The Fitness for Service (FFS) analysis is performed as a quantitative assessment to evaluate the integrity condition of a pipeline. Essentially, FFS assessment helps determine whether equipment components can operate safely despite existing deficiencies. This evaluation is carried out using the Finite Element Method (FEM). In the case of an underwater pipeline that experiences leak due to anchor pull at the flange connection, it undergoes plastic deformation and is lifted approximately 1 meter. The increasing demand for sustainable practices in the metal casting industry has driven the use of recycled aluminum alloys such as ADC12. However, the addition of aluminum scrap tends to increase oxide inclusions, which reduce melt fluidity and compromise casting quality. This study utilizes the Inclusion and Fluidity Measurement Instrument (IFMI), equipped with mullite ceramic filters, to assess the melt quality of ADC12 aluminum alloy containing 30% scrap. Fluidity and inclusion characteristics were evaluated at five pouring temperatures (660°C, 680°C, 700°C, 720°C, and 740°C). This research investigates the production decline and cost increase in the X&Y oil and gas fields from 2022 to 2023. Production fell by 34%, while production costs per barrel rose by 79%. To address these issues, a series of optimization processes are proposed. These aim to restructure and enhance the production facilities to reduce current production costs. The optimizations include reducing pressure at the PPP Platform in 2024. This study investigates the potential of ceramic membranes derived from areca nut fiber as a cost-effective and environmentally sustainable material for the removal of iron (Fe) and manganese (Mn) from groundwater. Two types of membranes were fabricated: one without activation and one chemically activated using 10% sodium hydroxide (NaOH). The morphological and elemental characteristics of both membranes were analyzed using Scanning Electron Microscopy (SEM) and Energy Dispersive X-ray (EDX) mapping. The concentrations of Fe and Mn before and after treatment were measured using Atomic Absorption Spectroscopy (AAS). To develop Ga2O3 and TiOₓNᵧ thin films for optical applications, the process–structure–properties relationship was investigated using a comprehensive set of characterization techniques to establish the atomic structure and composition of the films in relation to the deposition conditions. The results provide critical insights into how deposition parameters influence the atomic structure and composition during magnetron sputtering. The overarching motivation of this work is the development of seedless, non-intrusive, laser-based diagnostics for supersonic airflow in wind tunnels. To advance this goal, this dissertation fo-cuses on three-photon excitation in a research-grade argon beam within a tabletop vacuum system, which offers a controlled environment for testing and refining the approach. The chosen excitation scheme drives argon atoms to the 3d[5/2]₃ state using a pulsed Ti:Sapphire laser system, enabling time-of-flight (ToF) measurements on atoms that subsequently undergo a multi-step decay to the metastable 4s[3/2]₂ state. Silver titanate (AgTO) was synthesized via an ion-exchange reaction between sodium titanate and silver nitrate. This study investigates the efficiency of AgTO in the combined adsorption-photodegradation process for removing Methylene Blue (MB). The synthesized material was characterized using FTIR, XRD, SEM, and EDX techniques. Batch adsorption experiments were conducted to assess the effects of AgTO dosage (0.1–1.0 g/L), initial MB concentration (5–20 mg/L), pH (3–11), and temperature (313–333 K). Indian River Lagoon Shorelines

The spatial dataset: UCF_shorelinemodel_IRL_LWL.shp, was created with support from the Indian River Lagoon National Estuaries Program and Florida Sea Grant. These projects cumulatively led to creation of a living shoreline restoration prioritization model and hydrodynamic habitat suitability models for 1,550 km (963 miles) of estuarine shorelines in Indian River Lagoon and Lake Worth Lagoon.

The dataset presented herein includes:

  1. shoreline data from >15,000 transects collected in the field,
  2. shoreline wave climate data created through hydrodynamic modeling and frequency analysis of historic data,

For decades, compression tests have been a standard method for identifying materials properties. However, these tests are slow, destructive, and require fabricating multiple potentially expensive bulk samples. Nanoindentation is fast, almost non-destructive, and requires only a single sample, to which large numbers of indents can be applied. Furthermore, nanoindentation is widely applicable, even to materials that are too brittle for traditional compression tests. Here, we show that nanoindentation is sufficient for extracting much of the same information that is measured in traditional sample compression tests in metallic glasses. We conduct turbulence-informed Euler–Lagrange simulations of droplet-laden flow to examine the effects of atmospheric turbulence on droplet impingement characteristics on a NACA 0012 airfoil. We implement statistical overloading, whereby we inject millions of droplets, ranging in diameter from 1 to 160 microns, upstream of the airfoil at an average streamwise velocity of 120 m/s. Within a fraction of a second, these droplets would have either impinged on the airfoil or continued downstream. While the incoming air flow is maintained uniform, droplet injection is informed by ambient turbulence, affecting both the droplet velocity distribution as well as the size-dependent preferential concentration …

Cadmium silicon phosphide (CdSiP2) is a nonlinear optical material widely used in optical parametric oscillators. Intrinsic defects (vacancies and antisites) are responsible for unwanted broad optical absorption bands in these crystals that degrade the performance of the devices. In the present work, optical absorption and electron paramagnetic resonance (EPR) spectra are acquired (at room temperature and 12 K, respectively) from a neutron-irradiated CdSiP2 crystal. After the irradiation, the crystal is highly absorbing from the band edge near 600 nm to beyond 1.3 μm because of overlapping defect-related absorption bands. Polar ring galaxies (PRGs) are peculiar galaxies that show a ring of stars, gas, and dust oriented roughly over the poles of the central ‘host’ galaxy (i.e. roughly orthogonal to the disc of the host galaxy). The formation models for these rings involve mergers or tidal interactions of the host galaxy with another galaxy. Although the identified PRGs look different from each other, they all have a ring that is not in the same plane as the disc of the host galaxy. This dissertation explores the combination of two sophisticated techniques for addressing computational fluid dynamics: the discrete velocity Boltzmann equation (DVBE) and the localized collocation meshless model with upwinding (U-LCMM). The DVBE is a high-level model that describes the foundations of transport phenomena by addressing the microscale motions of particles themselves and the effect of their aggregate behaviors on continuum principles. This equation integrates multiple scales of phenomena; while it can be used for fluid flow at Navier-Stokes scales, it can also resolve fine features that can only be described at the molecular level. The hypersonic regime poses numerous challenges that researchers face in the development of hypersonic flight vehicles. Due to their excellent thermomechanical properties, ultra-high-temperature ceramics (UHTCs) have risen as a promising solution to act as a protective barrier between the harsh environment and surface materials of these flight bodies. The mechanical operation of a portable hypersonic simulation device was developed in-house and tested at Argonne National Laboratories (ANL) to gather in-situ material response of prospective UHTC samples when exposed to a hypersonic regime. Rising cancer cases spurred advancements in radiation therapy modalities, including electron accelerators. In this report, descriptive and diagnostic analysis was utilized to develop and characterize a functional, low energy cold cathode table-top electron accelerator for radiation physics experimentation in Central Washington University’s (CWU) undergraduate radiation lab. The device features a tungsten cathode (TC), brass anode, and copper Faraday Cup (FC) in a vacuum, enclosed by blue-tinted polyvinyl chloride (PVC). Nanoplasmonic fiber optic sensors leverage the optical fiber’s inherent compactness and surface sensitivity via evanescent-field interactions with the localized surface plasmon resonance of the metallic nanoparticles. One of the popular approaches is self-assembly owing to its relatively low cost. Despite its low cost, the self-assembly technique has low reproducibility and exhibits low sensitivity because of low nanoplasmonic coverage on the fiber optic surface. In this study, several fabrication techniques were explored to assess the refractive index sensitivity by comparing three common organosilanes, i.e., (3-Aminopropyl)triethoxysilane (APTES), (3-Aminopropyl)trimethoxysilane, and (3-Mercaptopropyl)trimethoxysilane. We employed the innovative freeze–thaw method to investigate the effect of chitosan (CS) molecular weights incorporated into polyvinyl alcohol on the morphology and physical characteristics of the hydrogel. This study aimed to unravel the intricate relationships between CS molecular weight variations, CS concentration, and the number of freeze–thaw cycles (4 or 6 cycles) on hydrogel swelling degree and weight loss. Incorporating CS improved the hydrogel structure and enhanced its porosity, resulting in remarkable increases in swelling degree ranging from 2 to 6 times the initial weight.
