Human N-acetylated α-synuclein (NAcαSyn) has long been accepted as an intrinsically disordered protein (IDP) predominantly found in terminal nerve regions in healthy subjects. NAcαSyn structural fluidity enables the formation of transient or dynamic assemblies. Some of these assemblies may serve functional, yet undefined purposes. NAcαSyn is highly susceptible to the surrounding environment, leading to misfolding under high cytosolic oxidative stress conditions, which in turn assemble in the form of toxic oligomers or amyloid fibrils. The 14-3-3 family of phospho-binding proteins regulate a variety of major cellular processes through interaction with a network of dynamic proteins. Deregulation of the 14-3-3 interaction network contributes to a variety of degenerative disorders and cancers. Our lab focuses on identifying novel 14-3-3 interactions and understanding how 14-3-3 binding regulates protein function. A major gap in this process is that identifying the phospho-site where 14-3-3 docks on a given protein is time- and resource-consuming. Prediction algorithms have been developed to predict canonical 14-3-3 binding sites, however, there are many non-canonical sites that existing software is unable to predict. To fill …

The Resistant Nodulation Division (RND) super family member, tripartite AcrA-AcrB-TolC efflux pump, is a major contributor in conferring multidrug-resistance in Escherichia coli. The structure of the pump complex, and drug translocation by functional rotation mechanism have been widely studied. Despite of all these data, the dynamics of the assembly process of the pump and AcrB during functional rotation in the process of drug efflux remains poorly understood. My thesis focuses on understanding the pump assembly process, dynamics of AcrB in functional rotation mechanism, and also investigate the mechanism of degradation of AcrB facilitated by a C-terminal ssrA tag.

As DNA analysis has advanced and produced tests with higher sensitivities, attention has turned toward obtaining DNA profiles from cells left with fingermarks. Recent studies have reported that cells deposited within fingermarks can exhibit differences in autofluorescence emission in the ‘red’ region of the visible spectrum (e.g., between 650-670 nm), which can be used to differentiate contributor cell population and separate them before DNA profiles. Interestingly, this emission was not consistent to the individual day-to-day and likely not a genetically-controlled attribute of the contributor. With the likelihood of contracting an antibiotic resistant infection increasing, the need for novel anti-infective agents is critical. The development of novel anti-infective agents benefits from the identification of new target pathways necessary for the survival of the pathogen. One such target is the methylerythritol phosphate (MEP) pathway, responsible for the synthesis of the isoprenoid precursors isopentyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP) in bacteria and other pathogenic organisms. Electron bifurcation is considered as a third fundamental mode of energy conservation mechanism in addition to two well-known mechanisms, substrate level phosphorylation and Oxidative phosphorylation, in electron bifurcation endergonic and exergonic redox reactions are coupled. The newly discovered flavin based electron bifurcation in electron transfer flavoproteins (ETFs) helps to reduce low potential ferredoxin, which provides electrons to drive biologically demanding reactions such as atmospheric dinitrogen fixation in diazotroph and methane production in methanogens.

Peptide fibre formation via molecular self-assembly is a key step in a range of cellular processes and increasingly considered as an approach to produce supramolecular biomaterials. We previously demonstrated the self-assembly of the tripeptide lysine-dityrosine (KYY) as a substrate for the formation of proton-conducting melanin-like materials. ในงานวิจัยนี้ วงจรเครื่องกำเนิดพลาสมาถูกออกแบบโดยวางรากฐานอยู่บนการใช้ Flyback Transformer ซึ่งเป็นเทคโนโลยีการสวิตชิ่งกำลังไฟฟ้าของ MOSFET ในการคายประจุแบบพลาสมาไดอิเล็กตริกแบริเออร์ดิสชาร์จเชิงผิวเพื่อนำมาเปรียบเทียบกับพลาสมาไอออนไนเซอร์ซึ่งเป็นเทคโนโลยีของบริษัท บิทไว้ส์ (ประเทศไทย) จำกัด แรงดันไฟฟ้ากระแสสลับที่จ่ายให้กับเครื่องพลาสมาไดอิเล็กตริกแบริเออร์ดิสชาร์จเชิงผิวใช้แรงดันไฟฟ้า 5.38 kV วงจรกำเนิดพลาสมาได้รับการออกแบบมีการทำงานที่ความถี่ 0.2 Hz เมื่อใช้ Duty Cycle ที่ 5%, 10%, 20% และ 100% การศึกษาอุณหภูมิวัสดุไดอิเล็กตริกที่เปิดระบบไว้ 10 นาที อยู่ที่ 34.0 °C, 36.5 °C, 39.6 °C และ 67.0 °C ตามลำดับ และเมื่อตรวจวัดค่าโอโซนพบว่ามีความเข้มข้นโอโซน 174 ppb, 794 ppb, 1,820 ppb และ 9,849 ppb ตามลำดับ จากการศึกษาสเปกโตรสโกปีการเปล่งแสงของพลาสมาไดอิเล็กตริกแบริเออร์ดิสชาร์จพื้นผิวที่ความดันบรรยากาศเผยให้เห็นว่ามีกลุ่ม N2, NO และ OH ซึ่งสารอนุมูลอิสระเหล่านี้มีบทบาทสำคัญในการฆ่าเชื้อจุลินทรีย์ โดยการใช้ Optical Emission Spectrometer พบว่ามีอุณหภูมิพลาสมาที่ 0.82 eV การทดสอบประสิทธิภาพการกำจัดเชื้อใช้ Escherichia coli (E. coli) ATCC 25922 ในการทดสอบ เชื้อ E. coli ถูกฉายด้วยไดอิเล็กตริกแบริเออร์ดิสชาร์จเชิงผิวและพลาสมาไอออนไนเซอร์เป็นเวลา 10 นาที จากผลการทดลองแสดงให้เห็นว่าไดอิเล็กตริกแบริเออร์ดิสชาร์จเชิงผิวสามารถลดปริมาณเชื้อ E. coli ได้ถึง 100% ในขณะที่พลาสมาไอออนไนเซอร์ไม่สามารถลดปริมาณเชื้อ E. coli ได้

วิทยานิพนธ์ฉบับนี้เสนอแนวทางการเลือกใช้และพัฒนาอุปกรณ์หยิบจับ (Gripper) สำหรับผลิตภัณฑ์ยางแปรรูปให้เหมาะสมกับรูปร่าง ขนาด และคุณสมบัติทางกลของยางแปรรูป ซึ่งอุปกรณ์หยิบจับนี้ถูกนำมาใช้เพื่อนำผลิตภัณฑ์ยางที่ผ่านกระบวนการขึ้นรูปชิ้นงานด้วยแม่พิมพ์โดยการให้ความร้อนออกจากแม่พิมพ์ออกจากแม่พิมพ์โลหะ ซึ่งจำเป็นที่ต้องทราบถึงแรงที่ใช้นำชิ้นงานออกจากแม่พิมพ์โดยแรงนี้ถูกเรียกว่า Demolding force และโดยปกติแรงนี้จะประกอบไปแรงเสียดทาน (Friction Force) และแรงยึดติด (Adhesion force) ดังนั้นวิทยานิพนธ์ฉบับนี้จึงมุ่งเน้นไปที่ปัจจัยที่ส่งผลต่อ Demolding force ได้แก่ รูปทรง พื้นที่สัมผัส ความแข็งของยาง และความหยาบผิวของแม่พิมพ์ โดยได้มีการออกแบบการทดลองโดยใช้ Full factorial design และใช้การวิเคราะห์ถดถอยพหุคูณ (Multiple regression analysis) เพื่อใช้ในการพยากรณ์ นอกจากนี้ยังได้พัฒนาซอฟท์แวร์มาเพื่อแนะนำการออกแบบและพัฒนาอุปกรณ์หยิบจับ และสุดท้ายได้มีการพัฒนาอุปกรณ์หยิบจับตัวต้นเพื่อทดสอบประสิทธิภาพของซอฟท์แวร์นี้

การวัลคาไนซ์เซชั่นยางในอุตสาหกรรมต้องอาศัยอุณหภูมิและระยเวลาในการคงรูปชิ้นงานซึ่งระยะเวลาในการคงรูปชิ้นงานจะขึ้นกับค่าการนำความร้อนของยางคอมปาวด์ที่แต่ละสูตร โดยงานวิจัยนี้จึงมีจุดประสงค์เพื่อศึกษาหลักการและกระบวนการทดสอบค่าการนำความร้อน การประมาณค่าการนำความร้อนในยางคอมปาวด์และปัจจัยที่ส่งผลต่อค่าการนำความร้อนในกระบวนการคงรูป โดยขั้นแรกได้ศึกษาการประมาณค่าการนำความร้อน พบว่า การประมาณค่านำความร้อนของแบบจำลองขนานเทียบกับค่าการนำความร้อนที่ได้จากการทดสอบ HFM จะมีค่าความคาดเคลื่อนเกิดขึ้นไม่เกิน 5 % สำหรับยางคอมปาวด์ที่วัลคาไนซ์เซชั่นอย่างสมบูรณ์ ขั้นที่สองเป็นศึกษาปัจจัยที่ส่งผลกระทบต่อค่าการนำความร้อน พบว่า สัดส่วนโดยปริมาตรของสารตัวเติมมีผลกระทบต่อค่าการนำความร้อนที่จะเปลี่ยนแปลงไปรวมถึงปริมาณน้ำมันที่ส่งผลกระทบต่อค่าการนำความร้อนอย่างมาก ส่วนยางที่วัลคาไนซ์เซชั่นอย่างสมบูรณ์จะให้ค่าการนำความร้อนสูงกว่ายางคอมปาวด์และยางที่วัลคาไนซ์เซชั่นร้อยละ 50 จะมีฟองอากาศภายในยางส่งผลให้ค่าการนำความร้อนต่ำกว่ายางคอมปาวด์ ส่วนยางที่เกิด และขั้นที่สามออกแบบชุดทดสอบสำหรับหาค่าการนำความร้อน พบว่า ชุดต้นแบบใช้วิธีการหาค่าการนำความร้อนในสภาวะคงที่ซึ่งต้องควบคุมความสม่ำเสมอของความร้อนและป้องสูญเสียความร้อนของชุดต้นแบบ ดังนั้นชุดต้นแบบจะมีส่วนประกอบหลักเป็นฮีตเตอร์สำหรับเป็นแหล่งความร้อนโดยจะใช้น้ำหล่อเย็นสำหรับการควบคุมอุณหภูมิประกอบกับฉนวนกันความร้อนสำหรับป้องกันการสูญเสียความร้อนโดยจะใช้การไหลของความร้อนในหนึ่งหน่วยพื้นที่เป็นตัวกำหนดค่าการนำความร้อน

งานวิจัยนี้ศึกษาอัตราส่วนผสมยางโบรโมบิวทิล (BIIR) พอลิโพรพิลีน (PP) และเอทิลีนออกทีนโคพอลิเมอร์ (EOC) เพื่อปรับปรุงความแข็งให้เท่ากับความแข็งของจุกปิดหลอดเก็บเลือดทางการค้า จากผลการทดสอบสมบัติความแข็งพบว่าที่อัตราส่วนผสม B65P10E25 (BIIR 65 wt%, PP 10wt% และ EOC 25 wt%) มีค่าความแข็งเท่ากับ 43 Shore A ซึ่งอยู่ในเกณฑ์ค่าความแข็งของจุกปิดหลอดเก็บเลือด จึงได้เลือกอัตราส่วนนี้มาเตรียมเป็นเทอร์มอพลาสติกวัลคาไนเซต (TPV) โดยผสมสารวัลคาไนซ์ไดคิวมิลเพอร์ออกไซด์ 1.5 wt% และสารโคเอเจนต์ไตรเมทิลออลโพรเพนไตรเมทาอะคริเลท 1 wt% ผลการทดสอบสมบัติเชิงกลต่าง ๆ พบว่า TPV ที่เตรียมได้มีสมบัติความแข็ง ความต้านทานต่อแรงดึง ระยะยืดสูงสุด ณ จุดขาด และความต้านทานต่อแรงฉีกขาดสูงขึ้น ในขณะที่เปอร์เซ็นต์การยุบตัวหลังได้รับแรงกดลดลงเป็น 21.83% เมื่อเทียบกับ B65P10E25 ที่ไม่มีสารวัลคาไนซ์ ผลจากภาพกล้องจุลทรรศน์อิเล็กตรอนแบบส่องกราด (SEM Micrograph) พบว่าการเติม EOC ในสัดส่วนที่เพิ่มขึ้น จะช่วยปรับปรุงความเข้ากันได้และการกระจายตัวของ BIIR และ PP นอกจากนี้เมื่อผสม EOC ในสัดส่วนที่มากกว่า 25% วัสดุจะแสดงสมบัติการคืนตัวหลังแทงเข็มที่ดี โดยสูตรที่ผ่านการทดสอบนี้ ได้แก่ B65P10E25, B65E35 (BIIR 65 wt% และ EOC 35 wt%) และ B65P10E25 TPV (TPV ที่เตรียมขึ้นจากอัตราส่วน B65P10E25) เป็นต้น นอกจากนี้ความสามารถในการซึมผ่านแก๊สออกซิเจนของ B65P10E25 TPV มีค่าเท่ากับ 151.81 cm3·mm/m2·day·0.1MPa ซึ่งอยู่ในเกณฑ์ที่ใช้พิจารณาวัสดุที่จะนำมาใช้เป็นจุกปิดสำหรับผลิตภัณฑ์ที่ต้องการป้องกันการซึมผ่านของอากาศภายนอกได้ดี ดังนั้นงานวิจัยนี้จึงสามารถเตรียม TPV ที่มีสมบัติเชิงกลที่ดี มีอัตราการซึมผ่านแก๊สต่ำ และสามารถคืนตัวได้หลังจากดึงเข็มออก ซึ่งสมบัติเหล่านี้ผ่านเกณฑ์ที่ใช้พิจารณาสำหรับนำไปประยุกต์ใช้เป็นจุกปิดในหลอดเก็บเลือด นอกจากนี้ยังสามารถนำมาผ่านกระบวนการขึ้นรูปซ้ำใหม่ได้

Combining vibrating mesh nebulizers with additional new technologies leads to substantial improvements in pharmaceutical aerosol delivery to the lungs across therapeutic administration methods. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. The first focus of this work was to improve the poor delivery efficiency, e.g., 3.6% of nominal dose (Dugernier et al. 2017), of aerosolized medication administration to adult human subjects concurrent with high flow nasal cannula (HFNC) therapy, a form of continuous-flow non-invasive ventilation (NIV). Cancer is the second leading cause of death in the US with over 1.7 million new cases each year. Current cancer treatments tend to also target healthy tissues due to similarities with cancerous ones, resulting in acute side effects. Early detection is the best approach towards defeating cancer, however, modern imaging techniques require sizeable samples, often implying a late stage in the disease. Complex carbohydrates make up more than half the biomass on earth, and they are crucial to a wide range of processes in living organisms. When polysaccharides and glycans are enzymatically digested into manageable units called oligosaccharides, they typically consist of 2-20 linear or branched sugar units. The circadian rhythms arise as an adaptation to the environmental 24-hour day and night cycle due to Earth's rotation. These rhythms prepare organisms to align their internal biological activities and day to day behavior or events with the environmental change of the 24-hour day and night cycle. Circadian rhythms are found widely in all living kingdoms of life on Earth. Cyanobacteria are photosynthetic prokaryotes which first used to study these circadian rhythms. Melanin is s most widely distributed pigment and is found in bacteria, fungi, plants, and animals. Melanogenesis is under complex regulatory control by multiple agents interacting through pathways activated by hormonal and receptor-dependent and -independent mechanisms. There are about 20 genes that are involved in the biochemical pathway of melanogenesis and its regulation, which include: tyrosinase, microphthalmia-associated transcription factor, melanocortin1 receptor, adenylate cyclase, protein kinase A. The SuperCam instrument suite provides the Mars 2020 rover, Perseverance, with a number of versatile remote-sensing techniques that can be used at long distance as well as within the robotic-arm workspace. These include laser-induced breakdown spectroscopy (LIBS), remote time-resolved Raman and luminescence spectroscopies, and visible and infrared (VISIR; separately referred to as VIS and IR) reflectance spectroscopy. A remote micro-imager (RMI) provides high-resolution color context imaging, and a microphone can be used as a stand-alone tool for environmental studies or to determine physical properties of rocks and soils from shock waves of laser-produced plasmas. Molecular Dynamics simulations are a highly useful tool in helping understand the fundamental interactions present in a variety of chemical systems. The work discussed here illustrates it’s use in determining the conformational dynamics of the Zika and SARS-Cov-2 helicase in a physiological environment, largely in an effort to discover inhibitors capable of rendering the protein inert. Additionally, we show how it can be used to understand paradoxical trends in the anion-induced precipitation of Cucurbituril cavitands.

In recent years, carbon dots (C-dots) have gained appreciable interest owing to their unique optical properties, including tunable fluorescence, stability against photobleaching and photoblinking, and strong fluorescence. Simple and low-cost hydrothermal and electrochemical approaches have been widely used in the preparation of biocompatible and high-quality C-dots. Various C-dots have been used for the quantitation of small analytes, mostly based on analyte induced fluorescence quenching. Depending on the nature of precursors, synthetic conditions (such as reaction temperature and time), and surface conjugation, multi-function C-dots can be prepared and used in diagnostics and therapeutics. The interaction of a nucleoside analogue, bromodeoxyuridine (BrdU) with human serum albumin (HSA) was studied to investigate the binding phenomenon and analyze the protein conformation upon BrdU binding. Multiple spectroscopic techniques, viz. intrinsic and three-dimensional (3-D) fluorescence, ultraviolet-visible (UV-Vis) absorption and circular dichroism (CD) spectroscopy along with molecular docking were used. A decrease in the Stern-Volmer constant (Ksv) with the increase in temperature suggested BrdU-induced quenching of protein fluorescence as static quenching and thus implied BrdU−HSA complex formation. This was also supported by UV-Vis absorption spectral results, showing hyperchromism in the absorption spectrum of HSA upon BrdU addition. Intermediate binding …

Cyanobacteria are photosynthetic organisms that are known to be responsible for oxygenating Earth’s early atmosphere. Having evolved to ensure optimal survival in the periodic light/dark cycle on this planet, their genetic codes are packed with various tools, including a sophisticated biological timekeeping system. Among the cyanobacteria is Synechococcus elongatus PCC 7942, the simplest clock-harboring organism with a powerful genetic tool that enabled the identification of its intricate timekeeping mechanism. The United States Department of Agriculture downgrades on the order of 17% of all Swiss cheese produced in the United States due to defects. Many of these defects are related to improper eye formation, number, distribution, or size; leading to an industry loss of over $69 million per annum. The microbiome in Swiss-type cheeses plays a significant role in eye development due to production of organic acids and gaseous emissions contingent on bacterial abundance and phenotype.
