Drug metabolism is the biochemical process of modifying drugs to detoxify and remove them through enzymatic transformations. These biotransformation’s occur primarily in the liver and are critical to understanding how pharmaceutical compounds are chemically altered inside the human body. Human carboxylesterases (CESs) catalyze the hydrolysis of esters, amides, thioesters, and carbamates. CES-mediated hydrolysis plays an important role in the metabolism of many drugs including the first FDA approved antiviral treatment for COVID-19, remdesivir (Veklury), the seizure control medication rufinamide (Banzel), and the flu antiviral drug oseltamivir (Tamiflu). Coronavirus disease-2019 (COVID-19), which is caused by a novel coronavirus named severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), has ravaged the world for the past 3 years. Even today, there still exists a need for rapid, accurate, economical and non-invasive diagnostic testing platforms that yield high specificity and sensitivity towards the constantly mutating SARS-CoV-2. Much of our current understanding of how small-molecule ligands interact with proteins stems from X-ray crystal structures determined at cryogenic (cryo) temperature. For proteins alone, room-temperature (RT) crystallography can reveal previously hidden, biologically relevant alternate conformations. However, less is understood about how RT crystallography may impact the conformational landscapes of protein-ligand complexes. Previously, we showed that small-molecule fragments cluster in putative allosteric sites using a cryo crystallographic screen of the therapeutic target PTP1B (Keedy et al., 2018). งานวิจัยนี้ได้ทำการศึกษา วิเคราะห์การใช้พลังงานของโรงงาน เพื่อออกแบบระบบเซลล์สุริยะบนหลังคา การกำหนดแผนงานติดตั้ง ศึกษาปัจจัยหลักที่ส่งผลกระทบต่อการผลิตไฟฟ้า และการประเมินประสิทธิภาพการใช้พลังงานไฟฟ้าจากเซลล์สุริยะจากการใช้งานเป็นแหล่งพลังงานสะอาดสำหรับกระบวนการผลิตยางคอมปาว์ด โดยออกแบบการใช้พลังงานไฟฟ้าให้เกิดประสิทธิภาพสูงสุดในอุตสาหกรรมผลิตยางคอมปาว์ด 2 โครงการ ระบบไฟฟ้าเซลล์สุริยะแบบออนกริดที่มีขนาดกำลังไฟฟ้าติดตั้งขนาด 865 กิโลวัตต์ และ 630 กิโลวัตต์ จากการศึกษาพบว่าทั้ง 2 โรงงานมีการใช้พลังงานไฟฟ้าในกระบวนการผลิตเป็นหลัก และความต้องการใช้พลังงานไฟฟ้าแปรผันตามยอดผลิตยางคอมปาว์ดร้อยละ 90 จากการวิเคราะห์ติดตามผลการใช้งานไฟฟ้าเซลล์สุริยะมีประสิทธิภาพต่ำกว่าร้อยละ 75 ในช่วงระยะ 5 เดือนหลัง เนื่องจากความต้องการใช้พลังงานไฟฟ้าของโรงงานลดลงและการจัดการโหลดการผลิตไม่เหมาะสมในช่วงกลางวัน และพบฝุ่นที่สะสมบนเซลล์สุริยะเป็น ผงคาร์บอน แคลเซียมคาร์บอนเนต และซิลิกาซึ่งเป็นส่วนผสมของการผลิตยางคอมปาว์ด จากการศึกษาพบว่า ฝุ่นจากกระบวนการผลิตส่งผลกระทบต่อการผลิตไฟฟ้าของเซลล์สุริยะร้อยละ 4 ดังนั้นระยะเวลาการล้างแผงเซลล์สุริยะของทั้ง 2 โครงการที่จุดสมดุลคือ ทุกๆ 3 เดือน จากการศึกษาข้อมูลการใช้ไฟฟ้าจากระบบเซลล์สุริยะนี้ชี้ให้เห็นว่า การผลิตไฟฟ้าของระบบเซลล์สุริยะแบบออนกริดนี้ มีปัจจัยหลักต่อการผลิตไฟฟ้าคือความต้องการใช้ไฟฟ้าของโรงงานเป็นหลัก เนื่องจากทั้ง 2 โรงงานมีการใช้ไฟฟ้าน้อยกว่ากำลังติดตั้งจึงทำให้เซลล์สุริยะผลิตไฟฟ้าน้อย แม้จะมีค่ารังสีดวงอาทิตย์สูงก็ตาม อย่างไรก็ตามระบบไฟฟ้าเซลล์สุริยะไม่สามารถจ่ายไฟฟ้าได้ตลอดเวลา ดังนั้นการบริหารจัดการโหลดการผลิตให้เหมาะสมจึงจำเป็นมาก เพื่อการใช้ไฟฟ้าจากเซลล์สุริยะให้มีประสิทธิภาพสูงสุด

Huntington’s disease (HD) is a fatal neurodegenerative disease caused by the expansion of the polyglutamine (polyQ) domain of the huntingtin protein (htt). The expansion of the polyQ domain beyond a threshold of approximately 35 repeats triggers complex toxic aggregation mechanisms and results in altered interactions between htt and lipid membranes. Many factors modulate these processes. One such modulator includes sequences flanking the polyQ domain, most notably the first 17 amino acids at the N-terminus of the protein (Nt17), and environmental factors including the presence of membranous structures. Enzymes are biological systems that aid in specific biochemical reactions. They lower the reaction barrier, thus speeding up the reaction rate. A detailed knowledge of enzymes will not be achievable without computational modeling as it offers insight into atomistic details and catalytic species, which are crucial to designing enzyme-specific inhibitors and impossible to gain experimentally. Processing crude oil of variable composition, especially due to the need to process crude oil blends obtained from various sources, presents a tremendous process challenge. We set out to determine whether the C-terminus (amino acids 481–798) of peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α, UniProt Q9UBK2), a regulatory metabolic protein involved in mitochondrial biogenesis, and respiration, is an arginine methyltransferase substrate. Arginine methylation by protein arginine methyltransferases (PRMTs) alters protein function and thus contributes to various cellular processes. In addition to confirming methylation of the C-terminus by PRMT1 as described in the literature, we have identified methylation by another member of the PRMT family, PRMT7. We performed in vitro methylation reactions using recombinant mammalian PRMT7 and PRMT1 at 37, 30, 21, 18, and 4 °C. …

Biophysical phenomena are modeled using a combination of quantum and classical methods to interpret and supplement three distinct and diverse problems in this dissertation. In the first project, decarboxylation reactions are ubiquitous across chemical and biological disciplines, yet the origin of non-catalytic solvent effects remains elusive. Specific solvent structure and energetics have not been well described for the monoanion of malonate, nor corrected from the gas-phase charge-assisted intramolecular hydrogen bond model known as “pseudochair”. Mesoporous thin films are widely used for applications in need of high surface area and efficient mass and charge transport properties. A well-established fabrication process involves the supramolecular assembly of organic molecules (e.g., block copolymers and surfactants) with inorganic materials obtained by sol-gel chemistry. Typically, subsequent calcination in air removes the organic template and reveals the porous inorganic network. A significant challenge for such coatings is the anisotropic shrinkage due to the volume contraction related to solvent evaporation, inorganic condensation, and template removal, affecting the final porosity as well as pore shape, size, arrangement, and accessibility. Biochar, a lightweight black residue made of carbon, has proved to be an effective adsorbent to remove TCE from groundwater. Not only is it an economical substitute for conventional adsorbents but it can be created by using a variety of organic materials. For this project, it will rely on walnut shell feedstock to create biochar as this had the best ability to adsorb TCE.

Latent blood visualizers have been used in forensic science since 1937 beginning with Luminol. These visualizers through different types of reactions react with trace amounts of blood that cannot be seen with the naked human eye. These different reactions produce color that can then be seen by examiners. The four types of latent blood visualizers that were compared is luminol, leuco crystal violet, fluorescein, and bluestar. Luminol and bluestar use chemiluminescence, fluorescein uses florescent, and leuco crystal violet uses oxidation to react with iron in hemoglobin. Fragment-based drug discovery (FBDD) is a powerful tool for developing anticancer and antimicrobial agents. Within this, magnetic resonance spectroscopy (NMR) provides a comprehensive qualitative and quantitative approach to screening and validating weak and robust binders with targeted proteins, making NMR among the most attractive strategies in FBDD. Kinases are an important class of enzymes involved in the regulation of different cellular processes. The dysfunctional activity, either hyperactivity or inactivity, of kinases has been associated with many types of diseases, making kinases a major therapeutic target. Model compounds, 3,6,9-trithaiundecane-1,11-dicarboxylic acid (TTDPA), 2,5,8-trithianonane-1,9-dicarboxylic acid (TTDAA), and 1,11-diamide-3,6,9-trithiaundecane (TTDAce), closely related to the adducts formed by cysteine alkylation of the chemical weapon, sulfur mustard, were synthesized. It is shown that TTDPA forms complexes with key metal micronutrients: copper, nickel, cobalt, manganese, and zinc. Though the strength of binding to TTDPA varies, the complexes in many cases precipitate from solution. All metals produced a visible precipitate upon interaction with TTDPA under the conditions tested, however only Cu2+, Mn2+, and Zn2+ produced enough to be measured. Recent studies have challenged the interpretation of plant water isotopes obtained through cryogenic vacuum distillation (CVD) based on observations of a large 2H fractionation. These studies have hypothesized the existence of an H-atom exchange between water and organic tissue during CVD extraction with the magnitude of H exchange related to relative water content of the sample; however, clear evidence is lacking. Two quinidine-functionalized coumarin molecular probes have been synthesized and have been found to bind metal cations (Cd2+, Co2+, Cu2+, Fe2+, Hg2+, Ni2+, and Zn2+) with high affinity in organic–aqueous media (DMSO–HEPES). The chemodosimeters coordinate with the Zn2+ ions in a two-to-one ratio (molecular probe : Zn2+) with a log β of 10.0 M−2. Upon the addition of the closed-shell metal ions studied, a fluorescence turn-on via an excimer formation is seen at 542 nm due to the quinaldine moiety adopting a syn arrangement when coordinated to the metal Zn2+ ions. The assembly of mammalian SWI/SNF chromatin remodeling complexes is developmentally programed, and loss/mutations of SWI/SNF subunits alter the levels of other components through proteolysis, causing cancers. Here, we show that mouse Lsd1/Kdm1a deletion causes dramatic dissolution of SWI/SNF complexes and that LSD1 demethylates the methylated lysine residues in SMARCC1 and SMARCC2 to preserve the structural integrity of SWI/SNF complexes. The methylated SMARCC1/SMARCC2 are targeted for proteolysis by L3MBTL3 and the CRL4DCAF5 ubiquitin ligase complex. We identify SMARCC1 as the critical target of LSD1 and L3MBTL3 to maintain the pluripotency and self-renewal of embryonic stem cells. L3MBTL3 also regulates SMARCC1/SMARCC2 proteolysis …

My current work deals with the preparation and characterization of nanocomposites for effluent treatment applications. The expertise lies on the preparation of carbonaceous nanocomposites including graphitic materials. The prepared materials are characterized thoroughly by different methods like FE-SEM, TEM, FT-IR, XRD, VSM, surface area and porosity. The composite materials are used for the removal of conventional and emerging contaminants like antibiotics, pesticide residues, endocrine disruptors etc. In addition to the batch studies, fabrication of microcolumns to aid the scale-up of the process is also carried out. DNA is often referred to as the "molecule of life " since it contains the genetic blueprint for all forms of life on this planet. The core building blocks composing DNA are deoxynucleotides. While the deoxyribose sugar and phosphate group are ubiquitous, it is the composition and spatial arrangement of the four natural nucleobases, adenine (A), cytosine (C), guanine (G), and thymine (T), that provide diversity in the coding information present in DNA.
